The effects of LHRH on pituitary somatotropes in ovariectomized female rats

Introduction: Luteinizing hormone-releasing hormone (LHRH), found in the hypothalamus, primarily controls the release of gonadotropic hormones from the anterior pituitary gland, but also affects some other pituitary cells. The aim of this study was to examine the effects of high-dose LHRH on the immunohistological features of pituitary somatotropes (GH cells) and blood levels of growth hormone (GH) in ovariectomized (Ovx) Wistar rat females.
Methods: Adult (12 weeks old) female rats were divided into two groups, each comprising seven females. The first group of Ovx females daily received s.c. LHRH (Ovx+LHRH; 25 μg) during the final three days of the fourth week after the surgery, while the second group, Ovx controls, received an adequate volume of sterile olive oil during the same period. All females were sacrificed 24 h after the last injection. GH cells were studied using the peroxidase-antiperoxidase (PAP) immunohistochemical procedure. Serum concentrations of GH in all rats were measured by the hGH-Delfia kit.
Results: The absolute and relative pituitary weights in Ovx+ LHRH females were significantly decreased (p<0.05) by 8.5% and 12.3% respectively, in comparison with the Ovx controls. In the LHRH-treated Ovx females, GH cells were longer, irregularly shaped, with more intensely stained cytoplasm, compared to the controls. Concentration of GH hormone in the serum of LHRH-treated Ovx females was significantly increased (p<0.05) by 73.3% in comparison with the Ovx controls.
Conclusion: Our findings show that the LHRH application caused a change in the immunohistological features and secretory activity of GH cells in Ovx females.
