Soy extract and soy isoflavones do not affect serum cholesterol and triglyceride levels in orchidectomized young adult rats

Soy-based dietary supplements are promoted as a "natural" way to reduce menopausal symptoms, and to decrease the risk of heart disease, osteoporosis and breast cancer in women. Studies published so far provide contradictory information on these effects. Observed health benefits of using soy supplements are minimal. By preventing prostate cancer development, soy isoflavones actually have more impact on health of the male population; clinical and experimental data on serum cholesterol levels and bones in males are still incomplete. We have previously demonstrated that purified isoflavones, genistein (G) and daidzein (D), decrease serum cholesterol levels and increase serum triglycerides in middle-aged rats. To further examine whether the animals' age has an impact on the obtained results, we applied the same experimental design on young adult male rats. Orchidectomized (Orx) 3 month-old Wistar rats (which were fed soy-free diet) were subcutaneously treated with 10 mg/kg or 30mg/kg of G, D, or soy extract enriched with isoflavones (40%). The control groups received vehicle, testosterone or estradiol for three weeks, after which the total serum cholesterol (TC), low density lipoprotein, high density lipoprotein and total triglycerides (TT) were measured. Compared to the matching vehicletreated controls, the testosterone treatment significantly (p<0.05) lowered the TC levels, while estradiol significantly increased the TC levels. Estradiol treatment increased (p<0.05) the TT levels. Injections of neither G, D nor soy extract did not affect the serum cholesterol and triglyceride levels. In conclusion, cholesterollowering effect of soy isoflavones was lesser in young adult than in middle-aged rats.
