The influence of methanol and aqueous-acetone extracts from three Veronica species on wound healing process

The plant species from genus Veronica (Plantaginaceae) are traditionally used for the treatment of chronic skin conditions, itching and in promotion of wound healing. The aim of this research was to evaluate possible effects of three Veronica species (Veronica jacquinii Baumg., Veronica urticifolia Jacq. and Veronica teuctium L.) on the formation of new capillaries (angiogenesis) as a part of wound healing process. Angiogenesis involves the attachment, migration and differentiation of endothelial cells into tubule like structures. The influence of methanol and 70% acetone extracts of air-dried flowering aerial parts on adhesion, migration and capacity of EA.hy926 human endothelial cells to form capillary-like tubes on extracellular matrix was evaluated by cell adhesion, scratch and 3D tube formation assays, respectively. Tested extracts in the highest non-toxic concentration (25 μg/ml) inhibited spontaneous tube formation which occurs after plating endothelial cells on matrix rich in extracellular proteins, implying their possible anti-angiogenic activity. The most potent inhibitory effect exhibited methanol extract from Veronica jacquinii and aqueous-acetone extract from Veronica teucrium. On the other hand, investigated extracts did not significantly change adhesive or migratory capacity of EA.hy926 cells, according to cell adhesion and scratch assays. Considering traditional use of Veronica species, these results suggest a need for further assessment of their supposed wound healing properties This research was funded by Ministry of Education, Science and Technological Development of the Republic of Serbia (Project III 41030 and TR 31089).
