AB  - Introduction: The endemic species Nepeta cyrenaica Quézel & Zaffran, native to northeastern Libya, is valued as an important honey-bearing plant.

Objectives: This study was aimed to examine the micromorphology, phytochemistry, and bioactivity of in vitro-propagated N. cyrenaica for the first time.

Materials and methods: The leaf indumentum was examined using light and scanning electron microscopy and further characterised for histochemistry. The chemical composition of essential oil (EO) was performed using GC-MS analysis, while dichloromethane (DCM), methanol (ME), ethanol (ET), and aqueous (AQ) extracts were analysed using qualitative and quantitative LC/MS analyses. The antioxidant activities of EO and extracts were assessed using three parallel assays, while enzyme-inhibiting effects were evaluated against four enzymes.

Results: The leaves bear various types of glandular trichomes, with lipophilic secretion predominating. The main EO component of EO was 1,8-cineole. A considerable number of phenolics and iridoids were tentatively identified in the ME extract. Quantitative LC/MS analysis confirmed that ferulic acid, rosmarinic acid, and epigallocatechin gallate were present in the highest amount in the extracts, in which three iridoids were also quantified. Although the ME extract contained the highest amount of polyphenolics and iridoids, the DCM extract showed the best overall biological potential. Additionally, EO exerted the strongest acetylcholinesterase and tyrosinase inhibition.

Conclusion: This study demonstrated that the endemic N. cyrenaica can be efficiently grown under in vitro conditions, where it develops various glandular trichomes that are thought to secrete and/or accumulate bioactive compounds with valuable medicinal potential.

AB  - Pear (Pyrus communis L.) is consumed as fresh fruit, in numerous food products, and also used as a traditional remedy in various countries, including Serbia. In search of bioactive compounds, six traditional pear varieties (‘Vidovača’, ‘Lubeničarka’, ‘Karamanka’, ‘Jeribasma’, ‘Lončara’, ‘Takiša’) and wild pear from Serbia were investigated and compared with a commercial variety (‘Williams Bartlett’). The aim of this study was to determine the total phenolic and flavonoid contents, phenolic composition, antioxidant capacity, and antineurodegenerative activities of methanolic extracts of peel, flesh, and mixed peel and flesh of pear fruits. Phenolic composition of extracts was determined with HPLC-DAD, while the antioxidant activity of extracts was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and ferric-reducing antioxidant power (FRAP) assays. Bearing in mind that oxidative stress is closely linked to neurodegeneration, the antineurodegenerative potential of the extracts was assessed by the inhibition of acetycholineserase (AChE) and tyrosinase (TYR) activities. The extracts of traditional varieties, particularly peel extracts, had a high content of phenolics, as well as significant antioxidant and moderate antineurodegenerative potential, compared to the commercial variety. The highest contents of total flavonoids and individual compounds, such as arbutin and chlorogenic acid, as well as the strongest antioxidant and TYR inhibitory activities were reported for the ‘Takiša’ peel extract. The performed analyses have revealed that fruits of traditional Serbian pear varieties are rich in bioactive components and could be used as functional food and for possible nutraceutical applications, to prevent diseases induced by oxidative stress.

AB  - In this study, we examined Salvia amplexicaulis Lam., S. jurisicii Košanin and S. ringens Sibth. & Sm. collected in the Republic of North Macedonia, which are for the first time subjected to detailed micromorphological and structural analysis using light and scanning electron microscopy. The nutlets and mucilage were additionally subjected to spectroscopic analysis using Raman and Attenuated Total Reflectance Fourier Transform Infrared spectroscopy. The anatomical structure of stems and leaves is described and compared. The stems, leaves and calyces bear numerous one- and multi-cellular nonglandular trichomes, and various peltate, capitate and digitiform glandular trichomes. The nutlets differ in size and shape, as well as in myxocarpy. The nutlets predominantly contained α-linolenic and linoleic acid, whereas the mucilages are primarily formed of polysaccharides. The results obtained in this study confirmed the importance of micromorphological and anatomical analysis of Salvia L. spp. plant parts, particularly trichomes and nutlets, and hence contributing to the knowledge about the variety of micromorphological characteristics within the genus Salvia L.

Chemical characterization of Lamiaceae plant extracts – the quest for novel immunomodulators

AB  - The physiological imbalance between the production and accumulation of reactive oxygen species (ROS), known as oxidative stress, is responsible for developing and progression of a plethora of diseases – cancer, among others. Since plants are recognized as potential antioxidant agents, the aim of this study was to chemically characterize ethanolic extracts of 18 species from Lamiaceae family using liquid chromatography-mass spectrometry (LC-MS), followed by testing the antioxidant activity using ABTS assay. Biocompatibility of the investigated extracts was assessed by MTT assay on normal human lung fibroblasts (MRC-5). Finally, antiproliferative activity was evaluated using MTT assay on human colorectal cancer (HCT-116) cell line, while the effects of extracts on superoxide anion radical (O2•-) production were tested using NBT assay. Melissa officinalis, Mentha piperita, Origanum majorana, O. vulgare, and Thymus vulgaris showed the highest antioxidant activity against ABTS radicals. LC-MS analysis indicated that these extracts mostly contain caffeic, protocatechuic and p-coumaric acids. The extracts were biocompatible with MRC-5 cell line. Interestingly, Lavandula angustifolia and Ocimum basilicum extracts, which were less active against ABTS radicals, showed a significant antiproliferative effect against HCT-116. Moreover, L. angustifolia and T. vulgaris extracts significantly stimulated the production of O2•- in HCT-116, indicating potential antitumor activity. The extracts that expressed antiproliferative effect and stimulated ROS production contain higher amounts of rosmarinic, caffeic and chlorogenic acids, naringin, luteolin, apigenin and cirsimaritin, indicating that some of them, or more probably their synergistic effects, are responsible for the extracts’ ability to moderate the production of ROS in HCT-116 cells.

AB  - Chokeberries (Aronia melanocarpa (Michx.) Elliot have recently become one of the most popular and widely used berry fruits, and there are numerous studies dealing with the investigation of their chemical composition and biological activities. Unlike berries, chokeberry leaves were far less investigated. Therefore, the aim of this study was to conduct a bioactivity-based analysis of chokeberry leaves collected after berries were harvested. Chemical analysis of hydroethanolic chokeberry extract and its ethyl acetate, n-butanol, and water fractions was performed. Antioxidant, hypoglycemic, and antineurodegenerative activities of the obtained samples were assessed by in vitro tests. The highest total phenolics content was found in n-butanol fraction (221.5 mg gallic acid equivalents/g dw). Fourteen compounds (six phenolic acids and eight flavonoids) were identified in the samples. Caffeoylquinic acid derivatives and quercetin glycosides were dominant phenolic compounds. The highest content of phenolic acids, as well as flavonoids, was recorded in n-butanol fraction (80.75 mg/g dw and 47.79 mg/g dw, respectively). This fraction was the most active in 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS), and ferric reducing antioxidant power (FRAP) assays. Also, the n-butanol fraction was the most effective inhibitor of acetylcholinesterase (AChE) and tyrosinase (TYR) enzymes. All samples showed a better inhibitory effect on α-glucosidase enzyme with inhibition concentrations IC50 = 4.21 - 9.22 μg/mL, than α-amylase enzymes (IC50 = 14.36 - 88.61 μg/mL). Pearson’s correlation between the content of individual phenolics and the results of biological tests was performed. Thus, hyperoside and isoquercitrin showed a significant correlation with DPPH and ABTS tests, caffeoylquinic acids with FRAP test, quercetin 3-O-vicianoside with β-carotene assay, hyperoside and isoquercitrin with α-amylase inhibition activity, and isorhamnetin 3-O-rutinoside with α-glucosidase inhibition assay. Considering the obtained results, chokeberry leaves represent a valuable natural source of phenolic compounds with promising potential for the development of new products with beneficial effects on human health.
