Evaluation of Antioxidant, Antimicrobial and Potential Food Preserving Properties of Rubus Discolor (Rosaceae) Fruit Extracts

AB  - The presented study was aimed at exploring the bioactive properties (antioxidant, antimicrobial and food preserving) and chemical composition of R. discolor fruits collected from 2 geographically distinct wild-growing populations and extracted by using different solvents (methanol, ethanol, acetone and water). The total phenol (TPC), flavonoid (TFC) and anthocyanin (TAC) content varied from 52.7 to 186.8 mg GAE/g, 3.5 to 7.0 mg QE/g and 11.1 to 28.2 mg/g dry weight (dw), respectively. HPLC-DAD analysis identified cyanidin-3-glucoside (Cy-3-Glu) as the dominant anthocyanin that ranged in concentration from 18.4 to 32.2 mg/g dw. The acetone extract revealed the highest antioxidant activity through the ferric reducing antioxidant power (FRAP), total reducing power (TRC) and ß-carotene bleaching in vitro methods. The examined extracts showed antimicrobial activity, being more effective against Gram-positive (G+) than Gram-negative (G-) bacteria. Furthermore, Candida glabrata was the most susceptible among the pathogenic yeasts. The ethanol fruit extract was the most active against Listeria monocytogenes and thus chosen for incorporation in yoghurt and further analyses regarding its food preserving properties, which confirmed its efficacy towards this food-borne pathogen. Additionally, panelists gave high marks to the novel product regarding color, taste, texture flavor and overall acceptance. The obtained results indicated that R. discolor fruit extracts exhibits notable bioactivity and might therefore be considered as a good platform for the development of new preparations to be used in the food industry.

AB  - This study was designed to evaluate the genoprotective, antigenotoxic, as well as antitumor potential of methanolic, ethanolic, and aqueous extracts of Melissa officinalis, Mentha × piperita, Ocimum basilicum, Rosmarinus officinalis, Salvia officinalis, and Satureja montana (Lamiaceae), in different model systems. The polyphenols in these extracts were quantified both spectrophotometrically and using HPLC-DAD technique, while DPPH assay was used to assess the antioxidant activity. The genoprotective potential was tested on pUC19 Escherichia coli XL1-blue, and the antigenotoxicity on Salmonella typhimurium TA1535/pSK1002 and human lung fibroblasts, while the antitumor activity was assessed on colorectal cancer cells. Rosmarinic acid, quercetin, rutin, and luteolin-7-O-glucoside were among the identified compounds. Methanolic extracts had the best DPPH-scavenging and SOS-inducing activities, while ethanolic extracts exhibited the highest antigenotoxicity. Additionally, all extracts exhibited genoprotective potential on plasmid DNA. The antitumor effect was mediated by modulation of reactive oxygen species (ROS), nitric oxide (NO) production, and exhibition of genotoxic effects on tumor cells, especially with O. basilicum ethanolic extract. Generally, the investigated extracts were able to provide antioxidant protection for the acellular, prokaryotic, and normal human DNA, while also modulating the production of ROS and NO in tumor cells, leading to genotoxicity toward these cells and their decrease in proliferation.

Traditional Use of Medicinal Plants in South-Eastern Serbia (Pčinja District): Ethnopharmacological Investigation on the Current Status and Comparison With Half a Century Old Data.

AB  - Balkan Peninsula is one of the most important biodiversity centers in Europe. Despite that, the usage of plant species in the traditional medicine of some Balkan regions remained largely unexplored in the past. This study aimed to collect and document data on the traditional use of medicinal plants in Pčinja district in South-Eastern Serbia, which is among the least developed regions in Serbia. Also, comparison with data collected by Dr. Jovan Tucakov, in a book called Herbal therapy was conducted. The survey was carried out using semi-structured interviews and 113 informants were interviewed. Quantitative ethnobotany factors were calculated, allowing us discussing the results. The informants reported data on 86 medicinal plants belonging to 43 families in Pčinja district. Lamiaceae, Asteraceae, and Rosaceae were the dominant locally used families. The species with the highest number of use reports were Mentha piperita, Matricaria chamomilla, and Hypericum perforatum. Gastrointestinal ailments, respiratory problems and skin diseases were the most frequently reported indications. Usually, the administration was primarily oral followed by topical applications. Leaves were dominantly exploited plant parts and the most frequent preparation form was infusion. Medicinal plants in Pčinja district are mainly used as a mode of primary health care for treating minor health issues. After comparing our results with the ones collected half a century ago by Dr. Jovan Tucakov we can conclude that plant species mentioned in our investigation previously had a much wider spectrum of application.

AB  - Chokeberries (Aronia melanocarpa (Michx.) Elliot have recently become one of the most popular and widely used berry fruits, and there are numerous studies dealing with the investigation of their chemical composition and biological activities. Unlike berries, chokeberry leaves were far less investigated. Therefore, the aim of this study was to conduct a bioactivity-based analysis of chokeberry leaves collected after berries were harvested. Chemical analysis of hydroethanolic chokeberry extract and its ethyl acetate, n-butanol, and water fractions was performed. Antioxidant, hypoglycemic, and antineurodegenerative activities of the obtained samples were assessed by in vitro tests. The highest total phenolics content was found in n-butanol fraction (221.5 mg gallic acid equivalents/g dw). Fourteen compounds (six phenolic acids and eight flavonoids) were identified in the samples. Caffeoylquinic acid derivatives and quercetin glycosides were dominant phenolic compounds. The highest content of phenolic acids, as well as flavonoids, was recorded in n-butanol fraction (80.75 mg/g dw and 47.79 mg/g dw, respectively). This fraction was the most active in 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS), and ferric reducing antioxidant power (FRAP) assays. Also, the n-butanol fraction was the most effective inhibitor of acetylcholinesterase (AChE) and tyrosinase (TYR) enzymes. All samples showed a better inhibitory effect on α-glucosidase enzyme with inhibition concentrations IC50 = 4.21 - 9.22 μg/mL, than α-amylase enzymes (IC50 = 14.36 - 88.61 μg/mL). Pearson’s correlation between the content of individual phenolics and the results of biological tests was performed. Thus, hyperoside and isoquercitrin showed a significant correlation with DPPH and ABTS tests, caffeoylquinic acids with FRAP test, quercetin 3-O-vicianoside with β-carotene assay, hyperoside and isoquercitrin with α-amylase inhibition activity, and isorhamnetin 3-O-rutinoside with α-glucosidase inhibition assay. Considering the obtained results, chokeberry leaves represent a valuable natural source of phenolic compounds with promising potential for the development of new products with beneficial effects on human health.

AB  - Balkan Peninsula is one of the most important biodiversity centers in Europe. Despite 
that, the usage of plant species in traditional medicine of some Balkan regions remained 
largely unexplored in the past. This study aimed to collect and document data on the 
traditional use of medicinal plants in Pčinja district in South-Eastern Serbia, which is 
among the least developed regions in Serbia. The survey was carried out using semi structured interviews and 113 informants were interviewed. Quantitative ethnobotany 
factors were calculated, allowing us discussing the results.
The informants reported data on 86 medicinal plants belonging to 43 families. 
The most dominant botanical families were Lamiaceae (30% of species), Asteraceae 
(26% of species) and Rosaceae (19% of species). Species with the highest number of 
use reports were Mentha piperita, Hypericum perforatum, Matricaria chamomilla, Salvia
officinalis, Urtica dioica, Thymus serpyllum and Tilia cordata. The majority of the plants 
were reported to have 1-3 different usages and the species with most diverse uses were 
Hypericum perforatum and Urtica dioica. In Pčinja district folk medicine is primarily used 
for healing minor diseases with some exceptions. The most frequent medicinal uses 
were for treating diseases of the digestive system, respiratory system and diseases of 
the skin and subcutaneous tissue, followed by general and unspecified diseases (such 
as pain, fever and health prevention). Usually, the administration was primarily oral 
followed by topical applications. Leaves were dominantly exploited plant parts and 
the most frequent preparation form was infusion. The current study represents a useful 
documentation which can contribute to preserving ethnobotanical knowledge in 
South-Eastern Serbia. For some well-known plant species new applications have been 
recorded. This can present a good starting point for new investigations

AB  - Apoptosis is considered as a classical way of programmed cell death and important control mechanism of cell homeostasis. Cancer cells acquire different instruments to circumvent programmed cell death. This promotes uncontrolled growth and frequently confers chemoresistance to tumor cells. Activation of apoptotic signaling pathway has been a target of anti-cancer drugs in an induction of cytotoxicity. The mechanism of apoptosis is complex and includes many pathways. This chapter will focus on the current knowledge of apoptosis-triggering approaches in cancer therapy, as well as on mushroom extracts and isolated compounds acting as initiators of apoptosis through regulation of genes expression involved in cancer cell death. Furthermore, we pointed out directions for novel search for mushroom functionalities in this experimental field and discuss the possible further steps for exploration on the in vitro and in vivo levels.

AB  - Pomegranate peel extract (PPE) was obtained by aqueous-ethanol extraction and was abundant in punicalin, punicalagin and ellagic acid. As these compounds are known to act against inflammation and oxidative stress in synergistic fashion, effects of PPE were tested in vitro on immune cells and in animal models of multiple sclerosis and type 1 diabetes (T1D). In vitro, PPE inhibited interleukin-17 (IL-17) production from gut-associated lymphoid tissue (GALT) cells. PPE also reduced production of IL-17 in activated T cells isolated from animals with experimental autoimmune encephalomyelitis (EAE). It efficiently down-regulated clinical symptoms of EAE in DA rats and of streptozotocin-induced T1D in C57BL/6 mice. The effect of PPE in T1D was mediated by inhibition of immune cell infiltration into pancreatic islets and through interference with IL-17 and IFN-γ production in GALT. Our results suggest that PPE has the potential to be used as phytopharmaceutical for certain autoimmune and chronic inflammatory disorders.

AB  - Oxidative stress plays a central role in the onset of diabetes which is characterised by progressive pancreatic β-cell failure. We studied the ability of phenolic extracts prepared from chestnut leaf, catkin and spiny burs to prevent oxidative stress-induced β-cell (Rin-5F) death. The Rin-5F cell death was induced by treatment with the diabetogenic agent streptozotocin (STZ) and the protective effects of the chestnut extracts were assessed after cells treatment with a STZ/extracts mixture. The chestnut extracts increased cell viability by protecting DNA from oxidative damage and by enhancing the natural antioxidant system. Moreover, the chestnut extracts had a significant impact on the cell's redox-status which was observed as an increase in the levels of reduced glutathione. Chestnut extracts were capable of inhibiting lipid peroxidation, which was manifested as a lowering of malondialdehyde levels. We concluded that chestnut extracts increased cell viability after STZ treatment as a result of their antioxidative properties. © 2010 Elsevier Ltd.
