AB  - In the present study, we investigated in vitro the antimicrobial activity of the pygidial gland secretion of the guanophilic ground beetle Laemostenus (Pristonychus) punctatus (Dejean, 1828) and some of its chemicals against resistant and non-resistant bacteria and Candida species, the synergistic and additive potential of combinations of selected chemicals and antimicrobial drugs against resistant bacterial and fungal strains, anti-adhesive and anti-invasive potential of the secretion and formic acid alone and in selected combinations with antimicrobial drugs against methicillin-resistant Staphylococcus aureus (MRSA) toward spontaneously immortalized human keratinocyte cell line (HaCaT cells). In addition, we examined the antiproliferative activity of the secretion and formic acid in vitro. The tested secretion and the standards of formic and oleic acids possessed a significant level of antimicrobial potential against all tested strains (P < 0.05). The isolate from guano Pseudomonas monteilii showed the highest resistance to the secretion and formic acid, while MRSA achieved a significantly high level of susceptibility to all agents tested, particularly to the combinations of formic acid and antibiotics, but at the same time showed a certain level of resistance to the antibiotics tested individually. Candida albicans and C. tropicalis were found to be the most sensitive fungal strains to the secretion. Formic acid (MIC 0.0005 mg/mL) and gentamicin (MIC 0.0010 mg/mL) in the mixture achieved synergistic antibacterial activity against MRSA (FICI = 0.5, P < 0.05). The combination of formic acid, gentamicin and ampicillin accomplished an additive effect against this resistant bacterial strain (FICI = 1.5, P < 0.05). The secretion achieved a better inhibitory effect on the adhesion ability of MRSA toward HaCaT cells compared to formic acid alone, while formic acid showed better results regarding the invasion (P < 0.001). The combinations of gentamicin and ampicillin, as well as of formic acid and gentamicin and ampicillin achieved similar anti-adhesive and anti-invasive effects, with a slight advantage of formic acid and antibiotics in combination (P < 0.001). The secretion and formic acid were found to be non-toxic to HaCaT cells in vitro (IC50 ≥ 401 μg/mL).

AB  - Cistus creticus subsp. creticus is a shrubby Mediterranean plant used since ancient times in folk medicine for the treatment of different diseases. C. creticus extracts and resin contain different types of secondary metabolites , such as terpenoids (predominantly labdane type diterpenoids), and phenylpropanoids. Growth conditions seem to influence the content of labdane-type diterpenes and flavan-3-ols in leaves of C. creticus subsp. creticus . Histochemical staining of leaves' trichomes and comprehensive phytochemical characterization of resin, leaves and their exudates, indicated that long-stalked capitate trichomes of C. creticus subsp. creticus , grown both in vitro (IV) and in greenhouse (GH), are capable of producing bioactive oleoresin-related terpenoids and phenylpropanoids compounds. Bioactivity-guided approach was implemented in search for the major antibacterial compound in C. creticus resin against two Gram-negative ( Escherichia coli and Pseudomonas aeruginosa ) and two Gram-positive bacteria ( Bacillus cereus and Micrococcus flavus ). Bioautographic assay on TLC plates with separated components of Cistus resin extract, revealed a pronounced zone of microbial growth inhibition, corresponded to a highly active compound with Rf values of 0.45, structurally characterized and identified as ent -3 β -acetoxy-13- epi -manoyl oxide. This finding opens the route for focusing on isolation and functional characterization of genes involved in the biosynthesis of ent -3 β -acetoxy-13- epi -manoyl oxide and its precursor ent -3 β -hydroxy-13- epi -manoyl oxide, with the aim to establish sustainable in vitro biotechnological protocols for its large-scale production in homologous and heterologous plant and microbial hosts.

Terpene relationships among some soft and hard pine species

AB  - In this study we have reported the chemical composition of the essential oils obtained from the needles of twelve Pinus taxa, belonging to the subgenera Pinus (sections Pinus and Trifoliae) and Strobus (sect. Quinquefoliae). Monoterpenes dominated over sesquiterpenes in most of the investigated taxa of both subgenera, except in representatives of the subsection Pinaster, subgenus Pinus. alpha-Pinene was the most abundant terpene in eight pines, but other dominant terpenes were found in some taxa -trans-caryophyllene, germacrene D, abietadiene and beta-pinene. In the subsect. Pinaster, the most dominant compounds varied from species to species. In addition, P. halepensis and P. pinaster had the highest percentage of diterpenes, while P. heldreichii was the richest in germacrene D. In the PCA and cluster analyses, three species from the subsect. Pinaster emerged as the most distant: P. halepensis, P. pinaster, and P. heldreichii. In addition, the possible taxonomic implications of the terpene profile in the analysed Pinus taxa were also discussed.
AB  - 48 |vol. 46 (1) U ovom proučavanju objavili smo hemijski sastav etarskih ulja dobijenih iz iglica 12 taksona roda Pinus, koji pripadaju podrodovima Pinus (sekcije Pinus i Trifoliae) i Strobus (sekcija Quinquefoliae). U većini istraživanih taksona monoterpeni dominiraju nad seskviterpenima kod oba podroda, osim kod uzoraka podsekcije Pinaster podroda Pinus. α-Pinen je najobilniji kod 8 borova, ali kod nekih taksona dominantni su trans-kariofilen, germakren D, abietadien i β-pinen. U subcekciji Pinaster dominantne komponente variraju od vrste do vrste. Nadalje, P. ha- lepensis i P. pinaster imaju najveći procenat diterpena, dok je P. heldreichii najbogatiji u germakrenu D. U PCA i klaster analizama pokazale su se najudaljenije tri vrste subsekcije Pinaster: P. halepensis, P. pinaster i P. heldreichii. Nadalje, u analiziranim vrstama roda Pinus diskutovane su moguće taksonomske implikacije terpenskog profila.

abstract = "In this study we have reported the chemical composition of the essential oils obtained from the needles of twelve Pinus taxa, belonging to the subgenera Pinus (sections Pinus and Trifoliae) and Strobus (sect. Quinquefoliae). Monoterpenes dominated over sesquiterpenes in most of the investigated taxa of both subgenera, except in representatives of the subsection Pinaster, subgenus Pinus. alpha-Pinene was the most abundant terpene in eight pines, but other dominant terpenes were found in some taxa -trans-caryophyllene, germacrene D, abietadiene and beta-pinene. In the subsect. Pinaster, the most dominant compounds varied from species to species. In addition, P. halepensis and P. pinaster had the highest percentage of diterpenes, while P. heldreichii was the richest in germacrene D. In the PCA and cluster analyses, three species from the subsect. Pinaster emerged as the most distant: P. halepensis, P. pinaster, and P. heldreichii. In addition, the possible taxonomic implications of the terpene profile in the analysed Pinus taxa were also discussed., 48 |vol. 46 (1) U ovom proučavanju objavili smo hemijski sastav etarskih ulja dobijenih iz iglica 12 taksona roda Pinus, koji pripadaju podrodovima Pinus (sekcije Pinus i Trifoliae) i Strobus (sekcija Quinquefoliae). U većini istraživanih taksona monoterpeni dominiraju nad seskviterpenima kod oba podroda, osim kod uzoraka podsekcije Pinaster podroda Pinus. α-Pinen je najobilniji kod 8 borova, ali kod nekih taksona dominantni su trans-kariofilen, germakren D, abietadien i β-pinen. AB  - The variability of n-alkanes in the needle cuticular wax of Pinus heldreichii and P. peuce in two natural populations from the Scardo-Pindic mountains was investigated for the first time. Gas chromatography (GC) and gas chromatography/mass spectrometry (GC-MS) analyses of two-year-old needles were performed us-ing an Agilent 7890A GC equipped with an inert 5975C XL EI/CI mass spectrometer detector (MSD) and a flame ionization detector (FID) connected by a capillary flow technology 2-way splitter with make-up. An HP-5MS capillary column was used. n-Alkanes ranged from C19 to C33. In P. heldreichii the most abundant were C23, C25 and C27 (16.3, 15.6 and 12.8 % on average, respectively), while in P. peuce they were C29, C25, and C27 (16.5, 15.3 and 13.5 % on average, resp.). Mid-chain and long-chain n-alkanes prevailed in both species. Principle component analysis (PCA) and Cluster analyses of five and six n-alkanes, respectively, showed divergence of the Scardo-Pindic populations from the Dinaric ones.

AB  - Recently, various insect secretions have been tested as possible antimicrobial agents. In beetles, these secretions are essentially products of various exocrine glands, of which particular emphasis is on pygidial glands that are common for the suborder Adephaga. The antimicrobial potential of the pygidial gland secretion isolated from the adults of Calosoma sycophanta against human pathogens has been tested and compared with the potential of other carabid species, particularly within the tribe Carabini. The antimicrobial assay includes a microdilution method which was applied in order to determine the minimal inhibitory, minimal fungicidal and minimal bactericidal concentrations. It has been tested the effect of the secretion against eight strains of fungal and eight strains of bacterial species. The secretion sample of the tested carabid species has shown statistically significant antifungal effect against all strains of treated micromycetes, the highest in comparison with previously tested carabids (Carabus spp., Laemostenus punctatus and Abax parallelepipedus). Aspergillus versicolor proved to be the most sensitive micromycete, while the remaining seven fungal strains have shown the same level of sensibility. In comparison with commercial mycotics ketoconazole and bifonazole, applied as positive controls, the tested secretion showed much higher antifungal activity for all fungal strains. Antibacterial effect has been manifested only against one bacterial strain (Escherichia coli), contrary to other previously studied carabid species. These observations might have a significant impact on the ecological domain and possible purpose in biomedical studies and applications in the future. Additionally, morpho-histology of the pygidial glands of C. sycophanta is investigated and discussed.

AB  - The antifungal potential of the pygidial gland secretion of the troglophilic ground beetle Laemostenus
punctatus from a cave in Southeastern Serbia against cavedwelling micromycetes, isolated from the same habitat, has
been investigated. Eleven collected samples were analyzed
and 32 isolates of cave-dwelling fungi were documented. A
total of 14 fungal species were identified as members of the
genera Aspergillus, Penicillium, Alternaria, Cladosporium,
Rhizopus, Trichoderma, Arthrinium, Aureobasidium,
Epicoccum, Talaromyces, and Fusarium. Five isolates were
selected for testing the antifungal activity of the pygidial gland
se cr etio n: Talaromyces d uclauxi, Aspergillus
brunneouniseriatus, Penicillium sp., Rhizopus stolonifer, and
Trichoderma viride. The microdilution method has been applied to detect minimal inhibitory concentrations (MICs) and
minimal fungicidal concentrations (MFCs). The most sensitive isolate was Penicillium sp., while the other isolates demonstrated a high level of resistance to the tested agent.
L. punctatus has developed a special mechanism of producing
specific compounds that act synergistically within the
secretion mixture, which are responsible for the antifungal
action against pathogens from the cave. The results open opportunities for further research in the field of ground beetle
defense against pathogens, which could have an important
application in human medicine, in addition to the environmental impact, primarily.

Trichoderma viride. The microdilution method has been applied to detect minimal inhibitory concentrations (MICs) and
minimal fungicidal concentrations (MFCs). The most sensitive isolate was Penicillium sp., while the other isolates demonstrated a high level of resistance to the tested agent.
L. punctatus has developed a special mechanism of producing
specific compounds that act synergistically within the
secretion mixture, which are responsible for the antifungal
action against pathogens from the cave. The results open opportunities for further research in the field of ground beetle
defense against pathogens, which could have an important
application in human medicine, in addition to the environmental impact, primarily.",

AB  - Cuticular hydrocarbons (CHCs) in Drosophila melanogaster represent the basis of chemical communication being involved in
many important biological functions. The aim of this study was to characterize chemical composition and variation of cuticular
profiles in five D. melanogaster strains. These strains were reared for approximately 300 generations on five diets: standard
cornmeal medium and substrates prepared with apple, banana, tomato, and carrot. Differences in quantity and/or quality in
CHCs were assumed as a result of activation of different metabolic pathways involved in food digestion and adaptations to the
particular diet type.
In total, independently of sex and strain, 66 chemical compounds were identified. In females of all strains, 60 compounds were
identified, while, in males, 47 compounds were extracted. Certain new chemical compounds for D. melanogaster were found.
MANOVA confirmed that CHC amounts significantly depend on sex and substrates, as well as on their interactions. Discriminant
analysis revealed that flies belonging to ÐappleÏ and ÐcarrotÏ strains exhibited the most noticeable differences in CHC repertoires.
A non-hydrocarbon pheromone, cis-vaccenyl acetate (cVA) also contributed to the variation in the pheromone bouquet among
the strains. Variability detected in CHCs and cVAmay be used in the explanation of differences in mating behaviour previously
determined in analyzed fly strains.

Variability of n-Alkanes and Nonacosan-10-ol in Natural Populations of Picea omorika

AB  - This is the first report of population variability of the contents of n-alkanes and nonacosan-10-ol in the needle epicuticular waxes of Serbian spruce (Picea omorika). The hexane extracts of needle samples originated from three natural populations in Serbia (Vranjak, Zmajevaki potok, and Mileevka Canyon) were investigated by GC and GC/MS analyses. The amount of nonacosan-10-ol varied individually from 50.05 to 74.42% (65.74% in average), but the differences between the three investigated populations were not statistically confirmed. The results exhibited variability of the composition of n-alkanes in the epicuticular waxes with their size ranging from C18 to C35. The most abundant n-alkanes were C29, C31, and C27 (35.22, 13.77, and 12.28% in average, resp.). The carbon preference index of all the n-alkanes (CPItotal) of the P. omorika populations (average of populations IIII) ranged from 3.3 to 11.5 (mean of 5.9), while the average chain length (ACL) ranged from 26.6 to 29.2. The principal component and cluster analyses of the contents of nine n-alkanes showed the greatest difference for the population growing in the Mileevka Canyon. The obtained results were compared with previous literature data given for other Picea species, and this comparison was briefly discussed.

abstract = "This is the first report of population variability of the contents of n-alkanes and nonacosan-10-ol in the needle epicuticular waxes of Serbian spruce (Picea omorika). The hexane extracts of needle samples originated from three natural populations in Serbia (Vranjak, Zmajevaki potok, and Mileevka Canyon) were investigated by GC and GC/MS analyses. The amount of nonacosan-10-ol varied individually from 50.05 to 74.42% (65.74% in average), but the differences between the three investigated populations were not statistically confirmed. The results exhibited variability of the composition of n-alkanes in the epicuticular waxes with their size ranging from C18 to C35. The most abundant n-alkanes were C29, C31, and C27 (35.22, 13.77, and 12.28% in average, resp.). The carbon preference index of all the n-alkanes (CPItotal) of the P. omorika populations (average of populations IIII) ranged from 3.3 to 11.5 (mean of 5.9), while the average chain length (ACL) ranged from 26.6 to 29.2. The principal component and cluster analyses of the contents of nine n-alkanes showed the greatest difference for the population growing in the Mileevka Canyon. The obtained results were compared with previous literature data given for other Picea species, and this comparison was briefly discussed.",

AB  - This is the first report of individual variability and population diversity of the contents of nonacosan-10-ol and n-alkanes in the needle cuticular waxes of Bosnian pines originated from Montenegro, regarded as Pinus heldreichii var. leucodermis, and from Serbia, regarded as P. heldreichii var. pancici. The amount of nonacosan-10-ol varied individually from 27.4 to 73.2% (55.5% in average), but differences between the four investigated populations were not statistically confirmed. The size of the n-alkanes ranged from C18 to C33. The most abundant n-alkanes were C23, C27, and C25 (12.2, 11.2, and 10.8% in average, resp.). The carbon preference index (CPI) of the n-alkanes ranged from 0.8 to 3.1 (1.6 in average), while the average chain length (ACL) ranged from 20.9 to 26.5 (24.4 in average). Long-chain and mid-chain n-alkanes prevailed (49.6 and 37.9% in average, resp.). It was also found that the populations of P. heldreichii var. leucodermis had predominantly a narrower range of n-alkanes (C18?C31) than the trees of the variety pancici (C18?C33). AB  - This is the first report on the composition and variability of the needle-wax n-alkanes in natural populations of Pinus nigra in Serbia. Samples of 195 trees from seven populations belonging to several infraspecific taxa (ssp. nigra, var. gocensis, ssp. pallasiana, and var. banatica) were analyzed. In general, the size of the n-alkanes ranged from C16 to C33, with the exception of ssp. nigra, for which it ranged from C18 to C33. The most abundant were C23-, C25-, C27-, and C29-alkanes. The needle waxes of Populations IIII and V were characterized by a higher content of C23-, C25-, and C27-alkanes and a lower content of C24-, C26-, C28-, and C30-alkanes, compared to the other populations, and the trees of these populations could be assigned to ssp. nigra. The samples of Population VI were characterized by higher amounts of C22-, C24-, C30-, and C32-alkanes and lower amounts of C25- and C27-alkanes, and the trees could be considered as ssp. pallasiana. The samples of Population VII, consisting of trees belonging to var. banatica, were richer in C29-, C31-, and C33-alkanes. The wax compositions of Populations IV and V, both composed of trees previously determined as P. nigra var. gocensis, showed a tendency of splitting. Indeed, the alkane composition of Population IV was closer to that of ssp. pallasiana pines, while that of Population V was more similar to that of ssp. nigra pines. From the results presented here, it is obvious that in the central part of the Balkan Peninsula, significant diversification and differentiation of the populations of black pine exists, and these populations could be defined as different intraspecific taxa. Our results also indicate the validity of n-alkanes as chemotaxonomic characters within this aggregate.

AB  - Ovo je prvo saopštenje o n-alkanima u voskovima iglica varijeteta munike, Pinus heldreichii var. pančići. n-Heksanski ekstrakti uzoraka iglica koji potiču sa sedam izolovanih lokaliteta u Srbiji analizirani su gasnom hromatografijom (GC) i gasnom hromatografijom-masenom spektrometrijom (GC-MS). Rezultati su pokazali da se n-alkani nalaze u opsegu od C18 do C33. Među njima su najobilniji C27, C23, C25 i C29 (12,53, 12,46, 12,00 i 10,38 % u proseku, redom). Ugljenični preferencijalni indeks (CPItotal) Pinus heldreichii var. pančići je bio u opsegu od 1,1 do 2,1 (prosečno 1,6), a dužina niza ugljenikovih atoma (ACLtotal) u opsegu od 25,0 do 25,8 (prosečno 25,3).
