The phenolic constituents and antimicrobial activity of Xanthium spinosum (asteraceae) extracts

AB  - Xanthium spinosum is a cosmopolitan annual herb used in traditional medicine worldwide. Although known from ethnobotanical studies, the species is scarcely investigated from the aspects of phytochemistry and biological activity. Therefore, the phenolic composition and biological activity of X. spinosum were examined. Plant specialised metabolites (phenolics) extracted from the roots, leaves and fruits with dichloromethane:methanol (1:1) were analysed by liquid chromatography mass spectrometry (LC-MS). In total 10 phenolic compounds were identified and quantified. Six compounds were common to all the extracts. Chlorogenic acid was the most abundant constituent in all the extracts (4.262 mg/g in the fruit extract, 0.820 mg/g in the leaf extract, and 0.540 mg/g in the root extract). The biological activity (antimicrobial and antibiofilm) of the extracts was tested against 12 microfungi and 12 bacterial strains by the microdilution method. All the extracts exhibited moderate antimicrobial and antibiofilm activity and inhibited the growth of most of the examined microorganisms. The obtained results indicate the potential role of the tested extracts in pharmacy and medicine.

AB  - .Global health and modern medicine are facing a worldwide issue nowadays – antimicrobial resistance of microorganisms. A great interest in the pursuit of bioactive compounds from plants that can be antimicrobial drugs has been triggered due to multidrug resistance in pathogenic microorganisms and undesirable side effects of certain antibiotics (Alviano & Alviano, 2009). Ethnobotanical data suggest that many Centaurea species are traditionally used for treatment of various ailments in people and animals, as well as for nourishment (Khammar & Djeddi, 2012). Extracts obtained from Centaurea exhibit wide range of biological activity (Khammar & Djeddi, 2012) with a numerous studies concerning antimicrobial activity (Güven et al., 2005; Karamenderes et al., 2006; Özcan et al., 2019; Dimkić et al., 2020). Centaurea glaberrima Tausch (Asteraceae), Circum - Mediterranean Clade sensu Hilpold et al. (2014), is an endemic plant species that inhabits fields as well as a rocky cliffs and waste places in the West Balkans (Dostál, 1976). There is no information on biological activity of C. glaberrima therefore the aim of this study was to investigate antimicrobial potential of ethyl acetate extract of this endemic species.

Water soluble biomolecules from Nepeta nuda regulate microbial growth: A case study of apple juice preservation

AB  - The following study was designed to explore antimicrobial properties of the by-product obtained in a hydro-distillation process of essential oil from Nepeta nuda L. We strived to develop a novel drink with antimicrobial self-preserving properties based on two components, N. nuda decoct and apple juice. By using 96-well plate microdilution assay it was shown that the N. nuda decoct has antimicrobial potential towards 8 bacterial and 6 fungal species, with the range of minimal inhibitory concentrations 10-300 mg/mL. By using actual food system, such as apple juice, in combination with and without short thermal treatment, we have shown that the decoct of N. nuda can inhibit the growth of food contaminant fungus Penicillium aurantiogriseum. It was determined that 3 volumes of decoct (500 mg/mL) and 22 volumes of apple juice should be mixed in order to obtain self-preserving drink resistant to P. aurantiogriseum contamination. Likewise, when thermal treatment (80 °C for 10 s) is included, self-preserving mixture of decoct and apple juice should be made in volume ratios 3:47, respectively. The designed product maintained the pleasant taste as determined by panelists during the sensorial evaluation. Chemical investigations (UHPLC-Orbitrap MS analysis) of N. nuda decoct showed that the most abundant compound was 1,5,9-epideoxyloganic acid (0.410 mg/g of dried decoct). Since N. nuda is traditionally used as a tea, we presented the novel formulation of the drink with antimicrobial properties based on the its decoct and apple juice.

AB  - Arthemisia absinthium L., pelin, je višegodišnja žbunasta biljka sa brojnim lekovitim svojstvima. Pelinkovac je naziv 
za likere sa 28% alkohola, nazvane tako po pelinu koji im daje aromu. Ispitani su likeri Pelinkovac (Slatka kuća, 
Kragujevac, Srbija - SK) i Gorki List (Neodoljivo bilje, Subotica, Srbija - GL). Kako bi utvrdili da li dodatno 
dodavanje pelina u likere može povećati njihovo korisno dejstvo, mikrodilucionom metodom je ispitana njihova 
antimikrobna aktivnost i poređena sa rastvorom pelina u 28% etanolu (PE). Korišćeni su mikroorganizmi izolovani iz 
usne duplje pacijenata, gljive C. albicans, C. glabrata i C. krusei i bakterije S. pyogenes i E. faecalis. Najbolju 
antifungalnu aktivnost je pokazao PE sa minimalnom inhibitornom koncentracijom (MIK) 0.17 mg/mL. Likeri su 
imali sličnu antifungalnu aktivnost sa MIK (4.19–17.32 mg/mL). Najosetljiviji je bio klinički izolat C. glabrata, potom 
sojevi C. albicans (3 klinička izolata i jedan referentni soj C. albicans ATCC 10231), dok je najotporniji bio C. krusei 
(2 klinička izolata). Najbolju antibakterijsku aktivnost ispoljio je PE (MIK 0.34-1.36 mg/mL), a soj E. faecalis je bio 
osetljiviji od S. pyogenes. U sprečavanju formiranja biofilma C. albicans najefikasniji je SK (48.2% inhibicije), GL 
(43.6%), PE (26.4%). Formiranje biofilma E. faecalis bilo je redukovano za 86.0% sa SK, 74.6% (GL), odnosno 
13.6% sa PE. Biofilm S. spyogenes je bio rezistentniji, GL nije sprečio njegovo formiranje, SK je sprečio za 10.3% 
dok ga je PE redukovao za 74.5%. Oba likera mogu da spreče rast ispitanih mikroorganizama in vitro, ali je SK bolji 
u sprečavanju njihovog grupisanja, te bi se unapređivanju njegovog sadržaja moglo doprineti, naročito dodavanjem 
pelina koji se pokazao kao jak antimikrobni agens.
AB  - Arthemisia absinthium L., wormwood, is perennial bushy plant known for various health benefits. Pelinkovci is
Serbian name for the liqueurs with 28% alcohol that contain wormwood (pelin) as aromatic constituent. The study
examined two commercial liqueurs: Pelinkovac (Slatka kuća, Kragujevac, Serbia – PE) and Gorki list (Neodoljivo
bilje, Subotica, Srbija - GL). In order to determine if addition of plant extract could increase beneficial effects of these
strong drinks, microdilution method was used to examine their antimicrobial activity which has been compared with
the activity of wormwood extract dissolved in 28% ethanol (PE). Microorganisms, pathogens isolated from oral cavity,
were tested (fungi Candida albicans, Candida glabrata and Candida krusei as well as bacteria Streptococcus pyogenes
and Enterococcus faecalis). Highest antifungal potential was observed for PE with minimal inhibitory concentration
(MIC) 0.17 mg/mL. Liqueurs had similar antifungal activity with MIC in range 4.19–17.32 mg/mL. C. glabrata
clinical isolate was the most sensitive followed by strains of C. albicans (3 clinical isolates and one reference strain
C. albicans ATCC 10231), while C. krusei was the most resistant to antimicrobial treatment. Highest antibacterial
activity was observed for PE (MIC 0.34-1.36 mg/mL), with E. faecalis strain being more sensitive than S. pyogenes.
Best activity in preventing C. albicans biofilm formation was observed for SK (48.2% inhibition), GL (43.6%), PE
(26.4%). Formation of E. faecalis biofilm was inhibited for 86.0% with SK, 74.6% GL, 13.6% PE. Biofilm formed
by S. pyogenes was more resistant and GL could not prevent it from establishing, while SK prevented it for 10.3%.
PE had highest preventing ability since it inhibited S. pyogenes biofilm formation for 74.5%. Both liqueurs are able
to prevent growth of tested pathogenic microorganisms in vitro, but SK had better potential in prevention of grouping
into biofilm, indicating that we could further work on development of this drink, possibly through addition of more
wormwood since it has been proven as strong antimicrobial agent.

C. albicans ATCC 10231), while C. krusei was the most resistant to antimicrobial treatment. Highest antibacterial
activity was observed for PE (MIC 0.34-1.36 mg/mL), with E. faecalis strain being more sensitive than S. pyogenes.
Best activity in preventing C. albicans biofilm formation was observed for SK (48.2% inhibition), GL (43.6%), PE
(26.4%). Formation of E. faecalis biofilm was inhibited for 86.0% with SK, 74.6% GL, 13.6% PE. Biofilm formed
by S. pyogenes was more resistant and GL could not prevent it from establishing, while SK prevented it for 10.3%.
PE had highest preventing ability since it inhibited S. pyogenes biofilm formation for 74.5%. Both liqueurs are able
to prevent growth of tested pathogenic microorganisms in vitro, but SK had better potential in prevention of grouping
into biofilm, indicating that we could further work on development of this drink, possibly through addition of more
wormwood since it has been proven as strong antimicrobial agent.",
