Redox properties of transitional milk from mothers of preterm infants.

AB  - AIM There is a discrepancy between the amount of transitional milk produced by mothers of preterm infants and the low capacity of premature infants to consume it. This milk can be used in milk banks, but previous studies found that there are large variations in the level of host-defence proteins in individual samples of milk from mothers of premature infants, which implies that large individual variations in antioxidative defence composition are also possible. METHODS Milk samples were collected from 20 healthy mothers of preterm infants. We determined the values for non-enzymatic antioxidative capacity parameters (oxygen radical absorbance capacity (ORAC)), static oxidation-reduction potential (ORP), activities of antioxidant defence enzymes and the amount of vitamin C in whole milk, skim and whey fractions of transitional milk. RESULTS The main low-molecular-weight antioxidant in transitional milk is vitamin C and most of it is contained in whey. ORAC is higher in whole transitional milk than in skim milk and whey, and ORP is lower in whole transitional milk than that in skim milk and whey. Antioxidative enzyme activities are similar in all individual samples of transitional milk from mothers of preterm infants. CONCLUSIONS Our results indicate that transitional milk of mothers of preterm infants shows slow individual variations in antioxidative defence composition; therefore, it can be used in human milk banks.

Biochemical and pharmacological evaluation of 4-hydroxychromen-2-ones bearing polar C-3 substituents as anticoagulants

AB  - The objective of this study was to investigate in vitro and in vivo anticoagulant activity of sixteen 4-hydroxycoumarin derivatives bearing polar C-3 scaffolds. The activity was evaluated by measuring prothrombin time. Enhanced anticoagulant activity in vitro was observed for all tested compounds. Upon successive administration of 0.5 mg/kg of body weight to adult Wistar rats, over a period of five days, four derivatives (2b, 4c, 5c and 9c) presented anticoagulant activity in vivo. The most active compound was 2b, with PT = 30.0 s. Low or non-toxic effects in vivo were determined based on the catalytic activity of liver enzymes and the concentration of bilirubin, iron and proteins. Metabolic pathways of the most active compounds in vivo were determined after GC/MS analysis of collected rat urine samples. The excretion occurs by glucuronidation of 7-hydroxy forms of tested derivatives. In vivo results were described using PLS-based CoMFA and CoMSIA 3D-QSAR studies, which showed CoMFA-SE (q2 = 0.738) and CoMSIA-SEA (q2 = 0.763) to be the statistically most relevant models. Furthermore, molecular docking and DFT mechanistic studies performed on the rat VKORC1 homology model revealed interactions between the 4-OH coumarin group in the form of phenolic anion and the Cys135 catalytic site in the transition state.

Uticaj ubrzanog starenja na promene redukovanog i oksidovanog glutationa u klijancima kukuruza

AB  - A trial with four maize inbred lines with the ability to have different durations of seed germination in the course of the accelerated ageing (AA) treatment was set up. Changes of the content of total, reduced and oxidized glutathione (expressed as monomers) were observed in the seeds and seedlings before and after the treatment. For the first time, changes of glutathione in whole seedlings, as well as in the rest of the seed, were analyzed. It was noticed that maize in breds with a smaller decrease of the total glutathione but with an increase of the oxidized form had the ability of prolonged germination. In the control seed- lings, the amount of total glutathione was lower than in the treated ones. Maize seeds which lost germination faster had greater losses of total glutathione with an increased content of the oxidized form in seedlings. The ability of prolonged germination together with the possibility of glutathione synthesis in seedlings are genotypic traits.
AB  - Postavljen je ogled sa četiri linije kukuruza različite dužine očuvanja klijavosti semena tokom tretmana ubrznog starenja. Ispitivane su promene ukupnog, kao i redukovanog i oksidovanog glutationa (izraženi kao monomeri) u semenu i klijancima pre i nakon ubrzanog starenja. U istraživanjima su prvi put analizirane promene glutationa u celim klijancima, kao i ostatku semena. U semenu linija kukuruza koje imaju sposobnost dužeg očuvanja klijavosti bio je manji gubitak ukupnog glutationa, uz povećanje udela oksidovanog oblika. Kod njihovih klijanaca došlo je do povećanja sadržaja ukupnog glutationa u odnosu na kontrolne klijance. Seme kukuruza koje brže gubi klijavost imalo je veće gubitke ukupnog glutationa, uz veći sadržaj oksidovanog oblika kod formiranih klijanaca. Dužina očuvanja klijavosti, kao i sinteza glutationa kod klijanaca je genotipska osobina.

Vitamin E and serum lipid level in patients with rectal carcinoma

AB  - Background: Epidemiological studies indicated connection of the rectal carcinoma with the dietary lipid content, but the identification of the links associating the causes with the consequences are still far from being resolved. On the other hand, it has been experimentally confirmed that vitamin E expresses antitumor effects in some types of malignancies, but these effects strongly depend not only on the vitamin E form employed, but also on the presence of other antioxidants, e.g. selenium (Se). Materials and methods: In this work, we monitored the values of the following parameters in blood of patients diagnosed as having rectal carcinoma: cholesterol, HDL, LDL, triglycerides and vitamin E. The measurements were performed immediately before and after the surgery (rectal resection), as well as 1, 3 and 6 months postoperatively. One group of patients was postoperatively supplemented with dietary antioxidants and the values of the above parameters were monitored by the same schedule. Results: Ratio of vitamin E content and the sum of triglycerides and cholesterol (E/trg+chol) was lower in patients than in the population of adult healthy volunteers, suggesting it to be a more reliable indicator of the risk factor for rectal carcinoma development than the level of vitamin E alone. A decline of both vitamin E level and E/trg+chol ratio was noticed in patients as early as one month postoperatively, while in the group receiving antioxidants, a significant decrease of these parameters was recorded only 6 months postoperatively. Conclusion: The fact that the supplementation postpones reduction of vitamin E level postoperatively suggests that antioxidants could express some beneficial effects on the outcome of a standard therapy.
