AB  - Assessment of impact of pollution in the environmental studies requires multi-endpoints approach to properly
link cause and effects of pollution with focus on chemical pollution. In this sense systematic weight-of-evidence
approach (WoE) with multiple lines of evidence (LoEs) is preferable to apply. The WoE approach highlights the
importance to identify strengths and weaknesses of used LoEs. Therefore, in this study we have tested efficacy
of genotoxicological endpoints as one of the LoEs in the in situ assessment of pollution effects in the freshwater
ecosystems using Alburnus alburnus (bleak) as a bioindicator species. Additional LoEs that were used in the
study are: component-based methods for the assessment of SumTU in water based on monitoring data of the
Serbian Environmental Protection Agency (SEPA), effect based methods employing in vitro genotoxicological
analyses of Joint Danube Survey 4 (JDS4) water extracts and field derived species inventories for the
assessment and indication of ecological status/potential based on SEPA and JDS4 data. The study was
conducted within the JDS4 campaign at nine sampling sites at the Tisa, Sava, Velika Morava and Danube rivers
in the Republic of Serbia. In the case of three sampling sites, the results were uniform, meaning that all four
LoEs pointed to pollution pressure. The differences in the LoEs outcomes for other sites indicated the
importance of multiple LoEs approach for proper identification of ecological impact. In the case of current
study we have identified comet and micronucleus assay to be appropriate for the genotoxicological assessment
in the in situ studies due to high sensitivity in discrimination of sites in relation to pollution intensity, while
RAPD analysis to be more suitable for controlled ex situ investigations.

AB  - Environmental studies which aim to assess the ecological impact of chemical and other types of pollution should employ a complex weight-of-evidence approach with multiple lines of evidence (LoEs). This study focused on in situ genotoxicological methods such as the comet and micronucleus assays and randomly amplified polymorphic DNA analysis as one of the multiple LoEs (LoE3) on the fish species Alburnus alburnus (bleak) as a bioindicator. The study was carried out within the Joint Danube Survey 4 (JDS4) at nine sites in the Danube River Basin in the Republic of Serbia. Out of nine sampling sites, two were situated at the Tisa, Sava, and Velika Morava rivers, and three sites were at the Danube River. The three additionally employed LoEs were: SumTUwater calculated based on the monitoring data in the database of the Serbian Environmental Protection Agency (SEPA) (LoE1); in vitro analyses of JDS4 water extracts employing genotoxicological methods (LoE2); assessment of the ecological status/potential by SEPA and indication of the ecological status for the sites performed within the JDS4 (LoE4). The analyzed biomarker responses in the bleak were integrated into the unique integrated biomarker response index which was used to rank the sites. The highest pollution pressure was recorded at JDS4 39 and JDS4 36, while the lowest was at JDS4 35. The impact of pollution was confirmed at three sites, JDS4 33, 40, and 41, by all four LoEs. At other sampling sites, a difference was observed regarding the pollution depending on the employed LoEs. This indicates the importance of implementing a comprehensive weight-of-evidence approach to ensure the impact of pollution is not overlooked when using only one LoE as is often the case in environmental studies.

AB  - Within the Joint Danube Survey 4 (JDS4) (2019) surface water samples
were collected at 24 localities along the Danube River and were extracted by
horizon large volume solid-phase extraction (LVSPE) technique. LVSPE
technique provided 25,000x relative enrichment factor (REF) of the river
samples. The bioactivity of the LVSPE samples was investigated employing
prokaryotic SOS/umuC assay (S9+ and S9-) and battery of bioassays
analyzing different endpoints – cytotoxicity (MTS assay), genotoxicity
(comet assay), cell cycle analyses of ZFL cells and embryo toxicity (zFET) in
eukaryotic models (zebrafish liver cell line – ZFL and Danio rerio embryos –
zebrafish). In the case of the SOS/umuC assay none of the tested samples
(REF100) has induced genotoxic effect. The cytotoxic effect was detected for
11 samples at REF100 (MTS assay). The comet assay results indicated
genotoxic potential for nine samples – one in the Upper Danube, six in the
Middle Danube and two in the Lower Danube. The most potent were
JDS4-37 and JDS4-41 samples collected at two sites in the Serbian part of
the Danube River. These samples were analyzed at REF13. The analysis of
the cell cycle was performed in the case of eight samples for which were
previously recorded genotoxic effect. For one sample (JDS4-41) was found
the G1 phase cell cycle arrest. The zFET assay was performed for all the
samples at REF100 and there was no embryo toxicity or teratogenicity
recorded. The results obtained within this study indicate a higher sensitivity
of eukaryotic models compared to prokaryotic SOS/umuC assay. Likewise, it
was found that most of the genotoxic samples originate from the middle
part of the Danube.

AB  - BACKGROUND
Prokaryotic assays are of great interest in eco/geno-toxicology as they are short-term, simple and cost
effective. Application of strains of Salmonella typhimurium with modifications such as increased cell wall
permeability and lack of excision repair system is common for the detection of genotoxicity. Various eukaryotic models have been developed lately, such as fish derived cell lines, which can mimic the response of the aquatic organisms. Considering different level of cell organization among these models, difference in sensitivity is expected, yet only few studies address this topic.
OBJECTIVES
The major objective was to investigate genotoxic potential of extracts of surface water of the Danube
and to compare the results obtained in applied prokaryotic and eukaryotic models.
METHODS
Surface water samples from 24 sites along the Danube River were extracted by LVSPE Horizon sampler providing relative enrichment factor (REF) of the extracts of 25,000x. For representative prokaryotic model the SOS/umuC assay with S. typhimurium TA1535/pSK1002 was used. Eukaryotic model comprised assessment of cytotoxicity by MTS test and assessment of genotoxicity by comet assay in ZFL (ZebraFish Liver) cells.
RESULTS
While none of the tested samples has induced SOS response, the result of MTS assay indicated that 11 of 24 samples were cytotoxic. In comet assay, genotoxic potential was found in 9 of 24 samples. Results indicated higher sensitivity of used eukaryotic model in comparison with prokaryotic. Applied prokaryotic assay is often suggested to be used as prescreening tool in diagnostics of genotoxic potential which by our opinion could be misleading.

Prokaryotic assays are of great interest in eco/geno-toxicology as they are short-term, simple and cost
effective. Application of strains of Salmonella typhimurium with modifications such as increased cell wall
permeability and lack of excision repair system is common for the detection of genotoxicity. Various eukaryotic models have been developed lately, such as fish derived cell lines, which can mimic the response of the aquatic organisms. Considering different level of cell organization among these models, difference in sensitivity is expected, yet only few studies address this topic.
OBJECTIVES
The major objective was to investigate genotoxic potential of extracts of surface water of the Danube
and to compare the results obtained in applied prokaryotic and eukaryotic models.
METHODS
Surface water samples from 24 sites along the Danube River were extracted by LVSPE Horizon sampler providing relative enrichment factor (REF) of the extracts of 25,000x. For representative prokaryotic model the SOS/umuC assay with S. typhimurium TA1535/pSK1002 was used. Eukaryotic model comprised assessment of cytotoxicity by MTS test and assessment of genotoxicity by comet assay in ZFL (ZebraFish Liver) cells.
RESULTS
While none of the tested samples has induced SOS response, the result of MTS assay indicated that 11 of 24 samples were cytotoxic. In comet assay, genotoxic potential was found in 9 of 24 samples. Results indicated higher sensitivity of used eukaryotic model in comparison with prokaryotic. Applied prokaryotic assay is often suggested to be used as prescreening tool in diagnostics of genotoxic potential which by our opinion could be misleading.",

AB  - In this study, carried out within the Joint Danube Survey 4, a comprehensive microplastic screening in the water column within a large European river basin from its source to estuary, including major tributaries, was realized. The objective was to develop principles of a systematic and practicable microplastic monitoring strategy using sedimentation boxes for collection of suspended particulate matter followed by its subsequent analysis using thermal extraction desorption-gas chromatography/mass spectrometry. In total, 18 sampling sites in the Danube River Basin were investigated. The obtained suspended particulate matter samples were subdivided into the fractions of >100 mu m and <100 mu m and subsequently analyzed for microplastic mass contents. The results showed that microplastics were detected in all samples, with polyethylene being the predominant polymer with maximum contents of 22.24 mu g/mg, 3.23 mu g/mg for polystyrene, 1.03 mu g/mg for styrene-butadiene-rubber, and 0.45 mu g/mg for polypropylene. Further, polymers such as different sorts of polyester, polyacrylates, polylactide, and natural rubber were not detected or below the detection limit. Additional investigations on possible interference of polyethylene signals by algae-derived fatty acids were assessed. In the context of targeted monitoring, repeated measurements provide more certainty in the interpretation of the results for the individual sites. Nevertheless, it can be stated that the chosen approach using an integrative sampling and determination of total plastic content proved to be successful.

AB  - The presence of SARS-CoV-2 RNA in wastewater was first reported in March 2020. Over the subsequent months, the potential for wastewater surveillance to contribute to COVID-19 mitigation programmes has been the focus of intense national and international research activities, gaining the attention of policy makers and the public. As a new application of an established methodology, focused collaboration between public health practitioners and wastewater researchers is essential to developing a common understanding on how, when and where the outputs of this non-invasive community-level approach can deliver actionable outcomes for public health authorities. Within this context, the NORMAN SCORE “SARS-CoV-2 in sewage” database provides a platform for rapid, open access data sharing, validated by the uploading of 276 data sets from nine countries to-date. Through offering direct access to underpinning meta-data sets (and describing its use in data interpretation), the NORMAN SCORE database is a resource for the development of recommendations on minimum data requirements for wastewater pathogen surveillance. It is also a tool to engage public health practitioners in discussions on use of the approach, providing an opportunity to build mutual understanding of the demand and supply for data and facilitate the translation of this promising research application into public health practice.

AB  - In this study, we analyzed the impacts of different environmental conditions on aquatic
worm communities along the Danube River, based on two longitudinal surveys, the Joint Danube
Surveys 2 and 3 (JDS; 2007 and 2013). We identified the most important environmental factors
(among analyzed groups) that shape worm communities: hydromorphlogical alterations, flow
velocity and substrate (HYMO group), dissolved oxygen, nitrates and nitrites (physico-chemical
parameters), zinc and nickel (metals), monobutyltin cation, benzo(b) fluoranthene and
benzo(k)fluoranthene, polychlorinated biphenyls PCB 77 and PCB 118 (selected chemical
determinants—organotin compounds, Polycyclic aromatic hydrocarbons—PAHs and PCBs). A
homogenization of species composition of Oligochaeta assemblages along the Danube was
confirmed. As one of main factors related to biotic homogenization, hydromorphological
alterations represented by similar changes in flow velocity and substrates along Danube’s course
could be singled out. Our results indicate that Oligochaeta could be used for the identification of
the level of hydromorphological degradation in large rivers (homogenization), rather than for
stressors classified as nutrient and organic pollutants. Our results provide additional evidence in
risk assessment of the environment, contributing in water management and monitoring of the
ecological status as proposed by the Water Framework Directive.

AB  - This study was carried out to identify the relations between macroinvertebrate communities and river basin specific (RBS) pollutants in the Danube River. The investigation was performed at 68 sites along 2,500 km of the Danube. Forward selection (FS), canonical correspondence analyses (CCA), the Spearman correlation coefficient (SC) and BIO-ENV analysis (to detect synergistic effects) were used to identify the relations between the macroinvertebrate dataset and selected biological metrics with RBS pollutants. Of the 20 analysed pollutants (preselected based on NORMAN network methodology), seven (2,4-dinitrophenol, chloroxuron, bromacil, dimefuron, amoxicillin, bentazon and fluoranthene) were found to significantly correlate with macroinvertebrate communities. BIO-ENV analysis revealed 3 subsets of environmental variables that were in high correlation with the biota resemblance matrix, consisting mainly of a combination of the above-mentioned pollutants. Our results indicate that there are significant correlations between chemical determinants and aquatic biota. Moreover, this study contributes to the validation of the methodology used for prioritization of RBS pollutants proposed by the NORMAN network.

AB  - This paper discusses the use of different methods (multihabitat - MSH, Kick & Sweep 
method, Air lift sampling, multcorer, Van Veen grab, dredging) for aquatic 
macroinvertebrates sampling. Material was collected from the Danube River as a part of the 
three research programs: Joint Danube Survey 2, performed in period August-September 
2007 from Regensburg to Danube delta (96 sampling sites), Joint Danube Survey 3, 
performed in period August-September 2007 (68 sites) and national monitoring program 
performed in period September-November 2014 along Serbian stretch of the Danube River 
(12 sites). The aim of the study was to contribute to the standardization of the sampling 
procedures for the large lowland rivers. We tested the effectiveness of sampling methods in 
respect to macroinvertebrate taxa richness and metrics typically used for ecological status 
assessment. The selection of sampling approach is also discussed in relation to objectives of 
the survey – e.g. research, or routine monitoring, as well in respect to target taxa. Based on 
the our results we concluded that it is necessary to employ more than one sampling 
technique and to cover all available regions of large rivers, including the deeper parts in the 
case of research programs. For the routine monitoring we recommend to use standardized 
K&S method. Studies of the specific group of organisms require detailed monitoring of 
available habitats, and use of additional sampling methods (e.g. mussels or crayfish studies).

of Gastropoda and total number of taxa. To find synergistic influence of RBS pollutants on 
biota, BIO-ENV analysis was performed and reviled that 3 subsets of environmental variables 
were highly correlated with biota resemblance matrix, covering combination of same 
parameters singled out by FA.

AB  - In this study we assessed the level of genotoxic pollution along the Danube River by measuring the level of DNA damage in the haemocytes of freshwater mussels of Unio sp. (Unio pictorum/Unio tumidus) and Sinanodonta woodiana. The comet assay was used for the assessment of DNA damage. The research was performed on 34 out of 68 sites analysed within the Joint Danube Survey 3 - the world's biggest river research expedition of its kind in 2013. During research, 2285 river kilometres were covered with an average distance of 68km between the sites. The complex data set on concentrations of various substances present in water, suspended particulate matter and sediment on investigated sites gave the opportunity to identify the groups of xenobiotics which mostly affect the studied biomarker - DNA damage. The highest levels of DNA damage were recorded in the section VI (Panonnian Plain), which is under the impact of untreated wastewater discharges. Both positive and negative influences of the large tributaries on the level of genotoxicity in the Danube River were evident. Significant correlation in response was detected between the studied species of freshwater mussels. The level of DNA damage in mussels correlated with concentrations of compounds from the group of hazardous priority substances (polycyclic aromatic hydrocarbons), persistent organic pollutants (dioxins) and emerging pollutants (Oxazepam, Chloridazon-desphenyl).

abstract = "In this study we assessed the level of genotoxic pollution along the Danube River by measuring the level of DNA damage in the haemocytes of freshwater mussels of Unio sp. (Unio pictorum/Unio tumidus) and Sinanodonta woodiana. The comet assay was used for the assessment of DNA damage. The research was performed on 34 out of 68 sites analysed within the Joint Danube Survey 3 - the world's biggest river research expedition of its kind in 2013. During research, 2285 river kilometres were covered with an average distance of 68km between the sites. The complex data set on concentrations of various substances present in water, suspended particulate matter and sediment on investigated sites gave the opportunity to identify the groups of xenobiotics which mostly affect the studied biomarker - DNA damage. The highest levels of DNA damage were recorded in the section VI (Panonnian Plain), which is under the impact of untreated wastewater discharges. AB  - The Joint Danube Survey 3 (JDS3; the biggest river expedition in 2013) had offered the unique opportunity for a large-scale monitoring approach for biomarker response in feral fish collected along a Danube stretch from Kehlheim (DE) to Sulina (RO). The advantage of genotoxicity as a marker for pollution exposure in fish is the early detection of possible long-term effects such as cancer. Therefore, genotoxicity was in the focus of the biomarker investigations in fish during the expedition. Blood samples of common bleak (Alburnus alburnus) for the investigation of the micronucleus frequency and comet tail intensity of fragmented DNA material in erythrocytes were collected at 18 and 12 sampling sites, respectively. For 9 sampling sites same samples were used to compare the . in-situ data for the comparable genotoxic endpoint in the micronucleus (MN) and comet assay (CM). The data of both . in-situ assays showed a significant correlation, indicating the strength and comparability of the data sets. Significant variation in DNA damage in fish along the longitudinal profile of the Danube was demonstrated for both assays compared to reference sites. The results suggest that DNA damage in erythrocytes of fish was mainly affected by wastewater of highly populated regions. No linkage between the results and the general health/dietary status of the fish were revealed, whereas correlation with some genotoxicity drivers in the water phase, suspended particulate matter and sediments could be demonstrated.
