Potential of Thymus pannonicus All. in vitro culture: micropropagation, rosmarinic acid production and antimicrobial activity

AB  - In Serbia, citral chemotype of Thymus pannonicus All. (Lamiaceae) can be only found on Mt
Vršačke planine, as dominant form, where is traditionally used for medicinal purposes. To overcome
difficulties in harvesting due to distribution in inaccessible areas, and to prevent possible
overexploitation from its natural habitat, finding the means for alternative cultivation of this plant
for its prospective beneficial use in human health care was set as objective of this study. Efficient
system for in vitro shoot propagation of T. pannonicus was established for the first time. Different
types of auxin (indole-3-acetic acid, IAA; indole-3-butyric acid, IBA; 1-naphthaleneacetic acid, NAA)
were used for testing the process of rhizogenesis in vitro. Best response was detected on IBA and
NAA for rhizogenesis frequency and number of formed roots, respectively. Rooted plants were
successfully acclimatized. In vitro shoots cultivated on PGR free medium (E1) and medium with 1
mg L-1 IAA (E2) were extracted with 80% methanol and analysed by HPLC. Presence of rosmarinic
acid (RA) as dominant component was indicated in both extracts, with E2 containing higher RA
amount (45.116 mg g-1DW). Antimicrobial activity of extracts was tested using broth microdilution
method against nine strains of laboratory control pathogenic microorganisms. Both extracts
exerted significant antimicrobial activity against all tested pathogens with minimal inhibitory concen-
trations (MIC) in the range of 6.25-50 mg mL-1, while E2 was more effective against Gram positive
bacteria (6.25-12.5 mg mL-1). Presented results could give inducement for potential greater
commercial use of this significant species in the future.

AB  - Korovski suncokret (Helianthus annuus L.) je invazivna vrsta na teritoriji Republike Srbije koja izaziva značajno smanjenje prinosa u različitim poljoprivrednim kulturama. Tokom žetve suncokreta dolazi do rasipanja semena iz koga se u narednim godinama mogu razviti samonikle biljke. Ukoliko se, na određenoj površini, samonikle biljke suncokreta razvijaju duži vremenski period doći će do pojave korovskih formi ove vrste. Spontana hibridizacija korovskog suncokreta sa drugim formama suncokreta može prouzrokovati značajnu agresivnost ove invazivne vrste. Samonikle biljke suncokreta koje potiču od hibrida tolerantnih na ALS inhibitore su uglavnom nosioci gena tolerantnosti zbog čega je njihova osetljivost na herbicide značajno smanjena. Razmenom genetičkog materijala između različitih formi i tolerantnih hibrida suncokreta omogućen je transfer ALS gena koji je odgovoran za tolerantnost na herbicide ALS inhibitore na potomstvo. Poljski ogled je izveden na dva lokaliteta u okolini Požarevca, a tokom tri godine evidentirana je spontana hibridizacije između tolerantnih hibrida suncokreta (Sumo 1 PR i Rimi) i osetljivih hibrida suncokreta (hibrid Duško), samoniklog suncokreta (poreklom od osetljivog hibrida Sremac) i korovskog suncokreta.

AB  - Genetic and morphological interpopulation variability of weed species is often responsible for variable responses to herbicides. As weedy sunflower, an invasive form of Helianthus annuus L., possesses high morphological and genetic variability, very different responses of its populations to herbicides can be expected. This species is one of the dominant weed species in row crops, including maize, in many European countries in which nicosulfuron is intensively used for weed control. There are little available data about the response of this sunflower form to nicosulfuron or of the interpopulation variability of its response to other herbicides. The responses of three weedy sunflower populations to nicosulfuron were studied in field dose-response experiments, and acetolactate synthase (ALS) enzyme activity at different herbicide concentrations was determined in vitro. Interpopulation variability in the response to nicosulfuron was confirmed. Populations WS2 and WS3 were more that 20-fold and 30-fold less susceptible to nicosulfuron, respectively, than population WS1, based on fresh weight, whereas the differences were not so prominent based on other parameters, including plant height, leaf area and ALS activity, and ranged from 2 to 12-fold.
