AB  - The species of the genus Centaurium Hill are characterized by a pronounced phenotypic
plasticity, while interspecific hybridization readily occurs. Previous studies reported hybridization events between C. erythraea and C. tenuiflorum in the Mediterranean region,
but the status of a potentially hybrid taxon (termed "Centaurium ×tenuiflorum") remains
unclear. Therefore, molecular markers (EST-SSR, ITS, and chloroplast DNA markers) were
combined and their usefulness in estimating overall genetic variability and in identifying
phylogenetic relations between Centaurium taxa was scrutinized. Both EST-SSR and ITS
markers proved to be well suited to distinguish the parental taxa C. erythraea and C.
tenuiflorum, and a putative hybridogenic taxon "C. ×tenuiflorum", the last one being more
related to C. erythraea. Conversely, maternally inherited chloroplast markers (trnL-F, rbcL
and psbA-trnH) nominated C. tenuiflorum as a donor of plastid.

AB  - Utilization of microsatellite molecular markers was analyzed in Iris pumila and five other species 
belonging to the same genus: I. humilis, I. sibirica, I. pseudacorus, I. spuria and I. variegata, which 
inhabit the northern part of the Republic of Serbia. We selected 40 EST-SSR markers previously 
reported for the genus Iris. Sixteen markers that showed polymorphism were chosen for further 
research. To examine the potential of the selected microsatellite loci analysis in the detection of 
different I. pumila clones, we selected 18 individuals of this species that belonged to different 
genotypes. Optimization of amplification was performed for all 16 microsatellite loci: temperature 
program of the PCR protocol and number of amplification cycles were optimized as well as primer 
and DNA concentrations used in the reactions. Seven markers that were variable across the ana lyzed individuals were selected. The final analysis was performed using only 5 microsatellite loci 
since high-quality amplification for two loci was not possible. No greater deviations in the length 
of the fragments from the expected were observed, except for one primer. All microsatellite loci 
contained three-nucleotide repeats. I. pumila is tetraploid species and in several plants the presence of four alleles were observed. The observed number of alleles per locus ranged from 3 to 9. 
In the results obtained across 18 I. pumila individuals using 5 microsatellite markers, 17 multilocus 
genotypes were observed.

AB  - Species of the genus Centaurium are highly variable and clinal morphological variations caused
by environmental conditions are very noticeable. Nevertheless, it is also well documented that
species and subspecies of this genus hybridize readily in nature and it is considered that interspecific hybridization may be one of the leading causes of variability within the genus. Earlier studies
indicate that the species C. erythraea Rafn and C. tenuiflorum (Hoffmanns. et Link) Fritsch in areas
with a Mediterranean climate hybridize in nature. The taxonomic status of the hybridogenous
taxon, which probably originated through the hybridization of the mentioned species, here called
“C. ×tenuiflorum”, has not been resolved. Since it has been proven to be a useful genetic marker in
giving evidence of hybridization among several species of the genus Centaurium, the chloroplast
trnL - trnF region was used for the study of hybridization between these particular species. The
results of variation in the trnL - trnF region indicated that the taxon “C. ×tenuiflorum” shared a chloroplast haplogroup with the species C. tenuiflorum, which implies that C. tenuiflorum might be the
donor of chloroplast DNA. In subsequent research, it is necessary to combine the data obtained
from the analysis of the trnL - trnF region with the data that would be obtained from the analysis
of other regions of genomic and/or chloroplast DNA to resolve the status of this potentially hybridogenic taxon.

AB  - The majority of species belonging to genus Nepeta (fam. Lamiaceae) produce and accumulate nepetalactones as the main secondary metabolites. Nepetalactones are iridoid monoterpenoids recognized for their wide spectrum of high-value bioactive properties, namely antimicrobial, cytostatic, anti-inflammatory, phytotoxic, etc. However, biosynthetic pathway of these compounds in plants remains to a great extent unexplored. Most probably they derive through general iridoid biosynthetic pathway leading to nepetalactol, a common intermediate of all plant-produced iridoids, that is formed in a reaction catalyzed by iridoid synthase (IS, EC 1.3.1.99). Present investigation entails homology based degenerative primer approach which led to the identification of two promising iridoid synthase candidates (NrIS1 and NrIS2) in leaves of Nepeta rtanjensis Diklić & Milojević, rich in trans,cis-nepetalactone (trans,cis-NL) and dehydronepetalactone (DNL). Comparative UHPLC/qqqMS profiling of nepetalactones in detached trichomes and abraded leaves, complemented with Raman spectroscopy and dichloromethane dipping experiment highlighted trichomes as the main site of their accumulation. This was further supported by the significantly high trichome/abraded leaves expression ratio of NrIS1 (~31) and NrIS2 (~25), which strongly indicated the involvement of these genes in nepetalactone biosynthesis. High correlation between NrIS1 expression profile and trans,cis-NL and DNL contents in five developmental stages of N. rtanjensis leaves confirmed developmental regulation of nepetalactone biosynthesis. Concentrations of trans,cis-NL and DNL were 22-fold and 80-fold lower in the oldest leaves investigated than in the youngest leaves, respectively. In addition, the expression pattern of the NrIS1 followed the same trend exhibiting 7.4-fold higher expression in the oldest leaves compared to the youngest ones. The present study on spatial and developmental regulation of the nepetalactones production and accumulation in N. rtanjensis leaves, as well as the identification of two IS candidate genes is a proof-of-the-concept research that serves as a cornerstone for future studies on elucidation of the nepetalactones biosynthesis.

AB  - Nepetalactones are accumulated as the main secondary metabolites in the majority of Nepeta species; however, their biosynthetic pathway remains to a great extent unknown, as well as their relationship to the overall physiology of these plants. Nepetalactones most certainly arise through general iridoid biosynthetic pathway leading to nepetalactol, a common precursor of all plant-derived iridoids that is formed in a reaction catalysed by iridoid synthase (IS, EC 1.3.1.99). In this paper, we aimed to localize the sites of nepetalactone biosynthesis and accumulation by comparatively analysing the nepetalactones content and the expression of putative iridoid synthase (NrIS) in leaves of Nepeta rtanjensis Diklić & Milojević, a trans,cis-nepetalactone and dehydronepetalactone rich species. Another aim was to give an insight into the developmental regulation of nepetalactone biosynthesis in leaves of cultivated N. rtanjensis plants. Knowledge on the spatial-, and developmental- regulation of nepetalactone metabolism could greatly benefit potential growers by enabling them to adequately plan and manage plant cultivation, harvesting, and handling and thus increase yield of bioactive compounds. The expression profile of putative NrIS1 is highly correlated with trans,cis-nepetalactone and dehydronepetalactone content/ratio in leaves of different developmental stages. The results strongly indicate that developmental regulation of nepetalactone biosynthesis in leaves resides largely at the level of gene expression, and suggest an important role of NrIS1. Young leaves exhibited the most intensive nepetalactone biosynthesis and accumulation, but also its conversion to dehydronepetalactone. Thus, the pattern of NL accumulation in leaves can be explained by both the rate of its biosynthesis and metabolic degradation. The gene expression patterns of NrIS1 and NrIS2 transcripts in trichomes and abraded leaves revealed trichomes as the main site of their activity. UHPLC/qqqMS profiling of nepetalactones in trichomes and abraded leaves, complemented with Raman spectroscopy and dichloromethane dipping experiment, supported glandular trichome-specific localization of nepetalactone biosynthesis and accumulation.

AB  - Background. The diversity of wild trout stocks in Montenegro is compromised by insufficiently controlled stocking. Adriatic and Mediterranean areas show a high degree of endemism of the salmonid species, with numerous native Salmo taxa described. The invasive effect of brown trout, Salmo trutta Linnaeus, 1758, of the Atlantic haplogroup on the endemic Adriatic trout, Salmo farioides Karaman, 1938, in the drainage area of the Adriatic Sea basin is lower compared to the effect inflicted by S. trutta has on Salmo labrax Pallas, 1814 of the Danubian haplogroup. The presently reported study was intended to describe the genetic structure of the population of Adriatic trout, Salmo farioides, from the Mrtvica River, a right tributary of the Morača River, Montenegro, in order to get an insight into the spread of non-native strains and their introgression into the native Adriatic trout gene pool. Materials and methods. Trout specimens sampled from the lower section of the Mrtvica River from 2004 to 2007, and from its upper section in the spring of 2014, were analysed for their mtDNA haplotypes. Nuclear DNA markers (microsatellites, LDH-C1* gene) were also included in genetic analyses since mtDNA is inherited only maternally. After statistical analyses, relations between individuals from the upper and lower Mrtvica River were reconstructed. Results. Using both LDH-C1* and seven microsatellite loci with several alleles exclusive to S. trutta, an introgression was detected only in the upper reaches of the Mrtvica River, where only stream-dwelling trout form occurs. In the lower reaches no introgression was detected at all, as revealed by the absence of both the Atlantic mitochondrial haplotype and microsatellite alleles exclusive to S. trutta. Conclusion. The allelic frequency at seven microsatellite loci of Salmo spp. from the two river sections revealed very different population structure, as a consequence of a low rate of gene flow between them. In addition to the physical barrier that prevents Salmo sp. from the lower Mrtvica River to migrate to the upper river section, it seems that the opportunity to mate with the large pool of conspecifics, including lake-dwelling S. farioides (i.e., Salmo cf. dentex), prevents Salmo trutta from surviving at the lower stretch and spread to the rest of the Morača River and Lake Skadar systems.

Effect of childhood general traumas on suicide attempt depends on TPH2 and ADARB1 variants in psychiatric patients

AB  - Suicidal behavior has been associated with a deficient serotonin neurotransmission which is likely a consequence of individual genetic architecture, exposure to environmental factors and interactions of those factors. We examined whether the interaction of child abuse, TPH2 (tryptophan hydroxylase 2) variant rs4290270, affecting alternative splicing and editing of TPH2 pre-mRNAs, and ADARB1 (adenosine deaminase acting on RNA B1) variants rs4819035 and rs9983925 may influence the risk for suicide attempt in psychiatric patients. TPH2 rs4290270 was genotyped in 165 suicide attempters and 188 suicide non-attempters diagnosed with major depressive disorder, bipolar disorder and schizophrenia. Genotyping data for ADARB1 variants were taken over from our previous study. Child abuse before the age of 18 years was assessed using the Early Trauma Inventory-Self Report. Generalized linear models and backward selection were applied to identify the main and interacting effects of environmental and genetic factors, including psychiatric diagnoses, patients’ gender and age as covariates. Childhood general traumas were independently associated with suicide attempt. Two-way interaction between TPH2 rs4290270 and general traumas revealed that TT homozygotes with a history of general traumas had an increased risk for suicide attempt. Three-way interaction of general traumas, TPH2 rs4290270 and ADARB1 rs4819035 indicated that the highest predisposition to suicide attempt was observed in individuals who experienced general traumas and were TT homozygote for rs4290270 and TT homozygote for rs4819035. Our findings suggest that the risk for suicide attempt in psychiatric patients exposed to an adverse childhood environment may depend on TPH2 and ADARB1 variants.
