Expression of stress-related genes in diapause of European corn borer (Ostrinia nubilalis Hbn.)

AB  - Diapause is a state of arrested development during which insects cope with many external and internal stressful factors. European corn borer, Ostrinia nubilalis, overwinters as a fifth instar freeze-tolerant diapausing larva. In order to explore diapause-linked stress tolerance processes, the expression of selected genes coding for stress related proteins—glutathione S-transferase (Gst), thioredoxin (Trx), glutaredoxin (Grx), ferritin (Fer), metallothionein (Mtn), and heat shock proteins Hsp90, Hsc70, Hsp20.4, and Hsp20.1—was assessed in the fat body of diapause-destined, warm (22 °C) and cold (5 °C) acclimated diapausing larvae using the quantitative real-time PCR. Gene expression was normalised to mRNA transcripts for Actin and Rps03, and relative expression was calculated using non-diapausing larvae as a control group. During the initiation phase of diapause, the abundance of mRNA transcripts of Grx, Hsp90, Hsc70, andHsp20.1 was significantly upregulated, Trx, Fer, Mtn, andHsp20.1 were unchanged, while only Gst was clearly downregulated in comparison to non-diapause control. Later, in the early phase of diapause, the expression of most genes (except Trx and Hsp20.1) was upregulated in warm-acclimated
larvae, while only Trx and Hsp90 were upregulated in cold-acclimated larvae. Furthermore, the relative expression of all genes (except Trx) increased gradually throughout the diapause in cold-acclimated larvae. This result indicates that the half-life of mRNAs is prolonged during diapause at low temperature, which may lead to a gradual accumulation of mRNA transcripts. Our results show that both diapause programming and temperatures affect the expression of stress-related genes in Ostrinia nubilalis.

AB  - Dimethyl sulfoxide (DMSO) is a widely used solvent and cryoprotectant that can cause impaired blood flow, reduction in intracranial pressure, tissue edema, inflammatory reactions, inhibition of vascular smooth muscle cell migration and proliferation, processes which can lead to atherosclerosis of the coronary, peripheral and cerebral circulation. Although the adverse effects are rare when DMSO is administered in clinically established concentrations, there is no safe antagonist for an overdose. In this work, we treated isolated spontaneous and calcium-induced contractile active rat uteri (Wistar, virgo intacta), with DMSO and fullerenol C-60(OH)(24) nanoparticle (FNP) in DMSO. FNP is a water-soluble derivative of fullerene C-60. Its size is a 1.1 nm in diameter and is a very promising candidate for a drug carrier in nanomedicine. FNP also displays free radical scavenging activity. DMSO decreased both spontaneous and calcium-induced contractions. In contrast, FNP only decreased spontaneous contraction. FNP decreased copper-zinc superoxide dismutase activity and prevented the DMSO-induced increase in glutathione reductase activity. Atomic force microscopy detected that FNP aggregated with calcium ions. Our results indicate that FNP has properties that make it a good candidate to be a modulator of DMSO activity which could minimize side effects of the latter.

AB  - The uteri, spontaneously active or Ca2+ (6 mM) induced, were allowed to equilibrate, and to inhibit voltage-gated potassium (KV) channels 1 mM 4-amino pyridine (4-AP) was applied for 15 min before adding H2O2. H2O2 was added cumulatively: 2 mu M, 20 mu M, 200 mu M, 400 mu M, and 3 mM. Average time for H2O2 concentrations (2, 20, 200, and 400) mu M to reach its full effect was 15 min. H2O2 3 mM had a prolonged effect and therefore was left to act for 30 min. Two-way ANOVA showed significant differences in time dependency between spontaneous and Ca2+-induced rat uteri after applying 3 mM H2O2 (type of contraction, P = 0.0280), but not 400 mu M H2O2 (P = 0.9271). Our results indicate that H2O2 oxidises channel intracellular thiol groups and activates the channel, inducing relaxation. Cell antioxidative defence system quickly activates glutathione peroxidase (GSHPx) defence mechanism but not catalase (CAT) defence mechanism. Intracellular redox mechanisms repair the oxidised sites and again establish deactivation of KV channels, recuperating contractility. In conclusion, our results demonstrate that KV channels can be altered in a time-dependent manner by reversible redox-dependent intracellular alterations.

AB  - Changes in fitness components including larval stage duration, relative growth rate (RGR), and mass of the gypsy moth, Lymantria dispar L. (Lepidoptera: Lymantriidae), were investigated in caterpillars fed a synthetic diet with or without a cadmium supplement (10, 30,100,250 mu g Cd/g dry food weight). Morphometric changes of large protocerebral dorsomedial A2 neurosecretory neurons, their nuclei and the electrophoresis profiles of brain proteins were analyzed in the 4(th) instar gypsy moths fed the examined diets. The duration of the fourth larval instars were prolonged and RGR and body mass reduced if the caterpillars were fed diets containing high concentrations of cadmium (100 and 250 mu g). The size of large A2 dorsomedial neurosecretory neurons and their nuclei were significantly higher in larvae fed the diets supplemented with 10, 100 and 250 mu g Cd. A large amount of neurosecretory material appeared in dorsomedial neurosccretory neurons in larvae fed diets with 100 and 250 mu g Cd. Differences in larval brain protein profiles in the region of molecular mass ranges(Mr) of 98kDa, 46kDa and 3.4-6.1kDa were identified in the experimental groups.

AB  - Background and purpose: The effects of hydrogen peroxide (H(2)O(2)) on uterine smooth muscle are not well studied. We have investigated the effect and the mechanism of action of exogenous hydrogen peroxide on rat uteri contractile activity [spontaneous and calcium ion (Ca2+)-induced] and the effect of such treatment on anti-oxidative enzyme activities. Experimental approach: Uteri were isolated from virgin Wistar rats and suspended in an organ bath. Uteri were allowed to contract spontaneously or in the presence of Ca2+ (6 mM) and treated with H(2)O(2) (2 mu M-3 mM) over 2 h. Anti-oxidative enzyme activities (manganese superoxide dismutase-MnSOD, copper-zinc superoxide dismutase-CuZnSOD, catalase-CAT, glutathione peroxidase-GSHPx and glutathione reductase-GR) in H(2)O(2)-treated uteri were compared with those in uteri immediately frozen after isolation or undergoing spontaneous or Ca2+-induced contractions, without treatment with H(2)O(2). The effect of inhibitors (propranolol, methylene blue, L-NAME, tetraethylamonium, glibenclamide and 4-aminopyridine) on H(2)O(2)-mediated relaxation was explored. Key results: H(2)O(2) caused concentration-dependent relaxation of both spontaneous and Ca2+-induced uterine contractions. After H(2)O(2) treatment, GSHPx and MnSOD activities were increased, while CuZnSOD and GR (In Ca2+-induced rat uteri) were decreased. N omega-nitro-L-arginine methyl ester antagonized the effect of H(2)O(2) on Ca2+-induced contractions. Diethyldithiocarbamate (DDC) potentiated the effect of protamine sulphate. A mechanism for DDC's action was postulated on the basis of its interactions with divalent iron ions and Cui Zn-SOD. DDC chelates divalent iron ions creating DDC-iron (Fe-DDC) complexes. Fe-DDC forms stable NO-Fe-DDC(2) complexes by NO scavenging and de-nitrosylation processes, which in combination with DDC (5 mM) provoke inhibition of Cui Zn-SOD resulting in specific oxidative conditions culminating in potassium ion channel opening, membrane hyperpolarisation, inhibition of calcium ion influx and subsequent muscle relaxation. As Fe-DDC and NO-Fe-DDC(2) complexes exclude divalent iron ions from participating in the hydroxy radical generating Fenton reaction, DDC can also prevent iron-related pathophysiological manifestations. Such permissive roles of DDC open the possibility for application of its pharmacological form (disulfiram) to a wider spectrum of pathophysiological conditions related to smooth muscles.

AB  - Protamine sulphate (PS) effect on spontaneous and calcium-induced rhythmic contractions of isolated virgin rat uteri was studied. PS caused dose-dependent relaxation of both types of contractions (two-way ANOVA, significant dose effects). Pretreatment with NG-nitro-L-arginine methyl ester (L-NAME; 10(-5) mol/l), methylene blue (MB; 0.9 x 10(-6) mol/l) or propranolol (1.7 x 10(-5) mol/l) enhanced PS-mediated uterine muscle relaxation of spontaneous contractions. Dose-dependent relaxation of spontaneous active isolated rat uterus with PS was lower in uteri pretreated with single dose of tetraethylammonium (TEA; 6 x 10(-3) mol/l), glibenclamide (2 x 10(-6) mol/l) and 4-aminopyridine (4-AP; 10(-3) mol/l). Calcium-induced activity of the isolated rat uterus pretreated with the same concentration of L-NAME, MB, or propranolol modified the kinetic of PS-induced relaxation without changes in EC(50) values. Pre-treatment with glibenclamide, TEA and 4-AP significantly reduce PS relaxing effect of calcium-induced activity and according to EC(50) values the order of magnitude was glibenclamide > TEA > 4-AP. PS is mixture of polyamines and may activate different signal-transduction pathways. Our results cleary demonstrate that in uterine smooth muscle PS act dominantly through potassium chanels and marginaly through beta-adrenergic receptos or nitric oxide-dependent pathways.

AB  - After enzymic biotransformation, molsidomine (MO) acts via the metabolite 3-morpholmosydnonimine (SIN-1) through spontaneous liberation of nitric oxide (NO) and superoxide (O-2(.-)). The aim of this study was to compare the effects of MO and its active metabolite SIN-1 on the redox status of rat erythrocytes and reticulocytes. Rat erythrocyte as well as reticulocyte-rich red blood cell (RBC) suspensions were aerobically incubated (2h, 37 degrees C) without (control) or in the presence of different concentrations of MO or SIN-1. In rat erythrocytes, biotransformation of MO resulted in the production of NO and nitroxyl (NO-). Endogenous superoxide anion (O-2(.-)) participated in peroxynitrite generation. SIN-1 simultaneously liberated NO and O-2(.-), which formed peroxynitrite (at least in part), but the liberated NO predominantly reacted with haemoglobin, forming methaemoglobin in erythrocytes. In reticulocytes, MO and SIN-1 caused an increase in the levels of both nitrite and 3-nitrotyrosine (an indicator of peroxynitrite), whereas they decreased the level of O-2(.-). In reticulocytes, MO was metabolized into SIN-1 which led to the generation of NO, which reacted with O-2(.-) (endogenous or exogenous) forming reactive nitrogen species. In conclusion, there are two metabolic pathways for MO biotransformation: one causing NO and NO- generation predominantly in erythrocytes and the other, via SINA metabolism, in reticulocytes. AB  - Background and Purpose: In order to assess exposure to effects of environmental pollutants on aquatic oiganisms, several different biochemical and physiological parameters were studied. The activity of antioxidant defense enzymes was measured in the Mediterranean Sea shrimp (Parapenaeus longirostris) and compared with the presence of polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) in their sea water environment. Materials and Methods: The activity of the antioxidant defence enzymes superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione reductase (GR), as well as glutathione-S transferase (GST) as a phase II detoxification enzyme, was measured in the abdominal muscle of the shrimp in winter and spring at four different locations in the Montenegrine costal region of the Adriatic Sea. The results were correlated to the presence of PCBs and PAHs in the sea water at the time of sampling. Results and Conclusions: There was no obvious direct correlation between antioxidant defense activity and the presence and concentration of PCBs and PAHs in the sea water: The activities of SOD, GSH-Px and GR showed seasonal changes and therefore seasonality should be incorporated into the interpretation of antioxidant changes in future biomonitoring studies. Generally, SOD and GR activities were lower and GSH-Px was higher in winter when compared to spring. However these effects were not uniform and statistical trends were influenced by local environment. There were no seasonal changes in GST activity, but it's activity varied depending on the location, but without correlation to the presence of PCBs and PAHs.

abstract = "Background and Purpose: In order to assess exposure to effects of environmental pollutants on aquatic oiganisms, several different biochemical and physiological parameters were studied. The activity of antioxidant defense enzymes was measured in the Mediterranean Sea shrimp (Parapenaeus longirostris) and compared with the presence of polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) in their sea water environment. Materials and Methods: The activity of the antioxidant defence enzymes superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione reductase (GR), as well as glutathione-S transferase (GST) as a phase II detoxification enzyme, was measured in the abdominal muscle of the shrimp in winter and spring at four different locations in the Montenegrine costal region of the Adriatic Sea. The results were correlated to the presence of PCBs and PAHs in the sea water at the time of sampling. Results and Conclusions: There was no obvious direct correlation between antioxidant defense activity and the presence and concentration of PCBs and PAHs in the sea water: The activities of SOD, GSH-Px and GR showed seasonal changes and therefore seasonality should be incorporated into the interpretation of antioxidant changes in future biomonitoring studies. Generally, SOD and GR activities were lower and GSH-Px was higher in winter when compared to spring. However these effects were not uniform and statistical trends were influenced by local environment. There were no seasonal changes in GST activity, but it's activity varied depending on the location, but without correlation to the presence of PCBs and PAHs.",

AB  - We examined the concentrations of vitamin E ( vit E), sulphydryl groups ( - SH), total protein and protein electrophoretic profiles in the hepatopancreas, the gills and muscle of three freshwater crayfish species: Noble crayfish ( Astacus astacus), stone crayfish ( Austropotamobius torrentium) and spiny cheek crayfish ( Orconectes limosus). Vit E concentration in the hepatopancreas of O. limosus was lower compared to A. astacus, while in the gills of O. limosus it was lower compared to both A. astacus and A. torrentium. The concentration of - SH groups was lower in the hepatopancreas of A. astacus compared to A. torrentium and O. limosus. In the gills of A. astacus and A. torrentium the concentration of - SH groups was higher compared to O. limosus. Protein concentration was higher in the hepatopancreas of A. torrentium compared to A. astacus and O. limosus. A lower protein concentration in muscle of A. torrentium was found compared to O. limosus and A. astacus. Electrophoretic analysis of proteins indicated species and tissue specifities between investigated crayfish species. Our results represent the first study of its kind and provide the basis for future studies that will consider our reported parameters as potential biomarkers for biomonitoring of basic environmental conditions and some anthropogenic impacts.

AB  - The effects of nitroglycerine (NTG) are mediated by liberated nitric oxide (NO) after NTG enzymatic biotransformation in cells. The aim of this study was to evaluate some products of NTG bio-transformation and their consequences on the redox status of rat erythrocytes and reticulocytes, considering the absence and presence of functional mitochondria in these cells, respectively. Rat erythrocyte and reticulocyte-rich red blood cell (RBC) suspensions were aerobically incubated (2h, 37 degrees C) without (control) or in the presence of different concentrations of NTG (0.1, 0.25, 0.5, 1.0 and 1.5 mM). In rat erythrocytes, NTG did not elevate the concentrations of any reactive nitrogen species (RNS). However, NTG robustly increased concentration of methemoglobin (MetHb), suggesting that NTG bio-transformation was primarily connected with hemoglobin (Hb). NTG-induced MetHb formation was followed by the induction of lipid peroxidation. In rat reticulocytes, NTG caused an increase in the levels of nitrite, peroxinitrite, hydrogen peroxide, MetHb and lipid peroxide levels, but it decreased the level of the superoxide anion radical. Millimolar concentrations of NTG caused oxidative damage of both erythrocytes and reticulocytes. These data indicate that two pathways of NTG bio-transformation exist in reticulocytes: one generating RNS and the other connected with Hb (as in erythrocytes). In conclusion, NTG bio-transformation is different in erythrocytes and reticulocytes due to the presence of mitochondria in the latter.

AB  - The aim of our study was to investigate the copper and zinc containing superoxide dismutase (CuZn SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cells (RBCs), as well as phase II biotransformation enzyme glutathione-S-transferase (GST) activity in the plasma of male, two months old Wistar albino rats. The rats were divided into two experimental groups: control animals and animals treated with olive oil (100 μL of virgin olive oil, i.m., every fifth day during 30 days). The obtained results show that olive oil induced a significant decrease of CuZn SOD, CAT and GSH-Px activities in RBCs (p<0.05) and significant increase of GST activity in the plasma (p<0.05) in relation to the control animals. The results show that olive oil influences the activity of investigated enzymes in the blood of rats and in that way improves the overall antioxidative defense.
AB  - Cilj našeg rada bio je da se odrede aktivnosti bakar cink sadržavajuće superoksid-dismutaze (CuZn SOD), katalaze (CAT) i glutation-peroksidaze (GSH-Px) u eritrocitima, kao i enzima faze II biotransformacije glutation-S-transferaze (GST) u plazmi mužjaka dva meseca starih Wistar albino pacova tretiranih maslinovim uljem. Pacovi su podeljeni u dve grupe i to kontrolna i grupa tretirana maslinovim uljem (100 μL of maslinovog ulja, i.m., svaki peti dan tokom 30 dana). Rezultati pokazuju da maslinovo ulje izaziva značajno smanjenje aktivnosti CuZn SOD, CAT i GSH-Px (p<0.05) u eritrocitima, kao i povećanje aktivnosti GST u plazmi pacova (p<0.05) u odnosu na kontrolu. Dobijeni rezultati pokazuju da maslinovo ulje utiče na aktivnost ispitivanih enzima u smislu poboljšanja antioksidacione zaštite.

AB  - The effect of cadmium (Cd), coenzyme Q10 (CoQ10) and Cd+CoQ10 on the activities of superoxide dismutases (total SOD), manganese containing superoxide dismutase (Mn SOD) and copper-zinc containing superoxide dismutase (Cu,Zn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-Stransferase (GST), glutathione reductase (GR) and concentrations of ascorbic acid (AsA) and vitamin E (Vit E) in the heart of male Wistar albino rats were studied in comparison to the controls and cadmium treated animals. Cd induces a significant increase of total SOD, Cu, Zn SOD and GSH-Px activities, as well as AsA and Vit E concentrations, but leads to a significant decrease of CAT and GR activities. CoQ10 induces a significant increase of total SOD, Mn SOD, Cu, Zn SOD and GSH-Px activities, as well as AsA and Vit E concentrations. In the same group of animals the activities of CAT, GST and GR were significantly decreased. By concomitant treatment of rats with Cd+CoQ10 the activities of total SOD, Mn SOD and GSH-Px, as well as concentrations of AsA and Vit E were markedly increased. In the same group of animals the activities of Cu, Zn SOD, CAT and GR were significantly decreased. In respect to the Cd treated rats in Cd+CoQ10 partially are reversed changes (Cu,Zn SOD) of antioxidant defense system in the heart.
AB  - Ispitivan je uticaj kadmijuma (Cd), koenzima Q10 (CoQ10) i Cd+ CoQ10 na aktivnost ukupne superoksid dismutaze (Uk SOD), mangan sadržavajuće superoksid dismutaze (Mn SOD), bakar cink sadržavajuće superoksid dismutaze (Cu,Zn SOD), katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-Stransferaze (GST) i glutation reduktaze (GR), kao i na koncentracije askorbinske kiseline (AsA) i vitamina E (Vit E) u srcu mužjaka Wistar albino pacova u poređenju sa kontrolnom grupom i grupom tretiranom sa kadmijumom. Cd u srcu indukuje značajno povećanje aktivnosti Uk SOD, Cu,Zn SOD i GSH-Px, kao i koncentracija AsA Vit E uz značajno smanjenje aktivnosti CAT i GR. CoQ10 izaziva značajno povećanje aktivnosti Uk SOD, Mn SOD, Cu,Zn SOD i GSH-Px, kao i koncentracija AsA i Vit E. U istoj grupi životinja aktivnosti CAT, GST i GR su značajno smanjene. Istovremeni tretman sa Cd+CoQ10 dovodi do značajnog povećanja aktivnosti Uk SOD, Mn SOD i GSH-Px, kao i koncentracija AsA i Vit E. U istoj grupi životinja aktivnosti Cu,Zn SOD, CAT i GR su značajno smanjene. U poređenju sa pacovima tretiranim sa kadmijumom kod Cd+CoQ10 grupe dobijen je delimični zaštitni efekat (Cu,Zn SOD) na antioksidacioni zaštitni sistem u srcu pacova.

Concentrations of vitamin E and sulphydryl group in some tissues of freshwater crayfish species

AB  - The freshwater crayfishes can be useful organisms in the biomonitoring of environmental conditions. In our experiments we studied vitamin E (Vit E) and sulphydryl group (-SH)  concentrations  in  hepatopancreas,  gills  and  abdominal muscle of three crayfish species: Noble crayfish  (Astacus  astacus)  from  South  Morava river, Stone crayfish (Austropotamobius torrentium) from Krajkovacka river  and Spinycheek crayfish (Orconectes limosus) from Danube river. All species were collected in summer season (August). The  speciments  were  collected  by deep  nets and supematants for analysis were prepared according to the standard methods. Vit E concentration was assayed by standard method using bathophenantroline as reagent, while -SH groups concentration was  measured  by  standard  spectrophotometric method using dithionitrobenzoic acid (DTNB). The  obtained  results  show  significantly lower concentration of vit E in hepatopancreas of Orconectes limosus in respect to Astacus astacus (p<0.05). At the same time, significantly decreased concentration of vit E was detected in gills of Orconectes limosus comparing  to  Astacus astacus and Austropotamobius torrentium (p<0.005). Concentration of -SH groups was significantly increased  in  hepatopancreas  of  Noble  crayfish  in comparison to Stone crayfish (p<0.05) and Spinycheek crayfish (p<0.005). In conclusion,  the concentrations of the  investigated  antioxidant  compounds  (vit  E and
-SH groups) in the selected tissues of freshwater crayfishes are good markers for biomonitoring of environmental conditions. Differences in the investigated nonenzymatic oxidative stress parameters in tissues reflect different metabolic and antioxidative activities, which  can  be the consequence  of species  or tissue specifity,  as well as the consequence  of different environmental  conditions.

AB  - The aim of this study is to establish the effects of cadmium (Cd), coenzyme Q10 (CoQ10) and Cd + CoQ10 on the activities of: Superoxide dismutases (total SOD, manganese containing, Mn SOD, copper zinc containing, CuZn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST), as well as glutathione reductase (GR) in the kidneys of male Wistar albino rats in comparison with control animals. Significantly increased activities of Tot SOD and CuZn SOD in the kidneys of Cd-treated animals were observed. At the same time, the activity of Mn SOD was significantly decreased in the kidneys of animals treated with Cd, as well as with CoQ10. The activity of CAT was significantly lower in rats exposed to CoQ10 and Cd+CoQ10 in respect to the control animals. GSH-Px and GST activities were significantly decreased in all treated groups of animals (Cd, CoQ10 and Cd+CoQ10), while GR activity was not changed. Presented results reveal, that CoQ10 only partially reversed Cd-induced changes of antioxidant defense enzyme activities in kidney by reversing the effect of Cd on Tot SOD and CuZn SOD activities.
AB  - Cilj ovog rada bio je da se objasni efekat kadmijuma (Cd), koenzima Q10 (CoQ10) i Cd + CoQ10 na aktivnost: superoksid dismutaza (ukupne, Uk SOD, mangan sadržavajuće, Mn SOD, bakar cink sadržavajuće, CuZn SOD, katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-S-transferaze (GST) kao i glutation reduktaze (GR) u bubrezima mužjaka Wistar albino pacova u poređenju sa kontrolnom grupom životinja. Dobijeni rezultati pokazuju značajno povećanje aktivnosti Uk SOD i CuZn SOD u bubrezima pacova tretiranih sa Cd. U isto vreme aktivnost Mn SOD je bila značajno smanjena u bubrezima pacova tretiranih sa Cd, kao i tretiranih sa Cd+CoQ10. U poređenju sa kontrolnim životinjama, aktivnost CAT je bila značajno smanjena u bubrezima pacova tretiranih sa CoQ10, kao i tretiranih kombinacijom Cd+CoQ10. Aktivnosti GSH-Px i GST su bile značajno smanjene u bubrezima svih tretiranih grupa životinja (Cd, CoQ10 i Cd+CoQ10), dok se aktivnost GR nije statistički značajno menjala. Iz prikazanih rezultata može se zaključiti, da CoQ10 samo delimično utiče na promené aktivnosti antioksidacionih zaštitnih enzima nastale pod uticajem Cd, kao i da vraća aktivnosti Uk SOD i CuZn SOD na normalne vrednosti.

Aktivnosti GSH-Px i GST su bile značajno smanjene u bubrezima svih tretiranih grupa životinja (Cd, CoQ10 i Cd+CoQ10), dok se aktivnost GR nije statistički značajno menjala. Iz prikazanih rezultata može se zaključiti, da CoQ10 samo delimično utiče na promené aktivnosti antioksidacionih zaštitnih enzima nastale pod uticajem Cd, kao i da vraća aktivnosti Uk SOD i CuZn SOD na normalne vrednosti.",

AB  - The activity of cytosolic antioxidative defence enzymes in the liver and white muscle of thinlip gray mullet (Liza ramada Risso) were compared in winter and spring in the Adriatic Sea.. Activity of antioxidative enzymes is functionally organized due to metabolic demands: analyses of variance and correlation analysis revealed tissue- and seasonal- specific organization of antioxidative enzymes. In winter GST activity increased in both tissues compared with spring. At the same time decreased GSH-Px and GR activities were observed and this effect was more pronounced in liver then in white muscle. From correlation analyses it is concluded that the antioxidative components correlate, but the composition of the antioxidative defence system is different in respect to season and tissue. This means that the antioxidative defence system reorganizes its structure due to oxidative demands and to protect the tissues against reactive oxygen species and to establish homeostasis. Discriminant analyses separated groups according to the complete organization of individual components of the system very well and identified individual components (CAT, GST and GR) which contribute most to the differences. Statistical differences were observed between enzyme activities in tissues (liver and muscle) in both winter and spring, and between seasons (winter and spring) for liver tissue only. Since environmental parameters, such as temperature and oxygen concentration in the sea differ with season, we conclude that in this species the tissues examined expressed their antioxidative defence systems in different ways in respect of external/environmental conditions. We propose that tissue- and seasonal- specific levels of antioxidant enzyme activities should be considered in the interpretation of data from future biomonitoring field studies, especially in relation to low temperature.

AB  - The C3 clone of B-16 mouse melanoma was cultured for 1, 6 and 9 days and analyzed. The changes which are not directly linked to melanogenesis in the B-16 / C3 cultures during their maturation were characterized. Early (1 day) confluent (6 days) and old (9 days) cell cultures are distinguished by their leucine aminopeptidase (LAP) and α-naphthyl acetate esterase (ANAE) isoenzyme patterns. Both quantitative and qualitative changes in LAP and ANAE isoenzyme can be observed during culture maturation. There is an increase in the activity of the enzyme copper, zinc-containing superoxide-dismutase (CuZn SOD). The increaase in the CuZn SOD enzyme activity might be related to B-16/C3 cell melanogenesis and / or to differentiation.
AB  - C3 klonovi B-16 melanoma miša su gajeni u kulturi 1, 6 i 9 dana i potom analizirani. Izvršena je karakterizacija promena B-16/C3 ćelijskih kultura tokom njihove maturacije koje nisu direktno povezane sa melanogenezom. Pokazano je da se kulture u početnoj fazi (1 dan), konfluentnoj fazi (6 dana) i kasnoj fazi (9 dana) ćelijskog rasta razlikuju na osnovu izoenzimskih spektara leucin-aminopeptidaze i alfa-naftil-acetat-esteraze. Tokom razvitka ćelijskih kultura B-16/C3 zapažene su kvantitativne i kvalitativne promene leucin-aminopeptidaze i alfa-naftil-acetat-esteraze u njihovim izoenzimskim spektrima. Utvrđeno je da postoji porast aktivnosti bakar-cink superoksid-dismutaze tokom starenja kulture. Porast aktivnosti bakar-cink superoksid-dismutaze može biti povezan sa melanogenezom i/ili diferencijacijom B-16/C3 ćelija.

AB  - B-I6/C3 mouse melanoma cells undergo melanogenesis and differentiation 9 days after plating under usual conditions. In our experiments the effect of exogenous bovine CuZn superoxide dismutase (CuZn SOD) on B-I6/C3 cells in cultures was studied. The exogenous CuZn SOD was added, 24 hours after cell plating, in growth medium, which either contain or not Fetal Calf Serum (FCS). B-I6/C3 melanoma cells growth was followed over 5 days. Different effect of CuZn SOD on the culture was observed in relation to FCS present in growth medium. CuZn SOD induced a dose dependent increase in melanin content of B-I6/C3 cells in serum deprived medium. Our results are discussed in respect to dismutating CuZn SOD activity, which may act to an enhanced level of oxidative stress, related to the higher metabolism in melanoma cells. On the other hand we supposed that CuZn SOD produced an elevated level of hydrogen peroxide. Therefore hydrogen peroxide than may play some role in cell differentiation as transmembrane messenger.
AB  - Ćelije B-I6/C3 melanoma miša melaniziraju i diferenciraju devetog dana posle zasađivanja pod uobičajenim uslovima. U našim eksperimentima smo proučavali uticaj egzogeno dodavane CuZn superoksid-dismutaze ( CuZn SOD) na ćelije B-I6/C3 u kulturi. Egzogena CuZn SOD je dodavana 24 časa posle zasađivanja ćelija u medijume za rast koji su sadržavali ili ne goveđi fetalni serum. Rast ćelija B-I6/C3 je praćen pet dana. Zapažen je različit uticaj CuZn SOD na ćelijske kulture gajene sa serumom ili bez njega. CuZn SOD izaziva dozno zavisnu sintezu i nakupljanje melanina u ćelijama koje su rasle u medijumu koji nije sadržavao goveđi fetalni serum. Naši rezultatu se mogu diskutovati u svetlu dismutacione aktivnosti CuZn SOD koja deluje na povećani nivo oksidacionog stresa izazvanog visokim metabolizmom melanoma ćelija. S druge strane, pretpostavili smo da CuZn SOD svojom aktivnošću proizvodi više vodonik peroksida. Vodonik peroksid, nadalje, može da ima izvesnu ulogu u diferencijaciji ćelija kao transmembranski signalni molekul.

Antioxidant defense enzyme activities in the interscapular brown adipose tissue of rats treated with cadmium and coenzyme Q10

AB  - In our study we investigated the effects of exogenous cadmium (Cd,  17 mg /day /kg  b.m.  in drinking  water), coenzyme  Q10 (CoQ 10, 16 mg/kg/dose CoQ10 dissolved in  olive  oil,  i.m.,  every  fifth  day) and Cd+CoQ10 (i n above mentioned amou nt s)  on  antioxidant  de­ fense enzyme activities (Total SOD, Mn SOD, CuZn SOD, CAT, GSH-Px, GST and  GR)  in  interscapular  hrown  adipose  tis sue (IBAT) of  male two  months  old  Wistar albin o rats during  30 days.
Cd induces significantly increased Mn SOD activity, while concomitant treatment of animals with  Cd+CoQ10  reversed  this change.   The  activity   of  CuZn   SOD   was  significantly  decreased
both in Cd and Cd+CoQ10 treated animals. CAT activity was sig­ nificantly increased in rats treated with Cd, whereas Cd+CoQ 10 normalized the activity of this enzyme. The activity of GSH-Px was significantly increased in all investigated groups of animals. Cd in­ duces an increased activity of GST,  but  by concomitant  treatment of rats with Cd+CoQ10 the GST activity was retained . The activity of GR was significantly increased in Cd treated animals, while in rats treated with Cd+CoQ10  was signi fican tl y decre_ased.
It can be concluded that Cd  induces  oxidativ  e stress  and  a l ter the activities of some antioxidant  defense  enzymes  in  IBAT of  rats. It is also shown that CoQ 10 can  normal ized  Mn  SOD,  CAT  a nd GST  activities  after  Cd-induced  changes .

abstract = "In our study we investigated the effects of exogenous cadmium (Cd,  17 mg /day /kg  b.m.  in drinking  water), coenzyme  Q10 (CoQ 10, 16 mg/kg/dose CoQ10 dissolved in  olive  oil,  i.m.,  every  fifth  day) and Cd+CoQ10 (i n above mentioned amou nt s)  on  antioxidant  de­ fense enzyme activities (Total SOD, Mn SOD, CuZn SOD, CAT, GSH-Px, GST and  GR)  in  interscapular  hrown  adipose  tis sue (IBAT) of  male two  months  old  Wistar albin o rats during  30 days.
Cd induces significantly increased Mn SOD activity, while concomitant treatment of animals with  Cd+CoQ10  reversed  this change.   The  activity   of  CuZn   SOD   was  significantly  decreased
both in Cd and Cd+CoQ10 treated animals. CAT activity was sig­ nificantly increased in rats treated with Cd, whereas Cd+CoQ 10 normalized the activity of this enzyme. The activity of GSH-Px was significantly increased in all investigated groups of animals. Cd in­ duces an increased activity of GST,  but  by concomitant  treatment of rats with Cd+CoQ10 the GST activity was retained . The activity of GR was significantly increased in Cd treated animals, while in rats treated with Cd+CoQ10  was signi fican tl y decre_ased.
It can be concluded that Cd  induces  oxidativ  e stress  and  a l ter the activities of some antioxidant  defense  enzymes  in  IBAT of  rats. It is also shown that CoQ 10 can  normal ized  Mn  SOD,  CAT  a nd GST  activities  after  Cd-induced  changes .",

AB  - The effects of acute exposure to cadmium (Cd) on the blood antioxidant defense system, lipid peroxide concentration and hematological parameters, as well as the possible protective role of vitamin E were studied. Male Wistar albino rats (3 months old) were treated with cadmium (0.4 mg Cd/kg b.m., i.p., 24 h before the experiment) or with vitamin E+Cd (20 IU Vit E/kg b.m., i.m., 48 h + 0.4 mg Cd/kg b.m., i.p., 24 h before the experiment). The hematological parameters were assessed: red blood cell counts, hematocrit value and hemoglobin concentration were significantly decreased in the blood of Cd-treated rats. Intoxication with cadmium was also followed by significantly increased lipid peroxide concentrations. We also observed increased activity of antioxidant defense enzymes: copper zinc containing superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase and glutathione-S-transferase as well as concentrations of non-enzymatic components of antioxidant defense system: reduced glutathione, vitamin C and vitamin E. Pretreatment with vitamin E exhibited a protective role on the toxic effects of cadmium on the hematological values, lipid peroxide concentration as well as on enzymatic and non-enzymatic components of antioxidant defense system.

AB  - In our experiments male two months old Wistar albino rats were used. Third group (3) was control  (C).  In  the experiment  with chro Cd intoxication the animals were treated as follov,s: (1) Cd (14 mg/day/kg b.m. CdCb in drink water during 15 days (Cd), (2) Vit E+CoQ 1o+Cd (pretreated with 20 IU/kg/dose of Vit E and mg/kg/dose  of  CoQ10  i.m., every  fifth day during  15 days  and  then  treated  with  14   mg/day b.m. of Cd during next 15 days Third group (3) was control (C). After the treatment the rats w sacrificed by decapitation and blood samples were collected and analyzed. In the blood of rats conce ntration of lipide peroxides (LP) were evaluated, while the activity  of  gluta tione transferase was assayed in the plasma after separation from the    blood cells.
The results obtained in our experiments show that acute treatment with Cd leads to significantly increased LP concentration (p<0.01) in the blood of rats in respect to the con animals. Pretreatment  with  Vit  E+CoQ 10  pryor  to  Cd exposure  normalyzed  the concentration LP   in   the· blood.   Contrary   to   that,   in   chronic   Cd   intoxication   concentration   of LP significantly   decreased   (p<0.005)   in   comparison   to   the   control   value.   Pretreatment   Vit E+CoQ 10 pryor to Cd intoxication influences an additional decrease of LP concentra (p<0.005) compared with controls. It appears that in chronic Cd treatment the period of 15 day sufficient for the development of efficient red blood cell protection against Cd (per)oxida  damage. The activity of GST in the plasma of rats acutely treated with Cd was  signi fica  increased in respect to the controls (p<0.02). Pretreatment of rats with Vit E+CoQ 10 influenc significant decrease of GST activity in the plasma (p<0.05).  Similar  results  were  obtained chronic Cd intoxication where Cd induces a significantly higher activity of GST in the pla (p<0.01), while pretreatment  with Vit E+CoQ 10 reversed this value to the control   level.
From  the   presented   results,  it  can  be  concluded   that  Cd   both   in  acute  and  chr
intoxication influences blood LP concentration and plasma GST activi ty . Pretreatment antioxidants, such as Vit E and CoQ 10 diminishes the toxi c effects of Cd on both evalu parameters. At the same time, we were also noticed a different mechanisms of defence against toxicity in acute and in chronic intoxication, especially in the case of blood LP con cen tration.

AB  - In our experiments conunon carps (Cyprinus carpio   L.) were    acclimated    to    an    oxygen    concentration  of 7.3 lmg/L water (100%) - control (C) group. Two ex­ perimental groups (H1, H2) were exposed to acute hy­ poxia. One of them was exposed to a progressive reduc­tion of 02 to 4.1 O2 mg/L (55%) for 1 ¼ hour s and another 1.95mg/L (25%) for 2 hours, respectively. The fourth experimental group (HP) was exposed to a progressive increase of oxygen concentration O2 to 10 mg/L (150%) for I5 minutes, respectively. In our exper iment was determinated antioxidant defense· s yste m enzymes activity (superoxide-dismutase   (SOD),   catalase   (CAT) glutathione-peroxidase (GSH-Px) , and glutathione-reductase (GR) and glutathione-S-transferase (GST) ac­ tivity (Misra, H.P. and Fridovich, I. (19 72), Beutler (1982), Tamure et al. ( I982), Habig et  al.  (1974), Glatzle et  al. (1974).
Our  res ult s  show  that  stress  caused  by  acute  hypoxia and hyperoxia significantly changes the activities of antioxidant defense system enzymes, superoxide-dis­mutase   (SOD),  glutathione-peroxidase   (GSH-Px),   and glutathione-reductase (GR) in carps gills. Acute pro­ longed hypoxia (H2) caused a cetrain  degree  of  adapta­tion in respect  to  catalase  (CAT)  and  glutathione-S­ transferase  (GST) activity .

AB  - Cadmium (Cd) is an important pollutant of environment which accwnulates in the liver and the kidneys, as well as in other tissues. Cd induces oxidative damage causing many metabolic, histological and pathological changes, such as nephrotoxicity, cardiotoxicity, increased lipid peroxidation (LP) and alteration of the antioxidant defense system (AOS). Recent studies have shown that coenzyme Q10 (CoQ10) and vitamin E (Vit E) are the powerful liposoluble antioxidants, which inhibit lipid peroxidation and thus prevent free radical med.iated oxidative injuries. It is well known that CoQ10 and Vit E may act synergistically as antioxidants. The aim of this study was to investigate a possible protective influence of CoQ10 and Vit E in Cd induced oxidative stress in the rat hearts.

AB  - Diethyldithiocarbamate (DDC) exhibits a variety of effects including neurotoxic, radio-protective and sensitizing activity. It is a potent copper chelating agent used for the treatment of oxygen toxicity, as an imunomodulator in cancer therapy, as well as in HIV infected patients. In this study we examined the effect of DDC, a superoxide dismutase (SOD) inhibitor, on the activities of copper-zinc containing superoxide dismutase (CuZn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and glutathione-S-transferase (GST). Three hours after diethyldithiocar-bamate treatment (1 g/kg b.m, i.p) a significant decrease of SOD and increase of GR and GST activities were found in the blood of rats. A negative correlation between SOD and GR and a positive correlation between GR and GST activities were also obtained. DDC induced a concentration dependent increase of GR activity and NADPH consumption in an enzymatic assay in vitro. The obtained results may be interpreted to indicate that a decrease in SOD activity in the blood of rats, after DDC administration, may be compensated for by changes in the activity of some other compounds suggesting that regulation of antioxidative defence is very complex.
AB  - Dietilditiokarbamat (DDC) ispoljava različite uticaje kao što su neurotoksičnost, radioprotekcija i senzibilizacja. On je snažan helirajući agens za bakar i koristi se u tretiranju kiseonične toksičnosti imunomodulaciji kao i kod pacijenata zaraženih HIV virusom. U ovom radu smo proučavali uticaj DDC, inhibitora superoksid dismutaze (SOD), na aktivnost bakar cink sadržavajuće superoksid dismutaze (CuZn SOD), katalaze (CAT) glutation peroksidaze (GSH-Px), glutation reduktaze (GR) i glutation-S-transferaze (GST).Tretiranje pacova dietilditiokarbamatom u trajanju od tri časa (1g/kgt.m, i.p) dovodi do značajnogsmanjenja aktivnosti CuZn SOD i povećanja aktivnosti GR i GST u krvi pacova. Takodje je dobijena negativna korelacija izmedju SOD i GR aktivnosti. DDC indukuje povećanje GR aktivnosti koje je koncentraciono zavisno kao i NADPH potrošnju u in vitro enzimatskom eseju. Dobijeni rezultati ukazuju na činjenicu da smanjivanje aktivnosti SOD u krvi pacova posle tretiranja DDCom može biti kompenzovano promenom aktivnosti drugih komponenti što dalje sugeriše da je regulacija antioksidacione zaštite veoma kompleksna.
