"Scent of a fruit fly": Cuticular chemoprofiles after mating in differently fed Drosophila melanogaster (Diptera: Drosophilidae) strains.

AB  - In the world of complex smells in natural environment, feeding and mating represent two important olfactory-guided behaviors in Drosophila melanogaster (Diptera: Drosophilidae). Diet affects the chemoprofile composition of the individuals, which, indirectly, may significantly affect their mating success. In this study, chemoprofiles of recently mated flies belonging to four D. melanogaster strains, which were fed for many generations on different substrates (standard cornmeal-S strain; banana-B strain; carrot-C strain; tomato-T strain) were identified and quantified. In total, 67 chemical compounds were identified: 48 compounds were extracted from males maintained on banana and carrot, and 47 compounds from males maintained on cornmeal and tomato substrates, while total of 60 compounds were identified in females from all strains. The strains and the sexes significantly differed in qualitative nature of their chemoprofiles after mating. Significant differences in the relative amount of three major male pheromones (cis-vaccenyl acetate-cVA, (Z)-7-pentacosene, and (Z)-7-tricosene) and in female pheromone (Z,Z)-7,11-nonacosadiene among strains were also recorded. Furthermore, multivariate analysis of variance (MANOVA) pointed to significant differences between virgin and mated individuals of all strains and within both sexes. Differences in some of the well known sex pheromones were also identified when comparing their relative amount before and after mating. The presence of typical male pheromones in females, and vice versa may indicate their bidirectional transfer during copulation. Our results confirm significant effect of mating status on cuticular hydrocarbon (CHC) phenotypes in differently fed D. melanogaster flies.

AB  - Phytochemistry deals with the study of secondary metabolites produced by plants that synthesize these compounds for many reasons, including their own protection against attack of herbivores and plant diseases. Secondary metabolites are believed to represent plant adaptation to various environmental factors and that they enabled the survival of the species. Secondary metabolites of plants can have curative or toxic effects in humans and animals. Herbal medicine has a long tradition in folk medicine and until the early 20th century, when synthetic organic chemistry began to develop, plants were the main source of medicines. The two basic goals of our phytochemical research are: isolation and identification of new (biologically active) compounds ? potential drugs, and chemotaxonomy (chemosystematics). In the following text through one selected example, the genus Amphoricarpos Vis., our phytochemical research is shown on both aspects.

Application of LC-MS/MS with ion mobility for chemical analysis of propolis extracts with antimicrobial potential

AB  - The objective of this study was to test four-dimensional LC-ESI-MS/ /MS chromatography in analysis of complex mixture such as ethanol extracts of different propolis samples. In total more than 1200 picks were identified and only for 185 literature conformation was found. The given data represent the result of tentative identification, and summarized results are given in the text. Comparing the samples, from different altitudes, 96 components were detected as characteristic in high altitude samples and 18 in samples collected at low altitudes. Antimicrobial activity of ethanol extracts of propolis (EEP) and propylene glycol extracts of propolis (PGEP) were carried out on S. aureus, B. cereus, M. flavus, L. monocytogenes, P. aeruginosa, S. typhimurium, E. coli and E. cloacae bacterial strains and compared with broad-spectrum antibiotics, streptomycin and ampicillin. Anti-quorum sensing activity was performed on P. aeruginosa by testing the effect of representative propolis extracts on biofilm formation, twitching and motility activity and production of pyocyanin. We demonstrated that the majority of explored propolis extracts have greater or equal minimal inhibitory concentration and minimum bactericidal concentration values compared to antibiotics, independently of the solvent used for the extraction. The samples collected from the highest altitude emerged as least active antimicrobial agents but with the greatest potential as anti-quorum sensing agents.

Comparative studies on the antimicrobial and cytotoxic activities of Tanacetum vulgare L. essential oil and methanol extracts

AB  - Chemical composition of essential oil (EO) and methanol extracts (MEs) from different parts of Tanacetum vulgare L. plant was analyzed and investigated for potential biological activities and correlated with the main constituents detected in EO and MEs. The EO was characterized by a high content of oxygenated monoterpenes with trans-chrysanthenyl acetate as major compound. All MEs were characterized by neochlorogenic, 3,5-O-dicaffeoylquinic and caffeoylquinic acids. High phenolic content in MEs correlated to high antioxidant capacity, especially for roots. Tansy EO showed strong activity against the most of tested fungi and was efficient as bifonazole and ketoconazole, while the most sensitive bacteria were Gram-negative E. coli and E. cloacae. All MEs showed fungistatic and fungicidal effects against all of the eight tested fungi, but the antimicrobial activity was higher against Gram-positive bacteria. Additionally, shown for the first time, MEs of leaves and flowers exhibited a strong antiproliferative effect on human cervical adenocarcinoma (HeLa) cells, causing cell shrinkage and detachment. In the present study, the Tansy extracts and essential oil with low thujone content can provide a very promising and effective alternative in the field of antimicrobial applications and food preservation. The Tansy MEs possess a high antioxidant potential with phenolic acids being a major radical scavenging contributor with the proved antiproliferative activity to HeLa cells.
