AB  - Serum levels of somatostatin were determined by RIA method in 50 patients with liver cirrhosis, 15 with pernicious anemia and atrophic gastritis, 31 with inflamatory bowel disease, 32 with colorectal tumors and in 40 control persons. In patients with liver cirrhosis somatostatin levels were significantly higher than in control group (p<0.01). Patients with pernicious anemia and ulcerative colitis in acute phase of the disease had significantly lower levels of somatostatin (p<0.01) (p<0.005). In patients with colorectal tumors somatostatin were significantly lower then in control group (p<0.01). Our results show that the liver is involved in somatostatin metabolism. Atrophic gastritis, ulcerative colitis and M. Crohn, so as colorectal neoplasia were associated with significantly changes in somatostatin levels which suggest the potential pathophysiologic and terapeutic role of somatostatin in those disease.
AB  - Somatostatin u serumu određen je RIA metodom, kod 50 pacijenata sa cirozom jetre, 15 sa pernicioznom anemijom i atrofičnim gastritisom, 31 sa inflamatornim oboljenjem creva, 32 sa kolorektalnim tumorima i 40 kontrolnih osoba. Kod pacijenata sa cirozom jetre nađene su značajno više vrednosti somatostatina u odnosu na kontrolnu grupu (p<0,01). Pacijenti sa pernicioznom anemijom i ulceroznim kolitisom u akutnoj fazi bolesti imali su značajno niže vrednosti somatostatina (p<0,005). Kod pacijenta sa kolorektalnim tumorima somatostatin je bio značajno niži nego u kontrolnoj grupi (p<0,01). Dobijeni rezultati pokazuju da je jetra uključena u metabolizam somatostatina. Atrofični gastritis, ulcerozni kolitis i M. Crohn, kao i kolorektalni tumori bili su udruženi sa značajnim promenama u nivou somatostatina što sugeriše moguću patofiziološku i terapeutsku ulogu somatostatina u ovim oboljenjima.

AB  - The results on the effects of synthetic somatostatins on the secretion of ACTH have been rather conflicting, but some authors reported that somatostain is involved in the regulation of ACTH secretion in rats of both sexes. The inhibitory mechanism of ACTH secretion could operate through the inhibition of CRF release from the hypothalamus. In the present study, we have investigated the effects of Octreotide, a somatostain analogue given intracerebroventricularly (i.c.v) in low doses, on morphometric characteristics of pituitary adrenocorticotropes (ACTH cells) in adult Wistar rat males. The animals were i.c.v. given three 1.0 µg doses of Octreotide dissolved in 10 µL saline every second day. The controls were treated in the same way with the same volume of saline. ACTH-producing cells were studied using the peroxidase-antiperoxidase (PAP) immunohistochemical procedure. Body weight, as well as absolute and relative pituitary weights were not significantly changed in comparison with the corresponding controls (p>0.05). The characteristics of immunohistochemicaly labelled ACTH cells in intact rat males were as follows: localization between the capillaries stellate in shape with the cytoplasmatic processes among neighbouring cells. The nucleus follows the shape of the cell body. Small specific secretory granules were distributed mainly at the periphery of the cytoplasm. The shape, and localization of ACTH immunoreactive cells in Octreotide-treated animals were not significantly changed in comparison with the controls. Octreotide treatment significantly decreased all morphometric parameters measured, i.e. volume of ACTH cells and their nuclei by 44% and 18% respectively, as compared to the controls (p<0.05). Volume densities were also significantly decreased (by 31%; p<0.05) comparing to the corresponding controls. These results indicate that Octreotide applied i.c.v. exerts a significant inhibitory effect on the immunohistochemical and morphometric characteristics of ACTH cells in rat males.
AB  - Postoje veoma različita, a često i suprotstavljena mišljenja o efektima sintetskih somatostatina na lučenje adrenokortikotropnog hormona (ACTH) iz hipofize. Mogući mehanizam inhibicije lučenja ACTH je preko inhibicije lučenja CRF iz hipotalamusa. U ovom radu su ispitivani efekti intracerebroventrikularno (i.c.v) ubrizganog analoga somatostatina Oktreotida, na rast ACTH ćelija adenohipofize mužjaka Wistar pacova. Sve eksperimentalne životinje su primile tri doze od po 1.0 µg Oktreotida rastvorenog u 10 µL fiziološkog rastvora, svakog drugog dana. Kontrole su tretirane na isti način fiziološkim rastvorom. Pacovi su žrtvovani pet dana posle poslednje injekcije. ACTH ćelije su imunocitohemijski bojene metodom peroksidaza-antiperoksidaza (PAP). Telesna masa, apsolutna i relativna masa hipofize nisu bile statistički značajno promenjene u poređenju sa odgovarajućom kontrolom (p>0.05). Karakteristike imunohistohemijski obojenih ACTH ćelija kontrolnih pacova su: lokalizacija između kapilara, zvezdast oblik sa citoplazmatičnim produžecima između drugih ćelija. AB  - The effects of chronic exposure to light of adult female Wistar rats on growth and function of pituitary adrenocorticotropes (ACTH cells) were examined. The animals were exposed to continuous light of 600 lux for 95 days, starting on day 30 of age. Control rats were kept under a 12:12 h light-dark cycle, at ambient temperature. ACTH-producing cells were studied using the peroxidase-antiperoxidase immunohistochemical procedure and blood samples were collected for hormone analyses. In animals exposed to a chronic light-treatment all morphometric parameters measured throughout the present study i.e.: ACTH cell volume, nuclear volume and relative volume density were increased by 22% and the differences between this group and the controls were statistically significant (p<0.05). The concentration of plasma ACTH was elevated by 13% in light-exposed group in comparison with the control and this difference was statistically significant (p<0.05), as well. These findings suggest that continuous exposure to light is specifically involved in growth and secretory activity of ACTH cells of adenohypophysis of rat females.
AB  - Ispitivani su efekti dugotrajnog izlaganja ženki Wistar pacova stalnom svetlu, na adrenokortikotropne (ACTH) ćelije. Eksperimentalne životinje stare 30 dana izlagane su konstantnom svetlu (600 luksa) tokom 95 dana. Kontrolne životinje odgovarajuće starosti su držane na normalnom, 12:12 h dnevno-noćnom režimu i sobnoj temperaturi. ACTH ćelije su imunohistohemijski bojene metodom peroksidaza-antiperoksidaza (PAP), a istovremeno je sakupljana krv za određivanje koncentracije ACTH u plazmi. Životinje izložene stalnom svetlu imale su za 22% povećane sve morfometrijske parametre ACTH ćelija merene tokom ovog rada (zapremina ćelija, njihovih jedara i volumenska gustina) u odnosu na kontrole i uočene razlike su bile statistički značajne (p < 0.05). Koncentracija ACTH u plazmi je takođe bila veća za 13% (p < 0.05) u odnosu na kontrolne vrednosti. Ovi rezultati ukazuju da dugotrajno izlaganje stalnom svetlu na specifičan način stimuliše rast i sekretornu aktivnost ne samo ACTH ćelija adenohipofize već i ćelija pars intermedia ženki pacova.

These findings suggest that continuous exposure to light is specifically involved in growth and secretory activity of ACTH cells of adenohypophysis of rat females., Ispitivani su efekti dugotrajnog izlaganja ženki Wistar pacova stalnom svetlu, na adrenokortikotropne (ACTH) ćelije. Eksperimentalne životinje stare 30 dana izlagane su konstantnom svetlu (600 luksa) tokom 95 dana. Kontrolne životinje odgovarajuće starosti su držane na normalnom, 12:12 h dnevno-noćnom režimu i sobnoj temperaturi. ACTH ćelije su imunohistohemijski bojene metodom peroksidaza-antiperoksidaza (PAP), a istovremeno je sakupljana krv za određivanje koncentracije ACTH u plazmi. Životinje izložene stalnom svetlu imale su za 22% povećane sve morfometrijske parametre ACTH ćelija merene tokom ovog rada (zapremina ćelija, njihovih jedara i volumenska gustina) u odnosu na kontrole i uočene razlike su bile statistički značajne (p < 0.05). Koncentracija ACTH u plazmi je takođe bila veća za 13% (p < 0.05) u odnosu na kontrolne vrednosti. Ovi rezultati ukazuju da dugotrajno izlaganje stalnom svetlu na specifičan način stimuliše rast i sekretornu aktivnost ne samo ACTH ćelija adenohipofize već i ćelija pars intermedia ženki pacova.",

AB  - The effects of intracerebroventricular (i.c.v.) application of a somatostatin analogue, Octreotide, on growth of pituitary adrenocorticotropes (ACTH cells) were examined in adult female Wistar rats. The animals were subjected to i.c.v. administration of three 1.0μg doses of Octreotide dissolved in 10μ1 saline every second day. Controls were treated in the same way with the same volume of saline. ACTH-producing cells were studied using the peroxidase -antiperoxidase (PAP) immunohistochemical procedure. Octreotide treatment of rat females, significantly decreased (p<0.05) all morphometric parameters measured, i.e. volume of ACTH cells and their nuclei by 52% and 11 %, respectively, as compared to the controls. The volume densities were also significantly decreased (by 47%; p<0.05) in comparison with the corresponding controls. These findings suggest that centrally administered somatostatin analogue (Octreotide) is specifically involved in the control of growth and secretory activity of ACTH cells in female rats.
AB  - Ispitivani su efekti intracerebroventrikularno (i.c.v.) ubrizganog analoga somatostatina, Oktreotida, na rast adrenokortikalnih (ACTH) ćelija adenohipofize ženki Wistar pacova. Sve eksperimentalne životinje su primile tri 1μg doze Oktreotida rastvorenog u 10μL fiziološkog rastvora, svaki drugi dan. Kontrole su tretirane na isti način fiziološkim rastvorom. Pacovi su žrtvovani petog dana po poslednjoj primljenoj dozi Oktreotida. ACTH ćelije su imunocitohemijski bojene PAP metodom. The animals were implanted with an intracerebroventricular cannula and after a recovery period, administered three 1 mug doses of SRIH-14 or -28 dissolved in 5 mul saline every second day. Controls were treated in the same way with the same volume of saline only. TSH-producing cells were studied using the peroxidase-antiperoxidase immunohistochemical procedure. Blood samples were collected for hormone (TSH) analyses 5 days after the last injection. Both SRIH-treatments significantly decreased (p < 0.05) all morphometric parameters obtained for TSH-cells in comparison with controls. The volume of TSH-cells decreased by 27%, nuclei by 44% and volume density by 33% in animals treated with SRIH-14. In animals treated with SRIH-28, these parameters were also significantly decreased (p < 0.05) (22%, 31%, and 25% respectively) compared to control rats. Serum concentrations of TSH were significantly decreased (p < 0.05) by 15% in SRIH-14- and by 12% in SRIH-28-treated animals in comparison with the controls. These observations suggest that centrally administered SRIH-14 or -28 is specifically involved in the control of growth and secretory activity of TSH cells.

AB  - This study examined the effects of intracerebroventricularly (i.c.v.) administered somatostatin (SRIH-14 orSRIH-28) on growth and function of pituitary somatotropes (GH cells). Female rats were subjected to intracerebroventricular administration of three 1g doses of SRIH-14 or SRIH-28 every second day. Blood samples were collected for hormone analyses and pituitaries were removed for histological and morphometric evaluation, five days after the last injection. Compared to control animals, SRIH treatment decreased (p<0.05) all morphometric measurements obtained for GH cells. The concentration of serum growth hormone (GH) in SRIH-14- or SRIH-28-treated groups was lower (p<0.05) by 69% and by 61% respectively, in comparison to control rats. These findings suggest that centrally administered somatostatin is specifically involved in the control of growth and secretory activity of GH cells. Thus, pharmacological manipulation of SRIH receptors reached from cerebrospinal fluid may alter the systemic effects of GH.

Na osnovu ovako dobijenih rezultata može se zaključiti da oba aplikovana somatostatina inhibiraju rast i sekretornu aktivnost somatotropnih ćelija hipofize.
