AB  - Preeclampsia represents a pregnancy specific syndrome characterized by elevated arterial blood pressure and proteinuria. The aim of this study was to evaluate whole blood and erythrocytes energy status in healthy and preeclamptic women. Our study involved 18 healthy (control) and 19 preeclamptic women. Diagnosis of preeclampsia was done according to the recommendation of International Society for the Study of Hypertension in Pregnancy. After blood collection and erythrocytes extraction, the next parameters were followed: concentrations of lactate and adenine nucleotides (ATP, ADP and AMP). Results showed increased concentrations of lactate and ATP in whole blood of preeclamptic, vs. control pregnant women. All parameters of energy status were also elevated in erythrocites of preeclamptic women (elevation of ADP level was significant). This study suggests that preeclamsia was followed by an elevation of all parameters of energy status in whole blood and in erythrocytes, as a result of increased rate of glycolysis. .
AB  - Preeklampsija predstavlja za trudnoću specifičan sindrom koji se karakteriše povišenim krvnim pritiskom i proteinurijom. Cilj istraživanja je određivanje energetskog statusa pune krvi i eritrocita zdravih i trudnica sa preeklampsijom. Ispitivanjem je bilo obuhvaćeno 18 zdravih i 19 trudnica sa preeklampsijom. Dijagnoza preeklampsije je postavljena prema preporukama International Society for the Study of Hypertension in Pregnancy. Nakon uzimanja krvi i ekstrakcije eritrocita praćeni su sledeći parametri: koncentracije laktata i adenin nukleotida (ATP, ADP i AMP). Rezultati su pokazali povećanje koncentracija laktata i ATP u punoj krvi trudnica sa preeklampsijom u odnosu na kontrolne trudnice. U eritrocitima, svi parametri energetskog statusa su bili povećani u preeklampsiji (povećanje ADP nivoa je bilo značajno). Ova studija pokazuje da je preeklampsija praćena povećanjem svih parametara energetskog statusa u punoj krvi i eritrocitima, kao posledica stimulisanog intenziteta procesa glikolize. .

AB  - The effects of the sodium nitroprusside (SNP), a nitric oxide (NO) donor clinically used in the treatment of hypertensive emergencies on the energy production of rat reticulocytes were investigated. Rat reticulocyte-rich red blood cell suspensions were aerobically incubated without (control) or in the presence of different concentrations of SNP (0.1, 0.25, 0.5, 1.0 mM). SNP decreased total and coupled, but increased uncoupled oxygen consumption. This was accompanied by the stimulation of glycolysis, as measured by increased glucose consumption and lactate accumulation. Levels of all glycolytic intermediates indicate stimulation of hexokinase-phosphofructo kinase (HK-PFK), glyceraldehyde 3-phosphate dehydrogenase (GAPD) and pyruvate kinase (PK) activities in the presence of SNP. Due to the decrease of coupled oxygen consumption in the presence of SNP, ATP production via oxidative phosphorylation was significantly diminished. Simultaneous increase of glycolytic ATP production was not enough to provide constant ATP production. In addition, SNP significantly decreased ATP level, which was accompanied with increased ADP and AMP levels. However, the level of total adenine nucleotides was significantly lower, which was the consequence of increased catabolism of adenine nucleotides (increased hypoxanthine level). ATP/ADP ratio and adenylate energy charge level were significantly decreased. In conclusion, SNP induced inhibition of oxidative phosphorylation, stimulation of glycolysis, but depletion of total energy production in rat reticulocytes. These alterations were accompanied with instability of energy status.

AB  - The influence of acute hypoxia on glucose, pyruvate, lipid peroxide (LP) reduced glutathione (GSH) concentrations and lactate level in the whole blood of carp (Cyprinus carpio L) under aquarium conditions were studied. The activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), the concentrations of ATP and ADP and ATP/ADP ratio in the red blood cells (RBCs) were analyzed. Glutathione-S-transferase (GST) activity was determined in the plasma. Our experiments showed that short-term and long-term hypoxia causes significant changes of all examined haema-tological parameters. Increased concentration of LP and increased SOD CAT and GST activities, as well as a decreased GSH-Px activity showed that under hypoxic conditions oxidative stress and RBCs damage were produced.
AB  - Uticaj akutne hipoksije na koncentraciju glukoze, piruvata, lipidnih peroksida (LP), redukovanog glutationa (GSH) i na nivo laktata u krvi šarana (Cyprinu carpio L) je analiziran u akvarijumskim uslovima. U crvenim krvnim zrncima (RBCs) je analizirana aktivnost enzima superoksid-peroksidaze (GSH-Px) kao i koncentracije ATP, ADP i odnos ATP/ADP. U plazmi je analizirana aktivnost glutation S-transferaze (GST). U našim eksperimentima je utvrđeno da kratkotrajna progresivna hipoksija izaziva značajne promene svih ispitivanih hematoloških parametara. Povećanje koncentracije LP i povećanje aktivnosti enzima SOD, CAT i GST uz istovremeno smanjenje aktivnosti GSH-Px potvrđuju nastanak oksidacionog stresa i oštećenje RBCs pod hipoksičnim uslovima.

AB  - Exogenous hydrogen peroxide (H2O2) in concentrations of 2 and 5 mmol/l, but not 0.2 mmol/l and 1 mmol/l, induces significant changes of energy metabolism in retictiloeytes of rats. It induces reduction of total and coupled oxygen consumption accompanied by stimulation of glycolysis and increased degradation of adenine nucleotides. Due to inhibition of mitochondrial oxidative phosphorylation appears a significant decline of ATP production. However, simultaneous stimulation of glycolysis (Pasteur effect) is not sufficient to prevent the decline of ATP concentration. The loss of ATP is not compensated with an expected increase of ADP and AMP concentrations, but with an increase of hypoxanthine concentrations. Despite reduced overall ATP	production and ATP concentrations, average energy turnover was not significantly changed. Activity of oxidative pentose phosphate shunt, metabolic pathway serving primarily antioxidative function in reticulocytes, was also unchanged under the influence of H2O2.
