Age-dependent modulation of central ghrelin effects on food intake and lipid metabolism in rats

AB  - Ghrelin is an endogenous peptide potentially useful in therapy of anorexia and other age-related metabolic disturbances. We evaluated the influence of age on the orexigenic and lipid metabolism-altering effects of ghrelin. Peripubertal, young, adult and middle-aged rats (1, 2, 7 and 12 months old, respectively) were treated with 5 daily intracerebroventricular injections of ghrelin (0.15 nmol) or saline (control). The food intake was measured daily before treatment, while white adipose tissue and serum/plasma samples for detection of lipid metabolites/hormones were collected at the end of the experiment. The values of cumulative food intake and body weight gain declined, while the white adipose tissue deposits and blood concentrations of triglycerides, cholesterol and free fatty acids all increased with age. Ghrelin significantly increased all parameters, but the stimulatory effects on body weight gain and food intake were more pronounced in peripubertal/young rats, while the increase in white adipose mass, triglyceride and low-density lipoprotein cholesterol levels was more noticeable in adult/middle-aged animals. The decrease in sensitivity to ghrelin-mediated stimulation of food intake in older animals could not be explained by alterations in ghrelin's ability to reduce anorexigenic hormones insulin and leptin. However, the higher responsiveness of aged rats to ghrelin-mediated increase in lipid metabolites was accompanied by an increase in adrenocorticotropic hormone and corticosterone levels. These results indicate that aging, while reducing sensitivity to ghrelin-mediated increase in body weight gain and food intake, might enhance the responsiveness to the stimulatory effects of ghrelin on lipid metabolites and hypothalamic-pituitary-adrenal axis activity. AB  - Changes in feeding regime represent serious stress, while ghrelin is considered a key player in energy balance. We investigated the effects of intracerebroventricular (ICV) ghrelin application on pituitary adrenocorticotropic (ACTH) cells in rats fed diets differing in energy content. Before the ICV treatment, male Wistar rats were subjected to three different feeding regimes for 4 weeks: normal-fed (NF), food-restricted (FR) or high-fed (HF) (n = 3 x 14). At the age of 8 weeks, rats from each group were divided into two subgroups and given ICV, either ghrelin (G; 1 mu g ghrelin/5 mu l PBS, n = 7) or solvent alone (5 mu l PBS, n = 7) every 24 h for 5 days. The immunohistochemical appearance and quantitative morphology of pituitary ACTH cells were evaluated, as well as peripheral ACTH and corticosterone levels. Central ghrelin administration increased (p < 0.05) ACTH cell volumes in GNF, GFR and GHF rats by 8.1%, 11.8% and 9.1%, respectively, compared to the controls, while significant increases in ACTH cell volume density were observed in GNF and GHF rats. Circulating ACTH and corticosterone levels were elevated (p < 0.05) in GNF and GFR rats by 72.8% and 80.8%, respectively, when compared to the corresponding controls. AB  - We investigated the effects of centrally administered orexigenic hormone ghrelin on energy imbalance-induced inflammation. Rats were subjected for four weeks to three different dietary regimes: normal (standard food), high-fat (standard food with 30% lard) or food-restricted (70%, 50%, 40% and 40% of the expected food intake in 1st, 2nd, 3rd and 4th week, respectively). Compared to normal-weight controls, starved, but not obese rats had significantly higher levels of proinflammatory cytokines (TNF, IL-1 beta, IFN-gamma) in the blood. When compared to normally fed animals, the hearts of starved and obese animals expressed higher levels of mRNAs encoding proinflammatory mediators (TNF, IL-1 beta, IL-6, IFN-gamma, IL-17, IL-12, iNOS), while mRNA levels of the anti-inflammatory TGF-beta remained unchanged. Intracerebroventricular (ICV) injection of ghrelin (1 mu g/day) for five consecutive days significantly reduced TNF, IL-1 beta and IFN-gamma levels in the blood of starved rats, as well as TNF, IL-17 and IL-12p40 mRNA expression in the hearts of obese rats. Conversely, ICV ghrelin increased the levels of 1FN-gamma, IL-17,1L-12p35 and IL-12p40 mRNA in the heart tissue of food-restricted animals. This was associated with an increase of immunosuppressive ACTH/corticosterone production in starved animals and a decrease of the immunostimulatory adipokine leptin both in food-restricted and high-fat groups. Ghrelin activated the energy sensor AMP-activated protein kinase (AMPK) in the hypothalamus and inhibited extracellular signal-regulated kinase (ERK) in the hearts of obese, but not starved rats. Therefore, central ghrelin may play a complex role in energy imbalance-induced inflammation by modulating HPA axis, leptin and AMPK/ERK signaling pathways. (C) 2011 Elsevier Inc. All rights reserved.

AB  - Background/Aims: The antihyperglycaemic drug metformin reduces food consumption through mechanisms that are not fully elucidated. The present study investigated the effects of intracerebroventricular administration of metformin on food intake and hypothalamic appetite-regulating signalling pathways induced by the orexigenic peptide ghrelin. Methods: Rats were injected intracerebroventricularly with ghrelin (5 mu g), metformin (50, 100 or 200 mu g), 5-amino-imidazole-4-carboxamide 1-beta-D-ribofuranoside (AICAR, 25 mu g) and L-Ieucine (1 mu g) in different combinations. Food intake was monitored during the next 4 h. Hypothalamic activation of AMP-activated protein kinase (AMPK), acetyl-CoA carboxylase (ACC), regulatory-associated protein of mTOR (Raptor), mammalian target of rapamycin (mTOR) and p70 S6 kinase 1 (S6K) after 1 h of treatment was analysed by immunoblotting. Results: Metformin suppressed the increase in food consumption induced by intracerebroventricular ghrelin in a dose-dependent manner. Ghrelin increased phosphorylation of hypothalamic AMPK and its targets ACC and Raptor, which was associated with the reduced phosphorylation of mTOR. The mTOR substrate, 56K, was activated by intracerebroventricular ghrelin despite the inhibition of mTOR. Metformin treatment blocked ghrelin-induced activation of hypothalamic AMPK/ACC/Raptor and restored mTOR activity without affecting 56K phosphorylation. Metformin also reduced food consumption induced by the AMPK activator AICAR while the ghrelin-triggered food intake was inhibited by the mTOR activator L-leucine. Conclusion: Metformin could reduce food intake by preventing ghrelin-induced AMPK signalling and mTOR inhibition in the hypotalamus. Copyright (c) 2012 S. Karger AG, Basel

AB  - Ghrelin, the endogenous ligand of growth hormone secretagogue receptor type la (GHS-R1a), has emerged as pleiotropic modulator of diverse biological functions, including energy homeostasis and recently, reproduction. The influence of intracerebroventricularly (ICV) administered ghrelin (1 mu g/day/rat for 5 days) to rats of different ages, i.e., peripubertal (38 days), adult (60 days) and middle-aged (180 days) on the ventral prostate size and morphology, serum testosterone levels and testis weight was examined. Ghrelin treatment significantly increased (p < 0.05) absolute ventral prostate weight in peripubertal and middle-aged rats, by 27% and 37% respectively, due to enhancement of epithelial and/or luminal compartment of the gland. In adult rats, both absolute and relative volumes of the acinar lumen were significantly decreased (p < 0.05), by 38% and 44% respectively, which was associated with significant increases (p < 0.05) in relative and absolute volumes of interacinar stroma, whereas ventral prostate weigh was unchanged. Irrespective of animal age, ghrelin did not affect serum testosterone levels. These are the first results of ghrelin treatment effects on healthy prostate appearance, which allow us to conclude that the rat ventral prostate response to ghrelin depends on the developmental stage of animals. Our results merit further investigations and may have clinical implications, especially in the light of data on possible role of ghrelin in prostate hypertrophy and adenomas.

AB  - Ghrelin, a growth hormone secretagogue that exerts an important role in appetite and weight regulation, participates in the activation of the hypothalamo-pituitary-adrenal (HPA) axis. Male Wistar rats (5/group) received daily for 5 days, via an ICV (intracerebroventricular) cannula, 5 ill phosphate buffered saline with or without 1 mu g of rat ghrelin. Two hours after the last injection, blood and adrenal glands were collected from decapitated rats for blood hormone analyses and histologic and morphometric processing. Ghrelin treatment resulted in increased (p < 0.05) body weight (13%), absolute whole adrenal gland weight (18%) and whole adrenal gland volume (20%). The absolute volumes of the entire adrenal cortex, ZG, ZF, and ZR also increased (p < 0.05) after ghrelin by 20%, 21%, 21% and 11%, respectively. Ghrelin-treated rats had elevated (p < 0.05) blood concentrations of ACTH, aldosterone and corticosterone (68%, 32% and 67%, respectively). The data clearly provide both morphological and hormonal status that ghrelin acts centrally to exert a global stimulatory effect on the adrenal cortex. Clarifying of the ghrelin precise role in the multiple networks affecting the stress hormone release, besides its well known energy and metabolic disbalance effects, remains a very important research goal.

AB  - Body weight depends on the balance between energy intake and consumption. An interaction between ghrelin and thyroid function has been reported only in pathophysiological states. We examined whether intracerebroventricular (ICV) administration of ghrelin affects the structure and function of the pituitary-thyroid axis in young adult male rats. Ghrelin (0.3 nmol/5 mu l PBS) or an equal volume of PBS were injected every 24 h into the lateral cerebral ventricle for 5 days. Two hours after the last treatment the animals were killed, their pituitaries and thyroids excised and prepared for further histological, immunohistochemical and morphometric investigation. Serum TSH levels were measured by RIA, while the total T(4) and T(3) levels were examined by ECLIA. Ghrelin treatment increased pituitary weight (p < 0.05) when compared to the controls, with no effect on the thyroid weight. Smaller, degranulated TSH-immunopositive cells were noticed within the pituitaries of ghrelin-treated animals; their cellular and nuclear volume as well as the relative volume density of thyrotrophs decreased (p < 0.05) in comparison to the control values. The level of serum TSH was reduced (p < 0.05). In the thyroid parenchyma of ghrelin-treated rats, an increased number of hypofunctioning follicles was noticed, characterized by flattened, weakly Tg-immunoreactive epithelium and colloid distension. The relative volume densities of the follicles and colloid increased (p < 0.05), while the thyroid index of activation rate and the serum level of total T(4) decreased (p < 0.05). In conclusion, centrally applied ghrelin modulated the immunohistomorphometric features of pituitary TSH cells and decreased the level of serum TSH, consequently changing thyroid morphology and function, by reducing the T(4) hormone level in the serum. It has central nervous system actions that affect appetite, body mass and energy balance. An intracerebroventricular (ICV) injection protocol of sub-nanomolar doses of ghrelin, known to alter the morphology of ACTH and GH producing pituicytes and plasma levels of these hormones, was used to provide an overview of metabolic changes linked to energy metabolism. Variables measured were: food intake (FI), water intake (WI), fecal mass, urine volume, body weight (BW), retroperitoneal (RP) and epididymal (EPI) white adipose tissue (WAT), and changes in serum leptin, insulin, triglycerides, cholesterol, and glucose. Five injections of rat ghrelin or PBS (n=8 per group) were given ICV every 24 h (1 mu g/5 mu L PBS) to adult male rats. Ghrelin had a positive and cumulative effect on FI, WI and BW (p<0.05), but not feces mass or urine volume (p>0.05). Centrally applied ghrelin clearly increased RP WAT (by 235%, p<0.001), EPI WAT (by 85%, p<0.05) and serum insulin levels (by 43%, p<0.05), and decreased serum leptin levels (by 77%, p<0.05) without (p>0.05) evoking changes in blood triglyceride cholesterol, or glucose levels. These data and the available literature clearly document that exposure of the brain of normal rats, over time, to sub-nanomolar doses of ghrelin results in metabolic dysregulation culminating in increased body mass, consummatory behavior, and lipid stores as well as changes in blood leptin/insulin levels. Thus, modulation of central ghrelin receptors may represent a pharmacological approach for controlling multiple factors involved in energy balance and obesity. (C) 2008 Elsevier B.V. This natural GH secretagogue (GHS) was originally purified from rat stomach, but it is expressed widely in different tissues where it may have endocrine and paracrine effects. The central effects of ghrelin on adrenocorticotropic, hormone (ACTH) cells, ACTH release and subsequent corticosterone release from adrenal glands remains to be clarified. The aim of this study was to specifically determine the morphological features of ACTH-producing pituicytes and blood concentration of ACTH and corticosterone after central administration of ghrelin. Five doses of rat ghrelin or PBS (n = 10 per group) were injected every 24 h (1 mu g of ghrelin in 5 mu L PBS), into the lateral cerebral ventricle of male rats. Results showed that ghrelin increased (p < 0.05) absolute and relative pituitary weights compared to controls (58% and 41% respectively). Morphometric parameters, i.e. the volume of the ACTH cells, nuclear volume, and volume density were all increased (p < 0.05), by 17%, 6% and 13%, respectively, 2 h after the last ghrelin treatment. Ghrelin increased circulating concentrations of ACTH and corticosterone (p < 0.05) by 62% and 66%, respectively. The data provide clear documentation that intracerebroventricular ghrelin stimulates ACTH cell hypertrophy and proliferation, and promotes ACTH and corticosterone release. Determining the role of ghrelin in physiological stress responses and whether control of the peptide's activity would be useful for prevention and/or treatment of stress-induced diseases remain important research goals. Five injections of rat ghrelin or phosphate-buffered saline (PBS; n = 10 rats/group) were given every 24 hrs (1 mu g of ghrelin in 5 mu l of PBS) into the lateral cerebral ventricle of male rats. Significant (P < 0.05) increases in absolute and relative pituitary weights occurred in ghrelin-treated rats versus controls (58% and 41%, respectively). Morphometric parameters (i.e., the volume of GH cells, volume of their nuclei, and volume density) all significantly (P < 0.05) increased by 17%, 18%, and 19%, respectively, in the ghrelin-treated group versus controls. Terminal serum concentration of GH was significantly (P < 0.05) increased by 15% with ghrelin treatment. The results clearly document that daily nanomolar doses of ghrelin into the lateral cerebral ventricle stimulate GH cell proliferation and promote GH release. Thus, achieving pharmacologic control of central ghrelin receptors is a promising modality to modulate the actions of GH.

AB  - The effect of chronic exposure to light of adult Wistar rats on growth and function of adrenal zona glomerulosa (ZG) and zona fasciculata (ZF) were examined. The females were exposed to continuous light of 600 lux for 95 days, starting on day 30 of age. The controls were kept under a 12 : 12 h light-dark cycle, at ambient temperature. The rats were sacrificed by decapitation and the left adrenal gland of each animal was dissected out and prepared for morphometric analyses. In animals exposed to chronic lighting, the absolute and relative volume of ZG were insignificantly increased by 5% (p > 0.05) compared to controls. The volume of ZG cells and their nuclei were insignificantly changed by 1% (p > 0.05) in comparison with corresponding controls. The absolute and relative volume of ZF were significantly increased (by 14 and 9%, respectively, p < 0.05), as compared to controls. The volume of ZF cells and their nuclei were significantly increased (by 12 and 9%, respectively; p < 0.05). Serum concentration of corticosterone was also significantly (p < 0.05) increased by 13 % in light-exposed group in comparison with control rats. These findings suggest that continuous exposure of female rats to constant light increased growth and secretory activity of ZF cells.

AB  - The results on the effects of synthetic somatostatins on the secretion of ACTH have been rather conflicting, but some authors reported that somatostain is involved in the regulation of ACTH secretion in rats of both sexes. The inhibitory mechanism of ACTH secretion could operate through the inhibition of CRF release from the hypothalamus. In the present study, we have investigated the effects of Octreotide, a somatostain analogue given intracerebroventricularly (i.c.v) in low doses, on morphometric characteristics of pituitary adrenocorticotropes (ACTH cells) in adult Wistar rat males. The animals were i.c.v. given three 1.0 µg doses of Octreotide dissolved in 10 µL saline every second day. The controls were treated in the same way with the same volume of saline. ACTH-producing cells were studied using the peroxidase-antiperoxidase (PAP) immunohistochemical procedure. Body weight, as well as absolute and relative pituitary weights were not significantly changed in comparison with the corresponding controls (p>0.05). The characteristics of immunohistochemicaly labelled ACTH cells in intact rat males were as follows: localization between the capillaries stellate in shape with the cytoplasmatic processes among neighbouring cells. The nucleus follows the shape of the cell body. Small specific secretory granules were distributed mainly at the periphery of the cytoplasm. The shape, and localization of ACTH immunoreactive cells in Octreotide-treated animals were not significantly changed in comparison with the controls. Octreotide treatment significantly decreased all morphometric parameters measured, i.e. volume of ACTH cells and their nuclei by 44% and 18% respectively, as compared to the controls (p<0.05). Volume densities were also significantly decreased (by 31%; p<0.05) comparing to the corresponding controls. These results indicate that Octreotide applied i.c.v. exerts a significant inhibitory effect on the immunohistochemical and morphometric characteristics of ACTH cells in rat males.
AB  - Postoje veoma različita, a često i suprotstavljena mišljenja o efektima sintetskih somatostatina na lučenje adrenokortikotropnog hormona (ACTH) iz hipofize. Mogući mehanizam inhibicije lučenja ACTH je preko inhibicije lučenja CRF iz hipotalamusa. U ovom radu su ispitivani efekti intracerebroventrikularno (i.c.v) ubrizganog analoga somatostatina Oktreotida, na rast ACTH ćelija adenohipofize mužjaka Wistar pacova. Sve eksperimentalne životinje su primile tri doze od po 1.0 µg Oktreotida rastvorenog u 10 µL fiziološkog rastvora, svakog drugog dana. Kontrole su tretirane na isti način fiziološkim rastvorom. Pacovi su žrtvovani pet dana posle poslednje injekcije. ACTH ćelije su imunocitohemijski bojene metodom peroksidaza-antiperoksidaza (PAP). Telesna masa, apsolutna i relativna masa hipofize nisu bile statistički značajno promenjene u poređenju sa odgovarajućom kontrolom (p>0.05). Zapremina ćelija je bila smanjena za 44%, jedara za 18%, a volumenska gustina za 31% (p<0.05) u poređenju sa odgovarajućom kontrolom. Na osnovu dobijenih rezultata može se zaključiti za Oktreotid deluje inhibitorno na rast ACTH ćelija adenohipofize mužjaka pacova.

AB  - The effects of chronic exposure to light of adult female Wistar rats on growth and function of pituitary adrenocorticotropes (ACTH cells) were examined. The animals were exposed to continuous light of 600 lux for 95 days, starting on day 30 of age. Control rats were kept under a 12:12 h light-dark cycle, at ambient temperature. ACTH-producing cells were studied using the peroxidase-antiperoxidase immunohistochemical procedure and blood samples were collected for hormone analyses. In animals exposed to a chronic light-treatment all morphometric parameters measured throughout the present study i.e.: ACTH cell volume, nuclear volume and relative volume density were increased by 22% and the differences between this group and the controls were statistically significant (p<0.05). The concentration of plasma ACTH was elevated by 13% in light-exposed group in comparison with the control and this difference was statistically significant (p<0.05), as well. These findings suggest that continuous exposure to light is specifically involved in growth and secretory activity of ACTH cells of adenohypophysis of rat females.
AB  - Ispitivani su efekti dugotrajnog izlaganja ženki Wistar pacova stalnom svetlu, na adrenokortikotropne (ACTH) ćelije. Eksperimentalne životinje stare 30 dana izlagane su konstantnom svetlu (600 luksa) tokom 95 dana. Kontrolne životinje odgovarajuće starosti su držane na normalnom, 12:12 h dnevno-noćnom režimu i sobnoj temperaturi. ACTH ćelije su imunohistohemijski bojene metodom peroksidaza-antiperoksidaza (PAP), a istovremeno je sakupljana krv za određivanje koncentracije ACTH u plazmi. Životinje izložene stalnom svetlu imale su za 22% povećane sve morfometrijske parametre ACTH ćelija merene tokom ovog rada (zapremina ćelija, njihovih jedara i volumenska gustina) u odnosu na kontrole i uočene razlike su bile statistički značajne (p < 0.05). Koncentracija ACTH u plazmi je takođe bila veća za 13% (p < 0.05) u odnosu na kontrolne vrednosti. Ovi rezultati ukazuju da dugotrajno izlaganje stalnom svetlu na specifičan način stimuliše rast i sekretornu aktivnost ne samo ACTH ćelija adenohipofize već i ćelija pars intermedia ženki pacova.

Povećana agregabilnost trombocita posle aterogene dijete u kunića

AB  - In atherosclerosis researches different animal models are used but the most common is the rabbit, because of the easy development of atherosclerotic lesions. Atherosclerosis is a multicellular process and platelets play an important role in atherogenesis. Excessive plasma lipids stimulate platelet aggregability and thus atherosclerosis development. The effects of an atherogenic diet on lipid status, abdominal aorta wall structure, and platelet aggregability were studied in rabbits. Adult male Chinchilla rabbits were fed an atherogenic diet (2% edible oil solution of crystaline cholesterol at 8 mg cholesterol per kg b.wt.daily) for 8 weeks. Plasma lipid levels (triglycerides, total cholesterol, LDL-cholesterol, HDL-cholesterol and LDL/HDL ratio) and platelet aggregability were then measured and compared to the values obtained for control animals. Histological analysis showed atherogenic changes in the abdominal aorta wall of the experimental animals. Total plasma cholesterol level, plasma LDL-cholesterol, and LDL/HDL ratio were significantly increased compared to the controls (p<0.01), as well as triglyceride and HDL-cholesterol levels (p <0.05). Platelet aggregability was also significantly increased (p<0.01) after the atherogenic diet in comparison with controls when ADP, at concentrations both of 5 and 10 (mol/L, was used as the aggregant. Disaggregation was common in control animals but this phenomenon was not recorded in the experimental animals. It can be concluded that an 8-week atherogenic diet in rabbits induced marked changes of the lipid parameters, provoked atherogenic changes in the abdominal aorta wall and increased platelet aggregability.
AB  - U istraživanjima ateroskleroze korišćeni su razlčiti animalni modeli, ali najčešće korišćene životinje su kunići zbog toga što lako razvijaju aterosklerozne lezije. Ateroskleroza je multicelularni proces i trombociti igraju važnu ulogu u aterogenezi. Prekomerna koncentracija lipida u plazmi izaziva povećanu agregaciju trombocita i na taj način pospešuje razvoj ateroskleroze. Istraživali smo efekte aterogene dijete na lipidni status promene na" abdominalnoj aorti i agregabilnost trombocita u kunića. Odrasli mužjaci Cinčila kunića bill su podvrgnuti aterogenoj dijeti koja se sastojala od 2% rastvora kristalnog holesterola u jestivom ulju u toku 8 nedelja. Dnevna doza holesterola bila je 8 mg/kg telesne mase. Po završetku dijete, određivane su koncentracije triglicerida, ukupnog holesterola, LDL holesterola i HDL holesterola, odnos LDL/HDL, kao i agregabilnost trombocita. Vrednosti su poređene sa kontrolnim životinjama. Histološka analiza je pokazala aterogene promene u zidu abdominalne aorte eksperimentalnih životinja. Koncentracije ukupnog i LDL holesterola u plazmi kao i odnos LDL/HDL bili su visoko statisticki značajno povećane (p<0,01). Koncentracije triglicerida i HDL-holesterola u plazmi takođe su bile statističk značajno povećane (p<0,05). Agregabilnost trombocita bila je statisticki visoko značajno povećana (p<0,01) posle aterogene dijete, bez obzira na to da II je agregant ADP korišćen u koncentraciji 5 ili 10 (mol/L. Fenomen dezagregacije bio je uobičajen za kontrolne životinje, ali nije registrovan kod eksperimentalnih životinja. Može se zaključiti da je osmonedeljna aterogena dijeta u kunića izazvala značajne promene lipidnih varijabli, izazvala nastanak aterogenih promena u zidu abdominalne aorte i povećala agregabilnost trombocita.

It can be concluded that an 8-week atherogenic diet in rabbits induced marked changes of the lipid parameters, provoked atherogenic changes in the abdominal aorta wall and increased platelet aggregability., U istraživanjima ateroskleroze korišćeni su razlčiti animalni modeli, ali najčešće korišćene životinje su kunići zbog toga što lako razvijaju aterosklerozne lezije. Ateroskleroza je multicelularni proces i trombociti igraju važnu ulogu u aterogenezi. Prekomerna koncentracija lipida u plazmi izaziva povećanu agregaciju trombocita i na taj način pospešuje razvoj ateroskleroze. Istraživali smo efekte aterogene dijete na lipidni status promene na" abdominalnoj aorti i agregabilnost trombocita u kunića. Odrasli mužjaci Cinčila kunića bill su podvrgnuti aterogenoj dijeti koja se sastojala od 2% rastvora kristalnog holesterola u jestivom ulju u toku 8 nedelja. Dnevna doza holesterola bila je 8 mg/kg telesne mase. Po završetku dijete, određivane su koncentracije triglicerida, ukupnog holesterola, LDL holesterola i HDL holesterola, odnos LDL/HDL, kao i agregabilnost trombocita. Vrednosti su poređene sa kontrolnim životinjama. Histološka analiza je pokazala aterogene promene u zidu abdominalne aorte eksperimentalnih životinja. Koncentracije ukupnog i LDL holesterola u plazmi kao i odnos LDL/HDL bili su visoko statisticki značajno povećane (p<0,01). Koncentracije triglicerida i HDL-holesterola u plazmi takođe su bile statističk značajno povećane (p<0,05). Agregabilnost trombocita bila je statisticki visoko značajno povećana (p<0,01) posle aterogene dijete, bez obzira na to da II je agregant ADP korišćen u koncentraciji 5 ili 10 (mol/L. Fenomen dezagregacije bio je uobičajen za kontrolne životinje, ali nije registrovan kod eksperimentalnih životinja. Može se zaključiti da je osmonedeljna aterogena dijeta u kunića izazvala značajne promene lipidnih varijabli, izazvala nastanak aterogenih promena u zidu abdominalne aorte i povećala agregabilnost trombocita.",

AB  - In this study we measured the effects of eight-week atherogenic diet on plasma lipid status, glutathione-S-transferase activity, red blood cells-catalase activity, and glutathione and vitamin E plasma level adult male Wistar albino rats. The animals were sacrificed by decapitation 24 h after the end of the diet. Triglycerides, total cholesterol, and LDL cholesterol plasma levels, as well as LDL/HDL ratio, and vitamin E plasma level significantly raised (p<0,05) after the diet in comparison with the controls. HDL cholesterol plasma level, glutathione-S-transferase and red blood cells-catalase activities, and glutathione plasma level remained unchanged (p>0,05). These findings demonstrated that prolonged specific atherogenic diet in rats induced significant raise of the lipid parameters, but did not affect blood antioxidant status.

AB  - The effects of intracerebroventricularly administered somatostatin (SRIH-14 or -28) on growth and function of pituitary thyrotropes (TSH-cells) were examined in adult male Wistar rats. The animals were implanted with an intracerebroventricular cannula and after a recovery period, administered three 1 mug doses of SRIH-14 or -28 dissolved in 5 mul saline every second day. Controls were treated in the same way with the same volume of saline only. TSH-producing cells were studied using the peroxidase-antiperoxidase immunohistochemical procedure. Blood samples were collected for hormone (TSH) analyses 5 days after the last injection. Both SRIH-treatments significantly decreased (p < 0.05) all morphometric parameters obtained for TSH-cells in comparison with controls. The volume of TSH-cells decreased by 27%, nuclei by 44% and volume density by 33% in animals treated with SRIH-14. In animals treated with SRIH-28, these parameters were also significantly decreased (p < 0.05) (22%, 31%, and 25% respectively) compared to control rats. Serum concentrations of TSH were significantly decreased (p < 0.05) by 15% in SRIH-14- and by 12% in SRIH-28-treated animals in comparison with the controls. These observations suggest that centrally administered SRIH-14 or -28 is specifically involved in the control of growth and secretory activity of TSH cells.
