Antioxidant defence enzyme activities in hepatopancreas, gills and muscle of Spiny cheek crayfish (Orconectes limosus) from the River Danube

AB  - The aim of our study was to determine the activity of antioxidant defence (AD) enzymes: superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and the phase II biotransformation enzyme glutathione-S-transferase (GST) in the hepatopancreas, the gills and muscle of Spiny cheek crayfish (Orconectes limosus) from the River Danube and to compare tissue specificities of investigated enzymes. Our results indicated that both specific and total SOD activities in the hepatopancreas were lower compared to the gills and muscle. Total SOD activity in the gills was lower with respect to that in muscle. CAT and GSH-Px (both specific and total) activities were higher in the hepatopancreas compared to those in the gills and muscle. In the gills the specific and total GR activities were higher than in the hepatopancreas and muscle. The specific and total GST activities were higher in the hepatopancreas compared with the gills and muscle. Our study represents the first comprehensive report of AD enzymes in tissues of O. limosus caught in the River Danube. The noted tissue distributions of the investigated AD enzyme activities most likely reflected different metabolic activities and different responses to environmental conditions in the examined tissues. AB  - We investigated the activities of glutathione peroxidase (GSH-Px) glutathione reductase (GR) and phase II biotransformation enzyme glutathione-S-transferase (GST) in the hepatopancreas, the gills and muscle of Noble crayfish (Astacus astacus-AST), Stone crayfish (Austropotamobius torrentium-AUS) and Spynicheek crayfish (Orconectes limosus-ORC) from the South Morava River, Krajkovačka River and Serbian part of the Danube. Specific GSH-Px activity was lower in the hepatopancreas of AUS than in ORC and in the muscle of AST than in AUS and ORC. We obtained differences of total GSH-Px activity in the muscle between AST and AUS and between AST and ORC. Specific and total GR activity was higher in the hepatopancreas of AST than in other species. Specific and total activity of GR in the gills and in the muscle of crayfish species was higher in AUS then in AST and ORC. The specific and total GST activity was lower in the hepatopancreas of AST and ORC than in AUS. Specific and total GST activity in the gills was higher in AUS than in AST. The lowest specific and total GST activity was detected in the muscle of ORC compared to the AST and AUS. Our results show that specific and total enzyme activities of GSH-Px, GR and GST have species and tissue specifics between crayfish species and represent biomarkers of environmental influences on these organisms.
AB  - Određivali smo aktivnost glutation-peroksidaze (GSH-Px), glutation-reduktaze (GR) i enzima faze II biotransformacije glutation-S-transferaze (GST) u hepatopankreasu, škrgama i mišiću rečnog raka (Astacus astacus-AST), raka kamenjara (Austropotamobius torrentium-AUS) i američkog raka (Orconectes limosus-ORC) iz Južne Morave, Krajkovačke reke i Dunava. Specifična aktivnost GSH-Px je manja u hepatopankreasu kod AUS nego kod ORC, kao i u mišiću AST u poređenju sa AUS i ORC. Postoji razlika u aktivnosti između ukupne GSH-Px u mišiću kod AST i AUS, kao i između AST i ORC. Specifična i totalna aktivnost GR je veća u hepatopankreasu kod AST nego kod druge dve vrste, dok je u škrgama i mišiću rakova veća kod AUS nego kod AST i ORC. Specifična i totalna aktivnost GST je manja u hepatopankreasu kod AST i ORC u poređenju sa AUS, a u škrgama je veća kod AUS nego kod AST. AB  - In our experiments, we studied superoxide dismutase (SOD) and catalase (CAT) activities in the hepatopancreas, the gills and the abdominal muscle of Noble crayfish (Astacus astacus), Stone crayfish (Austropotamobius torrentium) and Spynicheek crayfish (Orconectes limosus) from the South Morava River, Krajkovačka River and Serbian part of the Danube respectively. The specific SOD activity in the hepatopancreas of the A. astacus was significantly higher in respect to other two species (p<0.005) as well as in the hepatopancreas of A. torrentium then in O. limosus (p<0.02). The same trend was obtained for the total SOD activity in the hepatopancreas, which was significantly decreased in O. limosus compared to two other investigated species (p<0.005). The specific CAT activity in the muscle of the Austropotamobius torrentium was significantly higher in respect to Astacus astacus (p<0.005) and Orconectes limosus (p<0.005). Specific CAT activity was considerably higher in the Astacus astacus then in Orconectes limosus (p<0.02). The same trend was detected in the case of the total CAT activity in the muscle but, total CAT activity was significantly decrease in Orconectes limosus, compare to Astacus astacus (p<0.01). The obtained results show strong tissue and species specificities, indicating different metabolic activities and various environmental influences.
AB  - U našim eksperimentima određivali smo aktivnost superoksid-dismutaze (SOD) i katalaze (CAT) u hepatopankreasu, škrgama i abdominalnom mišiću rečnog raka (Astacus astacus L) sa lokaliteta Južna Morava, raka kamenjara (Austropotamobius torrentium Schrank) sa lokaliteta Krajkovačka reka i američkog raka (Orconectes limosus Raf) sa lokaliteta Dunav. Specifična aktivnost SOD u hepatopankreasu rečnog raka bila je značajno veća u hepatopankreasu u poređenju sa druge dve vrste (p<0.005), kao i u hepatopankreasu Austropotamobius torrentium u odnosu na Orconectes limosus (p<0.02). AB  - Fish are the most important organisms used in biomonitoring of aquatic ecosystems. Parameters of oxidative stress in fish tissues represent significant biomarkers in the assessment of the status of environment. Specimens of hake (Merluccius merluccius L.) and sea bream (Pagellus erythrinus L.) were collected at the end of May 2005 from the locality in front of sea-port Bar (South Adriatic Sea). Parameters of oxidative stress (concentration of lipid peroxidase - LP and reduced glutathione - GSH) were determined in the liver and white muscle of hake and sea bream. Physical-chemical parameters (salinity, concentration of dissolved oxygen and temperature) were determined in water of the investigated locality. Obtained results showed that the concentration of LP was higher in liver of hake and sea bream in comparison to white muscle. However, significantly higher concentration of GSH was found in white muscle of hake and sea bream in comparison to liver. These changes of oxidative stress parameters point to tissue specificity, which is the consequence of different metabolic and antioxidative activity. Inter-specific differences were also established. Higher concentrations of LP and GSH in liver of hake and in white muscle of sea bream were recorded, due to the changes of physical-chemical parameters of environment. The intensity of oxidative stress in liver of hake and sea bream was higher in comparison to muscle.
AB  - Ribe predstavljaju najznačajnije organizme koji se koriste u biomonitoringu vodenih ekosistema. Parametri oksidacionog stresa u tkivima riba predstavljaju značajne biomarkere u proceni stanja životne sredine. Krajem maja 2005. godine uzorkovane su jedinke oslića (Merluccius merluccius L.) i arbuna (Pagellus erythrinus L.) sa lokaliteta ispred luke Bar (Južni Jadran). Parametri oksidacionog stresa (koncentracija lipidnih peroksida - LP i redukovani glutationa - GSH) su određivani u tkivima jetre i belog mišića oslića i arbuna. U vodi ispitivanog lokaliteta određivani su i fizičko-hemijski parametri (salinitet, koncentracija rastvorenog kiseonika i temperatura). Dobijeni rezultati su pokazali da je koncentracija LP veća u jetri oslića i arbuna u odnosu na belo mišićno tkivo. Međutim, zabeležena je znatno veća koncentracija GSH u belom mišiću i oslića i arbuna u odnosu na jetru. Ovakve promene parametara oksidacionog stresa ukazuju na tkivnu specifičnost, kao posledica različite metaboličke i antioksidacione aktivnosti. Ustanovljene su i interspecijske razlike, zabeležena je veća koncentracija LP i GSH u jetri oslića, kao i u belom mišiću arbuna, kao posledica promena fizičko-hemijskih parametara sredine i prisustva polutanata. Intenzitet oksidacionog stresa u jetri oslića i arbuna je bio viši u odnosu na mišiće.

AB  - The effects of cadmium (Cd, 15 mg/kg/day) on the blood antioxidant defense system and the possible protective effects of selenium (Se, 7 μg/kg/day) were studied. Chronic Cd administration resulted in increased activities of copper, zinc superoxide dismutase (CuZn SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) in red blood cells (RBC), as well as glutathione S-transferase (GST) activity and vitamin E (Vit E) concentration in the plasma. Se induced a significant increase in GSH-Px and GST activities, as well as concentrations of vitamin C and E. Thus, the mechanism by which Se decreases Cd toxicity in rats, seems to rely on the protection of the antioxidant defense system in blood, possibly by forming non-toxic cadmium selenide. The accumulation of Cd in RBC also decreased significantly upon co-exposure to Se. The obtained results show, that Cd exhibit toxic effects on the antioxidant defense system in the blood of rats and that Se demonstrates the protective effect against Cd toxicity.
AB  - U ovom radu ispitivan je efekat kadmijuma (Cd, 15 mg/kg/danu) na antioksidacioni zaštitni sistem i moguća zaštitna uloga selena (Se, 7 μg/kg/danu). Posle hroničnog tretmana Cd značajno se povećavaju aktivnosti bakar, cink superoksid dismutaze (CuZn SOD), katalaze (CAT) i glutation peroksidaze (GSH-Px) u crvenim krvnim ćelijama (RBC), kao i aktivnost glutation S-transferaze (GST) i koncentracije vitamina E (Vit E) u plazmi. Selen značajno povećava aktivnosti enzima GSH-Px i GST, kao i koncentracije vitamina C i E. Mehanizam kojim selen smanjuje toksičnost Cd kod pacova odnosi se na zaštitu antioksidacionog sistema u krvi, posebno formiranje ne-toksičnih kadmium-selenita. Akumulacija Cd u RBC takođe je značajno smanjena posle ko-tretmana selenom. Dobijeni rezultati pokazuju da Cd ispoljava toksične efekte na antioksidacioni zaštitni sistem u krvi pacova i da Se pokazuje zaštitni efekat protiv toksičnosti Cd.

AB  - The aim of our study was to investigate the copper and zinc containing superoxide dismutase (CuZn SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cells (RBCs), as well as phase II biotransformation enzyme glutathione-S-transferase (GST) activity in the plasma of male, two months old Wistar albino rats. The rats were divided into two experimental groups: control animals and animals treated with olive oil (100 μL of virgin olive oil, i.m., every fifth day during 30 days). The obtained results show that olive oil induced a significant decrease of CuZn SOD, CAT and GSH-Px activities in RBCs (p<0.05) and significant increase of GST activity in the plasma (p<0.05) in relation to the control animals. The results show that olive oil influences the activity of investigated enzymes in the blood of rats and in that way improves the overall antioxidative defense.
AB  - Cilj našeg rada bio je da se odrede aktivnosti bakar cink sadržavajuće superoksid-dismutaze (CuZn SOD), katalaze (CAT) i glutation-peroksidaze (GSH-Px) u eritrocitima, kao i enzima faze II biotransformacije glutation-S-transferaze (GST) u plazmi mužjaka dva meseca starih Wistar albino pacova tretiranih maslinovim uljem. Pacovi su podeljeni u dve grupe i to kontrolna i grupa tretirana maslinovim uljem (100 μL of maslinovog ulja, i.m., svaki peti dan tokom 30 dana). Rezultati pokazuju da maslinovo ulje izaziva značajno smanjenje aktivnosti CuZn SOD, CAT i GSH-Px (p<0.05) u eritrocitima, kao i povećanje aktivnosti GST u plazmi pacova (p<0.05) u odnosu na kontrolu. Dobijeni rezultati pokazuju da maslinovo ulje utiče na aktivnost ispitivanih enzima u smislu poboljšanja antioksidacione zaštite.

Specimens of hake (Merluccius merluccius L) and red mullet (Mullus barbatus L) were collected from the locality in front of sea-port Bar (South Adriatic Sea) at the end of May 2005. Activity of the enzymes of the antioxidative system (superoxide dismutase - SOD and catalase - CAT) and parameters of oxidative stress (lipid peroxidation - LP and reduced glutathione - GSH content) were determined in the liver of hake and red mullet. Obtained results showed that the activities of SOD and CAT were significantly lower in the liver of hake than in red mullet. Higher concentrations of LP and GSH in liver of hake were recorded, due to the changes of physical-chemical parameters of environment. The intensity of oxidative stress in liver of hake was higher in comparison to red mullet.
AB  - Ribe predstavljaju najznačajnije organizme koji se koriste u biomonitoringu vodenih ekosistema. Parametri oksidacionog stresa u tkivima riba predstavljaju značajne biomarkere u proceni stanja životne sredine. Krajem maja 2005. godine uzorkovane su jedinke oslića (Merluccius merluccius L) i trlje (Mullus barbatus L) sa lokaliteta ispred luke Bar (Južni Jadran). Aktivnost enzima antioksidacionog sistema (superoksid dismutaze - SOD i katalaze - CAT) i parametri oksidacionog stresa (koncentracija lipidnih peroksida - LP i redukovanog glutationa - GSH) su određivani u jetri oslića i trlje. Dobijeni rezultati su pokazali da su aktivnosti SOD i CAT značajno manje u jetri oslića nego kod trlje. Zabeležena je veća koncentracija LP i GSH u jetri oslića, kao posledica promena fizičko-hemijskih parametara sredine i prisustva polutanata. Intenzitet oksidacionog stresa u jetri oslića je bio viši u odnosu na trlju.

AB  - Preeclampsia represents a pregnancy specific syndrome characterized by elevated arterial blood pressure and proteinuria. The aim of this study was to evaluate whole blood and erythrocytes energy status in healthy and preeclamptic women. Our study involved 18 healthy (control) and 19 preeclamptic women. Diagnosis of preeclampsia was done according to the recommendation of International Society for the Study of Hypertension in Pregnancy. After blood collection and erythrocytes extraction, the next parameters were followed: concentrations of lactate and adenine nucleotides (ATP, ADP and AMP). Results showed increased concentrations of lactate and ATP in whole blood of preeclamptic, vs. control pregnant women. All parameters of energy status were also elevated in erythrocites of preeclamptic women (elevation of ADP level was significant). This study suggests that preeclamsia was followed by an elevation of all parameters of energy status in whole blood and in erythrocytes, as a result of increased rate of glycolysis. .
AB  - Preeklampsija predstavlja za trudnoću specifičan sindrom koji se karakteriše povišenim krvnim pritiskom i proteinurijom. Cilj istraživanja je određivanje energetskog statusa pune krvi i eritrocita zdravih i trudnica sa preeklampsijom. Ispitivanjem je bilo obuhvaćeno 18 zdravih i 19 trudnica sa preeklampsijom. Dijagnoza preeklampsije je postavljena prema preporukama International Society for the Study of Hypertension in Pregnancy. Nakon uzimanja krvi i ekstrakcije eritrocita praćeni su sledeći parametri: koncentracije laktata i adenin nukleotida (ATP, ADP i AMP). Rezultati su pokazali povećanje koncentracija laktata i ATP u punoj krvi trudnica sa preeklampsijom u odnosu na kontrolne trudnice. U eritrocitima, svi parametri energetskog statusa su bili povećani u preeklampsiji (povećanje ADP nivoa je bilo značajno). Ova studija pokazuje da je preeklampsija praćena povećanjem svih parametara energetskog statusa u punoj krvi i eritrocitima, kao posledica stimulisanog intenziteta procesa glikolize. .

AB  - Preeclampsia represents a pregnancy specific syndrome with high rate of both maternal and neonatal morbidity and mortality. The maternal syndrome is characterized by elevated arterial blood pressure and proteinuria. Several recent studies have reported that there is a disbalance of oxidant/antioxidant system in women with preeclampsia, and this factor might contribute to the pathogenesis of the disease. Our study involved 22 healthy (control) and 20 preeclamptic women. Diagnosis of preeclampsia was done according to the recommendation of International Society for the Study of Hypertension in Pregnancy. After blood collection, the next parameters were followed in erythrocytes: concentration of lipid peroxides (TBARS) non-enzymatic components (reduced glutathione - GSH, oxidized glutathione GSSG) and activities of enzymatic components of AOS (glutathione peroxidase GSH-Px, glutathione reductase - GR and glutathione-S-transferase - GST). Results of this study showed that concentration of TBARS and activity of GST were significantly higher, while activity of GR was significantly lower in erythrocytes of preeclamptic, vs. healthy women. Concentrations of GSH, GSSG and activity GSH-Px did not change significantly. Increased concentration of TBARS and activity of GST, as well as disorder in GSH and GSH-dependent enzymes status were indicators of oxidative stress in erythrocytes of preeclamptic women.
AB  - Preeklampsija je sindrom specifičan za trudnoću sa visokim stepenom morbiditeta i mortaliteta fetusa i majke. Klinički se manifestuje posle 20. nedelje gestacije. Kod majke sindrom se manifestuje povišenim krvnim pritiskom i proteinurijom. Brojne savremene studije pokazuju da je disbalans oksidacionog i antioksidacionog sistema značajan faktor u patogenezi preeklampsije. Istraživanjem je obuhvaćeno 22 zdrave i 20 trudnica sa preeklampsijom. Dijagnoza preeklampsije je postavljena prema preporukama International Society for the Study of Hypertension in Pregnancy. U eritrocitima su praćeni sledeći parametri: koncentracija lipidnih peroksida (TBARS), neenzimskih komponenti (redukovani glutation - GSH, oksidovani glutation - GSSG) i aktivnost enzimskih komponenti AOS (glutation-peroksidaza, GSH-Px, glutation-reduktaza, GR i glutation-S-transferaza, GST). Accumulation of cadmium in some tissues of rats after chronic treatment with CdCl2

AB  - Cadmium is the important environmental pollutant and major routes of Cd intake are oral exposure and inhalation. From the total amount of food Cd intake, only about 6% is absorbed in the gastrointestinal tract of animals. The aim of our experiment was to determine the rate of Cd accumulation in liver, kidneys, skeletal muscle (musculus gastrocnemius) and testes of two months old Wistar albino male rats. Animals were divided into 2 experimental groups consisting 7 animals and treated in 30 days courses. The first group of animals functioned as controls (C) and the second group was treated with 200 mg CdCh x 5H20 in drinking water during 30 days (Cd). The average intake of 17 mg Cd/mg b.m. was calculated from the water consumed. Cadmium concentration was determined by atomic absorption spectrophotometry in the mixture of nitric and perchloric acid at the wavelength of 228.8 nm, slit 0.1 nm and lamp current 4 mA in the mixture of air and acetylene. The obtained results show a significant accumulation of Cd in all examined tissues of rats (p<0.005) in respect to controls. These results are in accordance with the findings of other authors and show that Cd mainly accumulated in the tissues of high metabolic rate, such as liver and kidneys. In these organs Cd accumulates mainly in the hepatocytes and cells of the proximal and distal renal tubules. In the cells Cd mainly deposits in the cytoplasm, nuclei and nucleoli where disturbs the cell metabolism, protein synthesis and sulfhydryl homeostasis.

The first group of animals functioned as controls (C) and the second group was treated with 200 mg CdCh x 5H20 in drinking water during 30 days (Cd). The average intake of 17 mg Cd/mg b.m. was calculated from the water consumed. Cadmium concentration was determined by atomic absorption spectrophotometry in the mixture of nitric and perchloric acid at the wavelength of 228.8 nm, slit 0.1 nm and lamp current 4 mA in the mixture of air and acetylene. The obtained results show a significant accumulation of Cd in all examined tissues of rats (p<0.005) in respect to controls. These results are in accordance with the findings of other authors and show that Cd mainly accumulated in the tissues of high metabolic rate, such as liver and kidneys. In these organs Cd accumulates mainly in the hepatocytes and cells of the proximal and distal renal tubules. In the cells Cd mainly deposits in the cytoplasm, nuclei and nucleoli where disturbs the cell metabolism, protein synthesis and sulfhydryl homeostasis.",

AB  - Cadmium (Cd) is an ubiquitous toxic metal that may induce oxidative damage by disturbing the prooxidant-antioxidant balance in the tissues. Among antioxidant micronutrients, selenium (Se) is an essential dietary trace element which ·plays an important role in a number of biological processess. The effect of cadmium on the activity of glutathione peroxidase (GSH-Px) and glutathione S-transferase (GST) which play an important role in the detoxification of xenobiotics, was studied in the liver, kidney and heart of rats. Furthermore, the role of selenium in the modulation of Cd toxicity with respect to GSH-Px and GST was also evaluated. AB  - Effect of thyroxine on antioxidant enzyme activities and GSH content in brain (cortex and rest of the brain) of different maturated rats were studied. Male Mill Hill hybrid hooded rats aged 15, 45 and 75 days were treated with L-thyroxine, T4 (40 μg/100 g body mass), s.c, one dose per day, 14 days (finally aged 30, 60 and 90 days, respectively). Effect of T4 on antioxidant defense (AD) enzyme activities and GSH content in brain differs with respect to age. T4 treatment decrease activities of Mn SOD and CuZn SOD in the rest of the brain of 60 days old rats in comparison to controls and CAT activity in cortex of 30 and 60 day old rats. GSH-Px activity in cortex of 90 days old rats was decreased in T4 treated group in respect to controls, while activity remains unchanged in 30 and 60 day old rats. The activity of GSH-Px in the rest of the brain was unaffected by the treatment in all groups. GSH content in cortex of animals of 30 and 60 days old rats were higher in comparison with corresponding controls, while in 90 days old rats effects were opposite. L- thyroxine treatment significantly decrease GSH content in rest of the brain of 30 days old rats in respect with corresponding controls, while in 60 and 90 days old animals such changes were not detected.
AB  - U ovom radu ispitivan je efekat tiroksina na aktivnost antioksidacionih zaštitnih enzima i količinu glutatationa (GSH) u mozgu (moždana kora i ostatak mozga) pacova različite starosti. Mužjaci pacova soja Mili Hill hybrid hooded starosti 15, 45 and 75 dana tretirani su sa L-tiroksinom, T4 (40 μg/100 g telesne mase), s.c, jedna doza dnevno, 14 dana (do finalne starosti 30, 60 i 90 dana). Efekat T4 na aktivnost antioksidacionih zaštitnih enzima i količinu GSH u mozgu razlikuje se sa starošću pacova. Tretman sa T4 smanjuje aktivnost Mn SOD i CuZn SOD u ostatku mozga pacova starih 60 dana u poređenju sa kontrolama i aktivnost CAT u moždanoj kori pacova starih 30 i 60 dana. Male Mill Hill hybrid hooded rats aged 15, 45 and 75 days were treated with L-thyroxine, T4 (40 mg/100 g body mass), s.c., one dose per day, 14 days (finally aged 30, 60 and 90 days, respectively). Effect of T4 on GSH-dependent antioxidant enzyme activities in the IBAT differs with respect to age. T4 treatment gradually decrease activities of all GSH-dependent antioxidant enzymes in 60 and 90 days old rats in comparison to young ones. GSH content in animals of 30 and 60 days old rats are lower in comparison with 90 days old rats, but the effects are oppposite. L-thyroxine treatment significantly increase GSH content in 30 days old rats (p<0.001) in respect with coresponding controls, while decrease in 60 and 90 days old animals were detected (p<0.01). Different response of non-mature rats to thyroxine comparing to older rats could be attributed to the difference in thyroxine metabolism and developmental phase of regulatory physiological systems maturation including antioxidative.

AB  - In our study we investigated the effects of exogenous cadmium (Cd,  17 mg /day /kg  b.m.  in drinking  water), coenzyme  Q10 (CoQ 10, 16 mg/kg/dose CoQ10 dissolved in  olive  oil,  i.m.,  every  fifth  day) and Cd+CoQ10 (i n above mentioned amou nt s)  on  antioxidant  de­ fense enzyme activities (Total SOD, Mn SOD, CuZn SOD, CAT, GSH-Px, GST and  GR)  in  interscapular  hrown  adipose  tis sue (IBAT) of  male two  months  old  Wistar albin o rats during  30 days.
Cd induces significantly increased Mn SOD activity, while concomitant treatment of animals with  Cd+CoQ10  reversed  this change.   The  activity   of  CuZn   SOD   was  significantly  decreased
both in Cd and Cd+CoQ10 treated animals. CAT activity was sig­ nificantly increased in rats treated with Cd, whereas Cd+CoQ 10 normalized the activity of this enzyme. The activity of GSH-Px was significantly increased in all investigated groups of animals. Cd in­ duces an increased activity of GST,  but  by concomitant  treatment of rats with Cd+CoQ10 the GST activity was retained . The activity of GR was significantly increased in Cd treated animals, while in rats treated with Cd+CoQ10  was signi fican tl y decre_ased.
It can be concluded that Cd  induces  oxidativ  e stress  and  a l ter the activities of some antioxidant  defense  enzymes  in  IBAT of  rats. It is also shown that CoQ 10 can  normal ized  Mn  SOD,  CAT  a nd GST  activities  after  Cd-induced  changes .

AB  - The brain is widely responsive to gonadal hormones. The functional significance of ovarian hormones in the brain is evident from biochemical studies indicating that estradiol or progesterone treatment of testectomized rats produces changes of antioxidant enzyme activities. The effect of estradiol benzoate (EB) and progesterone (P) in the control of antioxidant (AO) enzyme activities was studied in the brain of adult male Wistar rats. The activities of catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) and glutathione reductase (GR) were measured in appropriate subcellular fractions, prepared from brains of animals belonging to various experimental groups. These groups were designed with the intention to follow changes in enzyme activities 2 h or 24 h after systemic administration of 5 mug EB or 2 mg P to testectomized (TX) animals. The obtained results show that both EB and P increase CAT activity, whereas EB decreases GSH-Px, GST and GR activities. These findings clearly show the modulatory role of EB and P in the control of enzymes responsible for the protection of rat nerve cells against oxidative damage caused by free oxygen radicals.

AB  - The effects of acute exposure to cadmium (Cd) on the blood antioxidant defense system, lipid peroxide concentration and hematological parameters, as well as the possible protective role of vitamin E were studied. Male Wistar albino rats (3 months old) were treated with cadmium (0.4 mg Cd/kg b.m., i.p., 24 h before the experiment) or with vitamin E+Cd (20 IU Vit E/kg b.m., i.m., 48 h + 0.4 mg Cd/kg b.m., i.p., 24 h before the experiment). The hematological parameters were assessed: red blood cell counts, hematocrit value and hemoglobin concentration were significantly decreased in the blood of Cd-treated rats. Intoxication with cadmium was also followed by significantly increased lipid peroxide concentrations. We also observed increased activity of antioxidant defense enzymes: copper zinc containing superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase and glutathione-S-transferase as well as concentrations of non-enzymatic components of antioxidant defense system: reduced glutathione, vitamin C and vitamin E. Pretreatment with vitamin E exhibited a protective role on the toxic effects of cadmium on the hematological values, lipid peroxide concentration as well as on enzymatic and non-enzymatic components of antioxidant defense system.

CdCb in drink water during 15 days (Cd), (2) Vit E+CoQ 1o+Cd (pretreated with 20 IU/kg/dose of Vit E and mg/kg/dose  of  CoQ10  i.m., every  fifth day during  15 days  and  then  treated  with  14   mg/day b.m. of Cd during next 15 days Third group (3) was control (C). After the treatment the rats w sacrificed by decapitation and blood samples were collected and analyzed. In the blood of rats conce ntration of lipide peroxides (LP) were evaluated, while the activity  of  gluta tione transferase was assayed in the plasma after separation from the    blood cells.
The results obtained in our experiments show that acute treatment with Cd leads to significantly increased LP concentration (p<0.01) in the blood of rats in respect to the con animals. Pretreatment  with  Vit  E+CoQ 10  pryor  to  Cd exposure  normalyzed  the concentration LP   in   the· blood.   Contrary   to   that,   in   chronic   Cd   intoxication   concentration   of LP significantly   decreased   (p<0.005)   in   comparison   to   the   control   value.   Pretreatment   Vit E+CoQ 10 pryor to Cd intoxication influences an additional decrease of LP concentra (p<0.005) compared with controls. It appears that in chronic Cd treatment the period of 15 day sufficient for the development of efficient red blood cell protection against Cd (per)oxida  damage. The activity of GST in the plasma of rats acutely treated with Cd was  signi fica  increased in respect to the controls (p<0.02). Pretreatment of rats with Vit E+CoQ 10 influenc significant decrease of GST activity in the plasma (p<0.05).  Similar  results  were  obtained chronic Cd intoxication where Cd induces a significantly higher activity of GST in the pla (p<0.01), while pretreatment  with Vit E+CoQ 10 reversed this value to the control   level.
From  the   presented   results,  it  can  be  concluded   that  Cd   both   in  acute  and  chr

AB  - Cadmium (Cd) is an important pollutant of environment which accwnulates in the liver and the kidneys, as well as in other tissues. Cd induces oxidative damage causing many metabolic, histological and pathological changes, such as nephrotoxicity, cardiotoxicity, increased lipid peroxidation (LP) and alteration of the antioxidant defense system (AOS). Recent studies have shown that coenzyme Q10 (CoQ10) and vitamin E (Vit E) are the powerful liposoluble antioxidants, which inhibit lipid peroxidation and thus prevent free radical med.iated oxidative injuries. It is well known that CoQ10 and Vit E may act synergistically as antioxidants. The aim of this study was to investigate a possible protective influence of CoQ10 and Vit E in Cd induced oxidative stress in the rat hearts.

Njihovo dosledno postovanje je neophodno ne  samo zbog zadovoljenja kvaliteta hrane koja se distribuira na domacem tzistu vec je i preduslov nase uspesne izvome orijemacije i konkurenmosti na inostranom trzistu prehrambenih proizvoda. Istrazivanja oksidacione stabilnosti hrane koja se sprovode u mnogim istrazivackim laboratorijama u svetu namecu potrebu uvodjenja novih kriterijuma i standarda koji bi obuhvatili i odredjivanje antioksidacionih komponenata u hrani. Rezultati dobijeni u nasim istrazivanjima pokazuju da naknadnim dodavanjem pojedinih komponenata antioksidacionog sistema (selen) moze znacajno da se poveca ukupni nivo ostalih antioksidanata u tkivima eksperimentalnih zivotinja.

AB  - Rapid  urbanization  and  eutrophization,  disturbing  normal  ecological equilibrium of water ecosystems and directly endangering living organisms is the main problem in Boka Kotorska bay. Mediterranean mussel (Mytilus galloprovincialis, Bivalvia, Mollusca) is a species which naturally inhabits the waters of Boka Kotorska bay, but it is artificially bred and increasingly used in human diet. We established that the presence of heavy metals in sea water even in allowed concentrations caused increased accumulation in tissues of mussel (Cu, Mn, Cr, Ni, Al, Mg, Zn, Cd, Pb) pointing to a numerous negative consequences not only in those organisms but in humans, potential consumers of mussels as well.

AB  - Coenzyme Q (CoQ) is present in almost all living organisms where it exhibits antioxidant properties and also represents an obligatory component of the mitochondrial respiratory chain (2). CoQ may prevent lipide peroxidation, protects DNA from oxidation and protects organism from oxidative stress induced by various toxic agents (1). The scope of our study was to investigate the concentrations of  ascorbic acid (AsA) and vitamin  E (Vit E) in the liver, kidneys, heart and testes,   as well as CoQ concentration in the liver, kidneys and testes of male, two months old Wistar albino rats treated with 20 mg/kg of coenzyme Q10 (CoQ10) dissolved in olive oil, i.m., every fifth  day, during 30 days. An average intake of CoQ10 was 16 mg/kg/dose. All  obtained  results  were  compared with control animals  (C).
Our results show that treatment with CoQ10 induced a significant increase of AsA and Vit E concentrations in all investigated tissues. The concentration  of  CoQ  was  significantly  increased only in the liver of rats treated with CoQ10. It is well known that CoQ10 in  its  reduced  form (CoQ10H2) is the powerful antioxidant which regenerate the active form of Vit E from the Vit E radicals (tocopheroxyl radicals, Vit E"), (4), stabilized extracellular ascorbate by its NADPH-CoQ10 reductase (3) and elevate the endogenous CoQ pool in the cells (1). At the other hand, dose and time dependent studies were shown that CoQ concentration after parenteral administration was increased only in the plasma and liver ofrats  (5).It can be concluded that all obtained  effects induced by CoQ10  led to the elevated concentrations  of non-enzymatic components of antioxidant defense system and increased antioxidant protection of all examined tissues.

Protective role of vitamin E and coenzyme Q10 on antioxidant defense system in the blood of rats treated with cadmium

AB  - The aim of our study was to investigate the activities of copper zinc containing superoxide dismutase (CuZn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) and glutathione reductase (GR), as well as ascorbic acid (AsA), vitamin E (Vit E) and lipide peroxide (LP) concentrations in the blood of male two months old Wistar albino rats. The animals were divided as follows: (1) untreated controls (C); (2) animals treated with 14 mg/day/kg b.m. cadmium in drinking water during 15 days (Cd); (3) rats treated with 20 IU/kg/dose of Vit E and 15 mg/kg/dose of coenzyme Q10 (CoQ10), i.m., every fifth day during 15 days (Vit E+CoQ10) and (4) animals pretreated with Vit E and CoQ10 in above mentioned amounts during 15 days and then treated with 14 mg/day/kg b.m. of Cd during next 15 days (Cd+Vit E+CoQ10). Each group consisted of seven animals.               All obtained results were compared in respect to the control animals.
   Cadmium induced a significant increase of GST activity, as well as Vit E concentration in the plasma. In Vit E+CoQ10 treated animals the activities of GSH-Px and GR in erythrocytes were significantly decreased, whereas plasma AsA and Vit E concentrations were markedly elevated.          In Cd+Vit E+CoQ10 treated rats erythrocyte GSH-Px and GR activities, as well as plasma Vit E concentration were significantly increased, while plasma AsA concentration was significantly decreased. In all examined groups of animals LP concentration in the whole blood was significantly decreased. The obtained results show that Cd in the dosages administered influenced antioxidant defense system in the blood of rats and the protective role of Vit E and CoQ10 against Cd toxicity was also observed.

AB  - The aim of our study was to investigate the effects of coenzyme Q10 (CoQ10) administration on the  activities  of  superoxide  dismutases: total  (Tot  SOD),  copper  zinc  containing  (CuZn  SOD) and manganese containing (Mn SOD), catalase (CAT), glutathione peroxidase (GSH-Px,) glutathione-S-transferase (GST) and glutathione reductase (GR) in liver, kidneys, heart and testes of male two months old Wistar albino rats. The animals were divided in two experimental groups: (1) control rats (C) and (2) rats treated with 20 mg/kg/dose of coenzyme Q10 by i.m. injections, every fifth day during 30 days  (CoQ10).
An average intake of CoQ10 was 16 mg/kg/dose. All obtained results were compared in respect to the control animals.
The obtained data shows that Tot SOD activity was significantly increased in the heart  of CoQ10 treated animals. At the same time, the activity of CuZn SOD was also increased in the heart, as well as in testes of CoQ10 treated rats. Mn SOD activity was markedly decreased in kidneys and testes, while in heart was significantly increased. The activity of CAT was significantly decreased in liver, kidneys and heart, whereas GSH-Px activity was decreased only in the kidneys of CoQ10 treated animals. At the same time, GSH-Px activity was markedly elevated in heart and testes. Finally, the activity of GR was significantly decreased in the heart of animals received CoQ10.
The obtained results show that CoQ1O in the dosage administered influenced antioxidant enzyme activities in examined tissues of rats. It is also shown that various tissues exhibit different response to CoQ10 depending of their metabolic activity.

AB  - We investigated the activity of antioxidant enzymes: superoxide dismutases - total (Tot SOD), copper zinc containing (CuZn SOD) and manganese containing (Mn SOD), catalase (CAT), glutathione peroxidase (GSH-PX) and glutathione-S-transferase (GST) in the liver, kidneys, heart and interscapular brown adipose tissue (IBAT), as well as contents of ascorbic acid (AsA) and vitamin E (Vit E) in the liver and kidneys of male two months old Wistar albino rats. The animals were divided in two experimental groups: control animals (C) and animals treated with selenium (Se). The rats treated with selenium were drank water _c ontaining 0.1 µg/mL selenium (Se) as Na-selenite during 30 days. The average daily intake of 7 µg Se/kg body mass was calculated from the water consumed. Both experimental groups consisted of seven animals. The obtained results were compared in respect to the control animals. .
Our results show that Tot SOD and CuZn SOD activities were significantly increased in the liver (p<0.02), kidneys (p<0.005), heart (p<0.005) and IBAT (p<0.05) of Se treated animals, while the activity of Mn SOD was significantly increased only in the liver (p<0.02). CAT-activitywas statistically increased in the liver (p<0.02) of animals treated with Se, while the GSH-Px activity was significantly increased only in the heart (p<0.02). The activity of GST was significantly increased in  the liver (p<0.02), kidneys (p<0.02) and heart (p<0.005) of Se treated  animals.
The obtained results show that AsA content in the liver and kidneys was significantly increased (p<0.02 and p<0.05, respectively). The content of Vit E was also significantly increased in the liver (p<0.005) and kidneys (p<0.005) of animals exposed to Se.
Presented results show that oral intake of Se as Na-selenite, improves antioxidant defense system by increasing antioxidant enzyme activities, as well as, by elevation of AsA and Vit E contents in the all examined tissues of rats.
