AB  - Thioacetamide (TAA) is widely used to study liver toxicity accompanied by oxidative stress, inflammation, cell necrosis, fibrosis, cholestasis, and hepatocellular carcinoma. As an efficient free radical’s scavenger, C60 fullerene is considered a potential liver-protective agent in chemically-induced liver injury. In the present work, we examined the hepatoprotective effects of two C60 doses dissolved in virgin olive oil against TAA-induced hepatotoxicity in rats. We showed that TAA-induced increase in liver oxidative stress, judged by the changes in the activities of SOD, CAT, GPx, GR, GST, the content of GSH and 4-HNE, and expression of HO-1, MnSOD, and CuZnSOD, was more effectively ameliorated with a lower C60 dose. Improvement in liver antioxidative status caused by C60 was accompanied by a decrease in liver HMGB1 expression and an increase in nuclear Nrf2/NF-κB p65 ratio, suggesting a reduction in inflammation, necrosis and fibrosis. These results were in accordance with liver histology analysis, liver comet assay, and changes in serum levels of ALT, AST, and AP. The changes observed in gut microbiome support detrimental effects of TAA and hepatoprotective effects of low C60 dose. Less protective effects of a higher C60 dose could be a consequence of its enhanced aggregation and related pro-oxidant role.

AB  - We investigated the biochemical and ultrastructural changes in the liver of the freshwater fish, European perch (Perca fluviatilis), in response to Aphanizomenon flos-aquae bloom in the Gruža Reservoir, Serbia. The activities of total manganese- and copper zinc-containing superoxide dismutase (Tot SOD, Mn-SOD, Cu/Zn-SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and biotransformation phase II enzyme glutathione-S-transferase (GST), as well as concentrations of total glutathione (GSH) and sulfhydryl (-SH) groups were examined before and during the bloom period. Mn-SOD activity was significantly higher, while the activities of Cu/Zn-SOD, CAT and GSH-Px and the concentration of the -SH groups were significantly lower during the bloom. The ultrastructure of the liver revealed necrotic and apoptotic damage to the hepatocytes during the bloom period. Our work represents the first study to report the influences of an Aphanizomenon flos-aquae bloom in the Gruža Reservoir on antioxidant biomarkers and on histopathological alterations in the liver of the freshwater fish European perch (Perca fluviatilis).

AB  - U nаšoj studiji ispitivali smo koncentraciju vitamina E i SH grupa u digestivnoj
žlezdi i škrgаmа slаtkovodne školjke Unio tumidus nа četiri lokаlitetа (Jаmenа, Sremskа
Mitrovicа, Šаbаc i Ostružnicа) nа reci Sаvi.
Kod slatkovodne školjke U. tumidus koncentracija vitamina E bila je statistički
značajno veća na lokalitetu Šabac i u digestivnoj žlezdi i škrgama u odnosu na druge
ispitivane lokalitete. Koncentracija SH grupa kod slаtkovodne školjke U. tumidus bila je
statistički značajno veća u digestivnoj žlezdi u poređenju sa škrgama na sva četiri
lokaliteta, a najveće vrednosti koncentracije SH grupa su zabeležene na lokalitetima
Šabac i Ostružnica. Dobijeni rezultаti u nаšoj studiji pokаzuju veliku specifičnost u
odnosu nа tkivo i lokаlitet, ukаzujući nа rаzličite metаboličke аktivnosti i rаznovrstаn
uticаj sredine. Naši podaci ukаzuju dа vаrijаcije u koncentraciji vitamina E i SH grupa
kod U. tumidus, takođe mogu biti korišćene zа molekularno fiziološki biomonitoring
životne sredine.
Nаša studija predstаvljа prvi obimаn izveštаj o koncentraciju vitamina E i SH
grupa kаo relevаntnih biohemijskih biomаrkera oksidаcionog stresа kod slаtkovodne
školjke U. tumidus nа rаzličitim ispitivanim lokаlitetimа nа reci Sаvi i obezbeđuje
osnovu zа budućа proučаvаnjа kojа će uzeti u obzir nаše pаrаmetre kаo potencijаlne
biohemijske biomаrkere zа molekularno fiziološki biomonitoring osnovnih sredinskih
uslovа i nekih аntropogenih uticаjа.

AB  - U našoj studiji određivali smo sezonski zavisnu promenu u koncentraciji
vitamina E i SH grupa u škrgama, bubregu i crevu rečne mrene (Barbus barbus) iz
Dunava, koja predstavlja bentosnu ribu široke geografske distribucije. Istraživanja su
izvedena u prigradskom području Beograda, između Višnjice i Grocke, u prolećnoj i
letnjoj sezoni. Koncentracije ispitivanih parametara merene su standardnim
spektrofotometrijskim metodama.
Rezultati dobijeni za koncentraciju SH grupa u u škrgama, bubregu i crevu
mrene pokazuju statistički značajno povećanje u letnjoj sezoni u poređenju sa prolećnom
sezonom. Koncentracija vitamina E pokazuje statistički značajno smanjenje u letnjoj
sezoni u odnosu na proleće i ovaj trend zabeležen je u svim ispitivanim tkivima. Naša
studija predstavlja prvu procenu koncentracije vitamina E i SH grupa u škrgama,
bubregu i crevu rečne mrene u srpskom delu Dunava.
Zaključuje se da je sezonska fluktuacija ispitivanih komponenti sistema zaštite
od oksidacionih oštećenja u korelaciji sa sezonskim temperaturnim varijacijama.

AB  - U našoj studiji istraživali smo promene aktivnosti enzima zaštite od oksidacionih oštećenja: ukupne superoksid-dismutaze (Uk SOD), bakar, cink sadržavajuće superoksid-dismutaze (CuZnSOD), mangan sadržavajuće superoksiddismutaze (MnSOD) i katalaze (CAT) u škrgama i mišiću slatkovodne ribe grgeča (Perca fluviatilis) kao odgovor na cvetanje cijanobakterije Aphanizomenon flos-aquae u Gružanskom jezeru. Takođe, ispitivana je koncentracija ukupnih proteina, kao i elektroforetska analiza proteina i SOD. Svi parametri određivani su tokom dva perioda, pre i za vreme cvetanja. Aktivnosti Uk SOD i MnSOD bile su niže i u škrgama i u mišiću, dok je aktivnost CuZnSOD pokazala niže vrednosti u mišiću za vreme cvetanja u odnosu na period pre cvetanja. Aktivnost CAT bila je značajno veća u oba ispitivana tkiva tokom cvetanja. U mišiću je zabeleženo statistički značajno povećanje koncentracije ukupnih proteina za vreme cvetanja, dok je elektroforetska analiza proteina ukazala na razlike u profilu proteina u škrgama i mišiću između dva ispitivana perioda. Elektroforetski profil enzima SOD pokazao je tipičan distribucioni obrazac SOD izoformi u oba tkiva. Naš rad predstavlja prvu studiju o uticaju cvetanja cijanobakterija na aktivnost enzima zaštite od oksidacionih oštećenja superoksid-dismutaze i katalaze u škrgama i mišiću slatkovodne ribe grgeča (Perca fluviatilis) iz Gružanskog jezera i ukazuje da cvetanje Aphanizomenon flos-aquae indukuje oksidacioni stres u ispitivanim tkivima. Ovo istraživanje daje osnovu za molekularno fiziološki biomonitoring uticaja cijanobakterija na različite slatkovodne organizme.

AB  - Data are presented in this study on the tissue glutathione (GSH) content and on the activity on the main enzymes involved in GSH metabolism ( glutathione peroxidase - GSH-Px, glutathione reductase - GR and phase II biotransfor­ mation enzyme glutathione-S-transferase - GST) in the whole body of fresh­ water snail, Viviparus acerosus of two different age groups - young and adults from the Danube River. The obtained results show, that there was no differences in content of GSH in both investigating groups of snails. We observed that the activity of GR was significantly lower in adults in comparison to young groups. The activity of GST was markedly higher in adults in respect to the young snails. Moreover, GSH-Px activity did not change in different age groups, this possible indicating no variation in organic peroxide production and/or metabolism during ageing process. Ageing seems to affect the activity of GSH related enzymes and, in particular, the activities of GR and phase II biotransformation enzyme GST. Our work represents the first study of its kind and these data indicate that, in age­ ing freshwater snails, Viviparus acerosus from the Danube river the increased susceptibility to oxidative stress is partly related in alteration in glutathione me­ tabolism.

AB  - We investigated alterations of antioxidant defence system (ADS) in the liver of freshwater fish, European perch (Perea fluviatilis) as response to toxic Apha­ nizomenon flos-aquae bloom in the Gruza Reservoir. Activities of total, manga­ nese, copper zinc containing superoxide dismutase (Tot SOD, Mn SOD, CuZn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR), phase II biotransformation enzyme glutathione S-transferase (GST), con­ centrations of total glutathione (GSH) and sulphydryl (-SH) groups were mea­ sured before and during bloom period. Histopathological examinations were studied using electron microscopy. Activities of CuZn SOD, CAT and GSH-Px and concentration of -SH groups were significantly lower, while Mn SOD activ­ ity was significantly higher during bloom period. No differences were observed for Tot SOD, GR and GST activities, as well as for GSH concentration. Electron microscopy showed alterations in hepatocytes, which results in damage of cells. Our work represents the first study of its kind and suggest on influence of toxic Aphanizomenon flos-aquae bloom in the Gruza Reservoir on ADS and on histo­ pathological changes in the liver of Perea fiuviatilis indicating on necrotic and apopototic damages of hepatocytes.

AB  - In the present study we have used the activity of aantioxidative defence enzymes: superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and the activity of glutathione S-transferase (GST), an enzyme of the phase II biotransformation process to evaluate their utility as biological toll for pollution monitoring in the Sava River. We also determined total protein content, as well as protein and SOD electrophoretic profiles in the digestive gland and the gills of the freshwater bivalve anodonta woodiana. Obtained results indicate a significant influence of tissue specifity of the all investigated enzyme activities. The activities of SOD, CAT, GSH-Px and phase II biotransformation enzyme GST were significantly higher in the digestive gland in respect to the gills, whereas GR activity exhibited higher levels in the gills. This was most likely a reflection of different metabolic activities and different responses to environmental conditions of the examined tissues. Our study also suggest, that the variation of SOD expression pattern in Anodonta woodiana could be used as a tool for the environmental monitoring. Our wo bivalve Anodonta woodiana friom the Sava Riverrk represents a first study of this kind and showed that the antioxidant defence enzymes can be considered as a potential biomarkers of the freshwater

abstract = "In the present study we have used the activity of aantioxidative defence enzymes: superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and the activity of glutathione S-transferase (GST), an enzyme of the phase II biotransformation process to evaluate their utility as biological toll for pollution monitoring in the Sava River. We also determined total protein content, as well as protein and SOD electrophoretic profiles in the digestive gland and the gills of the freshwater bivalve anodonta woodiana. Obtained results indicate a significant influence of tissue specifity of the all investigated enzyme activities. The activities of SOD, CAT, GSH-Px and phase II biotransformation enzyme GST were significantly higher in the digestive gland in respect to the gills, whereas GR activity exhibited higher levels in the gills. This was most likely a reflection of different metabolic activities and different responses to environmental conditions of the examined tissues. Our study also suggest, that the variation of SOD expression pattern in Anodonta woodiana could be used as a tool for the environmental monitoring. Our wo bivalve Anodonta woodiana friom the Sava Riverrk represents a first study of this kind and showed that the antioxidant defence enzymes can be considered as a potential biomarkers of the freshwater",

Specific GSH-Px activity was lower in the hepatopancreas of AUS than in ORC and in the muscle of AST than in AUS and ORC. We obtained differences of total GSH-Px activity in the muscle between AST and AUS and between AST and ORC. Specific and total GR activity was higher in the hepatopancreas of AST than in other species. Specific and total activity of GR in the gills and in the muscle of crayfish species was higher in AUS then in AST and ORC. The specific and total GST activity was lower in the hepatopancreas of AST and ORC than in AUS. Specific and total GST activity in the gills was higher in AUS than in AST. The lowest specific and total GST activity was detected in the muscle of ORC compared to the AST and AUS. Our results show that specific and total enzyme activities of GSH-Px, GR and GST have species and tissue specifics between crayfish species and represent biomarkers of environmental influences on these organisms.

Specifična i totalna aktivnost GR je veća u hepatopankreasu kod AST nego kod druge dve vrste, dok je u škrgama i mišiću rakova veća kod AUS nego kod AST i ORC. Specifična i totalna aktivnost GST je manja u hepatopankreasu kod AST i ORC u poređenju sa AUS, a u škrgama je veća kod AUS nego kod AST. Najmanja specifična i totalna GST aktivnost je u mišiću kod ORC u poređenju sa AST i AUS. Specifična i ukupna aktivnost GSH-Px, GR i GST pokazuje veliku specifičnost za vrstu i tkivo između ispitivanih vrsta rakova i predstavlja biomarker uticaja sredine na ove organizme.",
