AB  - Metallothioneins (MTs) constitute a family of low-molecular  weight, cysteine­ rich and heat stable 
proteins involved in the binding and regulation of essential metals, such as copper and zinc, and 
in the detoxification of these and other non-essential metals, such as cadmium and mercury. 
Previously we showed that the induction of MTs in Merluccius merluccius and Mui/us barbatus 
correlated with elevated concentrations of Cu and Pb, determined by chemical analysis of the 
seawater from Bar and Valdanos. In the present study we studied protein expression of MTs in the 
hepatopancreas of several seawater fish species: Red mullet (Mui/us barbatus), European hake 
(Merluccius merluccius), Tub gurnard (Trig/a lucerna) and Thinlip mullet  (Liza ramada), from the 
Monetengrin Adriatic coastline. Western blot analysis revealed the highest induction of MTs in the 
hepatopancreas in the following order: Merluccius merluccius, Trig/a lucerna, Mui/us barbatus and 
Liza ramada. Considering that MTs play a role in the metabolism of essential metals, they  are 
constitutively expressed. The presence of elevated concentrations of both essential and toxic 
metals provokes the induction of MTs. These results are in correlation with literature data showing 
that fish species differ in their detoxification capacities and amounts of accumulated  metals.  We 
 conclude that the level of induction of MTs in the hepatopancreas of the examined fish species 
correlates with the level of their reliability as bioindicator species in heavy metal 
biomonitoring.

AB  - Haptoglobin is a constitutively expressed protein which is predominantly synthesized in the liver. During the acute-phase (AP) response haptoglobin is upregulated along with other AP proteins. Its upregulation during the AP response is mediated by cis-trans interactions between the hormone-responsive element (HRE) residing in the haptoglobin gene and inducible transcription factors STAT3 and C/EBP beta. In male rats that have been subjected to chronic 50% dietary restriction (DR), the basal haptoglobin serum level is decreased. The aim of this study was to characterize the trans-acting factor(s) responsible for the reduction of haptoglobin expression in male rats subjected to 50% DR for 6 weeks. Protein-DNA interactions between C/EBP and STAT families of transcription factors and the HRE region of the haptoglobin gene were examined in livers of male rats subjected to DR, as well as during the AP response that was induced by turpentine administration. In DR rats, we observed associations between the HRE and C/EBPalpha/beta, STAT5b and NF-kappaB p50, and the absence of interactions between STAT3 and NF-kB p65. Subsequent induction of the AP response in DR rats by turpentine administration elicited a normal, almost 2-fold increase in the serum haptoglobin level that was accompanied by HRE-binding of C/EBPbeta, STAT3/5b and NF-kB p65/p50, and the establishment of interaction between STAT3 and NF-kappaB p65. These results suggest that STAT3 and NF-kappaB p65 crosstalk plays a central role while C/EBPbeta acquires an accessory role in establishing the level of haptoglobin gene expression in male rats exposed to DR and AP stimuli.

AB  - The enzyme CYP1A is an established biomarker of fish exposure to polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs). The metallothioneins (MT), a family of Cys-rich proteins, bind a wide range of metals and participate in their metabolism. The aim of the study was to examine the correlation between CYP1A and MT expression in commercially important fish species Mullus barbatus and Merluccius merluccius and contaminants (PAHs, PCBs, toxic metals) in seawater and sediment from three localities with different level of contamination in the Adriatic Sea in winter, i.e., Platamuni, Valdanos and the port of Bar. The relative concentration of CYP1A was the highest in both fish species from Bar. Increased concentrations of PCBs in the seawater were observed only in Bar. A species-specific higher increase in the protein concentration of CYP1A was observed in Mullus barbatus compared to Merluccius merluccius. The levels of MT were the highest in Merluccius merluccius from Bar and in Mullus barbatus from Valdanos. The induction of MT correlated with the elevated concentrations of Cu and Pb determined by chemical analysis of the seawater from Bar and Valdanos, respectively. According to the chemical analysis of the seawater and the biological response of the fish, the Platamuni locality exhibited the lowest level of contamination.
AB  - CYP1A predstavlja dobro okarakterisan biomarker kod riba pri izlaganju policikličnim aromatičnim ugljovodonicima (PAH) i policikličnim bifeholima (PCB). Metalotioneini (MT) pretstavljaju familiju proteina koji vezuju metale i učestvuju u njihovoj metabolizmu, transportu i regulaciji. Cilj ovog rada je bio da se ispitaju korelacije između promena u nivou CYP1A i MT u hepatopankreasu dve komercijalno važne ribe: Mullus barbatus (trlja) i Merluccius merluccius (oslić) i kontaminanatata: PAH, PCB i toksičnih metala) na tri lokaliteta u Jadrnskom moru (Platamuni, Valdanos i luka Bar) u zimu. CYP1A je u najvećem stepenu indukovan na lokalitetu Bar u obe ispitivane vrste, što je u korelaciji sa prisustvom povećane količine PCB u morskoj vodi u Baru. Nivo MT je najveći kod oslića izlovljenog u Baru, a kod trlje u Valdanosu. To je u korelaciji sa izmerenim povećanim koncentracijama bakra u Baru, a olova u Valdanosu. Na osnovu izučavanih parametara, Platamuni su lokalitet sa najmanjim stepenom kontaminacije.

AB  - CYP1A is an established biomarker of fish exposure to PAHs and PCBs. The metallothioneins (MT)constitute a family of Cys-rich proteins that bind a wide range of metals and participate in their uptake,transport and regulation. The aim of this work was to detect changes in CYP1A and MT protein levelsin the hepatopancreas of two commercially important fish species: Mullus barbatus and Merlucciusmerluccius from three different types of locality in the Adriatic Sea (the open sea area of Platamuni,the port of Bar and Valdanos,) in winter. The relative concentration of CYP1A was highest in bothfish species from Bar. Chemical analysis of sea water did not reveal the presence of PAHs. Increasedconcentrations of PCBs were observed only in Bar. A species-specific and higher increase in the proteinconcentration of CYP1A was observed in Mullus barbatus compared to Merluccius merluccius. Thelevels of MT were highest in Merluccius merluccius from Bar and in Mullus barbatus from Valdanos.Induction of MT correlated with elevated concentrations of Pb and Cu, determined by chemicalanalysis of sea water from Valdanos and Bar, respectively. According to the examined parameters, thePlatamuni locality exhibited the lowest level of contamination.

AB  - The organophosphorus compounds soman and paraoxon induce the acute-phase (AP) response. All phases of the AP response, from macrophage activation and stimulation of glucocorticoid secretion to AP protein expression appear to be under the control of similar molecular mechanisms to those during the turpentine-induced AP response. The AP protein content in the circulation 24 h after either soman, paraoxon or turpentine administration was injury-specific. Both soman and paraoxon poisoning were characterized by significantly increased synthesis of alpha(1)-acid glycoprotein (AGP) that displayed an immunomodulatory effect in vitro. This result suggests that after organophosphate poisoning AGP participates in vivo in a negative feedback mechanism that prevents over-activity of the immune system.

AB  - During  the  acute-phase  (AP)  response,  the synthesis  rate  and serum  concentration of
the AP protein a 2-macroglobulin (MG) increases in the rat. MG gene transcription  is
mainly  controlled  by the inflammatory  cytokine  IL-6 and  related  trans- factors,   the
Signal  Transducers  and  Activators  of  Transcription  (STAT)  and  CCAAT/Enhancer
Binding   Protein   (C/EBP) .  Nearly   every   aspect   of   the  body's  defense  system· is changed  in  malnutrition  (MN). We examined  whether  MN  affected  rat  MG  synthesis
and  STAT3  and  C/EBP  expression.  MN  was  induced  in  male  Wistar  rats .by 50%
food  restriction  during  six  weeks. The  AP  response was induced  by  turpentine oil injection (1 µl/g body weight). Using "rocket"  immuno electrophoresis we observed that,  compared   to   the  well-nourished   (WN)   control,  MN   was  associated  wit increased serum  MG. The  AP  response  promoted  a further rise  in  se rum  MG. The STAT3  and  C/EBP	levels  in  the  nuclear  extract  were  examined  by  immunoblote	analysis.  In  rats  with   MN,  STAT3  was  elevated  and  during  the   AP  response, increased  STAT3 levels  were observed  in both  WN and MN  rats. The C/EBP	levels were similar  in  MN  and  WN  rats  however  the  AP response  induced its significants, increase in rats with MN. Obtained results suggested that malnutrition contribute  to an  increased MG expression by  influencing  IL-6  signal  transduction  pathway.
Malnutrition-related increased of MG synthesis could be connected with positive regulatory role of STAT3 , while the additional AP-related MG increased synthesis in malnourished  rats require  activit y of C/EBP .

AB  - The cytochrome P450 isozymes that are responsible for the biotransformation of xenobiotic compounds such as polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs), are represented by the CYP1A subfamily of proteins in fish. CYP1A is well-established biomarker for environmental contamitantion in aquatic ecosystems. The aim of this study was to investigate CYP1A expression in the hepatic microsomal pellet of the European hake - Merluccius merluccius, the Tub gurnard - Trigla lucern and the Thinlip gray mullet Liza ramada during winter and spring in the sea-port Bar (Adriatic sea, Serbia and Montenegro), a region of intensive urban and industrial activities. Samples of sea water and sediment in this area were analysed for PAHs and PCBs. The presence of representative compounds from both groups was established in both seasons. Western blot analysis revealed the induction of CYP1A in all examined fish species, with increased relative amounts  of CYP1A in spring compared to winter. A species-specific response, characterized by the highest CYP1A level in the hepatopancreas of Liza ramada and the lowest level in Trigla lucerna was observed. Minimal concentrations of PAHs and PCBs in sea water that are considered capable of exerting a negative impact on marine organisms differ between countries. Therefore, the observed inducton of CYP1A could present a biological effect that links the bioavailability of the compounds of interest and their intrinsic toxicity. Measurements of biomarker responses in fish from contaminated sites could contribute to environmental monitoring and environmental risk assessment.

AB  - Homeostasis is the state of metabolic equilibrium supporting the optimal functioning of an organism that is actively maintained by complex biological mechanisms. An organism responds to deviations from this stable situation by a coordinated sequence of systemic and metabolic changes, or by local changes such as inflammatory reactions, collectively known as the acute phase response (APR) The APR is an evolutionarily conserved mechanism aimed at reestablishing an organisms normal functioning during infection, tissue injury, chemical irritation and neoplastic disease. The initiation and progression of the APR involves a coordinated series of events, including cytokine release, endothelial-cell activation, leukocyte chemotaxis, alteration of the temperature set-point and activation of the adrenal pituitary axis which provide the necessary metabolic signals to the responding organ . The liver is a 1 major partner in APR and the blood-borne inflammatory cytokines such as Tumor Necrosis Factor, lnterleukin-1and Interleukin-6 trigger transient changes in the synthesis of numerous proteins in this organ.

AB  - Oxidative stress was proposed as a key factor in the development of complications in diabetes. Antioxidant defense mechanisms pri- marly act through a broad spectrum of antioxidative enzymes, how- ever, other molecules could also express antioxidative properties. One such molecule is haptoglobin (Hp), a serum protein that func- tions as an antioxidant by virtue of its ability to bind hemoglobin and thereby prevent oxidative tissue damage. Elevation of serum Hp con- centration was obtained 12 h after acute tissue injury, and its level decreased after 24h to the control level. In our studies of streptozo- tocin-induced diabetes in adult male Wistar rats (i.p., 65mg/kg body weight), we found chronic but different degrees of increased serum Hp concentrations depending on time after treatment. The highest elevation (eight-fold), was obtained two weeks after treatment by rocket immunoelectrophoresys where the maximal activation of total SOD and superoxide anion was detected (ELISA) in the blood and in the liver, the central organ of Hp synthesis. At this time, most of the diabetic rats died suggesting that the early stage of diabetes was critical for survival. Four end seven weeks after treatment, expres- sion of Hp was still elevated, but less than after two weeks, as was the concentration of the superoxide anion and SOD activity. The strong correlation between the diabetes-related oxidative status and expression of Hp suggests that its antioxidative property plays an important role in the early stage of diabetes.

AB  - The serum concentrations of several acute-phase proteins are increased during diabetes mellitus as a consequence of the activity of the main inflammatory mediators of the acute-phase response. The diabetic state leads to themetabolic syndrome, characterized by many negative effects. Our study was focused on ·2-macroglobulin (MG), one of the most prominent acute-phase proteins in the rat. Changes in MG concentration were examined in the laboratory model ofstrepto- zotocin-induced (65 mg/kg body weight) diabetes in the rat. Rocket immunoelectrophoresis revealed increased MG concentration by the 7th week after diabetes induction. During the period leading up to the 7th week, the observed elevated concentrations of triglycerides and cortisol were symptomatic of the progression of the metabolic syn- drome. Previously, we showed that MG exerts an immunosuppressive effect.Whereas during acute stressMGplays a role in restoring home- ostasis, its raised concentration during protracted stress exerts a harm- ful effect; moreover,MGis an established biomarker of cardiac hyper- trophy in diabetesmellitus.

AB  - Expression of the rat α2-macroglobulin (MG) gene undergoes dynamic changes throughout an individual’s
life and during the acute-phase (AP) response. Details of the participation of the STAT
family of transcription factors in its control remain incompletely understood. Here we examined
the involvement of STAT5b in MG gene expression during development and the AP response.
Immuno-blot analysis revealed the highest nuclear level of STAT5b in the fetus and during postnatal
development, whereas in the adult it decreased. Stimulation of MG expression during the
AP response was accompanied by a decrease in STAT5b. Examination of STAT5b localization
revealed that the relative concentrations of STAT5b were higher in the nuclear matrix than in
the nuclear extract. Affinity chromatography with the extended promoter region of the MG gene
(–825/+12), followed by immuno-blot analysis, revealed dynamic changes in STAT5b binding.
The highest concentration of the promoter-binding form of STAT5b was observed in the fetus.
As postnatal development progressed, the level of promoter-bound STAT5b decreased and in
the adult liver it was the lowest. Stimulation of MG gene expression during the AP response
in both the fetus and adult was accompanied by significantly decreased STAT5b binding to the
MG promoter. The AP response was accompanied by lower levels of STAT5b serine and tyrosine
phosphorylation in both fetus and adult. In the nuclear matrix derived from adult tissues, tyrosine
phosphorylated species were completely absent. We conclude that developmental-stage differences
in the mechanisms that determine STAT5b nuclear localization contribute to its activity
in vivo.

AB  - Previously, we characterized the endonucleolytic activity of the nuclear matrix prepared from rat liver cryopreserved in liquid
nitrogen. The enzymic activity was attributed to a 23 kDa, Mg2þ
-dependent and sequence non-specific endonuclease (p23)
stably associated with the nuclear matrix. Here we show that p23 was absent from the nuclear matrix prepared from fresh
liver. Instead, both ex vivo (cryopreservation), as well as in vivo-induced necrosis by repeated freezing/thawing of liver tissue
in an anaesthetized rat, promoted the activation and translocation of p23 to the nuclear matrix. Considering that ex vivo and
in vivo freezing/thawing of the liver were accompanied by morphological (nuclear compaction) and biochemical events
(increased LDH activity, disorderly genomic DNA degradation, absence of lamin proteolysis, appearance of 62 and 50 kDa
necrotic cleavage products of PARP-1) commonly observed during necrosis, and because the association of p23 with the
nuclear matrix was saturable, reflecting the existence of a limited number of distinct high affinity sites on the nuclear matrix
for p23, we concluded that the activation of the nuclear matrix-associated endonuclease p23 is a feature of liver cryonecrosis.
Although cryonecrosis represents a typical example of acute cell damage, our results suggest that it is realized by ordered
molecular events.

AB  - Haptoglobin (Hp), an acute-phase protein with antioxidant proper- ties, is produced by liver cells primarily in response to IL-6 type cytokines. IL-6 cytokines mediate the induction of the Hp gene by the inducible form of the signal transducer and activator of tran- scription-3 protein (STAT3) and the CAAT/enhancer binding protein beta (C/EBPβ). This study was aimed at revealing the specificity of the IL-6 signal transduction pathways in the regulation of Hp gene expression in different diabetic phases. Diabetes was induced in male Wistar rats by single streptozotocin injection (65 mg/kg body weight). An 8-fold increase in the Hp serum level was observed two weeks after treatment (early phase) by rocket immunoelectrophore- sis. Four end seven weeks after treatment, Hp expression was still elevated but lower than after two weeks. The specificity by which the IL-6-related transcription factors regulate Hp gene transcription in diabetic rats was determined by examining of profiles (Western- immunoblot) and DNA binding activities of STAT3 and C/EBPα and
β isoforms expressed in hepatocytes. DNA-affinity chromatography with the Hp gene hormone responsive element (HRE) revealed per- manent HRE binding of inducible Tyr phosphorylated 91kD STAT3 isoform. C/EBPβ isoforms primarily mediated Hp gene regulation during the early phase of diabetes while C/EBPα was involved after four week of treatment, during progression of the illness. The obtained results suggest that the molecular machinery that mediates Hp gene regulation becomes slightly altered as diabetes progresses probably as the result of an impaired balance between pro- (IL-6) and anti-inflammatory (insulin) mediators.

AB  - CYP1A is a well-established biomarker of fish exposure to xenobiotics. Polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) are ubiquitous environmental pollutants in the aquatic environment. They induce a dose-dependent transcriptional induction of CYP1A genes and resulting increased concentrations of proteins. Our aim was to characterize the ecological impact of chemical contaminants in view of their toxicological potential. For that reasons the relative changes of CYP1A concentrations in the hepatopancreas of the Red mullet-Mullus barbatus, European hake-Merluccius merluccius and Tub gurnard-Trigla lucerna were examined in spring, at the mouth of the river Bojana. All species are of considerable commercial importance. In this work chemical analysis of seawater was performed and CYP1A induction was estimated by immunoblot analysis. Chemical analyses of seawater and sediment revealed the presence of anthracene, benzo(A)pyrene, pcb28 and pcb101. CYP1A was detected in the hepatic microsomal fraction in all examined fish species as an adaptive response to the slightly increased concentrations of PAHs and PCBs. It was shown in this study that CYP1A is a sensitive biomarker demonstrating that pollutants entered the organisms and elicited toxic effects at the critical target(s). The apparent initiation of the defense response in fish suggests that the contaminants present in seawater have an ecotoxicological potential. These findings could be interpreted as an early-warning signal for the deleterious effects of the pollutants.

AB  - We examined whether the transcriptional activation of the rat haptoglobin (Hp) gene during the acute phase (AP) response reflects the O-linked N-acetylglucosamine (O-GlcNAc) status of liver nucleoproteins (NPs) and their binding for the hormone responsive element (HRE). After deglycosylation with N-acetylglucosaininidase of the O-GlcNAc glycoproteins obtained by WGA, affinity chromatography and South-Western analysis, it was observed that only increased HRE binding ability of p64/p70 in control and p51 obtained from turpentine-treated rats can be directly attributed to the presence of O-GlcNAc residues. Therefore, expression of the rat Hp gene could be controlled by this modification of certain trans-acting NPs.

AB  - The acute inflammatory response of the liver associated with chronic food restriction was examined in adult female and male Wistar rats. The changes in the levels of serum markers of liver injury, AST and ALT and the appearance of a serum marker of inflammation, the acute phase protein (APP) haptoglobin (Hp) were assessed following turpentine treatment of well-nourished (WN) controls and undernourished (UN) rats. Undernutrition was induced by food restriction during a six week period by offering chow equivalent to 50% of the normal food intake. In the female rats undernutrition significantly potentiated liver injury and increased their sensitivity to the toxic effects of turpentine, which was opposite to the results obtained for males. Differences in the basal levels of AST, ALT and Hp between females and males imply that the effects of chronic food restriction on protein synthesis in the liver are gender related.
AB  - Ispitivan je akutni inflamatorni odgovor jetre odraslih ženki i mužjaka Wistar pacova u uslovima hroničnog izgladnjivanja. Praćene su promene koncentracije AST-a i ALT-a kao serumskih markera povrede jetre, i prisustvo akutno faznog proteina haptoglobina-serumskog markera inflamacije, nakon tretiranja normalno hranjenih (WN) i neuhranjenih pacova (UN) terpentinom. Neuhranjenost je bila indukovana restrikcijom hrane u periodu od šest nedelja količinom koja je za 50% bila manja od one koja se normalno uzima. Kod ženki pacova neuhranjenost je značajno potencirala inflamaciju jetre i povećava senzitivnost na toksične efekte terpentina što je suprotno od onog dobijenog za mužjake. Razlike u bazalnim nivoima AST-a, ALT-a i haptoglobina između ženki i mužjaka ukazuju da su efekti hroničnog izgladnjivanja na sistem za proteinsku sintezu u jetri specifični za pol.

AB  - Widespread use of organophosphates, especially in agriculture, led us to study their toxic effects. Our investigations were focused on paraoxon an oxygenated analog of a phosphothionate pesticide. This points to an analogy between the response to organophosphate intoxication and the acute phase reaction to injury. The capacity of paraoxon to elicit the acute phase response was assessed by studying two major processes characteristic of acute inflammation, the expression of acute phase proteins (APP) and the immunosuppressive activity of serum. After an LDso paraoxon administration to rats, the serum APP levels increased with time reaching a maximal level at the 24 h time point. The several-fold increases of AGP, MG, Hp and TST concentrations in the circulation of intoxicated rats, as well as a significant immunosuppressive activity of examined animal serum, pointed to the role of APP, especially AGP and MG, as immune modulators. These processes are analog to those observed during the acute phase response to injury and aimed at reestablishing homeostasis.
AB  - Široka upotreba organofosfatnih jedinjenja u industriji, a posebno u poljoprivredi, navela nas je da ispitujemo njihove toksične efekte. Istraživanja su bila fokusirana ka paraoksonu, kiseoničnom analogu fosfotionatnog pesticida, sa ciljem da se utvrdi analogija između odgovora organizma na intoksikaciju paraoksonom i akutno fazne reakcije na povredu. Sposobnost paraoksona da indukuje akutno fazni odgovor analizirana je ispitivanjem dva glavna procesa karakteristična za inflamaciju: ekspresije akutno faznih proteina i imunosupresivne aktivnosti seruma. Nakon ubrizgavanja LDso paraoksona u pacove, nivo akutno faznih proteina u serumu životinja se povećava dostižući maksimalni nivo u 24 satu. Višestruko povećanje koncentracije al- kiselog glikoproteina (AGP), a2-makroglobulina (MG), haptoglobina i tiostatina u cirkulaciji tretiranih pacova kao i značajna imunosupresivna aktivnost seruma tretiranih životinja ukazali su na ulogu ovih proteina, naročito AGP i MG, kao finih modulatora imunološkog odgovora. Ovi procesi su analogni procesima tokom akutno faznog odgovora organizma na povredu i odvijaju se u cilju uspostavljanja fiziološke ravnoteže.

Efekat nutritivnog statusa na nivo akutnog faznog odgovora proteina kod ženki pacova

AB  - In response to inflammation, infection and tissue injury, the sequence of events known as acute-phase (AP) response is induced and a number of plasma proteins, called acute-phase proteins (APPs) are synthesized in the liver as a part of the host defense mechanism. Any condition which impairs acute phase protein response (APPR) and hence organism's ability to restore homeostasis may limit its survival in various stress circumstances. Malnutrition is well-known causes of several metabolic, immune and neuroendocrine dysfunctions and this study was aimed at investigating its effect on the magnitude of normal APPR and in relation to turpentine injury. Malnutrition in female Wistar rats was induced by food restriction during six weeks period until they riched and kept body weight to 50% of that of ad libitum fed mates. Results of rocket immunoelectrophoresis revealed that malnutrition alone or in conjunction with turpentine was capable of elevating an expression of one of APPs, haptoglobin (Hp). Based on the studies of liver-specific regulatory proteins that bind to hormone regulatory elements of Hp gene and thus determine its expression we found that malnutrition probably modulated Hp response by changing the expression of active members of C/EBP, STAT and NF-kB transcription factor families.
AB  - U odgovoru na inflamaciju, infekciju i povredu tkiva, kao sastavni deo odbrambenih mehanizama u organizmu se pokreće sled događaja poznat kao akutno-fazni odgovor (AFO) u okviru koga se u jetri sintetiše grupa proteina plazme označena kao akutno fazni proteini (AFP). Svako stanje koje narušava akutno fazni proteinski odgovor organizma (AFPO), a time i njegovu sposobnost da ponovo uspostavi homeostazu može da deluje ograničavajuće na njegovo preživljavanje u uslovima različitih vrsta stresa. Izgladnjivanje je dobro poznat uzrok nekoliko metaboličkih, imunih i neuroendokrinih poremećaja te je ovaj rad imao za cilj da ispita uticaj neuhranjenosti na nivo normalnog AFPO-a i onog izazvanog akutnom povredom. Ženke pacova soja Wistar su izgladnjivane tokom perioda od šest nedelja sve do dostizanja telesne mase koja predstavlja 50% telesne mase ženki pacova koje su se u istom periodu hranile normalno. Rezultati "roket" imunoelektroforeze su pokazali da je izgladnjivanje, samo ili u sprezi sa terpentinskom povredom dovelo do uvecanja ekspresije jednog od AFP-a, haptoglobina (Hp). Izučavanjem za jetru specifičnih regulatornih proteina koji se vezuju za hormon regulatorne elemente gena Hp i time određuju nivo njegove ekspresije zaključili smo da izgladnjivanje najverovatnije moduliše odgovor Hp-a tako što menja ekspresiju aktivnih članova C/EBP, STAT and NF-kB familija transkripcionih faktora.

Binding of rat liver nucleoproteins to the distal regulatory element of the α1-acid glycoprotein gene

AB  - The binding of insoluble nuclear matrix and soluble nucleoproteinsto DRE, the distal responsive element (-5300/ -5150) that is required for the transcriptional regulation of the rat α1-acid glycoprotein (AGP) gene, was examined in the basal state when AGP transcription was low, and during the acute-phase response when its transcription was increased. By South-Western analysis of nuclear matrix preparations it was established that as a result of the acute-phase response, in addition to binding to a 40 kDa protein (p40), the distal responsive element exhibited a considerably increased binding to p45, and a slightly less prominent but enhanced binding to p29, p35, p37 and p55 proteins in relation to the control. South-Western analysis of the soluble nucleoprotein preparation revealed an increased binding to p35, p37 and p45, and an unchanged binding to p29 and p70. The presence of polypeptides with apparently identical molecular masses in the different nuclear components points to the existence of dynamic interactions between nuclear compartments that are related to the functional properties of the nucleus.
