Histochemical Localization of Alkaloids in the Bulbs of In Vitro-Regenerated Snake’s Head Fritillary (Fritillaria meleagris L.): The Effect of a Temperature Regime

AB  - The genus Fritillaria is characterized by the production of alkaloids, primarily of the isosteroidal type. The aim of this preliminary study was to perform a histolocalization screening aimed at revealing the presence of alkaloids in the bulbs of F. meleagris, cultured in vitro under different growth regimes and at different developmental stages, as a powerful tool to identify preferable in vitro plant material for alkaloid isolation. Histochemical localization of alkaloids was performed using Wagner’s and Dragendorff’s reagents in fresh sections of bulbs cultured at 24 °C or 7 °C for 4 weeks, as well as those cultured at 24 °C following the 4-week chilling treatment, which were sampled at the beginning of sprouting. A positive reaction was observed with both reagents and was particularly intense in cold-treated bulbs. Alkaloids were mainly distributed in the bulbs and partly in the sprouts of the in vitro grown F. meleagris. The most intense staining, indicative of high alkaloid content, was observed in the bulb scales of pre-chilled bulbs that sprouted at 24 °C, rendering them preferable as in vitro plant material for alkaloid isolation. The results suggest that alkaloid production in the bulbs of F. meleagris can be improved by manipulating growth in the microenvironment of in vitro cultures, in order to meet the increasing industrial demand for medicinally or commercially important metabolites used as traditional medicines and herbal remedies.

AB  - Fritillaria meleagris is a horticulturally and medicinally valuable bulbous plant that requires a period of low temperatures for proper growth and flowering. Since conventional methods of propagation are ineffective and very slow, tissue culture techniques offer an integrated approach to mass production of this valuable geophyte. In this study, we investigated the effects of various auxin–cytokinin combinations on different morphogenetic pathways in bulb scale culture. Bulbs obtained in vitro were cut longitudinally, and bulb scales were cultured for four weeks at 7 °C on MS medium supplemented with 6-benzylaminopurine (BAP) in combination with 2,4-dichlorophenoxyacetic acid (2,4-D) or α-naphthaleneacetic acid (NAA) at different concentrations in order to investigate the influence of plant growth regulators (PGRs) on different morphogenetic responses. Regeneration percentage, number of shoots per explant, shoot length, number of bulbs and number of somatic embryos were monitored weekly. After chilling, bulb scales were transferred to 24 °C, and all parameters were recorded again. Low PGR concentrations were very effective for shoot multiplication, yielding up to 5.5 shoots per explant. 2,4-D (at 2 mg/L) in combination with low BAP (0.25 mg/L) produced the highest number of bulbs (11.00 ± 0.00), while PGR-free medium was extremely effective in somatic embryo formation (13.50 ± 2.90). Detached somatic embryos and bulblets continued to grow and develop on fresh PGR-free medium. We present data demonstrating that low auxin–cytokinin concentrations and PGR-free medium provide an effective method for a combined morphogenetic pathway in F. meleagris that is suitable for large-scale propagation.

AB  - U prvom delu doktorske disertacije ispitivan je odgovor biljaka Impatiens walleriana na vodni stres indukovan polietilen glikolom (PEG) i uticaj salicilne kiseline (SA) u in vitro uslovima. Kulture izdanaka I. walleriana su gajene na hranljivoj podlozi koja je sadržala različite kombinacije i koncentracije PEG (1-3%) i SA (1-3 mM). Nakon dugotrajnog delovanja vodnog stresa na biljke I. walleriana utvrđen je uticaj PEG i SA na parametre hidratacije, indikatore oksidativnog stresa, kao i na parametre rastenja izdanaka. PEG indukovani vodni stres je uzrokovao redukciju rastenja, smanjenje mase svežih izdanaka, broja listova i izdanaka, relativnog sadržaja vode (RWC) i sadržaja hlorofila, kao i akumulaciju prolina, H2O2, i malondialdehida (MDA). Aktivnosti antioksidativnih enzima, katalaza (CAT), superoksid dismutaza (SOD) i peroksidaza (POX), su povećane kao odgovor na PEG tretman i to zavisno od primenjene koncentracije PEG. U drugom delu doktorske disertacije ispitivan je uticaj folijarne primene 2 mM SA na I. walleriana gajene ex vitro u uslovima suše. Biljke su podeljene u 4 grupe: zalivane biljke, zalivane biljke tretirane sa SA, nezalivane biljke i nezalivane biljke tretirane sa SA. Utvrđen je broj cvetnih pupoljaka i cvetova, RWC, sadržaj MDA, prolina, kao i aktivnosti SOD, CAT i POX u različitim vremenskim tačkama tokom trajanja eksperimenta. Suša je dovela do venjenja, odbacivanja cvetova, smanjenja RWC i porasta MDA. Kao odgovor na sušu kod I. walleraina je pojačana aktivnost hloroplastne Cu/ZnSOD i dve izoforme gvajakol peroksidaze. Identifikovane su tri de novo sekvence dehidrina korišćenjem transkriptoma iz listova I. walleriana: IwDhn1, IwDhn2.1 i IwDhn2.2. Suša je indukovala ekspresiju IwDhn2.1 i IwDhn2.2. Primena SA je pozitivno uticala na biljke koje su bile izložene suši, sprečavajući venjenje, održavajući RWC i aktivnost antioksidativnih enzima, ali nije imala uticaj na sprečavanje odbacivanja cvetova.
AB  - In the first part of dissertation the responses of Impatiens walleriana to polyethylene glycol (PEG) induced water stress and the potential of exogenous salicylic acid (SA) as stress-ameliorating agent was described. Impatiens shoot culture was established on medium containing different concentrations and combinations of PEG (1–3% ) and SA (1–3 mM). After prolonged drought, water relation parameters, oxidative stress indicators, and growth responses of the shoots to PEG and/or SA were recorded. PEG reduced growth, fresh weight of shoots, the number of developed leaves and shoots, relative water content (RWC), and chlorophyll content. PEG caused accumulation of proline, H2O2, and malondialdehyde (MDA). The activities of catalase (CAT), superoxide dismutase (SOD), and peroxidase (POX) were increased in response to PEG in a dose-dependent manner. In the second part of dissertation it was investigated whether the foliar application of 2 mM SA could protect ex vitro grown I. walleriana plants from drought stress. The plants were divided into 4 groups: watered plants, drought-stressed plants, watered plants treated with SA and drought-stressed plants treated with SA. The number of flower buds and flowers, RWC, contents of MDA, proline and the activities of superoxide dismutases, catalases and peroxidases were recorded at different time points. Three dehydrin sequences were identified in de novo assembled leaf transcriptome: IwDhn1, IwDhn2.1 and IwDhn2.2. Drought stress caused wilting, floral abortion, reduction of RWC and increased MDA. In response to drought, I. walleriana induced chloroplastic Cu/ZnSOD and two guaiacol peroxidase isoforms. The remarkable drought response was induction of IwDhn2.1 and IwDhn2.2. SA had ameliorating effects on plants exposed to drought, including prevention of wilting, preservation of RWC, modulation of antioxidative activities, but had no effects on flowers preservation.

Effects of Exogenous Salicylic Acid on Drought Response and Characterization of Dehydrins in Impatiens walleriana

AB  - Impatiens walleriana is a valued ornamental plant sensitive to drought stress. We investigated whether the foliar application of 2mM salicylic acid (SA) can protect potted I. walleriana plants from drought stress. The plants were divided into: watered plants, drought-stressed plants, watered plants treated with SA and drought-stressed plants treated with SA. The number of flowers and flower buds, relative water content (RWC), contents of malondialdehyde (MDA) and proline (Pro) and the activities of superoxide dismutases, catalases and peroxidases were recorded at different time points. Three dehydrin sequences were identified in de novo assembled leaf transcriptome: IwDhn1, IwDhn2.1 and IwDhn2.2. Drought stress caused wilting, floral abortion, reduction of RWC and increased MDA—an indicator of lipid peroxidation. In response to drought, Impatiens accumulated Pro and induced chloroplastic Cu/ZnSOD and two peroxidase isoforms. The most remarkable drought response was strong induction of IwDhn2.1 and IwDhn2.2. Rehydration restored RWC, Pro level, Cu/ZnSOD activity and dehydrins expression in drought-stressed plants approximately to the values of watered plants.SA had ameliorating effects on plants exposed to drought, including prevention of wilting, preservation of RWC, increased Pro accumulation, modulation of antioxidative activities and remarkable decrease of lipid peroxidation, but without effects on flowers’ preservation.

We investigated whether the foliar application of 2mM salicylic acid (SA) can protect potted I. walleriana plants from drought stress. The plants were divided into: watered plants, drought-stressed plants, watered plants treated with SA and drought-stressed plants treated with SA. The number of flowers and flower buds, relative water content (RWC), contents of malondialdehyde (MDA) and proline (Pro) and the activities of superoxide dismutases, catalases and peroxidases were recorded at different time points. Three dehydrin sequences were identified in de novo assembled leaf transcriptome: IwDhn1, IwDhn2.1 and IwDhn2.2. Drought stress caused wilting, floral abortion, reduction of RWC and increased MDA—an indicator of lipid peroxidation. In response to drought, Impatiens accumulated Pro and induced chloroplastic Cu/ZnSOD and two peroxidase isoforms. The most remarkable drought response was strong induction of IwDhn2.1 and IwDhn2.2. Rehydration restored RWC, Pro level, Cu/ZnSOD activity and dehydrins expression in drought-stressed plants approximately to the values of watered plants.SA had ameliorating effects on plants exposed to drought, including prevention of wilting, preservation of RWC, increased Pro accumulation, modulation of antioxidative activities and remarkable decrease of lipid peroxidation, but without effects on flowers’ preservation.",

AB  - The aim of this study was to develop a fast, reliable and true-to-type protocol for in vitro plant regeneration and long-term storage of horned pansy (Viola cornuta L). Seed germination over 60% was recorded after 12 weeks of growth at 10 °C or 4 °C. Calli formation and shoot induction were obtained in petiole and hypocotyl culture on half-strength MS mineral salts with full concentration of Na-FeEDTA and vitamins (½MS medium) with 2,4-dichlorophenoxyacetic acid (2,4-D, 0.1 mg/L) and 6-benzylaminopurine (BAP, 2.0 mg/L) and leaf culture on ½MS medium with thidiazuron (TDZ,1.0 mg/L). The highest frequency of adventitious shoot induction (50%) with six shoots/explant was achieved in hypocotyl culture from top hypocotyl segments, close to epicotyl which was grown 8 weeks at 16 h light/8 h dark photoperiod. Subsequent shoot multiplication was achieved on ½MS medium with α-naphthaleneacetic acid (NAA, 0.1 or 0.5 mg/L) and BAP (1.0 mg/L). Rooting of shoots was obtained on ½MS medium with low concentration (0.1 mg/L) of auxins: indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) or NAA, or without growth regulators. In vitro-derived plantlets were acclimatized under greenhouse conditions. All plants developed normally, bloomed and set seeds. Shoot tips were cryopreserved succssefully using modified plant vitrification 3 (PVS3-based vitrification procedure). Cold acclimation for 2 weeks significantly improved shoot regrowth (64%) after thawing in comparison to non-acclimated shoots (39%). Clonal fidelity of regenerated plantlets at ploidy level was confirmed by chromosome counting. The presented protocol can be useful for mass propagation, genetic transformation studies and long-term storage of valuable Viola spp.

AB  - Cryopreservation represents a suitable method for long term storage of different plant genetic
resources. The aim of this study was to develop protocol for cryopreservation of Viola cornuta
shoot tips using one step freezing method with chemical dehydration of tissue with modified
Plant Vitrification Solutions (PVS2 or PVS3). Shoot tips (1-2 mm) of two-week cold acclimated
shoots were cultured on ½MS medium with 0.3 M sucrose for one day before treatment with loading
solution (2 M glycerol, 0.4 M sucrose) for 30 min. Osmotic dehydration with PVS2 solution (30% glycerol, 15% ethylene glycol and 15% DMSO in liquid ½MS medium with 0.4 M sucrose) were
tested at 0 °C or 24 °C. Osmotic dehydration with PVS3 (50% sucrose, 50% glycerol in liquid ½MS
medium) were tested at 24 °C for 45 min. After the treatment the explants were directly immersed
in liquid nitrogen (LN) for at least one day. Re-warming was performed at 42 °C in water bath
for 2 min. After re-warming, the PVS solutions were replaced with unloading solution containing
1.2 M sucrose for 20 min. Re-warmed shoot tips were cultured on ½MS medium with 0.1 mg L-1
BAP. We observed that PVS2 solution is cytotoxic for V. cornuta shoot tips and cannot be used for
cryopreservation. However, cryopreservation with PVS3 solution was successful, where 71.9-100%
shoot tips survived treatment before immersion to LN and 31-40% survived after re-warming from
LN. Regrowth of cryopreserved shoot tips with new well-formed leaves was obtained after four
weeks of culture.

AB  - Viola cornuta L. ‘Lutea Splendens’ is a perennial ornamental plant with small yellow flowers that
naturally grows in the Pyrenees in Spain and France. To develop novel cultivars with orange and
red flower colors, we performed Agrobacterium tumefaciens (LBA4404)-mediated transformation
with the binary vector pWBVec10a/P35S::Llccs::TNos that harbored capsanthin-capsorubin synthase
(Llccs) gene from Lilium lancifolium under the control of CaMV35S constitutive promoter and
the nopaline synthase (Nos) terminator. Capsanthin-capsorubin synthase catalyzes the conversion
of anteraxanthin and violaxanthin, two yellow ubiquitous 5-6-epoxy-xanthophylls, into capsanthin
and capsorubin, two red xanthophylls, respectively. Starting with hypocotyl explants, we developed
a transformation protocol with 0.3% shoot regeneration efficiency. Histochemical assay for
β-glucuronidase (GUS) activity showed uidA reporter gene expression in all putative Llccs-transgenic
shoots. The presence of Llccs transgene, hygromycin phosphotransferase (hpt) selectable
marker gene and uidA (GUS) reporter gene in all putative Llccs-transgenic lines were confirmed
by PCR analysis. This is the first report on Agrobacterium-mediated genetic transformation of V.
cornuta L. with the aim to introduce desirable traits into this species.

AB  - Impatiens hawkeri (Balsaminaceae) je ukrasna biljna vrsta poreklom sa Nove Gvineje. Krioprezervacija predstavlja metod za dugotrajno čuvanje biljnog materijala na -196C u tečnom azotu koji se koristi i za eliminaciju patogena kod biljaka.1 Vrste roda Impatiens su izrazito osetljive na tretmane dehidratacije koji prethode zamrzavanju.2 Takođe, koncentracija saharoze u hranljivim podlogama utiče na morfogenetski potencijal tokom gajenja u kulturi in vitro.3 U ovom radu smo ispitivali uticaj prekulture vrhova izdanaka na povišenoj koncentraciji saharoze (6%), kao i vitamina C na preživljavanje i regeneraciju biljaka posle hemijske dehidratacije vrhova izdanaka sa rastvorima za vitrifikaciju (PVS2 i PVS3). Vrhovi izdanaka I. hawkeri su izrazito osetljivi na tretman sa 100% PVS2 rastvorom. Najveći morfogenetski potencijal posle izlaganja PVS2 rastvoru su pokazali vrhovi tretirani sa 50% PVS2 rastvorom kada je dolazilo samo do kalusiranja vrhova izdanaka. Potpuna regeneracija biljaka posle PVS2 tretmana postignuta je uz dodatak vitamina C u hranljivoj podlozi u koncentraciji od 100 mg/l. Najbolji rezultati preživljavanja i regeneracije biljaka posle odmrzavanja postignuti su korišćenjem metode vitrifikacije sa 100% PVS3 rastvorom u trajanju od 45 min. Izdanci gajeni na hranljivoj podlozi sa 6% saharoze su pokazali veće preživljavanje (49,4%) posle izlaganja ultra niskim temperaturama u odnosu na izdanke gajene na podlogama sa 3% saharoze (26,5 %).

Krioprezervacija predstavlja metod za dugotrajno čuvanje biljnog materijala na -196C u tečnom azotu koji se koristi i za eliminaciju patogena kod biljaka.1 Vrste roda Impatiens su izrazito osetljive na tretmane dehidratacije koji prethode zamrzavanju.2 Takođe, koncentracija saharoze u hranljivim podlogama utiče na morfogenetski potencijal tokom gajenja u kulturi in vitro.3 U ovom radu smo ispitivali uticaj prekulture vrhova izdanaka na povišenoj koncentraciji saharoze (6%), kao i vitamina C na preživljavanje i regeneraciju biljaka posle hemijske dehidratacije vrhova izdanaka sa rastvorima za vitrifikaciju (PVS2 i PVS3). Vrhovi izdanaka I. hawkeri su izrazito osetljivi na tretman sa 100% PVS2 rastvorom. Najveći morfogenetski potencijal posle izlaganja PVS2 rastvoru su pokazali vrhovi tretirani sa 50% PVS2 rastvorom kada je dolazilo samo do kalusiranja vrhova izdanaka. Potpuna regeneracija biljaka posle PVS2 tretmana postignuta je uz dodatak vitamina C u hranljivoj podlozi u koncentraciji od 100 mg/l. Najbolji rezultati preživljavanja i regeneracije biljaka posle odmrzavanja postignuti su korišćenjem metode vitrifikacije sa 100% PVS3 rastvorom u trajanju od 45 min. Izdanci gajeni na hranljivoj podlozi sa 6% saharoze su pokazali veće preživljavanje (49,4%) posle izlaganja ultra niskim temperaturama u odnosu na izdanke gajene na podlogama sa 3% saharoze (26,5 %).",

from water stress requires understanding the events associated with dehydration and rehydration
at the molecular level. Dehydrins (DHNs) are highly hydrophilic proteins whose expression in many
plant species has been correlated with several types of abiotic stresses, including drought, salinity,
and cold. Dehydrins comprise Group II of late embryogenesis abundant (LEA) proteins, known to
be associated with plant responses to water deficit. DHNs lack stable tridimensional structures, so
they are considered intrinsically disordered proteins. They are characterized by three conserved
sequence motifs marked as a K (Lys-rich), Y (Tyr-rich) and S (Ser-rich). To study the expression of
Dhn genes in response to drought, we have sequenced I. walleriana transcriptome and identified
IwDhn transcripts from RNA-seq data using HMMER (hmmscan, v3.1b2) and Pfam 31 database.
Quantitative RT-PCR revealed that two IwDhn genes are extremely upregulated in drought, while
one has constitutive expression. It can be speculated that fast rehydration after drought stress is at
least in part result of the protective role of dehydrins.

AB  - Impatiens walleriana is one of the most popular Impatiens species characterized by fleshy, succulent
leaves and a variety of flower colours. The major problem in production and sale of this
ornamental species is related to its tendency to quickly wilt when drought stressed. Since one of
the most devastating consequences of drought is the onset of oxidative stress, we have studied
the responses of antioxidative enzymes to drought and applied salicylic acid (SA) as a potential
stress-alleviating regulator. The I. walleriana seeds were germinated in a growth chamber under
controlled conditions. Plants (8 weeks old) were divided into four groups: (1) regularly watered
plants (W), (2) regularly watered plants with single application of 2 mM SA (WS), (3) plants exposed
to drought (D), and (4) drought-exposed plants with single application of 2 mM SA (DS). The activities
of antioxidative enzymes including superoxide dismutase (SOD), catalase (CAT) and guaiacol
peroxidase (POX) were assessed in all four groups. Our results show that drought and SA (WS, D,
and DS) increase activity of Cu/ZnSOD and MnSOD in comparison to control (W). Total CAT activity
was increased in D, but not in WS and DS groups; however additional CAT isoforms were observed
in these groups. POX activity was generally high in D and especially in DS group. It can be concluded
that SA improves drought tolerance in I. walleriana grown ex vitro probably by modulating the
activity of antioxidative enzymes and can be used as a drought-ameliorating agent.

Agrobacterium-mediated genetic transformation of Viola cornuta

AB  - Introduction: Viola cornuta is a valuable perennial ornamental plant. Development of new traits, such as new flower color with classical breeding suffers from many difficulties, which can be overcome using genetic engineering. With aim to develop protocol for Agrobacterium-mediated transformation of V. cornuta, we used Agrobacterium tumefaciens strain LBA4404 harbouring the superbinary vector pTOK233 carried a GUS reporter gene and hygromycin phosphotransferase selectable marker gene.
Methods: Hypocotyl explants obtained from seedlings were grown on ½MS medium supplemented with 0.1 mg/l 2,4-D and 2.0 mg/l BA for shoot induction. After two days of pre-cultivation, hypocotyl explants were inoculated in bacterial suspension for 15 min and placed on the same culture medium with addition of acetosyringone 100 μM at pH 5.2. After two days of co-cultivation, explants were transferred on shoot induction medium supplemented with cefotaxime and hygromycin B for selection. Regenerated putative transformants were analyzed by PCR for hygromycin phsphotransferase gene presence and by histochemical assay for β-glucuronidase (GUS) activity.
Results: Shoots were obtained within 8 weeks after explants were inoculated with A. tumefaciens, with 2.0% regeneration efficiency. PCR analysis confirmed selectable marker gene presence in twelve out of sixteen (75.0%) independently derived putatively transformed lines that were tested. Additionally, all analyzed lines exhibited a notable β-glucuronidase activity that was not present in untransformed plants.
Conclusion: This is the first report about V. cornuta susceptibility to A. tumefaciens. Presented protocol for genetic transformation can be used for further introduction of desirable traits in V. cornuta cultivars.

'Lutea Splendens' were cultured on half-strength Murashige and Skoog (MS) medium with several concentrations of 6-benzylaminopurin (BAP), 6-furfurylaminopurine (KIN), N-phenyl-N'(1,2,3-thidiazol-5-yl) urea (TDZ) or N-(2-chloro-4-pyridyl)-N'phenylurea (CPPU) alone or supplemented with indole-3-acetic acid (IAA) or a-naphthaleneacetic acid (NAA). The most efficient direct adventitious shoot induction (21%) without callus formation was obtained using petiole explants cultured on half-strength MS medium containing 2.02 μM CPPU. Significantly higher shoot multiplication was achieved on medium supplemented with TDZ (7.6 shoots) or CPPU (7.1 shoots) than on medium supplemented with NAA and BAP (2.9 shoots). Shoots were rooted on half-strength MS medium without plant growth regulators.

AB  - We describe the responses of Impatiens walleriana to polyethylene glycol (PEG)-induced physiological drought and the potential of exogenous salicylic acid (SA) as stress-ameliorating agent. Impatiens shoot culture was established on 16 different media containing 0-3% PEG and 0-3 mM SA. After prolonged drought (60 days), water relation parameters, oxidative stress indicators, and growth responses of the shoots to PEG and/or SA were recorded. PEG reduced growth, fresh weight, the number of developed leaves and shoots (proliferation rate, PR), relative water content, and chlorophyll content. PEG increased leaf water loss (LWL) and caused accumulation of proline, H2O2, and malondialdehyde. The activities of catalase, superoxide dismutase, and peroxidase were increased in response to PEG in a dose-dependent manner, with specific peroxidase isoforms induced by drought. Exogenous SA counteracted the effects of PEG on growth, physiological and biochemical parameters, except on proline accumulation. SA was particularly effective in enhancing PR, preserving LWL, and protecting photosynthetic pigments and membranes from oxidative damage. Proline accumulation was strongly enhanced by both PEG and SA. SA had differential effects on different peroxidase isoforms. SA may be safely used in 2-3 mM concentration for drought protection of Impatiens with no negative effects.

undesirable to customers. Salicylic acid (SA) is now considered as an endogenous growth regulator
that influences many plant physiological processes including regulation of plant defense, and plays a significant
role in plant protection against various abiotic stresses. To evaluate the role of exogenously applied SA
against the effect of drought simulated by polyethylene glycol (PEG 8000), growth and physiological parameters
of I. walleriana were analyzed. In vitro developed shoots of I. walleriana were grown on MS media containing
different levels of PEG (0, 1, 2 and 3%) and treated with SA at varying concentrations (0, 1, 2 and 3
mM). Increasing PEG concentrations progressively reduced plant growth parameters (plant height, number
of leaves, fresh weight and proliferation rate) and decreased relative water content, chlorophyll and total pigments
in I. walleriana leaves. In contrast, SA treatments under simulated water deficit conditions enhanced I.
walleriana growth performance. Additionally, a decline in the photosynthetic pigment contents and the level
of water loss in leaves of drought stressed plants were significantly reduced with SA application. Taken together,
these results show that SA improves performance of I. walleriana under stress conditions, suggesting
that exogenous application of SA can be used to increase tolerance of Impatiens plants against water deficit.

AB  - Impatiens walleriana is an ornamental plant sensitive to dehydration with tendency to wilt without water.
Proper tissue dehydration is a necessary step in cryopreservation protocols to avoid formation of intracellular
ice crystals which are lethal for plant cells. In order to increase dehydration tolerance of I. walleriana shoots,
we evaluated proline accumulation, a common osmoprotectant, in I. walleriana shoots after two weeks of
culture at several dehydration pretreatments: increased concentration of sucrose (3, 4, 5, 6, and 10%), at different
temperatures (25 ± 2 °C and 4 °C) and different concentration of abscisic acid (ABA, 0, 0.1, 0.2, 0.5, 1, 5
and 10 mg L-1). We observed that graduated concentration of sucrose in culture media increased proline accumulation
in I. walleriana shoots grown under standard conditions, as well as at 4 °C. The highest proline
concentration was recorded in shoots grown on media with 6% sucrose (100 ± 2.3 μmol g-1) at 25 °C which
was more than three times higher than on medium with 3% sucrose (30 ± 0.6 μmol g-1). The highest proline
accumulation in shoots grown at 4 °C was observed at 10% sucrose (88 ± 0.9 μmol g-1) in comparison to
37 ± 0.6 μmol g-1 at 3% sucrose. Addition of ABA in culture medium increased proline concentration in shoots
grown on media with 10 mg L-1 ABA (68 ± 1.2 μmol g-1). Our results suggest that applied pretreatments significantly
increase accumulation of proline and can be helpful in increasing dehydration tolerance of I. walleriana
shoots prior to cryopreservation.

AB  - We developed an efficient plant regeneration protocol for a yellow cultivar of Viola cornuta ‘Lutea Splendens’.
Calli and shoots were induced from seedling explants (petiole, leaf, hypocotyls) by one month of culture
on ½-strength MS medium supplemented with 2,4-D (0.1 mg L-1) and BA (2.0 mg L-1). The highest frequency
of shoot induction (12%) was obtained from petiole explants, while the highest frequency of callus formation
(97%) was achieved on hypocotyl explants. The effects of explant origin (top, center, and bottom of hypocotyl)
and light/dark conditions on shoot and callus induction in hypocotyl cultures were also investigated. The
highest shoot induction in hypocotyl cultures (30%) was achieved when explants from the upper section of
the hypocotyls (near the epicotyls) were cultured in light conditions. Shoot multiplication from all explants
was achieved by transferring initially formed shoots to ½strength MS medium supplemented with NAA (0.2
mg L-1), GA3 (2.0 mg L-1), and TDZ (1.0 mg L-1) for four weeks of culture. Developed shoots were then further
multiplied on ½strength MS medium supplemented with NAA (0.5 mg L-1) and BA (1.0 mg L-1). Regenerated
shoots successfully rooted on ½-strength MS medium without growth regulators, acclimatized in a greenhouse
in autumn, and bloomed the following spring. All the regenerated plants exhibited normal morphology
and flower color. This regeneration protocol could be useful for mass propagation using in vitro cultures
and genetic transformation studies using seedling explants.

Cryopreservation of Impatiens walleriana shoot tips using PVS2 vitrification procedure

AB  - Vitrification with plant vitrification solution 2 (PVS2) is commonly used procedure for chemical dehydration
of tissue prior to cryopreservation of the shoot tips. PVS2 contains 30% glycerol (w/v), 15% ethylene glycol
(w/v) and 15% dimethylsulfoxide (DMSO; w/v) in basal culture medium containing 0.4 M sucrose. In order
to develop PVS2-vitrification based protocol for cryopreservation of Impatiens walleriana shoot tips, we tested
the effect of different PVS2 concentrations (100, 80, 60, 50, and 40%), and time of shoot tips exposure to
PVS2 solution (5, 10, 15, 20, 25 and 30 minutes) on survival and plant regeneration prior and after exposure
to liquid nitrogen (LN). Nodal segments of I. walleriana were cultured on plant growth regulator-free MS medium
for three weeks before the shoot tips were isolated. The results have shown that survival of shoot tips
prior to exposure to liquid nitrogen depended on PVS2 concentration and time of exposure to PVS2 solution
and ranged from 100% (lower PVS2 concentrations) to 50% (100% of PVS2, 30 min exposure). After exposure
to LN, the highest survival (77%) was achieved when shoot tips were exposed 25 min to 50% PVS2 solution,
while full strength PVS2 solution with 30 min exposure was harmful when only 26% of cultured shoot tips
survived. The highest plant regeneration (10.3%) of the cryopreserved shoot tips was observed when shoot
tips were exposed 25 minutes to 50% PVS2 solution prior to immersing in LN. Viability of PVS2 treated shoot
tips was determined by staining with fluorescein diacetate.

AB  - An efficient propagation protocol by somatic embryogenesis and organogenesis of Balkan endemic iris, Iris reichenbachii, was achieved and clonal fidelity of regenerated plants evaluated. Both regeneration pathways were induced at the same time in zygotic embryo culture on Murashige & Skoog (MS) medium supplemented with 0.5-5.0 mM 2,4-dichlorophenoxy acetic acid (2,4-D) as the sole hormone. Embryogenic calli were further mantained on medium supplemented with 2,4-D and kinetin (Kn; 0.5 and 5.0 mM, respectively). Organogenic calli were selected and further cultured on MS media supplemented with 1-naphthaleneacetic acid (NAA) and benzyladenine (BA; 0.5 and 4.5 mM, respectively) for shoot initiation. Somatic embryos germinated and shoots rooted on MS plant growth regulator-free medium. Plants regenerated by both procceses were succesfully acclimatized in greenhouse conditions and flowered in the following flowering season. Some alterations in flower morphology were detected among plants regenerated by organogenesis. Flow cytometric analysis revealed that plants with altered morphology of flowers had the same ploidy level and genome size as plants collected from the natural habitat. A tetraploid plant was observed in the population of plants regenerated by somatic embryogenesis induced at a high concentration of 2,4-D (10.0 mM).
AB  - Prikazan je efikasan protokol za propagaciju somatskom embriogenezom i organogenezom Balkanske endemične perunike, Iris reichenbachii i procenjena je klonalna indentičnost dobijenih biljaka. Oba načina za regeneraciju u uslovima in vitro su postignuta u kulturi zrelih zigotskih embriona na Murashige & Skoog (MS) hranljivoj podlozi obogaćenoj sa 2,4-dihlorofenoksi sirćetnom kiselinom 0.5-5.0 mM (2,4-D) kao jedinim regulatorom rastenja. Dobijeni embriogeni kalusi su dalje gajeni na hranljivoj podlozi sa 2,4-D i kinetinom (Kn; 0.5 odnosno 5.0 mM). Changes of antioxidative enzymes in Impatiens walleriana L. shoots in response to genetic transformation

AB  - Impatiens walleriana L. shoots were inoculated with Agrobacterium rhizogenes
A4M70GUS and the effects of genetic transformation on the catalase (CAT), superoxide
dismutase (SOD) and peroxidase (POX) activities in wounded region of stems and
unwounded leaves were evaluated 10, 24, 240 and 720 hours after inoculation. Following
Agrobacterum infection activities of plant antioxidative enzymes changed in a timedependent manner indicating that dynamic processes occurred during plantAgrobacterium interaction, plant cell transformation and formation of hairy roots.
Appearance of hairy roots on wound sites of shoots was observed ten days after
inoculation with A. rhizogenes and the root induction frequency was 100%. Among
selected hairy root lines significant differences in growth rate and biomass production
were observed and an average 3-fold increase in biomass production was observed for the
best growing hairy root line compared with the untransformed roots. PCR analysis
showed presence of uidA, rolB, rolC and rolD genes in all analyzed I. walleriana L. hairy
root lines, while amplification fragment of rolA gene was detected in 83.3% transformed
lines. Efficient transformation protocol for I. walleriana L described in this work offer
possibilities to generate hairy root cultures for in vitro propagation of plant viruses.
AB  - Izdanci Impatiens walleriana L. su inokulisani sa Agrobacterium rhizogenes A4M70GUS i praćen
je efekat genetičke transformacije na akivnost katalaza (CAT), superoksid dismutaza (SOD) i
peroksidaza (POX) u regionu povrede izdanka kao i u nepovređenim listovima 10, 24, 240 i 720
sati nakon infekcije. Promene u aktivnosti antioksidativnih enzima biljke u zavisnosti od vremena
nakon infekcije ukazuju na dinamične procese koji se odigravaju tokom interakcije biljke i
Agrobakterije, transformacije biljnih ćelija i formiranja “hairy roots” (transformisanih korenova).
Pojava transformisanih korenova je uočena 10 dana nakon inokulacije i učestalost njihove
indukcije iznosila je 100%. Između izabranih “hairy roots” linija postojale su značajne razlike u
stopi rasta i uvećanju biomase, a za transformisanu liniju korenova koja je pokazivala najbolji rast
zabeležen je trostruko veći porast biomase u odnosu na netransformisane korenove. PCR analizom
je potvrđeno prisustvo uidA, rolB, rolC and rolD gena u svim analiziranim linijama
transformisanih korenova I. walleriana L. dok je fragment amplifikacije rolA gena detektovan kod
83.3% transformisanih linija. Efikasni protokol transformacije I. walleriana L koji je opisan u
ovom radu pruža mogućnost stvaranja kultura transformisanih korenova u cilju in vitro
propagacije biljnih virusa.

Production of hairy root cultures of lettuce (Lactuca sativa L.)

AB  - Hairy root cultures of lettuce (Lactuca sativa L.) were obtained by
   inoculation of cotyledonary leaves of in vitro lettuce seedlings (cvs.
   Nansen and Ljubljanska ledenka) with Agrobacterium rhizogenes A4M70GUS.
   Approximately in 96.7\% cvs. Nansen and in 91.2\% Ljubljanska ledenka
   inoculated explants produced hairy root when they were incubated on
   Murashige and Skoog (MS) half-strength medium without plant growth
   regulators. A total of 54\% of all hairy root cultures expressed GUS
   activity. Every hairy root represented an independent transformation
   event. Line Ljubljanska ledenka 18 showed the highest biomass (5.5 times
   the biomass of control root). A PCR analysis of the genomic DNA
   confirmed the presence of marker and target genes in 15 hairy roots
   examined.
