Plant regeneration of allium spp. in the culture of mature zygotic embryos
Abstract:
Plant regeneration in the culture of mature zygotic embryos of Allium aflatunense and A. fistulosum was investigated. Mature zygotic embryos after surface sterilization, were isolated and cultured on BDS or MS mineral solution, 30.0 gL-1 sucrose, solidified by 0.7 mgL"1 agar and with inositol 100.0 mgL-1, nicotinic acid 1.0 mgL-1, casein hydrolysate 100.0 mgL-1 L-proline 500.0 mgL-1, glicin 2.0 mgL-1, vitamin B1 1.0 mgL-1, vitamin B6 1.0 mgL-1, 2,4-dihydrophenoxyacetic acid (2,4-D, 9.0 mM) or thidiazuron (TDZ, 9.0 mM). Somatic embryos and bulblets were formed directly in the coleoptile and radicule region of zygotic embryos only on BDS medium with TDZ. The histological analysis shows that somatic embryos on BDS medium with 2,4-D are formed indirectly on the embryogenic callus. Germination of somatic embryos and bulblets was investigated on nine different culture media supplemented with activated carbon, IAA, NAA, IBA or BAP. Somatic embryos and bulblets were further cultivated under cool treatment at 4°C for breaking dormancy.
