Next generation of fullerol derivate in melanoma cell reprogram

dc.description.abstract | Nanotechnology, as a field of research related to the manifestation of special physicochemical properties and behavior of materials in nanoscale, has experienced significant progress in recent decades. It has led to the development of a distinct area - nanomedicine, where nanomaterials find a place in the prevention, monitoring, diagnosis and treatment of various diseases. Some types of nanomaterials show high efficacy in the elimination of tumor cells and have already undergone clinical trials. Fullerene is the third allotropic modification of carbon beside graphite and diamond. It is characterized by an icosahedral molecular crystal structure with 12 pentagons and a variable number of hexagons. The low solubility of fullerenes in polar solvents limited potential applications in biomedicine and served as a platform for the development of a range of compounds with better solubility and significantly improved properties. Among them, polyhydroxylated fullerenes-fullerols stand out; with spectrum of biological activities that attracts the attention of numerous researchers. In our study, the potential of a new fullerol derivate (3HFWC) to inhibit the growth of melanoma cells of different origins and invasiveness, in conjunction with hyperpolarized light, was examined. Suppression of melanoma cell growth was a consequence of the loss of division potential, triggered cellular senescence, and intensified melanocytic differentiation. In the syngeneic mouse model, the combined action of 3HFWC and hyperpolarized light significantly inhibited melanoma growth as a consequence of direct action on tumor cells but also modulation of the pro/anti-tumor immune response. The complex mechanism of action that combines the phenotypic transformation of melanoma cells and the effects on the immune system makes hyper-hydroxylated fullerene worth further research. | sr |
dc.description.abstract | Нанотехнологија, као област истраживања везана за испољавање посебних фи- зичко-хемијских својстава и понашања материјала на нано нивоу, је доживела значајан напредак последњих деценија. Довела је до развоја посебне гране-нано- медицине, где наноматеријали налазе место у превенцији, дијагностици и лечењу различитих болести. Поједине врсте наноматеријала показују веома високу ефи- касност у елиминацији туморских ћелија и већ су подвргнуте клиничким испи- тивањима. Фулерен је трећа алотропска модификација угљеника поред графита и дијаманта. Kарактерише га молекуларна кристална структура у форми икосаедра са 12 пентагона и променљивим бројем хексагона. Слаба растворљивост фулерена у поларним растварачима је ограничила потенцијалну примену у биомедицини и послужила као платформа за развој низа једињења са бољом растворљивошћу и значајно побољшаним својствима. Међу њима се свакако издвајају полихидрокси- ловани фулерени-фулероли чија палета биолошких активности привлачи пажњу бројних истраживача. У нашој студији испитиван је потенцијал деривата фуле- рола (3HFWC) да у садејству са хиперполаризованом светлошћу инхибира раст ћелија меланома, различитог порекла и нивоа инвазивности. Супресија раста ће- лија меланома је била последица губитка деобног потенцијала, увођења ћелија у стање сенесценције, односно ћелијског мировања и интензивиране меланоцитне диференцијације. У сингеном мишјем моделу, удруженим деловањем 3HFWC и хиперполаризоване светлости је значајно инхибиран раст меланома као последи- ца директног деловања на туморске ћелије али и модулације имунског одговора усмереног на/против тумора. Kомплексан механизам деловања који обједињује фенотипску трансформацију ћелија меланома и ефекте на имунски сyстем чини хиперхидроксиловани фулерен вредним даљег истраживања. | sr |
