Determination of phenolics in four fern species: a comparative analysis of sporophytes and gametophytes

Like all vascular plants, the fern life cycle alternates between two distinct phases: a diploid sporophytic and a haploid gametophytic, the former representing the asexual, spore-producing phase and the later the sexual, gamete-producing phase of the life cycle. In ferns and other primitive plants, the haploid gametophytic phase, from spore germination to fertilization of the gametes, is completely independent of the diplod sporophytic plant. Since ferns are peculiar in having independent sporophytic and gametophytic generations, there is a good chance for distinctive screening of bioactivity and bioactive compounds both in sporophytic and gametophytic plant bodies. In the present investigation, methanolic extract of gametophytes grown in vitro and aerial parts of the mature sporophytes (fronds) were identified and quantified using HPLC-PDA-MS and UHPLC-PDN-HESI-MS/MS analysis. Bioactive components of ferns mainly belong to the phenolic, flavonoid, alkaloid and terpenoid families, with quite different chemical characteristics of two life cycles. Although class and amount of identified substances varied between investigated fern species, an interesting phenomenon was observed: simple phenylpropanoids, like caffeic acid and its derivates (in particular quinate ester of caffeic acid-chlorogenic acid) were dominant phenolic components in sporophytes, while the most abundant phenolic compounds in gametophytic tissue were flavanol catechine and its derivatives. These results indicate that fern sporophytes and gametophytes have distinct bioactive molecules, and therefore different strategies to cope with their environment.
