Antioxidative enzymes responses during genetic transformation of Impatiens walleriana L. shoots

The antioxidative enzymes in Impatiens walleriana were determined during genetic transformaion with Agrobacterium rhizogenes A4M70GUS. All shoots infected with that bacteria produced hairy roots in wounded region after 10 days of inoculation. Fifty-five putative transformed hairy root lines from independent transformation events were established. Integration of uidA, rolA, rolB, rolC and rolD genes within I. walleriana L. genome was confirmed by PCR in twelve lines. I. walleriana hairy root lines on Murashige and Skoog (MS) medium without plant growth regulators exhibited elevated rate of biomass accumulation, up to three times more compared to the untransformed control roots. Also, we investigated changes of reactive oxygen species(ROS) in wounded region of stems and leaves before transformation and 10, 24,240 and 720 hours following inoculation. The lowest catalase (CAT) and peroxidase (POX) activities were observed at 10 h after inoculation of I. walleriana with A. rhizogenes in both parts of plant. However in wounded region superoxide dismutase (SOD) activity increased significantly 10 h following inoculation and after this time decreased, whereas in leaves SOD activity increased gradually with time. In all samples Mn-SOD, Fe-SOD and CuZn-SOD isoforms were detected. In wounded region of stems 720 hours after inoculation displayed an extra Mn-SOD isoform. These results indicate that antioxidative enzymes plays important role during transformation.The balance between ROS removal and biosynthesis is plant response during Agrobacterium-mediated transformation.
