Hesperetin affects some antioxidative enzymes activity in the liver of old-aged Wistar rats

Age-related decline in liver functioning is associated with increased free radical generation. If free radicals are not rapidly removed, accumulation of damaged products causes cellular senescence. Hesperetin, a flavanone most abundant in citrus fruits, was identified as the promising phyto-therapeutic agent due to its putative anti-oxidative properties. Therefore, the aim of this study was to test whether hesperetin affects liver antioxidant enzymes (AOE), namely, superoxide dismutase 1 and 2 (SOD 1 and 2), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) activities, gene and protein expressions in rat ageing model. Twenty-four-month-old rats received per os hesperetin mixed with sunflower oil (vehicle), at a dose of 15 mg/kg, once daily, for 4 weeks. Controls received the same amount of vehicle alone. The mixture was applied to the oral cavity, in order to achieve more nutritionally relevant study. Our results demonstrated that after hesperetin supplementation, activities of CAT and GR were increased by 56% and 28% (p<0.05), respectively. The protein expressions of GPx and SOD2 were significantly decreased by 28% and 51 % (p<0.05), respectively, while just CAT gene expression was increased by 2.9 fold (p<0.05) in comparison with the values obtained for controls. These results suggest that hesperetin treatment affect some AOE in the livers of old-aged rats. It had strongest effect on CAT by increasing its activity and gene expression, without more prominent effect on other evaluated AOE. Decreased or unchanged values obtained for other AOE might be outcome of hesperetin antioxidant and free radical scavenging properties in the livers of rat ageing model.
