AB  - Invasive alien fish species represent one of the greatest threats to the
biodiversity and ecological status of freshwater ecosystems. Within their
management, their application as bioindicators of pollution in monitoring
programs should be considered. The aim of this study was to examine the
use of black bullhead Ameiurus melas (Rafinesque, 1820), invasive alien fish
in Serbia, as a bioindicator of water genotoxicity by alkaline comet assay.
Sampling was performed from June to September 2021 on Markovačko
Lake, which is exposed to diffuse pollution from the application of pesticides
on the surrounding agricultural land. Monitoring also included the
assessment of water quality by basic physical and chemical parameters, as
well as microbiological indicators of faecal pollution. Alkaline comet assay
was performed on fish erythrocytes, on ten specimens per month. The study
also included ten control fish from the recirculating aquaculture system
(RAS), which were transferred from Markovačko Lake for breeding, and
treatment of their erythrocytes with H2O2 as a positive control. Scoring of
comets was performed by CometScore 2.0 software. The lowest level of
DNA damage was recorded in the control group and the highest in the
positive control group. August was the month with the lowest level of DNA
damage, and September with the highest. Statistically significant
differences were found between all groups indicating a high sensitivity of
the alkaline comet assay to discriminate fine differences in the genotoxic
potential of water. Knowing that black bullhead is highly abundant in
stagnant waters and relatively easy to sample, this study draws additional
attention to its use as a bioindicator of genotoxicity.

AB  - The middle section of the Danube River basin in the previous three Joint
Danube Surveys (JDS) was found to be under intensive pollution, mainly
affected by wastewaters. As a consequence of the lack of wastewater
treatment plants, complex xenobiotics mixtures may get into surface water
and decrease water quality and organisms’ health. Some xenobiotics may
induce different types of DNA damage. Hence, DNA damage could be a
useful biomarker in the detection of “early warning signals” of organisms’
exposure to genotoxic compounds, while the usage of bioassays batteries
can provide better insight into a genotoxic potential. This study was
conducted in 2019, during the JDS4 expedition, to assess the genotoxic
potential of nine sites on the section of the Danube River which goes
through Serbia and its major tributaries: the Sava, Tisza and Velika Morava.
Blood and muscle of Alburnus alburnus (bleak) were used for genotoxicity
assessment by comet, micronucleus and RAPD assays. In comet assay, the
highest level of DNA damage was recorded at the Velika Morava mouth (JDS
39) and on the Danube River, downstream Radujevac (JDS 41). The highest
frequency of micronucleus was recorded at the Sava mouth (JDS 36) and on
the Danube River, downstream Pančevo (JDS 37). The lowest level of DNA
damage in both tests was recorded on the Sava, site Jamena (JDS 35). The
RAPD analysis distinct three major groups: Tisza mouth (JDS 33) and sites
on the Sava (JDS 35, JDS 36), then two sites on the Danube River (JDS 37,
JDS 41) and one site on the Danube River, Ram (JDS 40) with the Velika
Morava mouth (JDS 39). Analyzed bioassays showed different sensitivity
where the comet assay had the highest potential in discrimination of sites
based on genotoxicity. Bleak was proved to be a reliable bioindicator in eco/
genotoxicological studies.

AB  - Within the past decade, our research group placed great effort in
exploration of the impact of treated/untreated wastewaters on aquatic
ecosystems as less than 13 % of collected municipal wastewaters are
processed before their release to receiving waters in Serbia. Having in mind
that more than 90% of the rivers within the territory of Serbia belong to the
Danube River Basin, our major focus is naturally on the large transnational
waters such as the Danube and Sava rivers. Depended on the concept of the
study and hydro-morphological characteristics of the sites, various
approaches have been employed for in situ assessment of eco/geno-toxicity
which use different organisms in passive and active biomonitoring. Aquatic
organisms of different trophic levels have diverse life strategies, metabolism
pathways, and consequently, they have a different response to pollutant
pressure. In eco/geno-toxicological studies we have successfully employed
several species of freshwater mussels (Unio tumidus, Unio pictorum and
Sinanodonta woodiana) as well as few economically important fish species
(common carp (Cyprinus carpio), freshwater bream (Abramis brama), bleak
(Alburnus alburnus, chub (Squalius cephalus)).Comet assay is our primary
choice for the assessment of DNA damage considering its sensitivity and
cost effectiveness. Selection of certain types of tissues for the assessment
(such as blood or haemolymph), which do not require sacrificing of the
animals or additional manipulation in preparation of cell suspension coupled
with mini-gel format of the slides enables high-throughput screening of the
genotoxic potential in situ. Additionally, cryopreservation of the blood
samples directly onsite expands the research area as it overcomes issues
related to safe transportation of the samples to the laboratory. Data
obtained so far indicated that the genotoxic response in the studied
indicator organisms significantly differ which is understandable considering
the difference in uptake, accumulation and physiological responses.

AB  - U Srbiji se prerađuje manje od 13% otpadnih voda,1 što za posledicu ima
ispuštanje velikih količina neprečišćenih voda u vodotokove. Ravničarske reke
su pod značajnim pritiscima ovih izvora zagađenja, s obzirom da delom ili
čitavim tokom protiču kroz naseljene oblasti, kao i oblasti u kojima se nalaze
industrijska postrojenja. U analizi uticaja zagađenja na akvatične organizme,
oštećenje DNК molekula predstavlja značajan biomarker ranog upozorenja jer
može ukazati na uticaj ksenobiotika na biološke sisteme pre nego što se odrazi
na čitav organizam i/ili populaciju. U skladu sa tim, cilj ovog istraživanja je
procena osetljivosti široko rasprostranjene vrste Alburnus alburnus (uklija)
prilikom detekcije genotoksičnog efekta, kako na prostorno bliskim
lokalitetima na Savi, tako na i prostorno udaljenim na Dunavu, Tisi, Savi i
Velikoj Moravi. Za poređenje lokaliteta korišćen je integrisani odgovor
biomarkera (IBR) koji je objedinio pojedinačne biomarkere: vijabilnost ćelija
krvi, nivo DNК oštećenja ćelija krvi detektovanog u alkalnom komet i
mikronukleus testu i indeks zagađenja metalima/prioritetnim supstancama.
Integrisanjem pojedinačnih odgovora biomarkera u jedinstvenu IBR vrednost
omogućeno je jednostavnije poređenje lokaliteta. Dobijeni rezultati su ukazali da
su odgovori biomarkera uklija specifični za lokalitet. U skladu sa rezultatima,
utvrđeno je da uklija može biti pouzdan bioindikator u diskriminaciji i
udaljenih i prostorno bliskih lokaliteta koji su pod različitim uticajem
zagađenja.
AB  - У Србији се прерађује мање од 13% отпадних вода,1 што за последицу има
испуштање великих количина непречишћених вода у водотокове. Равничарске реке
су под значајним притисцима ових извора загађења, с обзиром да делом или
читавим током протичу кроз насељене области, као и области у којима се налазе
индустријска постројења. У анализи утицаја загађења на акватичне организме,
оштећење ДНК молекула представља значајан биомаркер раног упозорења јер
може указати на утицај ксенобиотика на биолошке системе пре него што се одрази
на читав организам и/или популацију. У складу са тим, циљ овог истраживања је
процена осетљивости широко распрострањене врсте Alburnus alburnus (уклија)
приликом детекције генотоксичног ефекта, како на просторно блиским
локалитетима на Сави, тако на и просторно удаљеним на Дунаву, Тиси, Сави и
Великој Морави. За поређење локалитета коришћен је интегрисани одговор
биомаркера (ИБР) који је објединио појединачне биомаркере: вијабилност ћелија
крви, ниво ДНК оштећења ћелија крви детектованог у алкалном комет и
микронуклеус тесту и индекс загађења металима/приоритетним супстанцама.
Интегрисањем појединачних одговора биомаркера у јединствену ИБР вредност
омогућено је једноставније поређење локалитета. Добијени резултати су указали да
су одговори биомаркера уклија специфични за локалитет. У складу са резултатима,
утврђено је да уклија може бити поуздан биоиндикатор у дискриминацији и
удаљених и просторно блиских локалитета који су под различитим утицајем
загађења.

abstract = "U Srbiji se prerađuje manje od 13% otpadnih voda,1 što za posledicu ima
ispuštanje velikih količina neprečišćenih voda u vodotokove. Ravničarske reke
su pod značajnim pritiscima ovih izvora zagađenja, s obzirom da delom ili
čitavim tokom protiču kroz naseljene oblasti, kao i oblasti u kojima se nalaze
industrijska postrojenja. U analizi uticaja zagađenja na akvatične organizme,
oštećenje DNК molekula predstavlja značajan biomarker ranog upozorenja jer
može ukazati na uticaj ksenobiotika na biološke sisteme pre nego što se odrazi
na čitav organizam i/ili populaciju. U skladu sa tim, cilj ovog istraživanja je
procena osetljivosti široko rasprostranjene vrste Alburnus alburnus (uklija)
prilikom detekcije genotoksičnog efekta, kako na prostorno bliskim
lokalitetima na Savi, tako na i prostorno udaljenim na Dunavu, Tisi, Savi i
Velikoj Moravi. Za poređenje lokaliteta korišćen je integrisani odgovor
biomarkera (IBR) koji je objedinio pojedinačne biomarkere: vijabilnost ćelija
krvi, nivo DNК oštećenja ćelija krvi detektovanog u alkalnom komet i
mikronukleus testu i indeks zagađenja metalima/prioritetnim supstancama.
Integrisanjem pojedinačnih odgovora biomarkera u jedinstvenu IBR vrednost
omogućeno je jednostavnije poređenje lokaliteta. Dobijeni rezultati su ukazali da
su odgovori biomarkera uklija specifični za lokalitet. U skladu sa rezultatima,
utvrđeno je da uklija može biti pouzdan bioindikator u diskriminaciji i
udaljenih i prostorno bliskih lokaliteta koji su pod različitim uticajem
zagađenja., У Србији се прерађује мање од 13% отпадних вода,1 што за последицу има
испуштање великих количина непречишћених вода у водотокове. Равничарске реке
су под значајним притисцима ових извора загађења, с обзиром да делом или
читавим током протичу кроз насељене области, као и области у којима се налазе
индустријска постројења. У анализи утицаја загађења на акватичне организме,
оштећење ДНК молекула представља значајан биомаркер раног упозорења јер
може указати на утицај ксенобиотика на биолошке системе пре него што се одрази
на читав организам и/или популацију. У складу са тим, циљ овог истраживања је
процена осетљивости широко распрострањене врсте Alburnus alburnus (уклија)
приликом детекције генотоксичног ефекта, како на просторно блиским
локалитетима на Сави, тако на и просторно удаљеним на Дунаву, Тиси, Сави и
Великој Морави. За поређење локалитета коришћен је интегрисани одговор
биомаркера (ИБР) који је објединио појединачне биомаркере: вијабилност ћелија
крви, ниво ДНК оштећења ћелија крви детектованог у алкалном комет и
микронуклеус тесту и индекс загађења металима/приоритетним супстанцама.
Интегрисањем појединачних одговора биомаркера у јединствену ИБР вредност
омогућено је једноставније поређење локалитета. Добијени резултати су указали да
су одговори биомаркера уклија специфични за локалитет. У складу са резултатима,
утврђено је да уклија може бити поуздан биоиндикатор у дискриминацији и
удаљених и просторно блиских локалитета који су под различитим утицајем
загађења.",

AB  - This study was conducted on the Danube River locality Višnjica, exposed to the discharge of the largest wastewater collector in Serbia's capital, Belgrade. Concentrations of metals and metalloids (Al, Fe, Cr, As, Sr, Mn, Cd, Zn, Mo, Cu, Li, Ni, B, Co, Pb, and Ba) and histopathological alterations were investigated in different tissues of common bream during one representative month of each season in 2014. This is the first study in which these two biomarkers were assessed parallelly in common bream. The highest concentrations of examined elements were noticed in gills and the lowest in muscle. Statistically significant differences in element concentrations between different seasons were noticed only in gills for Al, Cu, and Fe. Concentrations of As and Pb in fish muscle were below the maximum acceptable concentrations (MAC). The histopathological index (HI) of the liver showed higher values in comparison to the HI of the gills. Histopathological index of the gills had a significantly higher score in November in comparison to August. The liver HI had the highest score in April, and the lowest in August, while the total histopathological index had the highest score in November, and the lowest in August, both without significant differences between the months. This study endorses gills and liver as reliable organs for studying accumulation and histopathology as biomarkers of environmental changes. A faster reaction of the gills was confirmed since seasonal variations of both biomarkers were observed in this organ. Common bream proved as a good indicator of the state of organisms in polluted environments.

AB  - Water quality in the Danube River Basin is largely influenced by the inputs of pollutants - particularly
excessive nutrients, organic material, and hazardous substances, which affects the whole ecosystem
at various levels. The major goal of this study was to evaluate the level of genotoxic potential at the
site at Danube River, Novi Banovci, which is under significant anthropogenic pressure of untreated
municipal and industrial waste waters.
DNA damage in situ was assessed in 5 specimens of white bream (Blicca bjoerkna) erythrocytes, liver
and gills cells with comet assay and in erythrocytes with micronucleus test. Additionally, genotoxicity
of native water samples collected from the site and upstream and downstream of the site was
evaluated by the comet assay on HepG2 cells. Microscopic images of comets were scored using Comet
IV Computer Software (Perceptive Instruments, UK) and tail intensity was chosen as parameters to
assess the DNA damage.
Comet assay on white bream showed that gills were the most affected tissue, while liver showed the
smallest values. Micronuclei frequency was low in general, with mean of 1 micronuclei in 5000 scored
erythrocytes, with no significant correlation to the values of comet assay in erythrocytes. Comet assay
on HepG2 cells showed significant difference between values of negative control samples and samples
of water from effluent site and downstream of wastewater discharge. There was no significant
difference between negative control and upstream samples of water.
Our results showed that great impact of municipal and industrial wastewaters on the river ecosystem.
The application of in vitro and in situ tests gives a better insight into genotoxic effects. More
importantly, because Serbia still doesn’t have waste water treatme nt plants, this kind of research
points out the imperative for implementation of this facilities.

AB  - In this study, few different evaluation concepts were used for the assessment of genotoxic potential at the stretch of the Danube River identified as a significant hotspot of pollution originated through the untreated wastewaters. Three sites were chosen: one site upstream of the wastewater outlet in Novi Sad (Serbia), one at the outlet of wastewaters, and one site few kilometer downstream. Ex situ approach comprised prokaryotic SOS/umuC test on Salmonella typhimurium TA1535/pSK1005 and comet assay on human hepatoma cell line (HepG2). In situ approach was based on the active monitoring (cage approach) using freshwater mussels Sinanodonta woodiana and fish Cyprinus carpio. The comet and micronucleus assays were selected for evaluation of DNA damage in mussel haemocytes and fish blood cells. Within the ex situ part of the study, our results indicated that the eukaryotic model system is more sensitive compared to the prokaryotic one. In situ bioassays are recommended for obtaining a better insight into ecosystem status and in the case of our study the complete insight of genotoxic pressure. However, the choice of animals as bioindicators also has a significant impact on the quality of the obtained information. Differential response between fish and mussels was observed at the highly polluted site suggesting possible involvement of additional protective mechanism such as valve closure in mussels.

AB  - Tert-butylquinone (TBQ) and its alkylamino and aralkylamino derivatives are of high interest as a potential antitumor agent. Therefore, it was necessary to investigate if the compounds exert undesirable activities such as interaction with DNA molecule which could result in negative side effects in the case of their use in the diseases treatment. The major aim of this study was to investigate genotoxic potential of TBQ and selected derivatives in an acellular model by using plasmid DNA, in the prokaryotic model by the SOS/umuC assay in Salmonella typhimurium TA1535/pSK1002 and in eukaryotic models by using comet assay in human fetal lung cell line (MRC-5) and human liver cancer cell line (HepG2). Results indicated that in the acellular model TBQ and its derivatives do not interact with plasmid pUC19. In the prokaryotic model, only TBQ exerted weak genotoxic potential and only at highly cytotoxic concentrations. In eukaryotic models, genotoxic potential was detected mainly at the highest concentrations of the tested substances but the effect was lower in both cell lines in comparison with benzo[a]pyrene and etoposide which were used as positive controls. Weak genotoxic potential of tested compounds recommends them as good candidates for further testing in development of new antitumor agents.

To emphasize the impact of wastewaters, this study was carried out in a season with low water discharge. The level of genotoxic potential at the sites was studied by alkaline and Fpg-modified comet assay in fish blood, while RAPD technique was employed on the fish muscle tissue. The variations in the genotoxic potential were observed along the Sava River based on the level of DNA damage in blood of the specimens collected at the selected sites. Clustering of the impacted sites was also observed based on RAPD fingerprints. Comparison of the data obtained in RAPD and the data from the comet assay indicated the possible reflection of genotoxicity on the genetic variability in chub collected from the sites with different pollution pressure. This is a preliminary study which reports the differentiation of the population of chub along the Sava River based on RAPD analysis.
