AB  - Uvod. Faktori sredine, kao što je visoka ili niska temperatura ambijenta, predstavljaju snažne stresore za organizam. Hipofizne ćelije koje luče hormone, posebno somatotropne (GH), intenzivno reaguju na ove faktore. Cilj ove studije bio je da ispita efekat dugotrajnog (60-dnevnog) izlaganja pacova umereno visokoj temperaturi ambijenta (35±1°C) na imunohistomorfometrijske parametre GH ćelija hipofize.
Metode. Eksperiment je izveden na odraslim Wistar pacovima, podeljenim u dve jednake grupe: sedam pacova je kontinuirano bilo izloženo temperaturi 35± l°C tokom 60 dana (eksperimentalna grupa), dok je sedam pacova tokom istog perioda čuvano na 20±2°C (kontrolna grupa). Sve su životinje čuvane u režimu od 12 sati dnevne svetlosti i 12 sati mraka, dok su hranu i vodu mogle da uzimaju ad libitum. GH ćelije vizuelizovane su pomoću imunohistohemijske procedure peroksidaze-antiperoksidaze (PAP). Morfometrijska analiza izvedena je korišćenjem višenamenskog M42 test-sistema.
Rezultati. Nakon 60 dana izlaganja povišenoj ambijentalnoj temperaturi, telesna masa životinja bila je značajno smanjena za 16,4%, u poređenju sa kontrolama. GH ćelije u kontrolnoj i eksperimentalnoj grupi bile su ovalnog oblika i locirane u blizini kapilara. Tamne granule, distribuirane po citoplazmi GH ćelija, bile su sebi svojstvenih univer­zalnih karakteristika u obe grupe. Kod eksperimentalnih pacova, celularni i nukleami volumeni GH ćelija, kao i volumenska gustina GH ćelija, značajno su redukovani za 19,1%, 10,6% i 15,8%, redom, u poređenju sa intaktnim kontrolama.
Zaključak. Izlaganje odraslih pacova muškog pola umereno visokoj ambijentalnoj temperaturi u trajanju od 60 dana dovelo je do smanjenja histomorfometrijskih parametara GH ćelija.

Corticosteroid secretion after the soy extract application to orchidectomized adult male rats

AB  - Herein, we have tested the effects of application of commercial soy extract, rich in estrogen-like isoflavones, to orchidectomized adult rats (the model of mild andropause), on the corticosteroid secretion. Exploited animal model provides the opportunity to examine the potential effects of steroid-like compounds in the hormonal milieu deprived of endogenous sex steroids along with preserved hippocampal and hypothalamic regulation of the adrenal gland function. The experimental groups included sham-operated (SO), orchidectomized (Orx) and soy extract-treated orchidectomized (Orx + Soy; 30 mg/kg b.m. s.c., for 3 weeks) rats. Plasma level of ACTH and serum levels of aldosterone, corticosterone and DHEA were determined by the immunoassays. Orchidectomy caused the increase (P<0.05) of circulating ACTH, aldosterone and DHEA by 57.0%. 2.6 and 2.0 folds respectively, compared to SO group. In Orx + Soy group, blood levels of ACTH and corticosterone were higher (P<0.05) by 99.5% and 79.6% respectively, in comparison with SO group. Also, the treatment of orchidectomized rats with soy extract elevated (P<0.05) the systemic levels of ACTH, aldosterone and corticosterone by 26.9 %, 2.7 folds and 56.4% respectively, while the circulating DHEA level was decreased (65.6%; P<0.05), all compared to the corresponding parameters in Orx rats. Summarily, soy extract raised the ACTH level which has stimulatory influence on the mineralocorticoid and glucocorticoid output, while the adrenal androgens consequently fallen, in the rat model of mild andropause. The data support impression of a beneficial effect of soy isoflavone mixture on the homeostatic response to stress, but also call for precautions in the case of cardiovascular issue presence.

AB  - Advanced age is frequently entwined with the adrenal cortex function impairment, glucocorticoid excess and following metabolic syndrome.
Also, age-dependent
disturbancies in aldosterone secretion and Na*/K homeostasis at least partly underlie the hypertension issues. Classical therapeutic approach in the treatment of some menopausal symptoms implies administration of estradiol or calcium, or their combination. Herein, we aimed to investigate the effects of chronic treatment with estradiol and calcium on some adrenal gland parameters in the ageing female rat model. Middle-aged (14-month-old) female Wistar rats were treated with estradiol dipropionate (EDP; 0.625 mg/kg b.m.) and calcium glucoheptonate (Ca; 11.4 mg/kg b.m.) daily for two weeks, while control ruts received vehicle alone by the same schedule. Combined treatment caused the decrease (p<0.05) of body mass by 21.5%, while the absolute and relative adrenal gland weights were increased (p<0.05) by16.2% and 43.8% respectively, compared to the adequate control values. The absolute adrenal gland volume, after treatment with EDP and Ca in the combination, was also increased (12.1%; p<0.05), in comparison with control group.
In parallel with that, circulating aldosterone was elevated (p<0.05) by 50.0%, compared to the control levels. However, combined treatment with EDP and Ca insignificantly (P>0.05) affected Na* and K' blood concentrations. It can be concluded that simultaneous estradiol ants calcium administration to middle-aged female rats enlarges the adrenal glands and have stimulatory influence on the mineralocorticoid output, which may represent unwanted effect from the perspective of blood pressure regulation.

AB  - Introduction: Genistein is widely used as an alternative therapeutic for various pathological conditions in ageing male and exerts an inhibitory effect on the steroidogenic enzyme families, thus affecting hormonal homeostasis. The aim of this study was to examine the effects of genistein on histological features of the adrenal cortex zona reticularis and the following dehydroepiandrosterone (DHEA) secretion in orchidectomized adult male rats, the most accessible model reflecting andropause.
Material and Methods: The experimental adult Wistar male rats were divided into three groups (n=7): sham-operated (SO), orchidectomized (Orx) and genistein-treated orchidectomized (Orx+G) groups. Genistein (30 mg/kg b.m./day) was administered subcutaneously for three weeks, while the control groups received the vehicle alone. All animal were sacrificed by decapitation 24h after the last injection. The adrenal cortex was analyzed histologically, while serum concentration of DHEA was determined by immunoassay.
Results: Histological analysis revealed that there were no significant changes in adrenal cortex zona reticularis after orchidectomy and orchidectomy followed with genistein treatment. Circulating DHEA concentration was significantly (p<0.005) increased by 97.2% in Orx animals compared to the SO group. In Orx+G group serum DHEA was significantly (p<0.005) decreased by 41.5% in comparison with Orx rats. Serum concentration of DHEA upon genistein application to orchidectomized animals was insignificantly (p>0.05) changed by 15.3% when compared to the SO group.
Conclusion: These data, observed in orchidectomized adult rats, indicate that genistein has an inhibitory effect on the adrenal cortex zona reticularis secretory function.

Material and Methods: The experimental adult Wistar male rats were divided into three groups (n=7): sham-operated (SO), orchidectomized (Orx) and genistein-treated orchidectomized (Orx+G) groups. Genistein (30 mg/kg b.m./day) was administered subcutaneously for three weeks, while the control groups received the vehicle alone. All animal were sacrificed by decapitation 24h after the last injection. The adrenal cortex was analyzed histologically, while serum concentration of DHEA was determined by immunoassay.
Results: Histological analysis revealed that there were no significant changes in adrenal cortex zona reticularis after orchidectomy and orchidectomy followed with genistein treatment. Circulating DHEA concentration was significantly (p<0.005) increased by 97.2% in Orx animals compared to the SO group. In Orx+G group serum DHEA was significantly (p<0.005) decreased by 41.5% in comparison with Orx rats. Serum concentration of DHEA upon genistein application to orchidectomized animals was insignificantly (p>0.05) changed by 15.3% when compared to the SO group.
Conclusion: These data, observed in orchidectomized adult rats, indicate that genistein has an inhibitory effect on the adrenal cortex zona reticularis secretory function.",

AB  - Thyroid C-cells produce calcitonin (CT), a hypocalcemic hormone, that
   acts as an inhibitor of bone resorption. In this study, we investigated
   the effects of tamoxifen (TAM) as a selective estrogen receptor
   modulator on thyroid C-cells, trabecular bone and biochemical markers of
   bone metabolism in an animal model of androgen deficiency, represented
   by middle-aged orchidectomized (Orx) rats. Fifteen-month-old male Wistar
   rats were divided into: Orx and sham-operated (SO) groups. Rats from one
   Orx group were injected subcutaneously with TAM citrate (Orx + TAM; 0.3
   mgkg(-1) b.w.), while the rats from SO and a second Orx group received
   vehicle alone, once a day for 3 weeks. The peroxidase-antiperoxidase
   method was applied for localization of CT in C-cells. Thyroid C-cells
   were morphometrically and ultrastructurally analyzed. An ImageJ
   image-processing program was used to measure bone histomorphometric
   parameters. Blood serum samples were analyzed for CT, osteocalcin (OC),
   calcium (Ca2+) and phosphorus (P). Urinary Ca2+ concentrations were
   measured. TAM treatment significantly increased thyroid C-cell volume
   (V-c) and serum CT when compared with vehicle-treated Orx rats. Analysis
   of trabecular microarchitecture of the tibia showed that administration
   of TAM significantly increased cancellous bone area, trabecular
   thickness and trabecular number, whereas trabecular separation was
   significantly decreased compared with vehicle-treated Orx rats. Serum OC
   and urinary Ca2+ concentrations were significantly lower in comparison
   with the control Orx group. These results indicate that in our rat model
   of androgen deficiency, TAM stimulated calcitonin-producing thyroid
   C-cells and increased trabecular bone mass.

The stimulatory effect of moderate heat on rat pituitary ACTH cells: histological and hormonal study

AB  - Pituitary ACTH producing cells are very sensitive towards changes in duration of the heat exposure. The aim of this study was to examine the effects of 4-day continuous exposure of rats to moderate heat on histological and secretory characteristics of ACTH cells in adult male rats. The experimental group was exposed to 35±1°C for 4 days, whereas the control group was kept at room temperature during the same period. ACTH cells were studied using the immunohistochemical and immunofluorescent procedures. The morphometry was conducted using M42 multipurpose test system, while the hormone concentrations were determined by ACTH IMMULITE assay and corticosterone ELISA test. Compared to the controls, a significant (p<0.05) decrease of the body weight by 19.8% and significant (p<0.05) increase of the absolute and relative volumes of the pituitary gland by 23.1% and 36.1% respectively, were registered. It was found that in experimental group neither the localization nor the shape of ACTH cells was significantly changed, but the intensity of immunofluorescence signal was significantly (p<0.05) decreased. The cellular and nuclear volumes of ACTH cells were increased (p<0.05) by 6.9% and 14.3% respectively, while the volume density of ACTH cells was decreased (p<0.05) by 20.0%, in comparison with the corresponding controls. The observed changes were in accordance with increased (p<0.05) values of blood ACTH and corticosterone concentrations by 23.7% and 11.8%, respectively. Based upon the histological and hormonal data, it can be concluded that 4-day continuous exposure to moderate heat causes stimulation of the pituitary ACTH cells in rats.
