Phlomis Fruticosa L. Exerts in Vitro Antineurodegenerative and Antioxidant Activities and Induces Prooxidant Effect in Glioblastoma Cell Line

AB  - Despite the significant advances in drug development we are witnessing the inability of health systems to combat both neurodegenerative diseases and cancers, especially glioblastoma. Hence, natural products are comprehen-sively studied in order to provide novel therapeutic options. This study aimed to explore anti-neurodegenerative and anti-glioblastoma potential of extract of Phlomis fruticosa L. using in vitro model systems. It was found that the methanol extract of P. fruticosa was able to efficiently reduce activities of enzymes linked to neurodegenera-tive disease including acetylcholinesterase, butyrylcholinesterase and tyrosinase. Furthermore, P. fruticosa extract has shown excellent antioxidant potential, as evidenced by six different methods. Analysis of cytotoxic effect of P. fruticosa extract on A172 glioblastoma cell line revealed that the concentration of the extract necessary for 50 % inhibition of A172 growth (IC50) was 710 μg/mL. The extract did not induce changes in proliferation and morphology of A172 glioblastoma cells. On the other side, production of ROS was increased in A172 cells treated with the extract. Observed cytotoxic effect of P. fruticosa extract might be based on increase in ROS generation upon treatment. Quantitative chemical analysis revealed the presence of twelve different polyphenols with the cis 3-O-caffeoylquinic acid being the most abundant. This study provided scientific evidence for further exploration of P. fruticosa as a promising natural anti-neurodegenerative therapeutic option.

Bioactivities of Salvia nemorosa L. inflorescences are influenced by the extraction solvents

AB  - Salvia nemorosa L. is a rich source of bioactive constituents. We have used different solvents to obtain 14 different extracts in order to evaluate solvents impact on S. nemorosa inflorescences bioactivities. Firstly, the extraction yield, chemical composition and cytotoxic properties towards HaCaT cell line were elaborated. Afterwards, we have studied the extracts bioactivity in terms of wound healing, antioxidant and antidiabetic potentials as well as antimicrobial and antibiofilm capacities; followed by the determination of the antimicrobial mechanism. The extracts yield ranged from 3.03% to 15.75% and they were dominated by epigallocatechin gallate and rosmarinic acid. The majority of extracts did not possess cytotoxic properties towards human keratinocytes. Butanol was the most appropriate solvent for the improvement of wound healing capacity. Combined extraction with ethanol and water resulted in the highest antioxidant potential of the extracts that could be attributed to the high abundance of detected polyphenols. Extraction with butanol led to the increased inhibition towards enzymes linked to type 2-diabetes and increase in antimicrobial capacities. The extracts have shown antibiofilm potential towards Staphylococcus lugdunensis biofilms with more than 90% inhibition for the most active ones. The selected extract EB7 reduced bacterial ability to adhere to human keratinocytes and provided immunomodulatory action in HaCaT cells exposed to S. lugdunensis. The antibacterial mechanism of action of the same extract was associated with moderate perturbation in the cell membrane integrity and with the reduction in bacterial mitochondrial activity. S. nemorosa inflorescences were abundant sources of bioactive polyphenols and with different extraction solvents their bioactivities were further improved. This study reported for the first time the effects of extraction solvents on various bioactivities of S. nemorosa inflorescences, supporting S. nemorosa role as a natural bioactive therapeutic agent with possible antioxidant, antidiabetic and antimicrobial applications.

Chemical profiling, antimicrobial, anti-enzymatic, and cytotoxic properties of Phlomis fruticosa L.

AB  - Structural diversity of biologically active compounds identified in plants after many years of storage is rarely reported in literature. Herein, we studied chemical profile and biological activities of Phlomis fruticosa L. after plant material storage for 20 years. Chemical analyzes were performed by UHPLC-LTQ-Orbitrap/MS, and revealed presence of 44 compounds: including 13 phenolic acids, 9 phenylethanoids, 20 flavonoids and 2 phenolic related compounds (a phenolic acid derivative and an aliphatic alcohol). The extract showed antimicrobial activity, being the most potent against Aspergillus fumigatus with minimum inhibitory concentration of 0.31 mg/mL. Also, the extract was able to inhibit biofilm formed by Candida species and to inhibit biofilm formation by Staphylococcus aureus. Obtained results revealed that the extract has potential to interfere with the cell membrane permeability of Candida albicans and to suppress production of virulence factor staphyloxanthin in S. aureus. Furthermore, the extract inhibited the activity of α-amylase which is one of the therapeutic targets for diabetes type II. Also, the antiproliferative effect of the extract was demonstrated on human cancer cell lines, while the extract did not exhibit any cytotoxic effect on primary human cells. Based on the obtained results, P. fruticosa could be an interesting source of biologically active compounds even after long term storage.

Prunus spinosa L. leaf extracts: polyphenol profile and bioactivities

The richest in phenols as well as in flavonoids was acetone extract with 181.19 mg GAE and 80.10 mg QE per gram of dry extract, respectively. Moreover, the quantity of anthocyanins obtained by HPLC analysis was also the highest in acetone sample. Examined samples possessed antioxidant properties evaluated through four in vitro assays (DPPH, ABTS, FRAP and TRC). The acetone extract was proved to be the best antioxidant among tested samples, which could be ascribed to polyphenols, especially anthocyanins. The aqueous and the ethanol extract exhibited antibacterial effects, being particularly active against B. cereus and E. cloacae. T. viride, P. funiculosum, P. ochrochloron, P. verrucosum var. cyclopium were the most susceptible among fungal microorganisms examined. Both, the aqueous and the ethanol extract expressed inhibitory activity towards enzymes linked to diabetes mellitus type II. Additionally, the ethanol extract showed significantly higher potential in inhibiting α-glucosidase than the drug used as the positive control. Furthermore, the aqueous sample revealed antitumor effects on following malignant cell lines: HeLa, K562 and MDA-MB-453. The results presented herein suggest that P. spinosa leaves should be considered as a natural source of bioactive compounds with potential application in phytopharmacy and food industry.

AB  - Anthriscus cerefolium (L.) Hoffm. is a plant traditionally used around the globe since antiquity. Although widely used in many traditional medicines in different cultures, from the scientific point of view it is poorly investigated. Glioblastoma, a tumor type with poor prognosis, is the most common and lethal brain tumor in adults. Current therapeutic strategies for glioblastoma include surgery, radiation and chemotherapy. On the other hand, it has been revealed that patients with cancers are highly susceptible to microbial infections due to the invasive nature of cancer treatment approaches. This study was designed to investigate the chemical profile of herba Anthriscii cerefoli methanolic extract by applying UHPLC-LTQ OrbiTrap MS4 analysis and to analyze its anti-glioblastoma and antimicrobial activities. This study revealed that methanolic extract of herba Anthrisc cerefolii contained phenolic acids and flavonoids, with 32 compounds being identified. Anti-glioblastoma activity was investigated in vitro using A172 glioblastoma cell line. The cytotoxic effects of the extract on A172 cells were compared to the same effect on primary human gingival fibroblast (HGF-1) cells. Decreased rate of proliferation and changes in cell morphology were detected upon treatment of A172 cells with the extract. The antimicrobial activity of extract was tested against Staphylococcus aureus and Candida species. The extract was active against the tested bacterium and yeasts, inhibiting free floating cells and microbial biofilms. This study is the first one to provide a detailed description of the chemical profile of A. cerefolium extract dealing with scientific insights into its anti-glioblastoma and antimicrobial activities.

AB  - This study explored the chemical profile of the aerial parts of Ononis spinosa and further investigated its biological activities. Chemical profiling of the extract revealed the presence of 63 different compounds: phenolic acids, flavonoid glycosides and aglycones, isoflavonoid glycosides and aglycones, and other related compounds. Our results revealed that the extract was active against 8 strains of free floating bacteria. It showed anti-biofilm potential against Staphylococcus aureus and was able to supress the production of staphyloxanthin in S. aureus at sub-minimal inhibitory concentrations. Its antioxidant activity was evaluated by using several assays (phosphomolybdenum, DPPH, ABTS, CUPRAC, FRAP, and metal chelating assay), which showed that the extract exhibited a dose dependent activity. Inhibition of AChE, BChE, amylase, glucosidase and tyrosinase was achieved by the extract, demonstrating its anti-enzymatic activity. The antiproliferative potential of the extract towards human cancer cell lines (HepG2, MCF-7, SiHa and A172) was determined by using the crystal violet assay. Ki67, a marker of proliferation was downregulated in the A172 glioblastoma cell line.

AB  - We investigated the total phenolic and flavonoid contents and the anthocyanin profiles in aqueous, ethanol and acetone extracts of Prunus spinosa (Rosaceae) fruit, and their antioxidant, antibacterial, antifungal, antidiabetic and antitumor properties. The contribution of polyphenol contents to the bioactivity of the extracts was calculated and observed through Pearson’s coefficient of correlation. The acetone extract was the richest in phenols and anthocyanins and the ethanol extract in flavonoids. Cyanidin was the most abundant anthocyanin compound in all examined extracts. The ethanol extract showed the most promising antioxidant activity in DPPH, ABTS and FRAP assays. Tested bacteria were more affected by the ethanol than by the aqueous extract. Both the ethanol and aqueous extracts exhibited potential antidiabetic effects, observed as inhibition of α-amylase and α-glucosidase, enzymes linked with diabetes mellitus type II. The ethanol extract was a potent α-glucosidase-inhibitor with a significantly lower IC50 value than the positive control, glucobay, used to treat diabetes mellitus type II. Neither the ethanol nor the aqueous extracts had any effects on tested human malignant cell lines. Our results indicate that the ethanol extract showed the most pronounced in vitro antioxidant and antimicrobial effects, and a potential antidiabetic activity, which can be ascribed to its high flavonoid content. Our results indicate that research of compounds, particularly of flavonoids present in the ethanol extract and their anti-diabetic properties should be examined further.

AB  - The variability of volatiles isolated from twigs by the static headspace (HS) method in seven natural populations of Picea omorika from Serbia was investigated for the first time. In the overall chemical profile, monoterpenes strongly dominated hydrocarbons as the most volatile compounds (95.7%). The dominant compounds were ?-pinene (29.5 %), ?-pinene (25.7%) and myrcene (13.0%), totaling 68.2% of the volatiles on average. The following nine volatiles were found to be present in medium-to-high amounts (0.5-10%): tricyclene, camphene, ?-phellandrene, ?-3-carene, p-cymene, ?-phellandrene, terpinolene, (E)-caryophyllene, and germacrene D. Out of the 78 volatiles detected, the six most abundant (?-pinene, ?-pinene, myrcene, ?-3-carene, ?-phellandrene and camphene) were selected for principle component analysis (PCA) and cluster analyses (CA). PCA revealed a high degree of similarity between populations, while CA showed a degree of separation of two populations from the others. The presented results are in agreement with previous phytochemical and molecular analyses of this species that confirm high variability in both specialized metabolites and genetic markers.

Eighteen arbitrary RAPD primers produced 123 fragments of which 107 were polymorphic (87%). The similarity coefficient values varied from 0.34 to 0.67. The highest similarity coefficient was detected between Pinus wallichiana and P. strobus as well as between Picea abies and P. orientalis, and the lowest was detected between threePinus species (P. heldreichii, P. peuce and P. wallichiana) and Picea omorika. The analysis of RAPD markers confirmed the genetic relationships among species. GenusPicea is clearly separated from genus Pinus and is closer to genus Abies (A. concolor) than to genus Pinus, what confirms up-to-date numerous comparative-morphological, anatomical, chemotaxonomic and molecular results of these closely related genera. Furthermore, on the basis of our results, pine species from different subgenera -Pinus and Strobus are clearly separated. This statement is in agreement with contemporary intrageneric classification of the genus Pinus.

Osamnaest proizvoljnih RAPD prajmera produkovalo je 123 fragmenata od kojih 107 polimorfnih (87%). Vrednosti koeficijenta sličnosti su varirale od 0.34 do 0.67. Najviši koeficijent sličnosti je detektovan između Pinus wallichiana i P. strobus, kao i između Picea abies i P. orientalis,a najniži između tri vrste roda Pinus (P. heldreichii,P. peuce i P. wallichiana)i Picea omorika. Analiza RAPD markera potvrdila je genetičku vezu između vrsta. Rod Picea se jasno razdvojio od roda Pinus i bliži je rodu Abies (A. concolor) nego rodu Pinus, što potvrđuje dosadašnje brojne uporedno-morfološke, anatomske, hemotaksonomske i molekularne rezultate ovih blisko srodnih rodova. Nadalje, na osnovu naših rezultata, jasno se razdvajaju analizirane vrste borova iz različitih podrodova-Pinus i Strobus, što je, takođe, u saglasnosti sa savremenom intrageneričkom klasifikacijom roda Pinus.

Rod Picea se jasno razdvojio od roda Pinus i bliži je rodu Abies (A. concolor) nego rodu Pinus, što potvrđuje dosadašnje brojne uporedno-morfološke, anatomske, hemotaksonomske i molekularne rezultate ovih blisko srodnih rodova. Nadalje, na osnovu naših rezultata, jasno se razdvajaju analizirane vrste borova iz različitih podrodova-Pinus i Strobus, što je, takođe, u saglasnosti sa savremenom intrageneričkom klasifikacijom roda Pinus.",
