AB  - The widespread use of pesticides and their adverse eff ects (through soil pollution, pest resistance, etc.) have created a need for turning to natural resources in search for new/old methods
of pest control. Essential oils belong to a group of volatile secondary metabolites of plants which
aff ect the growth and development of surrounding plants by spreading around in gaseous state.
The eff ect of an essential oil emulsion of Nepeta rtanjensis Diklić et Milojević on seed germination of redroot pigweed (Amaranthus retrofl exus L.) was tested in this experiment. Dynamic and
total germination of seeds were evaluated after fi ve concentrations of the essential oil emulsion
(ranging 0.006-0.1 %) were applied. Before treatment, seeds were stratifi ed for two and a half
weeks at 3±2 °C. The experiment was carried out in Petri dishes under constant temperature of
30±1 °C. Seeds were germinated in the dark over the initial 24 h and then illuminated during 10
min counting sessions at intervals of 1-3 days. Germination of A. retrofl exus seeds was inhibited
after all tested treatments. The fi nal percentage of germinated seeds treated with the lowest
concentration (0.006%) of N. rtanjensis essential oil did not deviate signifi cantly from data in the
control groups but inhibition through germination dynamic was noted. Seeds did not germinate
at all after receiving the highest concentration (0.1%) of the essential oil.

AB  - The effects of graded sodium chloride (NaCl) concentrations (0-, 50-, 100-, 150-, and 200-
mM) on photosynthetic pigment contents in in vitro grown shoots of important medicinal plant
species (Centaurium erythraea Rafn) were investigated. Non-transformed, one AtCKX1 and two
AtCKX2 transgenic centaury lines, with altered cytokinin profiles, were used in this study. The chlorophyll
(Chl) and carotenoid contents differed in the non-transformed and transgenic lines. In general,
salinity significantly reduced the Chl a and Chl b contents in comparison to the NaCl-free medium.
The lowest Chl content was observed in AtCKX2 transgenic shoots grown on all the culture
media. The total carotenoid content was increased in shoots of non-transformed and both AtCKX2
transgenic lines grown in 50-mM NaCl. On the other hand, in concentrations ˃50-mM NaCl, the
total carotenoid content was decreased in all analysed centaury shoots. The Chl a/Chl b ratio in all
the shoots increased progressively in the graded NaCl concentrations. Contrarily, the addition of
NaCl in the culture medium reduced the Chl/carotenoid ratio in centaury shoots. Taken together,
the results of this study partly explained the mode of centaury plant adaptations to salt stress in
vitro. Thus, the results on centaury shoots confirmed that the determination of the photosynthetic
pigment contents can be a very useful non-destructive screening method in order to discriminate
susceptible and resistant plant species/lines to salt stress conditions.

AB  - Plant hormones regulate numerous developmental and physiological processes. Abiotic stresses
considerably affect production and distribution of phytohormones as the stress signal triggers. The
homeostasis of plant hormones is controlled by their de novo synthesis and catabolism. The aim of this
work was to analyse the contents of total and individual groups of endogenous cytokinins (CKs) as well
as indole-3-acetic acid (IAA) in AtCKX overexpressing centaury plants grown in vitro on graded NaCl
concentrations (0, 50, 100, 150, 200 mM). The levels of endogenous stress hormones including abscisic
acid (ABA), salicylic acid (SA) and jasmonic acid (JA) were also detected. The elevated contents of total
CKs were found in all analysed centaury shoots. Furthermore, increased amounts of all five CK groups,
as well as enhanced total CKs were revealed on graded NaCl concentrations in non-transformed and
AtCKX roots. All analysed AtCKX centaury lines exhibited decreased amounts of endogenous IAA
in shoots and roots. Consequently, the IAA/bioactive CK forms ratios showed a significant variation
in the shoots and roots of all AtCKX lines. In shoots and roots of both non-transformed and AtCKX
transgenic centaury plants, salinity was associated with an increase of ABA and JA and a decrease of
SA content.

Effects of high temperature on in vitro tuberization and accumulation of stress-responsive proteins in potato

AB  - Potato (Solanum tuberosum L.) plants are highly vulnerable to heat stress. Even moderately elevated temperatures can disturb the process of tuberization in this important crop, causing a decline in tuber initiation, a reduction in tuber bulking, and tuber disorders. In the present study, we investigated the effects of heat stress on tuberization in two potato cultivars, the heat-sensitive cultivar Désirée and the heat-tolerant cultivar Festival, using an in vitro system. A temperature of 29 °C reduced tuber initiation and tuber bulking, and stimulated shoot elongation in cv. Désirée, while this temperature treatment did not significantly alter tuberization or shoot elongation in cv. Festival. In addition, high temperature interfered with the onset of microtuber dormancy and promoted growth of tuber apical buds during the tuber bulking stage in both cultivars. Stress-responsive proteins HSP17.6-CI, HSP101, and eEF1A showed heat-induced accumulation patterns in shoots and microtubers of these two cultivars, with the exception of a decline in the abundance of eEF1A in cv. Désirée microtubers under heat stress. High levels of HSP17.6-CI in microtubers of cv. Désirée did not ameliorate the effects of heat stress on tuberization of this relatively heat-sensitive cultivar. Conversely, a higher level of eEF1A under heat stress in microtubers of the heat-tolerant cv. Festival indicated a possible function of this protein in alleviating the negative effects of high temperature on potato tuberization. This study suggested that analysis of stress-responsive proteins in potato microtubers combined with assessment of tuberization parameters in vitro may represent a useful screening procedure for selection of heat-tolerant potato genotypes.

AB  - The hormonal control of tuberisation in potato is complex. It also depends on environmental conditions, especially light conditions. The present work deals with effects of various combinations of 6-benzylaminopurine (BAP) and the growth retardant, tetcyclacis (Tc), on in vitro tuberisation in potato with the emphasis on process of stolon initiation. Potato shoots cv. Désirée were grown in in vitro cultures, on the MS media with various combinations of 0.01 - 10 mM BAP and 3 mM Tc, under long day conditions (16 h light/8 h darkness) or in continuous darkness. The effects on stolon and tuber initiation as well as on tuber diameter were determined in 30-day-old shoots. The obtained results showed that a decrease in endogenous gibbereliin level is a prerequisite for tuberisation under tuber non-inducing conditions in vitro. The increase in the cytokinin/gibberellin relationship stimulated stolon initiation under tuber non-inducing conditions in vitro, but only up to the level achieved by individually applied 3 mM Tc. The BAP interaction with Tc was not observed in the process of tuber initiation. In the process of the tuber enlargement, interaction between BAP and Tc, i.e. the increase in tuber diameter in comparison to values obtained by the individual application of these plant growth regulators, was observed only under tuber non-inducing conditions, when combination of 10 mM BAP and 3 mM Tc was applied.
AB  - Tuberizacija kod krompira predstavlja jedan od najpogodnijih model-sistema za proučavanje interakcija fitohormona i korelacija rastenja i razvića kod biljaka. Hormonalna regulacija ovog procesa, kome se posvećuje velika pažnja zbog značaja krompira za poljoprivredu, je složena i zavisi od uslova spoljašnje sredine, a pre svega uslova osvetljenja. U ovom radu su proučavani efekti kombinovanja 6-benzil aminopurina (BAP) i retardanta rastenja, tetciklacisa (Tc), na proces tuberizacije kod krompira u uslovima in vitro, sa naročitim osvrtom na proces inicijacije stolona. Ustanovljeno je da sniženje nivoa endogenih giberelina predstavlja neophodan uslov za tuberizaciju in vitro u neinduktivnim uslovima (dug dan). Povišavanje odnosa citokinini/giberelini stimuliše inicijaciju stolona u neinduktivnim uslovima za tuberizaciju in vitro, ali samo do nivoa koji se postiže pojedinačnom primenom 3 mM Tc. Nije uočena interakcija BAP i Tc u procesu inicijacije krtola. U procesu uvećavanja krtola uočava se interakcija samo u neinduktivnim uslovima, kada se primene 10 mM BAP i 3 mM Tc.

The present work deals with effects of various combinations of 6-benzylaminopurine (BAP) and the growth retardant, tetcyclacis (Tc), on in vitro tuberisation in potato with the emphasis on process of stolon initiation. Potato shoots cv. Désirée were grown in in vitro cultures, on the MS media with various combinations of 0.01 - 10 mM BAP and 3 mM Tc, under long day conditions (16 h light/8 h darkness) or in continuous darkness. The effects on stolon and tuber initiation as well as on tuber diameter were determined in 30-day-old shoots. The obtained results showed that a decrease in endogenous gibbereliin level is a prerequisite for tuberisation under tuber non-inducing conditions in vitro. The increase in the cytokinin/gibberellin relationship stimulated stolon initiation under tuber non-inducing conditions in vitro, but only up to the level achieved by individually applied 3 mM Tc. The BAP interaction with Tc was not observed in the process of tuber initiation.
