Gibberellic acid promotes in vitro regeneration and shoot multiplication in Lotus corniculatus L.

AB  - Shoots of Lotus corniculatus L., previously transformed with Agrobacterium tumefaciens LBA4404/pTOK233, were grown in gibberellic acid (GA(3))-containing media in an attempt to improve their growth and multiplication. Nodal stem segments of four poorly multiplying clones, a clone with very good multiplication, and two non-transformed clones were incubated in media containing GA(3) for 3 weeks, and then returned to a hormone-free medium for a further 3-5 subculture periods. Gibberellic acid increased the number of axillary buds from one in controls to 2-13 shoots during the first 3 weeks of subculture in GA(3)-containing media. The multiplied buds, which apparently derived from additional axillaries, continued to multiply in hormone-free medium, producing bunches of more than 50 shoots after 9 weeks. In addition, many nodal segments also produced callus tissues at the basal end, or along the internode below the node, in which abundant shoot meristems formed de novo. Histological examination confirmed their origin via organogenesis. About 25% of regenerated shoots rooted spontaneously. The application of auxins significantly improved rooting in more than 75% of all clones. Well-developed plantlets were produced upon the transfer of rooted shoots to pots. No differences in responses to hormones were observed between LBA4404/pTOK233-transformed clones and non-transformed clones, indicating that the reaction to GA(3) was not the consequence of transformation events. The results point to a possible involvement of GA(3) in lateral branching. They might also be recommended as a procedure suitable for increasing the production of transformed plants in L. corniculatus.

The effects of IAA and tetcyclacis on tuberization in potato (Solanum tuberosum L.) shoot cultures in vitro

AB  - Potato (Solanum tuberosum cv. Desiree) shoots grown in vitro in continuous darkness or in long days (LDs), were used to investigate indole-3-acetic acid (IAA) effects on stolon initiation and tuber formation, combining IAA with increased or decreased gibberellin levels. An increased gibberellin (GA) level was achieved by the applying 1 mu M GA(3), while decreased gibberellin level was presumably realized by the adding 3 mu M tetcyclacis (Tc). About 15% of potato shoots developed stolons both in LDs and in darkness. Stolon initiation was stimulated by GA(3) in darkness and by Tc in LDs. Tuber formation was strongly inhibited in LDs and by GA(3) both in light and darkness, but stimulated in darkness at low GA level. Exceptionally, tuber formation occurred in LDs at the highest Tc concentrations, in about 25% of explants. Indole-3-acetic acid alone stimulated stolon formation in LDs, both in the presence or absence of GA(3). IAA alone also stimulated tuber formation in dark-grown shoots, but could not overcome the inhibitory effect of LDs. Indications that, depending on their concentration ratio, IAA may interact with GA(3) in different tuberization phases, have been discussed.

AB  - Cotyledons from 6-day-old Lotus corniculatus cv. Bokor seedlings, transversally cut into two halves, were capa­ble of regenerating buds without intervening callus formation. The explants were co-cultivated with the Agrobacterium tumefaciens LBA4404/pTOK233 superbinary vector carrying the uidA-intron gene and the genes hpt and nptII. They were cultured for 14 days on a regeneration medium, then subjected to a stepwise hygromycin B selection procedure consisting of gradually increasing antibiotic concentrations (5-15 mg L-1) over 21 weeks. Transformed shoots were obtained within 5 months after co-cultivation. Out of 124 initially co-cultivated explants, 52 (42%) plants survived hygromycin B selection. The presence of transgenes in regenerated plants was verified by β-glucuronidase histochemical assays and PCR analysis for the presence of uidA gene sequences. Hygromycin B-resistant and PCR-positive T0 plants were cultured in the greenhouse to produce flowers and seeds. The obtained data demonstrate that the reported transformation protocol could be useful for introducing agriculturally important genes into the new L. corniculatus cultivar Bokor.
AB  - Kotiledoni, izolovani sa 6 dana starih klijanaca Lotus corniculatus, sorta Bokor, su se pokazali kao pogodni eksplantati za regeneraciju biljaka bez intermedijarne kalusne faze. Kotiledoni su presecani transverzalno na dve polovine i ko-kultivisani sa Agrobacterium tumefaciens superbinarnim vektorom LBA4404/pTOK233, koji nosi uidA-intron, hpt i nptII gene. Posle ko-kultivacije eksplantati su prvih 14 dana gajeni na regenerativnom medijumu bez selektivnog agensa, a zatim su 21 nedelju bili podvrgnuti selektivnoj proceduri tokom koje je koncentracija antibiotika postepeno povećavana (5-15 mg L-1). Transformisani izdanci dobijeni su 5 meseci nakon ko-kultivacije. Od 124 inicijalno ko-kultivisana eksplantata, dobijene su 52 (42%) biljke koje su preživele selekciju na higromicinu B. Prisustvo uidA transgena u regenerisanim biljkama je potvrđeno pomoću β-glukuronidazne histohemijske reakcije i PCR analizom. T0 biljke, rezistentne na higromicin B i PCR pozitivne, gajene su u stakleniku u cilju produkcije i rastenja cvetova i semena. Dobijenim rezultatima je jasno ukazano da bi saopšteni protokol za transformaciju mogao nesumnjivo biti upotrebljen za introdukciju agronomski značajnih gena u Lotus corniculatus, sorta Bokor.

AB  - We studied the effects of various cytokinins and urea derivatives on germination of aged seeds of in Lotus corniculatus L. The following substances were applied: N6-isoprenoid cytokinins (isopentenyl adenine and zeatin), adenine sulfate, N6-aromatic cytokinins (kinetin, benzyladenine) and their N9-ribosides, N-benzyl-9-(2- tetrahydropyranyl)adenine, and urea derivatives (diphenylurea, thidiazuron, and chloro-pyridyl phenylurea). With the exception of adenine sulfate, all cytokinins increased the percentage of seed germination up to twofold, depending on their kind and concentration. It is concluded that cytokinins may be among the missing factors in aged seeds of L. corniculatus contributing to the implementation of their full germination potential. They could be used to improve germination of both freshly harvested and aged seed samples, if necessary. .
AB  - Ispitivan je efekat različitih citokinina i derivata uree na klijanje starih semena Lotus corniculatus L. Upotrebljene su sledeće supstance: N6-izoprenoidni citokinini (izopentenil adenin i zeatin), adenin sulfat, N6-aromatični citokinini (kinetin, benziladenin), i njihovi N9-ribozidi, N-benzil-9- (2-tetrahidropiranil)adenin i derivati uree (difenilurea, tidiazuron i hloro-piridil fenilurea). Svi citokinini su, izuzev adenin sulfata, povećavali procenat klijanja semena do dva puta u zavisnosti od vrste citokinina i koncentracije. Zaključeno je da citokinini mogu biti jedan od nedostajućih faktora u starim semenima L. corniculatus, koji doprinose ostvarenju njihovog punog potencijala za klijanje. Ako je potrebno, citokinini mogu biti iskorišćeni za poboljšanje klijanja sveže ubranih i starih semena. .

It is concluded that cytokinins may be among the missing factors in aged seeds of L. corniculatus contributing to the implementation of their full germination potential. They could be used to improve germination of both freshly harvested and aged seed samples, if necessary. ., Ispitivan je efekat različitih citokinina i derivata uree na klijanje starih semena Lotus corniculatus L. Upotrebljene su sledeće supstance: N6-izoprenoidni citokinini (izopentenil adenin i zeatin), adenin sulfat, N6-aromatični citokinini (kinetin, benziladenin), i njihovi N9-ribozidi, N-benzil-9- (2-tetrahidropiranil)adenin i derivati uree (difenilurea, tidiazuron i hloro-piridil fenilurea). Svi citokinini su, izuzev adenin sulfata, povećavali procenat klijanja semena do dva puta u zavisnosti od vrste citokinina i koncentracije. Zaključeno je da citokinini mogu biti jedan od nedostajućih faktora u starim semenima L. corniculatus, koji doprinose ostvarenju njihovog punog potencijala za klijanje. Ako je potrebno, citokinini mogu biti iskorišćeni za poboljšanje klijanja sveže ubranih i starih semena. .",

AB  - The use of two Agrobacterium tumefaciens strains for transformation of Triticum aestivum L. cv. Vesna was studied. Immature embryos, isolated 15 d after pollination, were co-cultivated with the super-binary LBA4404/pTOK233 and the binary AGL1/pDM805 vectors. While the transient GUS-intron expression was high (69.9 and 80.0%), the number of plants regenerated on selective media containing hygromycin or phosphinotricin did not exceed 0.4 and 0.13%, respectively. Nevertheless, the regenerated plants were fertile and produced seeds. The T-0 plants, as well as the T-1 seedlings, displayed the activity in the beta-glucuronidase histochemical assay and a positive signal in PCR analysis for the presence of uidA gene sequences in their genomes. The data suggest that the transformation of wheat cv. Vesna with both Agrobacterium strains is feasible.

AB  - Buckwheat ( Fagopyrum esculentum Moench) is a heterostylous plant displaying heteromorphic sporophytic self-incompatibility (SI). In order to detect proteins involved in SI, pistils from both long and short styles were isolated and then selfed or cross-pollinated. One-dimensional gel electrophoresis revealed that short pistils 2 h after selfing contained an unique 50 kDa protein. In the two-dimensional electrophoresis two distinct groups of proteins possibly involved in SI response were detected in the short, and one in the long pistils.

AB  - Carob (Ceratonia siliqua L.) has compound pinnate leaves consisting of 4 - 6 pairs of leaflets. However, in conditions of in vitro culture only one pair of leaflets develops. With increasing irradiance from 9.3 to 74.1 mu mol m(-2) s(-1), leaf area increased 5-fold. Sucrose also significantly increased leaf area and the maxima were at concentration 147 mM at high irradiance and 233.6 mM at low irradiance. Sucrose was superior to Fructose, glucose and combination of both in increasing leaf area. Decreasing concentration of KNO3 and NH4NO3 caused a 3-fold decline of leaf area.

AB  - Somatic embryogenesis was induced in embryogenic calli of Spinacia oleracea L., on a Murashige and Skoog (1962) medium, containing 4.6 μM kinetin as the sole growth regulator. Abscisic acid, gibberellic acid, and indole-3-acetic acid were supplemented to kinetin-containing medium and their effects on the initiation of somatic embryos was studied. Abscisic acid at a particular concentration (4 μM) dramatically increased the number of embryos per g fresh weight of callus, while both gibberellic acid and indole-3-acetic acid suppressed the embryo initiation. It is suggested that the promoting effect of abscisic acid on the embryo initiation may be explained as a stress response of the tissue. The relative number of globular embryos vs. the embryos in heart/torpedo and cotyledonary stages was increased at 4 μM abscisic acid and at all gibberellic acid concentrations (0.3–10 μM). In contrast, the ratio of globular to polar embryos was lower than in controls at 1 μM abscisic acid and indole-3-acetic acid (1 and 10 μM). The effects of growth regulators on the ratio of globular to polar embryos indicate that they interfere with the normal distribution of cell division and cell expansion during early embryogenesis.
