AB  - Long-term exposure of populations to pollution may result in enhanced ability to cope with environmental stress. To compare the responses of two Lymantria dispar populations living in unpolluted and polluted forests (UP and PP, respectively), we chronically exposed larvae to cadmium at concentrations of 50 and 100 μg Cd/g dry food (Cd1 and Cd2, respectively). We examined cadmium accumulation in the midgut and hemolymph, activities of superoxide dismutase (SOD), catalase (CAT), and alkaline phosphatases (ALP) in the midgut, as well as Hsp70 protein expression in the midgut, hemolymph, and brain and evaluated these parameters as biomarkers of cadmium contamination. Larvae from PP, fed a control diet, showed higher activity of SOD and increased Hsp70 expression compared with larvae from UP. Excessive amounts of Cd were accumulated in the midgut of all Cd-fed larvae, whereas Cd content in the hemolymph was elevated only in larvae from PP after Cd2 treatment. In larvae from UP, Cd2 treatment decreased the activity of CAT and induced the expression of Hsp70 in the midgut and hemolymph. In larvae from PP, exposure to both Cd concentrations strongly attenuated SOD and CAT activities, while Hsp70 expression was not induced in any organ/tissue. Cd did not affect ALP activity in either population. Midgut Cd content proved to be a suitable indicator of Cd contamination for both polluted and unpolluted habitats.

AB  - Кадмијум је глобално присутан токсични метал који изазива бројне штетне ефекте у организму и заузима седмо место на листи приоритетних супстанци од којих је неопходно пронаћи оптималне начине заштите. Наше пређашње студије су показале да егзополисахарид изолован из соја Lactiplantibacillus plantarum BGAN8 (EPS-AN8) показује висок афинитет за везивање јона кадмијума у воденом раствору и остварује значајан ниво in vitro заштите Caco-2 ћелија од његових токсичних ефеката. Имајући у виду да је за општу популацију најзаступљенији унос кадмијума исхраном, у овој студији је праћен паралелни ефекат уноса кадмијума (кроз воду) и EPS-AN8 (кроз храну) у DA пацовима. Након 30 дана третмана, утврђено је да је унос EPS-AN8 допринео сниженом нивоу депонованог метала у бубрезима, јетри и крви, а повећаном садржају у фекалном материјалу. Такође, микрографије дуоденума, бубрега и јетре су указале на ниже степене кадмијумом индукованих оштећења. Праћењем активности ензима који учествују у антиоксидативној заштити (CAT, GST), терминалних производа липидне пероксидације (MDA) и продукције проинфламаторних цитокина (IL-1β, TNFα, IFN-γ) у хомогенатима дуоденума, утврђен је нижи ниво оксидативног стреса и инфламације, што представља додатни показатељ заштите коју остварује EPS-AN8. С обзиром да орални унос кадмијума доводи до значајних промена у цревној микробиоти, изоловали смо тоталну DNK из дуоденума и секвенцирали 16S rDNK ампликон на Illumina NovaSeq платформи. Примећено је да унос EPS-AN8 ублажава појаву промена у релативној заступљености бактеријских родова и врста карактерисичних за излагање кадмијуму, попут пораста броја опортунистичких патогених бактерија и смањења бројности лактобацила. Остварени заштитни ефекти указују на снажан потенцијал примене EPS-AN8 у условима високе контаминације кадмијумом.

AB  - The mobility of chemical elements in the soil-orchid system has been poorly studied. The aim of this study is to evaluate the uptake and mobility of several trace (Li, Ba, Sr, Ag, Hg, and B) and macronutrients (Ca, Mg, and K) in the orchid Anacamptis morio (L.) R.M.Bateman, Pridgeon & M.W.Chase from soils in western Serbia. The sampling sites are characterized by three different bedrock types-cherts, limestones, and serpentines, which are the source of the significant chemical differences in the elemental status of the soil and plant tissues. The four-step Community Bureau of Reference sequential extraction procedure was used to determine the distribution of fractions and predict their potential phytoavailability. The orchid and soil samples were analyzed for total elemental content analysis using ICP-OES. The greatest potential for plant availability was determined for Ba and Sr, representing about 80% of the total soil content. More than 40% of Li in the soils was found to be potentially phytoavailable. Significant correlations were found between the total content of Li, B, and Sr in soils. Between 38 and 60% of Li content and more than 80% of Ba and Sr content were determined to be potentially phytoavailable by sequential analysis. The highest bioconcentration factor (> 1) was determined in the case of B and Sr for all orchid organs, while translocation factor for Li was highest in tubers and leaves. The studied elements were mainly stored in tubers and roots, indicating the exclusion strategy of A. morio as a metal tolerance mechanism. The data obtained showed significant differences in metal content in soils and plants originating from sites with different parent materials, suggesting that bedrock type and associated soil properties are important factors that determine chemical element mobility and uptake.

AB  - Cadmium (Cd) is a highly toxic metal that is distributed worldwide. Exposure to it is
correlated with a vast number of diseases and organism malfunctions. Exopolysaccharides (EPS)
derived from Lactiplantibacillus plantarum BGAN8, EPS-AN8, previously showed great potential for
the in vitro protection of intestinal cells from this metal. Here, we investigated the potential of food
supplemented with EPS-AN8 to protect rats from the hazardous effects of Cd exposure. After thirty
days of exposure to lower (5 ppm) and higher (50 ppm)-Cd doses, the administration of EPS-AN8 led
to decreased Cd content in the kidneys, liver, and blood compared to only Cd-treated groups, whereas
the fecal Cd content was strongly enriched. In addition, EPS-AN8 reversed Cd-provoked effects on
the most significant parameters of oxidative stress (MDA, CAT, GST, and GSH) and inflammation
(IL-1 , TNF- , and IFN-
) in the duodenum. Moreover, micrographs of the duodenum were in line
with these findings. As the gut microbiota has an important role in maintaining homeostasis, we used
16S rRNA amplicon sequencing and investigated the effects of Cd and EPS-AN8 on one part of the
microbiota presented in the duodenum. Although Cd decreased the growth of lactobacilli and mostly
favored the blooming of opportunistic pathogen bacteria, parallel intake of EPS-AN8 reversed those
changes. Therefore, our results imply that EPS-AN8 might be extremely noteworthy in combatting
this toxic environmental pollutant.

AB  - The study of the chemical composition of wines is nowadays a topic of great interest because of the importance of this market, especially in Italy, and also considering the numerous cases of falsification of famous and very expensive wines. The present paper focused on the analysis of metals and polyphenols in Italian wines belonging to different provenance and types. At this purpose 20 elements were quantified by inductively coupled plasma optical emission spectrometry (ICP-OES) and ICP mass spectrometry (ICP-MS). Regarding polyphenols, a total of 32 were quantified, among 6 were anthocyanins. Furthermore, in 4 samples (1 rosè and 3 red wines) 42 anthocyanins and related compounds were identified by ultra-high performance liquid chromatography (UHPLC)-Orbitrap MS technique (among these, 6 were also quantified). Non-anthocyanins were determined using UHPLC coupled with a diode array detector and triple-quadrupole mass spectrometer (UHPLC–DAD-QqQ-MS). Total phenolic content (TPC) and radical scavenging activity (RSA) were measured using spectrophotometric methods. The results obtained by elemental techniques were submitted to principal components analysis (PCA) allowing to get information on both geographical and botanical origin of the examined wine samples. Some polyphenols have been detected in higher concentrations only in a certain type of wine, as for example in the case of Grechetto wine. Most of the identified anthocyanin derivatives (pyranoanthocyanins) are formed during the aging of wine by reaction with the other wine components.

AB  - In this study concentration of toxic elements As, Cd, and Pb were determined in different soil types and belonging orchid species Anacamptis morio vital parts, in order to examine accumulation patterns and provide new insights about the potential use of this orchid in bioremediation technology. Soils developed on limestone, serpentine, and the chert were subjected to the BCR sequential extraction. Samples of orchid roots and tubers, as underground parts, and stems, leaves, and inflorescences, as above-ground organs, were also analyzed for the content of As, Cd and Pb. During this research, it was observed that metal content in soil is directly proportional to its content in the plant, more specifically in roots, which suggests that A. morio can potentially be used in the phytostabilization of contaminated sites. Values for BCF factors showed Cd immobilization in roots regardless of the soil type. A certain level of arsenic was transferred from roots to leaves indicating the potential for accumulation of this element into aboveground organs. Assessment of the phytoremediation potential of this orchid or another plant species from diverse environments is important as it provides information about the possibility of their future application in environmental remediation programs.

AB  - Adverse effects of non-occupational exposure to cadmium (Cd) are increasingly
acknowledged. Using a rat model of oral Cd exposure in drinking water we have shown
that skin is a target for this metal. Due to contribution of individual variability to the
intensity of cadmium toxicity, dermatotoxicity of two environmentally relevant Cd
doses (5 and 50 ppm) was examined in individuals of two rat strains, Albino Oxford
(AO) and Dark Agouti (DA), which differ in response to chemicals. A dose-dependent
accumulation of Cd in the skin/epidermal cells was noted in both strains, and although
there were no strain differences in the Cd accumulation, the degree of skin response to
the metal differed. Signs of skin damage were evident in both strains, but response to
injury was more pronounced in DA. Individuals of DA rats responded by an increase
in the levels of antioxidant defense enzymes in the skin already at lower dose, in
contrast to AO (which reacted to higher dose solely), implying higher sensitivity of DA
strain to Cd-induced toxicity. Epidermal cells from both strains developed stress
response, however increased GSH, and higher metallothionein/MT-1 and MT-2
mRNA, Nrf2 protein, apoptosis, Ahr and Cyp genes in AO, depicting this strain`s
ability to better defend against Cd insult. Epidermal cells` IL-1β, TNF and IL-6
response was induced by Cd in DA, while pro-inflammatory cytokine production was
unchanged in AO (though increased following stimulation with S. epidermidis), with
increased IL-10 as a possible underlying mechanism. T cells from non-exposed rats
produce more IFN-γ and IL-17 in co-culture with epidermal cell from Cd–exposed DA
rats what strengthens the view that this strain is more prone to metal’s dermatotoxicity.
These data give a new insight into repercussion of genetic variability to toxicity of
cadmium acquired by the skin via gut, bearing relevance for variations in the link
between dietary cadmium and inflammation-based skin pathologies.

Influence of dietary cadmium exposure on fitness traits and its accumulation (with an overview on trace elements) in Lymantria dispar larvae

AB  - Bioaccumulation and excretion of heavy metals in insects is only partially clarified. We have investigated cadmium accumulation in the feces, head and integument of Lymantria dispar (Lepidoptera: Lymantriidae) larvae exposed to chronic dietary intake of cadmium. The aim of the experiment was to establish modalities of metal accumulation, primarily cadmium, as well as changes in fitness traits in two insect populations receiving 50 or 100 μg Cd/g dry food. The egg-masses originated from two localities: a protected nature reserve (unpolluted population) and an area near a busy highway (polluted population) in Serbia. At both added dietary levels cadmium concentration was highest in feces. Small alterations in metal concentrations after both cadmium treatments were detected in the integument. It was established that irrespective of population origin, the modality of decline of larval mass and relative growth rate (RGR) was similar. Concentrations of Cu and Zn in the integument were approximately the same regardless of dietary cadmium intake. However, cadmium accumulation in the examined tissues, as well as variability of fitness traits depended on population origin and cadmium concentration. In larvae from both populations not given contaminated food the head was the organ with the greatest accumulation of trace elements. Our results reveal how invasive phytophagous insects cope with high metal concentrations in their food by body mass (RGR) reduction and energy allocation towards processes that enable accumulation of cadmium and other trace elements in different tissues.

Ecological potential of Epilobium dodonaei Vill. for restoration ofmetalliferous mine wastes

AB  - Metalliferous mine wastes represent one of the major sources of environmental contamination frommining activities. Bor region (Serbia) is one of the largest copper mine basins in Europe where long-termmining caused severe environmental deterioration and created one of the most degraded locations inSerbia and Europe. At the spontaneously colonized metalliferous mine wastes in Bor, plant species Epilo-bium dodonaei dominates in the mine slopes and mine waste surfaces. Epilobium dodonaei has the statusof endangered and protected species in parts of European range (i. e. plant is included in the Red listsof the countries in the Carpathian mountains region), primarily due to losses of natural gravel habitats.The main focus of this research was physico-chemical characterization of mine waste, assessment ofphytoremediation potential and plant metabolic stress response of Epilobium dodonaei at the hot spotmetalliferous mine site in order to evaluate the possibility for application of endangered species in eco-logical restoration. The Bor mine wastes are characterized by coarse soil texture, various pH (4.58–8.30),and elevated concentrations of arsenic (44.5–271 mg kg−1) and copper (311–2820 mg kg−1) that exceedthe Serbian limiting threshold and remediation values. Oxidation of metal-sulfide minerals on waste sur-face leads to increased acidity, followed by elevated metal mobility of the mine spoil solution. Contentof arsenic, copper, lead and zinc in roots of E. dodonaei was correlated with pseudo-total and EDTA-available concentrations in Bor mine spoils. Furthermore, the content of arsenic, copper, lead and zinc inroots (3.98 mg kg−1, 140 mg kg−1, 3.19 mg kg−1, and 72.8 mg kg−1, respectively) and shoots (4.69 mg kg−1,57.7 mg kg−1, 1.17 mg kg−1, and 59.3 mg kg−1, respectively) of E. dodonaei reflected the multi-metal pol-lution at the investigated site. Epilobium dodonaei largely retains copper, lead and zinc in roots thanin shoots and has the potential for phytoremediation of mine wastes. Epilobium dodonaei at Bor minespoil had a high content of malondialdehyde in roots and leaves as well as reduced chlorophylls andcarotenoids content in leaves, indicating great oxidative stress. However, elevated arsenic and coppercontent could promote biosynthesis of antioxidants in roots and leaves of E. dodonaei at mine spoil. Cre-ation of an endangered species habitat on mine waste rocks of the Bor mining area and similar sites ofCarpatho-Balkan metallogenic province could successfully contribute to the preservation of E. dodon-aei. Development of practical procedures for the selection and application of endangered plant species inreclamation should create stronger link between ecological restoration and conservation biology. Finally,the application of endangered plant species should take a more prominent role in the restoration processand ecosystem design
