AB  - BACKGROUND
The increasing bacterial resistance to antibiotics poses a serious global threat. For that reason, the research into plant-derived antibacterial agents has become progressively more interesting, particularly in regard to their capacity to prevent the delay or inability of wound closure and healing.
OBJECTIVES
The aim of this study was to evaluate i) the antibacterial potential of ethanolic extracts (70% ethanol) of 18
Lamiaceae species against four Gram-negative and three Gram-positive bacterial strains; ii) the effect of these extracts on Pseudomonas aeruginosa PAO1 infection of human lung fibroblasts (MRC-5), and iii) the effect on MRC-5 cell migration.
METHODS
The antibacterial activity was tested by minimal inhibitory concentration (MIC) assay. The effect on bacterial infection of MRC-5 cells was determined using the invasion assay, while the cell migration was assessed with Scratch assay.
RESULTS
The tested Gram-positive bacteria, especially Bacillus subtilis, are more sensitive to the effects of ethanolic extracts than the Gram-negative bacteria. Moreover, the tested extracts significantly inhibited the invasion of Pseudomonas aeruginosa PAO1 during MRC-5 infection, and most of them also displayed promising stimulating potential on MRC-5 migration in a disrupted cell monolayer. Finally, the results of IBR (integrated biomarker response) analysis highlighted Salvia officinalis ethanolic extract as the most active one, which is in accordance with the existing data suggesting that this plant represents an effective antibacterial and wound-healing agent commonly used in traditional medicine. Altogether, these results are expected to foster further studies that will focus on combating bacterial resistance to antibiotics using natural products.

AB  - Bioactive compounds from natural sources are of great importance because of their potential pharmacological activity and tremendous structural diversity. In this study, the chemical composition of different moss extracts of Hedwigia ciliata P. Beauv. have been examined, as well as their antioxidant, antineurodegenerative/anti-neuroinflammatory, antidiabetic, and antiproliferative potential. The extracts were prepared by Soxhlet extractor using solvents of different polarity. Chemical characterization of the extracts revealed the presence of phenolics and flavonoid compounds, together with triterpenoids as secondary metabolites of high biological activity. Significant antioxidant properties of all the extracts were exhibited using the β-carotene assay. The highest activities were found for water:ethanol extract (with the highest inhibition rate of 96%), but also significant inhibition was measured for ethanol and ethyl acetate extracts (80% and 70%, respectively). Confirmation of biocompatibility of investigated moss extracts has been performed using normal human fibroblast cell line, MRC-5. The H . ciliata extracts exhibited significant antiproliferative activity (~ 50%) against the MDA-MB-231 (human breast adenocarcinoma cell line), which has not previously been reported elsewhere. The Griess assay confirmed the potential anti-neuroinflammatory activity of the extracts, as significant effects in reducing NO production by LPS-stimulated BV2 (normal murine microglia cell line) was observed. This data is in line with noted antineurodegenerative potential measured by the inhibition of acetylcholinesterase (with the highest inhibition rate of 60% for ethyl acetate extract) and tyrosinase (with the highest inhibition rate of 70% for ethanol extract). Additionally, the H . ciliata extracts exhibited significant antidiabetic effect mediated by α-glucosidase inhibition (with the highest inhibition rate of 80% for ethyl acetate extract). The obtained data suggest the presence of immunomodulatory effects of the moss extracts in vitro , which allows the design of new experiments aimed at detecting and characterizing bioactive compounds of the extracts and additionally elucidate detailed mechanisms of their effects.

AB  - Pear (Pyrus communis L.) is consumed as fresh fruit, in numerous food products, and also used as a traditional remedy in various countries, including Serbia. In search of bioactive compounds, six traditional pear varieties (‘Vidovača’, ‘Lubeničarka’, ‘Karamanka’, ‘Jeribasma’, ‘Lončara’, ‘Takiša’) and wild pear from Serbia were investigated and compared with a commercial variety (‘Williams Bartlett’). The aim of this study was to determine the total phenolic and flavonoid contents, phenolic composition, antioxidant capacity, and antineurodegenerative activities of methanolic extracts of peel, flesh, and mixed peel and flesh of pear fruits. Phenolic composition of extracts was determined with HPLC-DAD, while the antioxidant activity of extracts was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and ferric-reducing antioxidant power (FRAP) assays. Bearing in mind that oxidative stress is closely linked to neurodegeneration, the antineurodegenerative potential of the extracts was assessed by the inhibition of acetycholineserase (AChE) and tyrosinase (TYR) activities. The extracts of traditional varieties, particularly peel extracts, had a high content of phenolics, as well as significant antioxidant and moderate antineurodegenerative potential, compared to the commercial variety. The highest contents of total flavonoids and individual compounds, such as arbutin and chlorogenic acid, as well as the strongest antioxidant and TYR inhibitory activities were reported for the ‘Takiša’ peel extract. The performed analyses have revealed that fruits of traditional Serbian pear varieties are rich in bioactive components and could be used as functional food and for possible nutraceutical applications, to prevent diseases induced by oxidative stress.

Different levels of exploring the antioxidant and antitumor potential of Salvia officinalis and Salvia rosmarinus

AB  - Salvia officinalis (sage) and Salvia rosmarinus (previously Rosmarinus officinalis, rosemary) are commonly used medicinal plants from the Lamiaceae family, with strong antioxidant abilities. This study was carried out in several stages to comprehensively and gradually explore the antioxidant and antitumor mechanisms of their ethanolic extracts using different experimental approaches. Their in vitro antioxidant activity was assessed using ABTS assay. Plasmid pUC19 E. coli XL1-Blue was used to evaluate their genoprotective activity, while SOS/umuC assay on Salmonella typhimurium TA1535/pSK1002 and Comet assay on human lung fibroblast cell line (MRC-5) were employed to examine their antigenotoxic potential. Furthermore, their antitumor mechanisms were analyzed on the colorectal cancer cell line (HCT-116) by testing their impact on the cells’ viability (MTT assay), as well as the production of reactive oxygen species (ROS) (NBT test) and nitric oxide (NO) (Griess assay). The results showed that sage extract had slightly higher ABTS-scavenging and antigenotoxic activities in the bacterial model when compared to the rosemary extract, however, opposite results were obtained for the acellular model. Moreover, both extracts protected the MRC-5 cells from the induced genomic damages. As for tumor cells, sage extract reduced their viability by approximately 10 %. Additionally, both extracts significantly reduced the ROS production, while also increasing the NO production, especially the rosemary extract. These results delivered novel information about the nature of antioxidant effects of sage and rosemary ethanolic extracts. They provided substantial protection of the acellular, bacterial and healthy human cellular models by employing radical-scavenging and antigenotoxic mechanisms. At the same time, both extracts exhibited antitumor activity towards colorectal cancer cells in vitro that could be mediated by inducing an increase in NO production. With that being said, sage and rosemary ethanolic extracts represent potent genoprotective, antioxidant and antitumor agents. This study was carried out in several stages to comprehensively and gradually explore the antioxidant and antitumor mechanisms of their ethanolic extracts using different experimental approaches. Their in vitro antioxidant activity was assessed using ABTS assay. Plasmid pUC19 E. coli XL1-Blue was used to evaluate their genoprotective activity, while SOS/umuC assay on Salmonella typhimurium TA1535/pSK1002 and Comet assay on human lung fibroblast cell line (MRC-5) were employed to examine their antigenotoxic potential. Furthermore, their antitumor mechanisms were analyzed on the colorectal cancer cell line (HCT-116) by testing their impact on the cells’ viability (MTT assay), as well as the production of reactive oxygen species (ROS) (NBT test) and nitric oxide (NO) (Griess assay). The results showed that sage extract had slightly higher ABTS-scavenging and antigenotoxic activities in the bacterial model when compared to the rosemary extract, however, opposite results were obtained for the acellular model. Moreover, both extracts protected the MRC-5 cells from the induced genomic damages. As for tumor cells, sage extract reduced their viability by approximately 10 %. Additionally, both extracts significantly reduced the ROS production, while also increasing the NO production, especially the rosemary extract. These results delivered novel information about the nature of antioxidant effects of sage and rosemary ethanolic extracts. They provided substantial protection of the acellular, bacterial and healthy human cellular models by employing radical-scavenging and antigenotoxic mechanisms. At the same time, both extracts exhibited antitumor activity towards colorectal cancer cells in vitro that could be mediated by inducing an increase in NO production. With that being said, sage and rosemary ethanolic extracts represent potent genoprotective, antioxidant and antitumor agents.
