AB  - Fungi and plants interact in different ways, creating a scale of associations. Hemibiotrophic
fungi represent the most interesting group, as they use sequential biotrophic and necrotrophic infection strategies. The co-evolution of plant and fungal life-styles has not been well characterized.
We present changes of enzymatic component of antioxidative system in succulent plant Tacitus
bellus that specifically correspond to subsequent phases (spore germination, biotrophic phase,
and necrotrophic phase) of hemibiotroph Fusarium verticillioides infection. T. bellus response to F.
verticillioides spore germination was characterized by transient increase in catalase (CAT), but decrease in superoxide dismutase (SOD) and peroxidase (POD) activity. During biotrophic phase of F.
verticillioides infection, when hyphae spread intercellularly in epidermal and mesophyll tissue, host
antioxidative system was suppressed. The transition from biotrophic to necrotrophic phase (inter
and intracellular colonization and sporulation) triggered the host plant cells to create a highly defensive environment: CAT, SOD and POD activities were significantly stimulated, slowing, or even
currently arresting, colonization of T. bellus mesophyll cells. CAT, showing the most pronounced
activity increase, could be suggested as the main enzyme responsible for slowing the progression
of necrotrophic phase of F. verticillioides growth. However, contrary to host CAT and SOD which
isoenzyme profile didn’t change, new highly acidic POD isoforms replaced the two mildly acidic
isoforms, suggesting their specific role in slowing the progression of infection. Presented results
add to knowledge of events and mechanisms related to hemibiotrophic fungi pathogenicity in
succulent plants grown under high relative humidity, similar to conditions in greenhouse.

AB  - Changes in antioxidative enzymes activities during Tacitus bellus direct shoot organogenesis from leaf explants were examined. During the early stages of shoot organogenesis there was a decrease in superoxide dismutase (SOD) and an increase in catalase (CAT) activity, and later during organogenesis there was an increase in peroxidase (POD) and polyphenol oxidase (PPO) activity. Two highly regulated turning points may be distinguished regarding activities and isoforms of antioxidative enzymes: the initiation of shoot organogenesis and the shoot bud formation. Our data suggest the role of specific CAT, POD, SOD and PPO isoforms in separate processes during T. bellus direct shoot organogenesis.

AB  - In order to establish an efficient system for in vitro plant regeneration of a short day plant Chenopodium rubrum L. and a long day plant Chenopodium murale L., optimum culture conditions for somatic embryogenesis were investigated. The effects of different growth regulators, their combination and their concentrations on somatic embryos induction in different explant types (root, hypocotyl, cotyledon and leaf) were tested. Somatic embryogenesis was induced in both plants on Murashige and Skoog (MS) medium supplemented with sucrose (3%), agar (0.7%) and 1-10 mu M 2,4-dichlorophenoxyacetic acid (2,4-D) as the sole growth regulator. The largest embryogenic capacity was found in root explants of Chenopodium rubrum on 1 mu M 2,4-D and in basal parts of cotyledons in C. murale plants on 10 mu M 2,4-D.

In vitro propagacija Tacitus bellus

AB  - Tacitus bellus Moran & J. Meyran, syn. Graptopetalum bellum, fam Crassulaceae was discovered in 1972 in a mountainous areas in Mexico and southwest of the USA. Rosettes, up to 10 cm in diameter, lie almost flat to the ground, while large clusters of star like, pink till deep red, flowers appear in May and June, which makes it one of the most fascinating new succulent plants. Sterilized lower rosette leaves were aseptically placed on medium containing, mineral and vitamin solution supplemented with sucrose (3 %), benzylaminopurine (BAP) (2.25 mg dm-3) and naphtalenacetic acid (NAA) (0.186 mg dm-3) arid gelled with agar (0.7 %), Murashige and Skoog, 1962,. Callus and a large number of buds were observed on the explants bases after six months of cultivation on the two-phase medium. The bud multiplications were performed on MS media supplemented with BAP (0.1-0.5 mg dm-3) and NAA (0.1 mg dm-3), while rooting of small rosettes were performed on NAA (0.1 - 0.2 mg dm-3). Spontaneous rooting (80 %) were noticed on hormone free medium while NAA (0.1 or 0.2 mg dm-3) stimulated it up to 100%, depending on clones. Rooted plants were easily acclimatized.
