Effects of prolonged alcohol exposure on somatotrophs and corticotrophs in adult rats: Stereological and hormonal study

AB  - Exposure to alcohol alters many physiological processes, including endocrine status. The present study examined whether prolonged alcohol (A) exposure could modulate selected stereological and hormonal aspects of pituitary somatotrophs (growth hormone-GH cells) and corticotrophs (adrenocorticotropic hormone-ACTH cells) in adult rats. Changes in pituitary gland volume; the volume density, total number and volume of GH and ACTH cells following alcohol exposure were evaluated using a stereological system (newCAST), while peripheral GH and ACTH levels were determined biochemically. Our results demonstrated the reduction (p < 0.05) of the volume density (37%) and volume of GH cells (29%) in the group A. Also, there was a tendency for the total number of GH cells to be smaller in the group A. Serum GH level was significantly decreased (p < 0.05; 70%) in the group A when compared to control values. Moreover, prolonged alcohol exposure induced declines (p < 0.05) in volume density (24%) and volume of ACTH cells (29%). The total number of ACTH cells and ACTH level were higher (p < 0.05; 42%) in the group A than in control rats. Collectively, these results indicate that prolonged alcohol exposure leads not only to changes in GH and ACTH hormone levels, but also to alterations of the morphological aspects of GH and ACTH cells within the pituitary.

Estradiol and GH cells: Immunohistomorphometric study in an animal model of andropause

AB  - Andropause, the culminating phase of ageing in males is characterized by the decline of the growth hormone (GH)/insulin-like growth factor 1 (IGF-1) axis that is responsible for somatic development. Estrogens on the other hand, assume a relevant somatic role, as well as the one in the pituitary. They are particularly interesting because of their therapeutic uses in certain common ageing-associated diseases. The aim of the present study was to examine the effects of subcutaneous treatment with estradiol, dipropionate on the immunohistomorphometric features of GH cells, in an animal model of the andropause. Sixteen-month-old Wistar rats were divided into sham-operated (SO), orchidectomized (Orx) and estradiol dipropionate-treated orchidectomized (Orx+Edp) groups. Estradiol dipropionate (0.625 mg/kg/day) was administered subcutaneously for three weeks while the SO and Orx groups received the vehicle alone. GH cells were identified by the peroxidase-antiperoxidase (PAP) immunohistochemical procedure. In the Orx+Edp group, GH cell volumes as well as the relative volume densities were significantly (p<0.05) decreased, by 27.4% and by 61.1%, respectively, in comparison with the same parameters of SO animals. Compared to the Orx animals, the cell volumes and relative volume densities of GH cells in Orx+Edp group were also significantly (p<0.05) decreased, by 23.2% and by 64.1%, respectively. It can be concluded that chronic estradiol dipropionate application in an animal in the andropause results in the suppression of immunohistochemical and morphometric parameters of pituitary GH cells.

AB  - As a result of the global warming, the average ambient temperature during summertime has increased in regions with moderate continental climate. The effects of 24 h exposure to heat stress (35±1 °C) on the morphology and function of pituitary adrenocorti cotropes were examined in adult male Wistar rats. Significant changes in the morphofunctional features of adrenocorti cotropes were found after the heat stress, with no differences noted in the cell shape or localization, com pared to controls. The adrenocorti cotropes cell volume, as well as the volume density, were significantly decreased (p<0.05) by 12.3% and 26.7%, respectively, in comparison with controls. The concentration of plasma adrenocorti cotropic hormone and serum corticosterone in the heat stressed group were significantly decreased (p<0.05) by 21.9% and 27.2%, respectively, compared to controls. These findings suggest that 24 h exposure of adult male rats to heat stress has an inhibitory effect on the morphofunctional characteristics of adrenocorticotropes.
AB  - Usled globalnog otopljavanja u regionima sa umereno kontinentalnom klimom dolazi do povećanja srednje vrednosti temperature vazduha to kom letnjih meseci. U ovoj studiji ispitivani su efekti 24-časovnog izlaganja to plotnom stresu (35±1 °C) na morfologiju i funkciju hipofiznih adrenokortikotropa kod odraslih pacova Wistar soja. Dobijeni rezultati ukazuju na značajne promene u morfofunkcionalnim osobinama adrenokortiko tropa nakon izlaganja toplotnom stresu, bez uočenih razlika u obliku i lokalizaciji ćelija u poređenju sa kontrolama. Volumen adrenokortikotropa kao i njihova volumenska gustina bili su značajno smanjeni (p<0,05) za 12,3% odnosno 26,7%, u poređenju sa kontrolom. Koncentracije adrenokortikotropnog hormona u plazmi i kortikosterona u serumu u eksperimentalnoj grupi bile su značajno smanjene za 21,9% odnosno 27,2%, u poređenju sa kontrolnim vrednostima. Navedeni rezultati pokazuju da 24-časovno izlaganje odraslih mužjaka pacova toplotnom stresu ima inhibirajući efekat na morfofunkcionalne osobine hipofiznih adrenokortikotropa.
