AB  - The middle section of the Danube River basin in the previous three Joint
Danube Surveys (JDS) was found to be under intensive pollution, mainly
affected by wastewaters. As a consequence of the lack of wastewater
treatment plants, complex xenobiotics mixtures may get into surface water
and decrease water quality and organisms’ health. Some xenobiotics may
induce different types of DNA damage. Hence, DNA damage could be a
useful biomarker in the detection of “early warning signals” of organisms’
exposure to genotoxic compounds, while the usage of bioassays batteries
can provide better insight into a genotoxic potential. This study was
conducted in 2019, during the JDS4 expedition, to assess the genotoxic
potential of nine sites on the section of the Danube River which goes
through Serbia and its major tributaries: the Sava, Tisza and Velika Morava.
Blood and muscle of Alburnus alburnus (bleak) were used for genotoxicity
assessment by comet, micronucleus and RAPD assays. In comet assay, the
highest level of DNA damage was recorded at the Velika Morava mouth (JDS
39) and on the Danube River, downstream Radujevac (JDS 41). The highest
frequency of micronucleus was recorded at the Sava mouth (JDS 36) and on
the Danube River, downstream Pančevo (JDS 37). The lowest level of DNA
damage in both tests was recorded on the Sava, site Jamena (JDS 35). The
RAPD analysis distinct three major groups: Tisza mouth (JDS 33) and sites
on the Sava (JDS 35, JDS 36), then two sites on the Danube River (JDS 37,
JDS 41) and one site on the Danube River, Ram (JDS 40) with the Velika
Morava mouth (JDS 39). Analyzed bioassays showed different sensitivity
where the comet assay had the highest potential in discrimination of sites
based on genotoxicity. Bleak was proved to be a reliable bioindicator in eco/
genotoxicological studies.

AB  - U Srbiji se prerađuje manje od 13% otpadnih voda,1 što za posledicu ima
ispuštanje velikih količina neprečišćenih voda u vodotokove. Ravničarske reke
su pod značajnim pritiscima ovih izvora zagađenja, s obzirom da delom ili
čitavim tokom protiču kroz naseljene oblasti, kao i oblasti u kojima se nalaze
industrijska postrojenja. U analizi uticaja zagađenja na akvatične organizme,
oštećenje DNК molekula predstavlja značajan biomarker ranog upozorenja jer
može ukazati na uticaj ksenobiotika na biološke sisteme pre nego što se odrazi
na čitav organizam i/ili populaciju. U skladu sa tim, cilj ovog istraživanja je
procena osetljivosti široko rasprostranjene vrste Alburnus alburnus (uklija)
prilikom detekcije genotoksičnog efekta, kako na prostorno bliskim
lokalitetima na Savi, tako na i prostorno udaljenim na Dunavu, Tisi, Savi i
Velikoj Moravi. Za poređenje lokaliteta korišćen je integrisani odgovor
biomarkera (IBR) koji je objedinio pojedinačne biomarkere: vijabilnost ćelija
krvi, nivo DNК oštećenja ćelija krvi detektovanog u alkalnom komet i
mikronukleus testu i indeks zagađenja metalima/prioritetnim supstancama.
Integrisanjem pojedinačnih odgovora biomarkera u jedinstvenu IBR vrednost
omogućeno je jednostavnije poređenje lokaliteta. Dobijeni rezultati su ukazali da
su odgovori biomarkera uklija specifični za lokalitet. U skladu sa rezultatima,
utvrđeno je da uklija može biti pouzdan bioindikator u diskriminaciji i
udaljenih i prostorno bliskih lokaliteta koji su pod različitim uticajem
zagađenja.
AB  - У Србији се прерађује мање од 13% отпадних вода,1 што за последицу има
испуштање великих количина непречишћених вода у водотокове. Равничарске реке
су под значајним притисцима ових извора загађења, с обзиром да делом или
читавим током протичу кроз насељене области, као и области у којима се налазе
индустријска постројења. У анализи утицаја загађења на акватичне организме,
оштећење ДНК молекула представља значајан биомаркер раног упозорења јер
може указати на утицај ксенобиотика на биолошке системе пре него што се одрази
на читав организам и/или популацију. У складу са тим, циљ овог истраживања је
процена осетљивости широко распрострањене врсте Alburnus alburnus (уклија)
приликом детекције генотоксичног ефекта, како на просторно блиским
локалитетима на Сави, тако на и просторно удаљеним на Дунаву, Тиси, Сави и

биомаркера (ИБР) који је објединио појединачне биомаркере: вијабилност ћелија
крви, ниво ДНК оштећења ћелија крви детектованог у алкалном комет и

Интегрисањем појединачних одговора биомаркера у јединствену ИБР вредност
омогућено је једноставније поређење локалитета. Добијени резултати су указали да

AB  - Metoda nasumično umnožene polimorfne DNK (RAPD) je poslednjih godina našla primenu u ekogenotoksikologiji. U okviru dve studije slučaja na Savi i Dunavu, za RAPD analizu korišćeno je mišićno tkivo jedinki Alburnus alburnus (uklija) uzorkovanih sa ukupno devet lokaliteta. RAPD rezultati istraživanja na Savi ukazuju na grupisanje jedinki sa zagađenih lokaliteta (UB i UK), u odnosu na jedinke sa manje zagađenog lokaliteta, SZ. RAPD rezultati studije slučaja na Dunavu ne pokazuju jasno grupisanje uklija u odnosu na jačinu pritisaka što ukazuje na neophodnost dodatnih analiza.
AB  - In recent years the randomly amplified polymorphic DNA method (RAPD) is frequently used in ecogenotoxicology. Within two case studies on the Sava River and the Danube River, muscle tissue of Alburnus alburnus (bleak) specimens sampled from nine sites was used for RAPD analysis. The RAPD results on the Sava River study indicate the grouping of specimens from polluted sites (UB and UK) as opposed to specimens from the less polluted site, SZ. The RAPD results of the Danube River case study do not show a clear bleak grouping in relation to pollution pressures, indicating that additional analyses are needed.

AB  - This study was designed to evaluate the genoprotective, antigenotoxic, as well as antitumor potential of methanolic, ethanolic, and aqueous extracts of Melissa officinalis, Mentha × piperita, Ocimum basilicum, Rosmarinus officinalis, Salvia officinalis, and Satureja montana (Lamiaceae), in different model systems. The polyphenols in these extracts were quantified both spectrophotometrically and using HPLC-DAD technique, while DPPH assay was used to assess the antioxidant activity. The genoprotective potential was tested on pUC19 Escherichia coli XL1-blue, and the antigenotoxicity on Salmonella typhimurium TA1535/pSK1002 and human lung fibroblasts, while the antitumor activity was assessed on colorectal cancer cells. Rosmarinic acid, quercetin, rutin, and luteolin-7-O-glucoside were among the identified compounds. Methanolic extracts had the best DPPH-scavenging and SOS-inducing activities, while ethanolic extracts exhibited the highest antigenotoxicity. Additionally, all extracts exhibited genoprotective potential on plasmid DNA. The antitumor effect was mediated by modulation of reactive oxygen species (ROS), nitric oxide (NO) production, and exhibition of genotoxic effects on tumor cells, especially with O. basilicum ethanolic extract. Generally, the investigated extracts were able to provide antioxidant protection for the acellular, prokaryotic, and normal human DNA, while also modulating the production of ROS and NO in tumor cells, leading to genotoxicity toward these cells and their decrease in proliferation.

AB  - In this study, few different evaluation concepts were used for the assessment of genotoxic potential at the stretch of the Danube River identified as a significant hotspot of pollution originated through the untreated wastewaters. Three sites were chosen: one site upstream of the wastewater outlet in Novi Sad (Serbia), one at the outlet of wastewaters, and one site few kilometer downstream. Ex situ approach comprised prokaryotic SOS/umuC test on Salmonella typhimurium TA1535/pSK1005 and comet assay on human hepatoma cell line (HepG2). In situ approach was based on the active monitoring (cage approach) using freshwater mussels Sinanodonta woodiana and fish Cyprinus carpio. The comet and micronucleus assays were selected for evaluation of DNA damage in mussel haemocytes and fish blood cells. Within the ex situ part of the study, our results indicated that the eukaryotic model system is more sensitive compared to the prokaryotic one. In situ bioassays are recommended for obtaining a better insight into ecosystem status and in the case of our study the complete insight of genotoxic pressure. However, the choice of animals as bioindicators also has a significant impact on the quality of the obtained information. Differential response between fish and mussels was observed at the highly polluted site suggesting possible involvement of additional protective mechanism such as valve closure in mussels.

The major aim of this study was to investigate genotoxic potential of TBQ and selected derivatives in an acellular model by using plasmid DNA, in the prokaryotic model by the SOS/umuC assay in Salmonella typhimurium TA1535/pSK1002 and in eukaryotic models by using comet assay in human fetal lung cell line (MRC-5) and human liver cancer cell line (HepG2). Results indicated that in the acellular model TBQ and its derivatives do not interact with plasmid pUC19. In the prokaryotic model, only TBQ exerted weak genotoxic potential and only at highly cytotoxic concentrations. In eukaryotic models, genotoxic potential was detected mainly at the highest concentrations of the tested substances but the effect was lower in both cell lines in comparison with benzo[a]pyrene and etoposide which were used as positive controls. Weak genotoxic potential of tested compounds recommends them as good candidates for further testing in development of new antitumor agents.

AB  - Due to the increasing industrialization, metals are discharged into all spheres of the environment, in particular, in river courses, which leads to the need for constant monitoring. Metals do not degrade into harmless end products; they are very persistent and have high potential for accumulation in biota. Metals in the fish body are accumulated in different amounts in the tissue specific matter. In relation to the biotic conditions and fish metabolism, the load of food, or the stage of the reproductive cycle, the seasonal variation of metal levels can be expected. Because of that, the objective of our present study was to analyze 15 metals and metalloids in liver, gills, muscle, and gonads of European chub (Squalius cephalus) throughout the 4 seasons, autumn, winter, spring, and summer. The specimens were collected from two rivers, Pestan and Beljanica at the Kolubara basin, and their concentrations were determined with inductively coupled plasma optical emission spectrometry (ICP-OES). Specimens from both rivers have shown similarities in metal accumulation like the highest accumulation of majority of elements in gills, lowest accumulation of majority of elements in muscle (except for Hg), and higher accumulation of some elements in summer (Cu, Fe, Zn). In addition, Cu and Fe showed affinity for liver, while Ba, Cr, Sr, and Zn were specific for gills. Also, Al, B, Fe, Ni, and Pb did not show significant differences in concentrations among different seasons in all investigated tissues.

AB  - The Velika Morava River is the greatest national Serbian river and the significant tributary of the Danube River. The major problems in the Velika Morava River Basin (VMRB) represent untreated industrial and municipal wastewaters. In this study, the level of genotoxic potential at the sites along the VMRB was evaluated by parallel in vitro and in situ approach. Within in vitro testing, genotoxicity of native water samples collected from the sites in VMRB was evaluated by SOS/umuC test on Salmonella typhimurium TA1535/pSK1002 and by the comet assay on HepG2 cells. DNA damage in situ was assessed in bleak (Alburnus alburnus) erythrocytes by the comet (alkaline and Fpg-modified comet) and micronucleus assays. Additionally, the concentration of heavy metals in fish tissue was measured and this data, compiled with the data of the physico-chemical parameters measured in water, was used as a measure of the pollution pressure at the sites. Results showed that applied in vitro tests with native water samples are less sensitive in comparison with in situ tests and should be taken with precaution when making predictions on the status of the ecosystem. Within applied battery of in situ assays differential sensitivity of assays was observed where alkaline comet assay showed the highest potential in differentiation of the sites based on genotoxic potential. Integrated biomarker response showed that usage of the battery of bioassays provides better insight in a genotoxic effects in animals, and consequently, that the holistic approach is more suitable for this type of study.

To emphasize the impact of wastewaters, this study was carried out in a season with low water discharge. The level of genotoxic potential at the sites was studied by alkaline and Fpg-modified comet assay in fish blood, while RAPD technique was employed on the fish muscle tissue. The variations in the genotoxic potential were observed along the Sava River based on the level of DNA damage in blood of the specimens collected at the selected sites. Clustering of the impacted sites was also observed based on RAPD fingerprints. Comparison of the data obtained in RAPD and the data from the comet assay indicated the possible reflection of genotoxicity on the genetic variability in chub collected from the sites with different pollution pressure. This is a preliminary study which reports the differentiation of the population of chub along the Sava River based on RAPD analysis.

AB  - Cyanobacteria are present in many aquatic ecosystems in Serbia. Lake Ludoš, a wetland area of international significance and an important habitat for waterbirds, has become the subject of intense research interest because of practically continuous blooming of cyanobacteria. Analyses of water samples indicated a deterioration of ecological condition and water quality, and the presence of toxin-producing cyanobacteria (the most abundant Limnothrix redekei, Pseudanabaena limnetica, Planktothrix agardhii and Microcystis spp.). Furthermore, microcystins were detected in plants and animals from the lake: in macrophyte rhizomes (Phragmites communis, Typha latifolia and Nymphaea elegans), and in the muscle, intestines, kidneys, gonads and gills of fish (Carassius gibelio). Moreover, histopathological deleterious effects (liver, kidney, gills and intestines) and DNA damage (liver and gills) were observed in fish. A potential treatment for the reduction of cyanobacterial populations employing hydrogen peroxide was tested during this study. The treatment was not effective in laboratory tests although further in-lake trials are needed to make final conclusions about the applicability of the method. Based on our observations of the cyanobacterial populations and cyanotoxins in the water, as well as other aquatic organisms and, a survey of historical data on Lake Ludoš, it can be concluded that the lake is continuously in a poor ecological state. Conservation of the lake in order to protect the waterbirds (without urgent control of eutrophication) actually endangers them and the rest of the biota in this wetland habitat, and possibly other ecosystems. Thus, urgent measures for restoration are required, so that the preservation of this Ramsar site would be meaningful.

AB  - In this study, mutagenic and genotoxic potential of anti-tumor compounds avarol, avarone, and its derivatives 3'-methoxyavarone, 4'-(methylamino)avarone and 3'-(methylamino)avarone was evaluated and compared to cytostatics commonly used in chemotherapy (5-fluorouracil, etoposid, and cisplatin). Mutagenic potential of selected hydroquinone and quinones was assessed in prokaryotic model by the SOS/umuC assay in Salmonella typhimurium TA1535/pSK1002. Genotoxic potential was also assessed in eukaryotic models using comet assay in human fetal lung cell line (MRC-5), human adenocarcinoma epithelial cell line (A549), and in human peripheral blood cells (HPBC). The results indicated that avarol and avarone do not exert mutagenic/genotoxic potential. Among the studied avarone derivatives, mutagenic potential was detected by SOS/umuC test for 3'-(methylamino)avarone, but only after metabolic activation. The results of comet assay indicated that 3'-methoxyavarone and 3'-(methylamino)avarone have a significant impact on the level of DNA damage in the MRC-5 cell line. Genotoxic potential was not observed in A549 cells or HPBC probably due to a different uptake rate for the compounds and lower in metabolism rate within these cells.

AB  - Biomarkers attract increasing attention in environmental studies, as a tool for detection of exposure and effects of pollution, from both natural and anthropogenic sources. This study aims to assess the impact of multiple stressors during distinctive seasons, covering also extreme hydrological events (extensive flooding in the mid May 2014), on different levels of biological organization in the liver and gills of three closely related freshwater breams. Our previous study on DNA damage in blood cells of these specimens showed increased DNA damage in June 2014, one month after the flooding event. As a continuation of that research, the present study was conducted. As a biomarker of exposure DNA damage was measured by applying the alkaline comet assay, while histopathological alterations were monitored as a biomarker of effect. Additionally, concentrations of metals and metalloids in gills, liver and muscle were assessed. Sampling of fish tissues was performed in 2014, during winter (January and February), spring (March and early June) and summer (late June, July and August). Significant seasonal difference in DNA damage was observed for both tissues. During spring and summer the level of DNA damage in gills was significantly higher when compared to the liver. Histopathological analyses showed higher frequency of alterations in gills during spring, and in liver during summer, but without a significant seasonal difference. Gills had the highest concentration of metals and metalloids during the spring and summer, and liver during winter. Muscle was the least affected tissue during all three seasons. This study highlighted the importance of the multiple biomarker approach and the use of different fish tissues in assessment of surface water pollution.

AB  - The success of antioxidant therapy in hyperthyroidism implies that disease is mediated by oxidative stress, which is known as one of the causing agents of ageing, degenerative diseases, and cancer. The main objective of our study was to determine possible protective effects of methanolic extract of N. rtanjensis in triiodothyronine (T3)-induced DNA breaks of human lymphocytes under in vitro conditions, based upon plant antioxidant capacity related to its phytochemical profile, mainly its polyphenolic complex. The total phenolic and flavonoid content and the antioxidant activity using in vitro 1,1-dyphenyl-2- picrylhydrazyl reagent (DPPH) was determined in methanolic extracts of plant leaves and flowers. The phenolic compound content of 62.73±1.80mg of GaA/g, exhibited solid antioxidant activity (IC50= 112.59±0.95μg/ml). The antigenotoxic activity of 0.2, 0.5 and 1.0mg/ml N. rtanjensis methanol extracts mixture with 100µM of T3 was studied in human lymphocytes in vitro using the Comet assay. It is supposed that the antigenotoxicity of N. rtanjensis methanol extracts was caused by high presence of chlorogenic acid, rosmarinic acid and rutin, all known as efficient antioxidant bioactive compounds, which were determined by ultrahigh-pressure liquid chromatograph with MS/MS Mass Spectroscopy (UHPLC/-HESI-MS / MS).

(Unio pictorum/Unio tumidus) and Sinanodonta woodiana. The comet assay was used for the assessment of DNA damage. The research was performed on 34 out of 68 sites analysed within the Joint Danube Survey 3 - the world's biggest river research expedition of its kind in 2013. During research, 2285 river kilometres were covered with an average distance of 68km between the sites. The complex data set on concentrations of various substances present in water, suspended particulate matter and sediment on investigated sites gave the opportunity to identify the groups of xenobiotics which mostly affect the studied biomarker - DNA damage. The highest levels of DNA damage were recorded in the section VI (Panonnian Plain), which is under the impact of untreated wastewater discharges. Both positive and negative influences of the large tributaries on the level of genotoxicity in the Danube River were evident. Significant correlation in response was detected between the studied species of freshwater mussels. The level of DNA damage in mussels correlated with concentrations of compounds from the group of hazardous priority substances (polycyclic aromatic hydrocarbons), persistent organic pollutants (dioxins) and emerging pollutants (Oxazepam, Chloridazon-desphenyl).

AB  - Extreme hydrological events, such as water scarcity and flooding, can modify the effect of other stressors present in aquatic environment, which could result in the significant changes in the ecosystem functioning. Presence and interaction of various stressors (genotoxic pollutants) in the environment can influence the integrity of DNA molecules in aquatic organisms which can be negatively reflected on the individual, population and community levels. Therefore, in this study we have investigated the impact of flooding, in terms of genotoxicity, on organisms belonging to different trophic levels. The study was carried out on the site situated in the lower stretch of the Sava River which faced devastating effects of severe flooding in May 2014. The flooding occurred during our field experiment and this event provided a unique opportunity to assess its influence to the environment. The in situ effects of this specific situation were monitored by measuring physical, chemical and microbiological parameters of water, and by comparing the level of DNA damage in coelomocytes and haemocytes of freshwater worms Branchiura sowerbyi, haemocytes of freshwater mussels Unio tumidus and blood cells of freshwater fish Abramis bjoerkna/Abramis sapa, by means of the comet assay. Our study indicated that the flooding had a significant impact on water quality by decreasing the amount and discharge rate of urban wastewaters but simultaneously introducing contaminants from the nearby fly ash disposal field into river by runoff, which had diverse effects on the level of DNA damage in the studied organisms. This indicates that the assessment of genotoxic pollution in situ is strongly affected by the choice of the bioindicator organism.

Atmospheric nitrate deposition or nitrate-containing fertilizers were found not to be significant sources of riverine nitrate in the SRB. The ranges of isotope values suggest that NO3- in the SRB derives from soil nitrification, sewage, and/or manure, which were further supported by MST analysis. Microbiological indicators show the existence of hotspots of fecal pollution in the SRB, which are human associated. Long-term observations indicate persistent fecal contamination at selected locations caused by continuous discharge of untreated wastewaters into the SRB.

AB  - In this study we have performed a comprehensive genotoxicological survey along the 900 rkm of the Sava River. In total, 12 sites were chosen in compliance with the goals of GLOBAQUA project dealing with the effects of multiple stressors on biodiversity and functioning of aquatic ecosystems. The genotoxic potential was assessed using a complex battery of bioassays performed in prokaryotes and aquatic eukaryotes (freshwater fish). Battery comprised evaluation of mutagenicity by SOS/umuC test in Salmonella typhimuriumTA1535/ pSK1002. The level of DNA damage as a biomarker of exposure (comet assay) and biomarker of effect (micronucleus assay) and the level of oxidative stress as well (Fpg-modified comet assay) was studied in blood cells of bleak and spirlin (Alburnus alburnus/ Alburnoides bipunctatus respectively). Result indicated differential sensitivity of applied bioassays in detection of genotoxic pressure. The standard and Fpg-modified comet assay showed higher potential in differentiation of the sites based on genotoxic potential in comparison with micronucleus assay and SOS/umuC test. Our data represent snapshot of the current status of the river which indicates the presence of genotoxic potential along the river which can be traced to the deterioration of quality of the Sava River by communal and industrialwastewaters. The major highlight of the study is that we have provided complex set of data obtained from a single source (homogeneity of analyses for all samples).

The advantage of genotoxicity as a marker for pollution exposure in fish is the early detection of possible long-term effects such as cancer. Therefore, genotoxicity was in the focus of the biomarker investigations in fish during the expedition. Blood samples of common bleak (Alburnus alburnus) for the investigation of the micronucleus frequency and comet tail intensity of fragmented DNA material in erythrocytes were collected at 18 and 12 sampling sites, respectively. For 9 sampling sites same samples were used to compare the . in-situ data for the comparable genotoxic endpoint in the micronucleus (MN) and comet assay (CM). The data of both . in-situ assays showed a significant correlation, indicating the strength and comparability of the data sets. Significant variation in DNA damage in fish along the longitudinal profile of the Danube was demonstrated for both assays compared to reference sites. The results suggest that DNA damage in erythrocytes of fish was mainly affected by wastewater of highly populated regions. No linkage between the results and the general health/dietary status of the fish were revealed, whereas correlation with some genotoxicity drivers in the water phase, suspended particulate matter and sediments could be demonstrated.

AB  - In this study a comprehensive genotoxicological survey throughout the upper and middle stretches of Adige river basin is presented. The study was carried out at 7 sites located along the Adige main course and one the most significant tributaries, the Noce creek, both presenting different levels of pollution pressure. To give an insight into the nature of the genotoxic activity we employed the battery of prokaryotic and eukaryotic assays. Mutagenicity in water samples was evaluated by SOS/umuC test in Salmonella typhimurium TA1535/pSK1002. The level of DNA damage as a biomarker of exposure (comet assay) and biomarker of effect (micronucleus assay) and the level of oxidative stress as well (Fpg - modified comet assay) were studied in blood cells of Salmo cenerinus Nardo, 1847 and Salmo marmoratus Cuvier, 1829. Within the applied bioassays, comet assay showed the highest potential for discriminating the sampling sites which are under lesser extent of pressure (sampling sites 1-Barnes at Bresimo and 4-Noce downstream S. Giustina) from the sites under high pressure (sampling sites 5-Noce at Mezzolombardo and 6/7-Adige upstream and downstream municipality of Trento). Significant correlation between the standard and Fpg - modified comet assay indicated that oxidative stress could be a major contributor to observed DNA damage in collected specimens.

AB  - The current study extends our previous findings concerning the sensitivity of Lymantria dispar larvae to
cadmium in light of ecotoxicological risk assessment. Here we report the results of the comet assay
performed for the first time on this species. We examined the chronic effects of two cadmium concentrations (50 and 100 μg Cd/g dry food) on DNA integrity and haemocyte viability, as well as on fitnessrelated traits (larval mass and development duration parameters). All parameters were assessed individually and then used to calculate the integrated biomarker response (IBR) index. Egg-masses of
L. dispar were collected from two locations in Serbia - the uncontaminated Homolje mountains and a
metal-polluted area near Bor copper mines, smelter and refinery. Distinctive patterns in the response of
these populations to cadmium exposure were noticed. In haemocytes of larvae from the pollution-free
location both cadmium treatments increased the level of DNA damage, although in a similar range. Haemocyte viability and larval mass were reduced, while duration of the fourth instar and total development time were prolonged in a concentration-dependent manner. Cadmium tolerance was noticeable in the population from the metal-contaminated site at all organizational levels. Nevertheless, haemocyte viability in that population was reduced by the stronger treatment. Haemocyte viability was recognized as a promising biomarker due to the evident response of both populations to dietary cadmium. Genotoxicity, fitness-related traits and the IBR index could be used for biomonitoring of sensitive populations not previously exposed to metals.

AB  - Genotoxicity monitoring of the lower stretch of the Sava River was performed by the combined approach of in situ assessment of genotoxicity and active biomonitoring of two species of mussels from the Unionidae family, Unio pictorum and Unio tumidus. Genotoxic response was studied using comet assay on hemocytes. For active biomonitoring, the mussels were acclimated to controlled laboratory conditions for 10 days and then exposed at two sites in the Sava River in the area of the city of Belgrade. Hemolymph of exposed specimens of each species was taken after 7, 14, and 30 days of exposure. For in situ assessment, the mussels were collected from five sites in the lower flow of the Sava River. The mussels were sampled immediately after the acclimation served as controls in both types of monitoring procedures. The results of our studies indicated the presence of genotoxic pollution at all studied sites at the Sava River. The level of DNA damage varied at different sites depending on the source and level of pollution. The response to genotoxic pollution was evident at the site in the urban area of Belgrade city, as well as at the sites far from the large urban settlements, suggesting that the lower flow of the Sava River is under pollution pressure.

AB  - Data on the microbiological quality of the freshwater systems under the anthropogenic influence, such as the Sava River, are of the major importance for the water resource management. Furthermore, analyses of the microbial quality of fish meat provide information of the fish as a valuable food resource from the investigated river basin. The health status of the fish, including dynamics of infection and biodiversity of endoparasites, is important bioindicator of changes in the ecosystem structure and function. For the ecosystem-based approach to the Sava River management, investigations of microbiological quality of the Sava River water and the meat of the European chub as the bioindicator organism, as well as dynamics of infection/biodiversity of intestinal parasites Acanthocephala, were performed. The survey comprised the data collected in periods 2005, 2006 and 2012. Microbiological investigation of water was performed in 2006 and 2012, while microbiological analyses of fish meat and ichthyo-parasitological investigation took place during 2005–2006. A high number of heterotrophic bacteria were recorded during 2006 survey, confirmed by the distinctly higher values of the three faecal indicators (total coliform, E. coli and enterococci), and indicated poor water quality downstream of the cities Zagreb and Velika Gorica, as a result of the municipal sewage outlets. The results from 2012 survey indicated the existence of moderate to critical faecal and organic pollution in all samples. Accumulation of the bacteria in the European chub meat was mainly uniform along the watercourse within standards and limitations for the human consumption. Sampling sites downstream cities of Zagreb and Velika Gorica were characterised with the lower prevalence and abundance of two common species of the chub intestinal acanthocephalan parasites, Pomphorhynchus laevis and Acanthocephalus anguillae. Poor microbiological quality of the water and lower distribution of chub intestinal parasites were related to the anthropogenic influence, downstream of the urban areas.

Effects of 5-Fluorouracil, Etoposide and CdCl2 in Aquatic Oligochaeta Limnodrilus udekemianus Claparede (Tubificidae) Measured by Comet Assay

AB  - Genotoxicity of 5-fluorouracil (5-FU), etoposide (ET) and cadmium
   chloride (CdCl2) was evaluated in Limnodrilus udekemianus, cosmopolitan
   tubificid species, by alkaline single-cell gel electrophoresis (comet
   assay). Groups of 50 individuals were exposed in vivo in water-only
   short-term (96 h) tests to 5-FU (0.004, 0.04, 0.4, 4 and 40 mu M), ET
   (0.004. 0.04, 0.4 and 4 mu M) and CdCl2 (0.004, 0.04, 0.4, 4 and 40 mu
   M). Mortality of worms was observed only for CdCl2 (4 and 40 mu M). Cell
   viability lower than 70 \% was detected for 5-FU (0.4, 4 and 40 mu M),
   ET (4 mu M) and CdCl2 (0.4 and 4 mu M). All tested substances induced
   significant increase of DNA damage except 0.004 mu M of ET. L.
   udekemianus being sensitive to all tested substances indicates that it
   can be used in ecogenotoxicology studies. Concern should be raised to
   cytostatics, especially to 5-FU, since concentration of 0.004 mu M
   induced DNA damage is similar to ones detected in wastewaters.

AB  - This study was undertaken to investigate the potential of the freshwater mussel Sinanodonta woodiana for detection of genotoxic pollution of the environment. Study was performed at two sites in the Velika Morava River, from May 2010 to February 2011. The alkaline comet assay on haemocytes was used, and the olive tail moment (OTM) was chosen as a measure of DNA damage. The specimens held on acclimation under controlled laboratory conditions for 10 d were used as a control. Chemical analysis revealed the presence of phosphates and increased concentrations of zinc, copper and nickel at both sites during the entire sampling period. The values of OTM in mussels collected from the environment, significantly correlated with the concentration of zinc (r = 0.6248), temperature (r = 0.7006) and dissolved oxygen (r = 0.7738). Seasonal variations in genotoxic response were observed, with the highest OTM values obtained during summer months. Preliminary results of the in vitro study indicated the effect of water temperature on genotoxic response to zinc and cadmium in S. woodiana suggesting that the presence of genotoxic pollutants during months with lower temperature could be under-estimated. Obtained results indicate that S. woodiana could be a valuable tool for active biomonitoring of aquatic environments and emphasizes the importance of seasonal genotoxic monitoring with this organism.
