Beyond Traditional Use of Alchemilla vulgaris: Genoprotective and Antitumor Activity In Vitro

AB  - Alchemilla vulgaris L. (lady’s mantle) was used for centuries in Europe and Balkan countries for treatments of numerous conditions and diseases of the reproductive system, yet some of the biological activities of lady’s mantle have been poorly studied and neglected. The present study aimed to estimate the potential of A. vulgaris ethanolic extract from Southeast Serbia to prevent and suppress tumor development in vitro, validated by antioxidant, genoprotective, and cytotoxic properties. A total of 45 compounds were detected by UHPLC–HRMS analysis in A. vulgaris ethanolic extract. Measurement of antioxidant activity revealed the significant potential of the tested extract to scavenge free radicals. In addition, the analysis of micronuclei showed an in vitro protective effect on chromosome aberrations in peripheral human lymphocytes. A. vulgaris extract strongly suppressed the growth of human cell lines derived from different types of tumors (MCF-7, A375, A549, and HCT116). The observed antitumor effect is realized through the blockade of cell division, caspase-dependent apoptosis, and autophagic cell death. Our study has shown that Alchemilla vulgaris L. is a valuable source of bioactive compounds able to protect the subcellular structure from damage, thus preventing tumorigenesis as well as suppressing tumor cell growth.

Antitumor properites of alchemilla vulgaris agg.

AB  - Alchemilla vulgaris is well known in traditional medicine especially for the treatment
of gynecological problems in women. Several ethnomedicinal studies for the territory
of Balkan reports its well known biological properties against many conditions such
as infertility, dysmenorrhea, cysts, menopausal complaints and endometriosis.
Concidering ethnomedicinal data on female illnesses, the aim of our study was to
determine whether ethanol extract of Alchemilla vulgaris agg. exert antitumor effect
against mouse breast cancer cells in vitro. Treatment with Alchemilla vulgaris agg. extract
decreased viability of mouse breast cancer cells (4T1) in dose-dependent manner after
72 h. The viability decrease was followed by loss of dividing potential after the treatment.
In parallel with this, certain percentage of 4T1 cells was subjected to programmed cell
death ̶ apoptosis. Detected apoptosis was followed with caspase activation while typical
apoptotic morphology of treated cells was observed by fluorescent microscopy. Apart
from inhibited cell division and induced apoptosis, decreased cell viability was due to
triggered autophagy cell death. In parallel, diminished metastatic potential of these
cells was confirmed by abrogated adhesion, invasion, migration and decreased colony
forming potential after the treatment. All mentioned effects can be connected with
enhanced production of ROS and intracellular NO after the treatment with Alchemilla
vulgaris agg. Taken together, the effect of Alchemilla vulgaris agg. against breast cancer
cells makes this plant worthwhile for further evaluation in the field of oncology.
AB  - Alchemilla vulgaris је добро позната у традиционалној медицини, посебно за
лечење гинеколошких тегоба код жена. Неколико етномедицинских студија за
територију Балкана описује благотворна дејства ове биљке против различитих
патолошких стања попут неплодности, дисменореје, циста, тегоба у менопаузи
и ендометриозе. Циљ нашег истраживања био је да се утврдe антитуморска свој-
ства етанолног екстракта Alchemilla vulgaris agg. на ћелијској линији тумора дојке
4Т1 in vitro. Третман екстрактом Alchemilla vulgaris agg. у трајању од 72 сата је
смањио вијабилитет 4Т1 ћелија на дозно-зависан начин. Смањење вијабилно-
сти праћено је губитком пролиферативног потенцијала ћелија. Поред тога, одре-
ђени проценат 4Т1 ћелија подлегао је програмираној ћелијској смрти познатој
као апоптоза. Детектована апоптоза је праћена активацијом каспаза и додатно
потврђена типичном морфологијом нуклеуса коришћењем флуоресцентне ми-
кроскопије. Свеукупној антитуморској активности екстракта Alchemilla vulgaris
agg. доприноси и ћелијска смрт аутофагијом. Паралелно, метастатски потенцијал
ових ћелија је смањен, што је потврђено смањеном адхезијом, инвазијом, мигра-
цијом и потенцијалом ћелија да формирају колоније. Сви поменути ефекти могу
бити у корелацији са повећаном продукцијом реактивних кисеоничних и азот-
них врста детектованом након третмана. У целини, добијени резултати о делова-
њу Alchemilla vulgaris agg. на ћелије рака дојке чине ову биљку вредном пажње за
даљу евалуацију у области експерименталне онкологије.

AB  - Several ethnobotanical reports on Alchemilla vulgaris L. pointed out diverse biological properties against problems such as dysmenorrhea, pruritus vulvae, menopausal complaints 
as  well  as  related  diseases  in  women.  Also  previous  studies  have  shown  that  Alchemilla  vulgaris  L.  extracts  are  exhibiting  antiinflammatory,  antioxidant,  wound  healing  and 
neuroprotective activity. The aim of this study was to evaluate the direct effect of Alchemilla vulgaris L. ethanol extract against melanoma cells in vitro and in vivo, as well as its effect 
on tumor microenvironment ex vivo. This study was performed on two different mouse melanoma cell lines, B16 and B16F10, and on syngeneic mouse melanoma model in vivo. 
Obtained results revealed dose‐dependent decrease of cell viability after 72 h‐ treatment with Alchemilla vulgaris L. extract. The observed effect was followed by loss of dividing 
potential in both tested cell lines. In parallel with this, certain percentage of B16F10 cells was subjected to programmed cell death in a caspase independent manner while in B16 cells 
estimation of the presence of autophagosomes by flow cytometry has shown that autophagy is occurring after the treatment and it is shown to be mechanism of death. Concerning in 
vivo studies Alchemilla vulgaris L. extract significantly reduced tumor growth in B16 melanoma model partly through stimulation of antitumor immune responce. It altered dendritic 
cells phenotype which activated cytotoxic and CD4+ T lymphocytes to successfully destroy tumor cells. In summary, these data indicate that Alchemilla vulgaris L. is valuable of further 
investigation in the field of experimental oncology.

AB  - In this study, we examined Salvia amplexicaulis Lam., S. jurisicii Košanin and S. ringens Sibth. & Sm. collected in the Republic of North Macedonia, which are for the first time subjected to detailed micromorphological and structural analysis using light and scanning electron microscopy. The nutlets and mucilage were additionally subjected to spectroscopic analysis using Raman and Attenuated Total Reflectance Fourier Transform Infrared spectroscopy. The anatomical structure of stems and leaves is described and compared. The stems, leaves and calyces bear numerous one- and multi-cellular nonglandular trichomes, and various peltate, capitate and digitiform glandular trichomes. The nutlets differ in size and shape, as well as in myxocarpy. The nutlets predominantly contained α-linolenic and linoleic acid, whereas the mucilages are primarily formed of polysaccharides. The results obtained in this study confirmed the importance of micromorphological and anatomical analysis of Salvia L. spp. plant parts, particularly trichomes and nutlets, and hence contributing to the knowledge about the variety of micromorphological characteristics within the genus Salvia L.

The nutlets and mucilage were additionally subjected to spectroscopic analysis using Raman and Attenuated Total Reflectance Fourier Transform Infrared spectroscopy. The anatomical structure of stems and leaves is described and compared. The stems, leaves and calyces bear numerous one- and multi-cellular nonglandular trichomes, and various peltate, capitate and digitiform glandular trichomes. The nutlets differ in size and shape, as well as in myxocarpy. The nutlets predominantly contained α-linolenic and linoleic acid, whereas the mucilages are primarily formed of polysaccharides. The results obtained in this study confirmed the importance of micromorphological and anatomical analysis of Salvia L. spp. plant parts, particularly trichomes and nutlets, and hence contributing to the knowledge about the variety of micromorphological characteristics within the genus Salvia L.",

AB  - U okviru projekta bilateralne saradnje Republike Srbije i Narodne Republike Kine testirana su antitumorska svojstva ukupnih ekstrakata izolovanih iz biljaka: rtanjski čaj (Satureja montana subsp. kitaibelii), trava iva (Teucrium montanum), virak (Alchemilla vulgaris agg.), bela imela (Viscum album subsp. album), zečiji trn (Ononis spinosa), detelina kamenjarka (Anthyllis vulneraria), čestoslavica (Veronica chamaedrys), različak (Centaurea cyanis), svećica (Gentiana asclepiadea), vilino sito (Carlina acaulis), mirisni zdravac (Geranium macrorrhyzum), kotrljan (Eryngium amethystinum), izop (Hyssopus officinalis) i slatinski pelin (Artemisia santonicum). Efekat ekstrakata ispitivan je na humanim ćelijskim linijama tumora dojke MCF-7, melanoma A375, karcinoma pluća A549 i kolona HCT116, kao i ćelijama peritonealnog eksudata miša. Uticaj na vijabilnost tumorskih ćelija praćen je sulforodamin (SRB) i testom mitohondrijalne dehidrogenaze (MTT). Ekstrakti biljaka svećice, različka, vilinog sita i izopa nisu redukovali vijabilitet tumorskih ćelija. Sa druge strane ekstrakti ive, virka, mirisnog zdravca i kotrljana su pokazali značajan potencijal da ograniče rast ćelijske linije karcinoma pluća inhibirajući deobu ćelija i indukujući kaspazama posredovanu apoptozu. Tumoricidna aktivnost ekstrakata deteline kamenjarke i rtanjskog čaja je bila najsnažnija na liniji kancera dojke dok je pad vijabilnosti najverovatnije posledica intenzivirane autofagije i smanjenog proliferativnog kapaciteta ćelija. Tretmani ćelija ispitivanim ekstraktima bili su praćeni smanjenjem produkcije slobodnih radikala kiseonika/ azota ukazujući na to da ekstrakti poseduju izvestan antioksidativni potencijal ali da ove reaktivne vrste nisu medijatori njihove tumoricidne aktivnosti. Testirani ekstrakti koji su ispoljili antitumorsku aktivnost, sa izuzetkom ekstrakta kotrljana, su bili u istom opsegu doza netoksični za ćelije peritonealnog eksudata zdravih miševa ukazujući na selektivnost prema malignom fenotipu.

AB  - The current study was designed to evaluate the phytochemical profile and biological properties (antioxidant, enzyme inhibitory and cytotoxic activity) of methanolic, ethanolic, ethyl-acetate and water extracts of Alchemilla vulgaris from Southeast Serbia (central Balkan) which was traditionally used to alleviate and treat many diseases. Bioactive compounds were characterized by LC-MS/MS technique and biological properties were evaluated using antioxidant, enzyme inhibitory and cytotoxic assays. Twenty-six phenolic compounds were quantified in methanolic, ethanolic, ethyl acetate and water extracts, whereas the highest yields were found in ethyl-acetate (EA) extract (gallic acid, caffeic acid, catchin, quercetin). This extract has also shown the greatest antioxidant, anticancer and inhibitory enzyme activities which were demonstrated for the first time in this study. The obtained results indicated that Alchemilla vulgaris from South Serbia possesses high potential for pharmaceutical applications.

AB  - The majority of species belonging to genus Nepeta (fam. Lamiaceae) produce and accumulate nepetalactones as the main secondary metabolites. Nepetalactones are iridoid monoterpenoids recognized for their wide spectrum of high-value bioactive properties, namely antimicrobial, cytostatic, anti-inflammatory, phytotoxic, etc. However, biosynthetic pathway of these compounds in plants remains to a great extent unexplored. Most probably they derive through general iridoid biosynthetic pathway leading to nepetalactol, a common intermediate of all plant-produced iridoids, that is formed in a reaction catalyzed by iridoid synthase (IS, EC 1.3.1.99). Present investigation entails homology based degenerative primer approach which led to the identification of two promising iridoid synthase candidates (NrIS1 and NrIS2) in leaves of Nepeta rtanjensis Diklić & Milojević, rich in trans,cis-nepetalactone (trans,cis-NL) and dehydronepetalactone (DNL). Comparative UHPLC/qqqMS profiling of nepetalactones in detached trichomes and abraded leaves, complemented with Raman spectroscopy and dichloromethane dipping experiment highlighted trichomes as the main site of their accumulation. This was further supported by the significantly high trichome/abraded leaves expression ratio of NrIS1 (~31) and NrIS2 (~25), which strongly indicated the involvement of these genes in nepetalactone biosynthesis. High correlation between NrIS1 expression profile and trans,cis-NL and DNL contents in five developmental stages of N. rtanjensis leaves confirmed developmental regulation of nepetalactone biosynthesis. Concentrations of trans,cis-NL and DNL were 22-fold and 80-fold lower in the oldest leaves investigated than in the youngest leaves, respectively. In addition, the expression pattern of the NrIS1 followed the same trend exhibiting 7.4-fold higher expression in the oldest leaves compared to the youngest ones. The present study on spatial and developmental regulation of the nepetalactones production and accumulation in N. rtanjensis leaves, as well as the identification of two IS candidate genes is a proof-of-the-concept research that serves as a cornerstone for future studies on elucidation of the nepetalactones biosynthesis.

AB  - The essential oils obtained by hydrodistillation from the aerial parts
   of Artemisia judaica L., Artemisia herbaalba Asso. and Artemisia
   arborescens L. (cultivated) from Libya, were analyzed by GC and GC-MS.
   The antimicrobial properties were determined using the broth
   microdilution method against eight bacterial species: Bacillus cereus
   (clinical isolate), Micrococcus flavus (ATCC10240), Listeria
   monocytogenes (NCTC7973), Staphylococcus aureus (ATCC6538), Escherichia
   coli (ATCC35210), Pseudomonas aeruginosa (ATCC27853), Salmonella
   typhimurium (ATCC13311), Enterobacter cloacae (human isolates) and eight
   fungal species: Aspergillus niger (ATCC6275), A. ochraceus (ATCC12066),
   A. versicolor (ATCC11730), A. fumigatus (ATCC1022), Penicillium
   ochrochloron (ATCC9112), P. funiculosum (ATCC10509), Trichoderma viride
   (IAM5061) and Candida albicans (human isolate). The major constituents
   of A. arborescens oil were sesquiterpene hydrocarbons (47.4\%).
   Oxygenated monoterpenes were the dominant constituents in the A. judaica
   and A. herba-alba oils (54.2\% and 77.3\%, respectively). Camphor
   (24.7\%) and chamazulene (20.9\%) were the major components in the
   essential oil of A. arborescens, chrysanthenone (20.8\%),
   cis-chrysanthenyl acetate (17.6\%) and cis-thujone (13.6\%) dominated in
   the A. herba-alba oil, and the major constituents in the A. judaica oil
   were piperitone (30.21\%) and cis-chrysanthenol (9.1\%). The best
   antimicrobial activity was obtained for A. judaica oil and the lowest
   effect was noticed in A. arborescens oil. The effect of the tested oils
   was higher against Gram (+) than Gram (-) bacteria. All three oils
   showed the best antibacterial activity against Listeria monocytogenes
   and the lowest against Pseudomonas aeruginosa, Escherichia coli,
   Enterobacter cloacae, compared to streptomycin and ampicillin. All three
   oils showed better antifungal activities than ketoconazole, except A.
   arborescens oil against Aspergillus niger.
