AB  - Long-term exposure of populations to pollution may result in enhanced ability to cope with environmental stress. To compare the responses of two Lymantria dispar populations living in unpolluted and polluted forests (UP and PP, respectively), we chronically exposed larvae to cadmium at concentrations of 50 and 100 μg Cd/g dry food (Cd1 and Cd2, respectively). We examined cadmium accumulation in the midgut and hemolymph, activities of superoxide dismutase (SOD), catalase (CAT), and alkaline phosphatases (ALP) in the midgut, as well as Hsp70 protein expression in the midgut, hemolymph, and brain and evaluated these parameters as biomarkers of cadmium contamination. Larvae from PP, fed a control diet, showed higher activity of SOD and increased Hsp70 expression compared with larvae from UP. Excessive amounts of Cd were accumulated in the midgut of all Cd-fed larvae, whereas Cd content in the hemolymph was elevated only in larvae from PP after Cd2 treatment. In larvae from UP, Cd2 treatment decreased the activity of CAT and induced the expression of Hsp70 in the midgut and hemolymph. In larvae from PP, exposure to both Cd concentrations strongly attenuated SOD and CAT activities, while Hsp70 expression was not induced in any organ/tissue. Cd did not affect ALP activity in either population. Midgut Cd content proved to be a suitable indicator of Cd contamination for both polluted and unpolluted habitats.

AB  - Кадмијум је глобално присутан токсични метал који изазива бројне штетне ефекте у организму и заузима седмо место на листи приоритетних супстанци од којих је неопходно пронаћи оптималне начине заштите. Наше пређашње студије су показале да егзополисахарид изолован из соја Lactiplantibacillus plantarum BGAN8 (EPS-AN8) показује висок афинитет за везивање јона кадмијума у воденом раствору и остварује значајан ниво in vitro заштите Caco-2 ћелија од његових токсичних ефеката. Имајући у виду да је за општу популацију најзаступљенији унос кадмијума исхраном, у овој студији је праћен паралелни ефекат уноса кадмијума (кроз воду) и EPS-AN8 (кроз храну) у DA пацовима. Након 30 дана третмана, утврђено је да је унос EPS-AN8 допринео сниженом нивоу депонованог метала у бубрезима, јетри и крви, а повећаном садржају у фекалном материјалу. Такође, микрографије дуоденума, бубрега и јетре су указале на ниже степене кадмијумом индукованих оштећења. Праћењем активности ензима који учествују у антиоксидативној заштити (CAT, GST), терминалних производа липидне пероксидације (MDA) и продукције проинфламаторних цитокина (IL-1β, TNFα, IFN-γ) у хомогенатима дуоденума, утврђен је нижи ниво оксидативног стреса и инфламације, што представља додатни показатељ заштите коју остварује EPS-AN8. С обзиром да орални унос кадмијума доводи до значајних промена у цревној микробиоти, изоловали смо тоталну DNK из дуоденума и секвенцирали 16S rDNK ампликон на Illumina NovaSeq платформи. Примећено је да унос EPS-AN8 ублажава појаву промена у релативној заступљености бактеријских родова и врста карактерисичних за излагање кадмијуму, попут пораста броја опортунистичких патогених бактерија и смањења бројности лактобацила. Остварени заштитни ефекти указују на снажан потенцијал примене EPS-AN8 у условима високе контаминације кадмијумом.
AB  - Kadmijum je globalno prisutan toksični metal koji izaziva brojne štetne efekte u organizmu i zauzima sedmo mesto na listi prioritetnih supstanci od kojih je neophodno pronaći optimalne načine zaštite. Naše pređašnje studije su pokazale da egzopolisaharid izolovan iz soja Lactiplantibacillus plantarum BGAN8 (EPS-AN8) pokazuje visok afinitet za vezivanje jona kadmijuma u vodenom rastvoru i ostvaruje značajan nivo in vitro zaštite Caco-2 ćelija od njegovih toksičnih efekata. Imajući u vidu da je za opštu populaciju najzastupljeniji unos kadmijuma ishranom, u ovoj studiji je praćen paralelni efekat unosa kadmijuma (kroz vodu) i EPS-AN8 (kroz hranu) u DA pacovima. Nakon 30 dana tretmana, utvrđeno je da je unos EPS-AN8 doprineo sniženom nivou deponovanog metala u bubrezima, jetri i krvi, a povećanom sadržaju u fekalnom materijalu. Primećeno je da unos EPS-AN8 ublažava pojavu promena u relativnoj zastupljenosti bakterijskih rodova i vrsta karakterističnih za ilaganje kadmijumu, poput porasta broja oportunističkih patogenih bakterija i smanjenja brojnosti laktobacila. Ostvareni zaštitni efekti ukazuju na snažan potencijal primene EPS-AN8 u uslovima visoke kontaminacije kadmijumom.

Orally Administrated Lactiplantibacillus plantarum BGAN8-Derived EPS-AN8 Ameliorates Cd Hazards in Rats

AB  - Cadmium (Cd) is a highly toxic metal that is distributed worldwide. Exposure to it is
correlated with a vast number of diseases and organism malfunctions. Exopolysaccharides (EPS)
derived from Lactiplantibacillus plantarum BGAN8, EPS-AN8, previously showed great potential for
the in vitro protection of intestinal cells from this metal. Here, we investigated the potential of food
supplemented with EPS-AN8 to protect rats from the hazardous effects of Cd exposure. After thirty
days of exposure to lower (5 ppm) and higher (50 ppm)-Cd doses, the administration of EPS-AN8 led
to decreased Cd content in the kidneys, liver, and blood compared to only Cd-treated groups, whereas
the fecal Cd content was strongly enriched. In addition, EPS-AN8 reversed Cd-provoked effects on
the most significant parameters of oxidative stress (MDA, CAT, GST, and GSH) and inflammation
(IL-1 , TNF- , and IFN-
) in the duodenum. Moreover, micrographs of the duodenum were in line
with these findings. As the gut microbiota has an important role in maintaining homeostasis, we used
16S rRNA amplicon sequencing and investigated the effects of Cd and EPS-AN8 on one part of the
microbiota presented in the duodenum. Although Cd decreased the growth of lactobacilli and mostly
favored the blooming of opportunistic pathogen bacteria, parallel intake of EPS-AN8 reversed those
changes. Therefore, our results imply that EPS-AN8 might be extremely noteworthy in combatting
this toxic environmental pollutant.

AB  - With the ever-growing concern for human health and wellbeing, the prenatal period of
development requires special attention since fetuses can be exposed to various metals through the
mother. Therefore, this study explored the status of selected toxic (Pb, Cd, Ni, As, Pt, Ce, Rb, Sr, U)
and essential trace metals (Mn, Co, Cu, Zn, Se) in the umbilical cord (UC) sera, maternal sera, and
placental tissue samples of 92 healthy women with normal pregnancies. A further aim focuses on the
potential transplacental transfer of these trace metals. Based on the obtained levels of investigated
elements in clinical samples, it was observed that all of the trace metals cross the placental barrier
and reach the fetus. Furthermore, statistical analysis revealed significant differences in levels of
toxic Ni, As, Cd, U, Sr, Rb, and essential Mn, Cu, and Zn between all three types of analyzed clinical
samples. Correlation analysis highlighted As to be an element with levels that differed significantly
between all tested samples. Principal component analysis (PCA) was used to enhance these findings.
PCA demonstrated that Cd, Mn, Zn, Rb, Ce, U, and Sr were the most influential trace metals in
distinguishing placenta from maternal and UC serum samples. As, Co, and Cu were responsible for
the clustering of maternal serum samples, and PCA demonstrated that the Pt level in UC sera was
responsible for the clustering of these samples. Overall, the findings of this study could contribute to
a better understanding of transplacental transfer of these trace metals, and shed a light on overall
levels of metal exposure in the population of healthy pregnant women and their fetuses.

AB  - Adverse effects of non-occupational exposure to cadmium (Cd) are increasingly
acknowledged. Using a rat model of oral Cd exposure in drinking water we have shown
that skin is a target for this metal. Due to contribution of individual variability to the
intensity of cadmium toxicity, dermatotoxicity of two environmentally relevant Cd
doses (5 and 50 ppm) was examined in individuals of two rat strains, Albino Oxford
(AO) and Dark Agouti (DA), which differ in response to chemicals. A dose-dependent
accumulation of Cd in the skin/epidermal cells was noted in both strains, and although
there were no strain differences in the Cd accumulation, the degree of skin response to
the metal differed. Signs of skin damage were evident in both strains, but response to
injury was more pronounced in DA. Individuals of DA rats responded by an increase
in the levels of antioxidant defense enzymes in the skin already at lower dose, in
contrast to AO (which reacted to higher dose solely), implying higher sensitivity of DA
strain to Cd-induced toxicity. Epidermal cells from both strains developed stress
response, however increased GSH, and higher metallothionein/MT-1 and MT-2
mRNA, Nrf2 protein, apoptosis, Ahr and Cyp genes in AO, depicting this strain`s
ability to better defend against Cd insult. Epidermal cells` IL-1β, TNF and IL-6
response was induced by Cd in DA, while pro-inflammatory cytokine production was
unchanged in AO (though increased following stimulation with S. epidermidis), with
increased IL-10 as a possible underlying mechanism. T cells from non-exposed rats
produce more IFN-γ and IL-17 in co-culture with epidermal cell from Cd–exposed DA
rats what strengthens the view that this strain is more prone to metal’s dermatotoxicity.
These data give a new insight into repercussion of genetic variability to toxicity of
cadmium acquired by the skin via gut, bearing relevance for variations in the link
between dietary cadmium and inflammation-based skin pathologies.

AB  - The mobility (fractionation) of rare earth elements (REEs) and their possible impacts on ecosystems are still relatively unknown. Soil samples were collected from two sites in central Serbia, an unpolluted mountain region (site 1) and a forest near a city (site 2). In order to investigate REE fractions (acid-soluble/exchangeable, reducible, oxidizable, and residual) in soils, BCR sequential extraction was performed. Additionally, the content of REEs was also determined in stipes and caps of the mushroom Macrolepiota procera, growing in the observed sites. Sc, Y, and lanthanide contents were determined by inductively coupled plasma mass spectrometry (ICP-MS), and results were subjected to multivariate data analysis. Application of pattern recognition technique revealed the existence of two distinguished clusters belonging to different geographical sites and determined by greater levels of Sc, Y, and lanthanides in Goč soil compared to Trstenik soil. Additionally, PCA analysis showed that REEs in soil were concentrated in two groups: the first consisted of elements belonging to light REEs and the second contained heavy REEs. These results suggest that the distribution of REEs in soils could indicate the geographical origin and type of soil. The bioconcentration factors and translocation factors for each REE were also calculated. This study provides baseline data on the rare earth element levels in the wild edible mushroom M. procera, growing in Serbia. In terms of bioconcentration and bioexclusion concept, Sc, Y, and REEs were bioexcluded in M. procera for both studied sites.

AB  - Epidemiological studies indicate a possible link between cadmium (Cd) ingested through food and various pathologies / diseases. There is no such data when it comes to skin, although the deposition of this metal in hair is used as a biomarker of human exposure. Only one study has shown the effect of orally administered Cd on wound healing, but the effects on other homeostatic skin activities, including immune, have not been investigated. In order to investigate the potential immunomodulatory effect of this heavy metal on the skin, DA strain rats were exposed to subchronic (30 days) oral intake of cadmium (in drinking water, 5 mg / l and 50 mg / l).Cd was deposited in this tissue and led to oxidative stress, judging by the increase in antioxidant enzymes (SOD and CAT), structural changes in the skin, as well as the presence / activation of innate immune cells. Higher concentrations of deposited Cd in the epidermis compared to the dermis resulted in a stress response at the epidermal cell (EC) level, manifested by increased mRNA expression for metallothioneins (MT-1 and MT-2) and decreased GSH after a higher dose of CdThis metal also led to the manifestation of signs of inflammation, judging by the increased expression of mRNA for iNOS and IL-1β and the production of NO and IL-1β, at a lower dose, and mRNA and IL-6 and TNF protein product at a higher dose. On the other hand, Cd led to increased production of the regulatory cytokine IL-10 (at higher doses). Increased IL-17 production was observed in EC cultures isolated from the skin of rats exposed to 50 mg / l, while the amount of cytokine produced further increased in EC co-culture with lymph node lymphocytes after ConA stimulation (both doses).Cd present in the skin also increased the ability of the EC to respond to the application of the contact allergen DNCB, judging by the increase in IL-6 and TNF (at both doses), while IL-1β remained unchanged. The presented results show a generally proinflammatory effect of Cd on epidermal cells and indicate the potential of orally ingested Cd to modulate immune responses in the skin.
