AB  - Ganoderma lucidum is a well-known medicinal mushroom species in which polysaccharides are one of the major sources of biological activity. The species was considered as a species-complex due to significant variations in morphological, biochemical, and genetic features among populations with a worldwide distribution. This fact was the basis for setting the aim of this research: to study intraspecific diversity in polysaccharide production and intracellular sugar composition among selected G. lucidum strains. The presence of intraspecific diversity among 10 G. lucidum strains, from different areas worldwide, was noted. Values of produced mycelia biomass and intracellular polysaccharides were found in wide ranges (3.1-28.2 g L-1 and 20.0-53.3 mg g(-1), respectively), while differences in extracellular polysaccharide amounts were minor (0.2-1.5 mg mL(-1)). The significant quantitative and qualitative differences in intracellular sugar composition were noted. Glucose was the predominant sugar in almost all strains except one (HAI 447), where sucrose was dominant. The potential of polysaccharide production and intracellular sugar composition could be one more taxonomic criterion for strain characterization within G. lucidum. The differences in intracellular sugar composition and proportions could be reflected in features of produced polysaccharides and also in their biological activities.

AB  - Comparison of the potential for laccase and Mn-oxidizing peroxidases synthesis by ten strains of Ganoderma lucidum, originating from different worldwide areas, during solid-state fermentation of selected plant raw materials was the aim of this study. The great intraspecific variability in the production of analyzed enzymes as well as the dependence of the enzyme activity on plant raw materials were reported. The strain HAI 957 was the best laccase producer in the presence of corn stem, as a unique carbon source (129.46 U/L). The highest level of Mn-dependent peroxidase activity was noted after wheat straw fermentation by G. lucidum HAI 246 (78.64 U/L), while the maximal versatile peroxidase production (59.72 U/L) was observed in strain HAI 957 in the medium with oak sawdust.

AB  - The purpose of this study was to resolve the question of whether various nitrogen sources and concentrations affect characteristics of selected G. lucidum ligninolytic enzymes participating in wheat straw fermentation. This is the first study reporting the presence of versatile peroxidase activity in crude extract of G. lucidum culture, as well as isoforms profile of Mn-oxidizing peroxidases. NH(4)NO(3) was the optimum nitrogen source for laccase and Mn-dependent peroxidase activity, while peptone was the optimum one for versatile peroxidase activity. Four bands with laccase activity were obtained by native PAGE and IEF separations from medium enriched with inorganic nitrogen source, and only two bands from medium containing organic source. Medium composition was not shown to affect isoenzyme patterns of Mn-oxidizing peroxidases. Four isoforms of Mn-dependent peroxidase and three of versatile peroxidase were obtained on native PAGE. By IEF separation, five isoforms of Mn-dependent peroxidase and only two of versatile peroxidase were observed. The results demonstrated that G. lucidum has potential for mineralization and transformation of various agricultural residues and should take more significant participation in large-scale biotechnological processes.

AB  - The potential antifungal effects of Thymus vulgaris L., Thymus tosevii L., Mentha spicata L., and Mentha piperita L. (Labiatae) essential oils and their components against 17 micromycetal food poisoning, plant, animal and human pathogens are presented. The essential oils were obtained by hydrodestillation of dried plant material. Their composition was determined by GC-MS. Identification of individual constituents was made by comparison with analytical standards, and by computer matching mass spectral data with those of the Wiley/NBS Library of Mass Spectra. MIC's and MFC's of the oils and their components were determined by dilution assays. Thymol (48.9%) and p-cymene (19.0%) were the main components of T. vulgaris, while carvacrol (12.8%), alpha-terpinyl acetate (12.3%), cis-myrtanol (11.2%) and thymol (10.4%) were dominant in T. tosevii. Both Thymus species showed very strong antifungal activities. In M. piperita oil menthol (37.4%), menthyl acetate (17.4%) and menthone (12.7%) were the main components, whereas those of M. spicata oil were carvone (69.5%) and menthone (21.9%). Mentha sp. showed strong antifungal activities, however lower than Thymus sp. The commercial fungicide, bifonazole, used as a control, had much lower antifungal activity than the oils and components investigated. It is concluded that essential oils of Thymus and Mentha species possess great antifungal potential and could be used as natural preservatives and fungicides.

AB  - Ganoderma carnosum has been found in Europe only, at coniferous trees and it is difficult to distinguish it morphologically from G. lucidum. Since G. carnosum has not been studied biochemically yet, the aim of this study was to analyse its ability to produce Mn-dependent peroxidase (MnP), versatile peroxidase (VP) and laccase (Lac) under conditions of solid-state fermentation of selected plant raw materials as carbon sources, in the presence of different nitrogen concentrations in the medium. Wheat straw, corn stem, oak and grapevine sawdust were the analysed plant raw materials. Nitrogen source in synthetic medium was NH4NO3 and its concentrations were: 10mM N and 20 mM N. Enzyme activity was determined spectrophotometrically, using ABTS and phenol red, as the substrates for Lac and Mn-oxidizing peroxidases, respectively. Maximum level of MnP activity (56.82 U/l) was obtained in the medium with wheat straw and nitrogen concentration of 10 mM. Best carbon source for VP production was grapevine sawdust at nitrogen concentration of 10 mM (80.80 U/l). The obtained Lac activity was very low in the medium with wheat straw (1.80 U/l), while it was not detected in the presence of other three analyzed plant raw materials. Maximum of total protein content (0.06 mgml-1) was noted in the medium where oak sawdust was carbon source and nitrogen concentration was 20 mM.
AB  - Vrste roda Ganoderma su producenti mnogih biološki aktivnih supstanci pa su objekti proučavanja mnogih medicinskih i farmaceutskih studija. U novije vreme se sve više pažnje poklanja upoznavanju njihovog ligninolitičkog enzimskog sistema, u cilju uspešne primene u različitim biotehnološkim procesima. Za razliku od G. lucidum, čiji je ligninolitički sistem intenzivno proučavan, ostale vrste ovog roda su retko bile objekti istraživanja. G. carnosum se teško morfološki razlikuje od G. lucidum. i još uvek nije biohemijski proučena, pa je cilj našeg istraživanja bio analiza produkcije Mn-oksidujućih peroksidaza i lakaza u uslovima čvrste kultivacije, na različitim biljnim ostacima (pšenična slama, stabljike kukuruza, piljevina hrasta i vinove loze) u prisustvu azota u obliku NH4NO3 i u koncentracijama od 10 mM, odnosno 20 mM. AB  - The in vivo evaluation of the toxicological and antifungal activity of the essential oil of Thymus vulgaris L. and its main component thymol was made on 2-month-old male Wistar rats. We examined the therapeutic potency against experimentally induced dermatomycoses in rats, using the most frequent dermatomycetes, Trichophyton mentagrophytes, T. rubrum, and T. tonsurans. The therapeutic efficacy of a 1% solution of the essential oil of Thymus vulgaris and thymol as well as the commercial preparation bifonazole was evaluated. During the 37-day observation period the oil-treated animals were cured.

AB  - The major disease of the cultivated mushroom Agaricus bisporus Lange (Imb) in Serbian mushroom farms is wet bubble caused by the fungus Mycogone perniciosa (Magnus) Delacr. In this study we report the morpho-physiological characteristics and inter-relationships between colonies of five isolates of M. perniciosa. The results suggest that mycelial compatibility could serve as an additional parameter for a more reliable determination of different pathotypes of M. perniciosa.

AB  - The chemical composition and antimicrobial activity of Cymbopogon winterianus Jowitt (syn. Cymbopogon nardus (L.) W. Watson var. mahapengiri Winter) (Poaceae) and Carum carvi L. (Apiaceae) essential oils were investigated against 19 fungal and 7 bacterial species. Among the tested species were food contaminants, spoilage fungi, as well as plant or fungi and animal pathogens. In order to determine minimal inhibitory concentrations (MICs) minimal fungicidal concentrations (MFCs) and minimal bactericidal concentrations (MBCs), microdilution tests were used. Citronellal and trans-geraniol were dominant in Cymbopogon winterianus oil, whereas carvone and limonene were the main components in Carum carvi oil. The essential oil of C. carvi possessed stronger antifungal and antibacterial potential than did citronella oil.

AB  - This study showed that cultivation conditions (medium composition, initial medium pH, aeration rate, inoculum density) affect biomass and polysaccharide production in Ganoderma lucidum strain HAI 447. The synthetic medium (pH 5.5) with maltose at the concentration of 65.0 g L-1, peptone of 0.2%, as well as in the presence of K, Na, and Mg at concentrations of 6.0, 3.0, and 2.0 mM, respectively, presented the best conditions for biomass (29.2 g L-1) and extra- and intracellular polysaccharide (2.9 mg mL(-1) and 73.3 mg g(-1), respectively) production in the analyzed strain. Constant aeration was optimal for biomass and extracellular polysaccharide production, whereas cultivation under stationary conditions was the best for intracellular polysaccharide production. A peak in biomass and polysaccharide production was observed at an inoculum density of 20.0 mL.

AB  - The in vivo evaluation of antifungal activity of the Lavandula angustifolia essential oil was made on two-month old male Wistar rats. We examined the therapeutic potency against experimentally induced dermatomycoses in rats using the most frequent dermatomycetes, Trichophyton mentagrophytes. The therapeutic efficacy of 1% solution of essential oil as well as commercial preparation-bifonazole, was evaluated. During the 13-day observation period the oil-treated animals were cured completely.
AB  - Infekcije ljudi uzrokovane gljivama su u znatnom porastu u poslednjih 10 godina, posebno kod imunokompromitovanih pacijenata. Čak 10% infekcija kod hospitalizovanih pacijenata izazvano je gljivama. S obzirom da je pojava rezistencije na komercijalne fungicide učestala, javlja se potreba za novim alternativnim antifungalnim agensima. Prirodni produkti dobijaju sve veći značaj u tretmanu infektivnih oboljenja zbog svog netoksičnog karaktera visoke biodegradibilnosti i efikasnosti. Etarska ulja izolovana iz biljaka pokazuju veoma dobru antimikrobnu aktivnost. Eksperimentalna dermatomikoza kod Wistar pacova u ovom radu izazvana je dermatomicetom Trichophyton mentagrophytes. Prvi simptomi pojavili su se nakon 5 dana od inokulacije. Tretman etarskim uljem vrste Lavandula angustifolia, 1% rastvorom počeo je odmah nakon pojave prvih simptoma. Poboljšanje simptoma primećeno je nakon dva dana tretmana, a posle 13 dana od početka tretmana životinje su u potpunosti izlečene, na SDA podlogama nije bilo zabeleženo prisustvo patogena. Bifonazol je doveo do izlečenja nakon 15 dana tretmana. Etarsko ulje Lavandula angustifolia pokazalo je izuzetno jak antifungalni potencijal u tretmanu eksperimentalno indukovane dermatomikoze kod pacova, bolje od komercijalnog mikotika, bifonazola, koji je korišćen kao pozitivna kontrola.

AB  - The antifungal activity of Nepeta rtanjensis Diklić et Milojević essential oil on mycelia growth has been performed by macrodilution method. The most efficient impact of N. rtanjensis essential oil on mycelia growth in vitro was found in Alternaria species with the same value of minimal inhibitory quantity (MIQ) of 0.6 μl/ml. Bipolaris spicifera and Cladosporium cladosporoioides had MIQ values of 1.0 μl/ml whereas Trichoderma viride with MIQ value of 1.6 μl/ml showed the most efficient defense against the essential oil examined. The values of minimal fungicidal quantity (MFQ) in Alternaria sp. 2, B. spicifera and C. cladosporioides match the MIQ values whereas MFQ values in Alternaria sp. 1 is 0.8 μl/ml, and in T. viride 1.8 μl/ml.
AB  - Makrodilucionom metodom testiran je uticaj etarskog ulja endemične biljke Nepeta rtanjensis (Lamiaceae) na micelijalni rast mikromiceta: Alternaria sp. 1, Alternaria sp. 2, Bipolaris spicifera, Cladosporium cladosporioides i Trichoderma viride. Hemijska analiza etarskog ulja N. rtanjensis pokazala je apsolutnu dominaciju 4aα, 7α, 7aβ nepetalaktona (79.89%). Najefikasniji uticaj na rast micelije in vitro zabeležen je kod vrsta roda Alternaria sa istom vrednošću minimalne inhibitorne količine (MIK) od 0.6 μl/ml. Bipolaris spicifera i Cladosporium cladosporioides su imali MIK 1.0 μl/ml, dok je Trichoderma viride, sa vrednošću MIK od 1.6 μl/ml, pokazala najveću otpornost na dejstvo ispitivanog ulja. Vrednosti minimalne fungicidne količine (MFK) se kod Alternaria sp. 2 (0.6 μl/ml), B. spicifera (1.0 μl/ml) i C. cladosporioides (1.0 μl/ml) poklapaju sa vrednostima MIK, dok je kod Alternaria sp. 1 MFK 0.8 μl/ml, a kod T. viride 1.8 μl/ml. AB  - Mycogone perniciosa (Magnus) Delacr., which causes wet bubble disease of Agaricus bisporus Lange (Imb), results in a considerable crop loss on mushroom farms in Serbia. The isolation and identification of five isolates of M. perniciosa from diseased fruit bodies of white button mushroom from mushroom units in Serbia, Bosnia and Herzegovina and Holland were made. Morpho-physiological characteristics and inter-relationships of the obtained isolates were studied. Macroscopic and microscopic investigations of different zones between colonies of the isolates of M. perniciosa revealed the phenomenon of the hyphal interference between different isolates. The obtained results suggest that hyphal interference could serve as an additional parameter for a more reliable determination of fungal specifity.
AB  - Mycogone perniciosa (Magnus) Delacr., izazivač oboljenja mokre truleži najčešći je uzročnik gubitaka u gajilištima Agaricus bisporus Lange (Imb) u Srbiji. Izvršena je izolacija i identifikacija 5 izolata M. perniciosa sa obolelih plodonosnih tela šampinjona iz gajilišta u Srbiji, Bosni i Hercegovini i Holandiji. Ispitivane su morfo-fiziološke karakteristike kao i stepen srodnosti proučavanih izolata na osnovu analize međusobnog delovanja kolonija, odnosno korišćenjem fenomena hifalne interferencije. Makroskopska i mikroskopska istraživanja odnosa izolata i dobijeni rezultati ukazuju da hifalna interferencija može predstavljati dodatni parametar u razlikovanju izolata M. perniciosa. Izolati dobijeni iz gajilišta u Srbiji slični su međusobno, kao i izolati iz Holandije koji su pokazali međusobnu sličnost ali se razlikuju od prethodnih. AB  - This work covers the chemical composition and antifungal activities of essential oils isolated from savory (Satureja thymbra) and sage (Salvia pomifera ssp. calycina) analyzed using GC/MS. The main components of S. thymbra oil were gamma-terpinene (23.2%) and carvacrol (48.5%). The main components in S. pomifera oil were alpha-thujone (20.4%) and beta-thujone (36.1%). The oils were assayed for antifungal activity against Mycogone perniciosa. This mushrooms pathogen is very contagious and results in severe crop losses. Compounds were investigated by the microatmosphere method. The oil of S. thymbra showed the best antifungal activity. Minimal inhibitory and fungicidal quantities were 0.05-0.25 mu L/mL.

AB  - The most commonly cultivated mushroom species is the Agaricus bisporus Lange (Imb). One of the major pathogenic diseases of the cultivated mushroom in Serbia is Mycogone perniciosa (Mang). Biological control systems are not much used in mushroom cultivation. Medical and aromatic plants have been placed in the focus of intense studies. Pure culture of the M. perniciosa was isolated from infected A. bisporus. The essential oil of Hyssopus officinalis L. is used as a potential antifungal agent. The most abundant components in oil are isopinocamphone (43.29%), pinocamphone (16.79%) and b-pinene (16.31%). Antifungal activity of Hyssop was investigated by the modified microatmosphere method. The minimal inhibitory quantity was 5 μL/mL and a minimal fungicidal quantity was 15-20 μL/mL. There is no report on the use of Hyssop essential oil in mushroom disease.
AB  - Agaricus bisporus Lange (Imb) je najčešće komercijalno gajena jestiva gljiva Različiti mikroorganizmi gljive, bakterije i virusi su izazivači bolesti u gajilištima šampinjona. Mycogone perniciosa (Mang) je izazivač bolesti poznate pod nazivom vlažni mehur i najčešći uzročnik gubitaka u gajilištima u Srbiji. Biološka kontrola, koja je uspešno primenjivana na nekim poljoprivrednim kulturama, nije korišćena prilikom uzgoja gljiva. Jedna od mogućnosti je primena biljnih sprejova. Lekovite i aromatične vrste biljaka se intenzivno istražuju kao mogući antifungalni agensi. Uzorci obolelih šampinjona su sakupljani u gajilištima u Srbiji. Kulture M. perniciosa su izolovane sa obolelih plodonosnih tela A. bisporus. Korišćeno je etarsko ulje Hyssopus officinalis. Najzastupljenije komponente ulja su izopinokamfon (43.29%) trans-pinokamfon (16.79%) i b-pinen (16.31%). Antifungalna aktivnost etarskog ulja izopa ispitivana je modifikovanom "mikroatmosfera"- metodom. Minimalna inhibitorna količina je bila 5 μL/mL, a minimalna fungicidna količina 15-20 μL/mL. Velik broj preparata je napravljen i primenjen za kontrolisanje oboljenja pečuraka: fungicidi, primena mikrotalasa ili dejstvo nekih antagonističkih bakterija. Dosad nije bilo saopštenja o primeni etarskog ulja izopa protiv izazivača bolesti gajenih gljiva.

AB  - The chemical composition and antimicrobial activities of essential oils isolated from Myrrhis odorata, Hypericum perforatum, and Helichrysum arenarium, were investigated against seven bacterial and six fungal species. Activity against the bacteria and C. albicans were investigated by a bioautographic test on TLC plates, while all the other fungi were tested by a microdilution test. The oil of H. arenarium, showed the best antibacterial activity, while M. odorata oil showed the lowest effect, to compare with commercial products. In contrast, the best antifungal activity was observed for M. odorata oil, while H. arenarium, showed the lowest antifungal potential. Minimal inhibitory and fungicidal concentrations for fungi were 0.5-120 mu g/mL. The oil of M. odorata showed better activity then commercial product and it can be seen that this oil is very effective against all fungi tested.

AB  - Yellow pheasant´s Eye is a herbaceous plant from dry ressy areas. Owing to habitat destruction and over- collection for ornamental and medical purposes A. vernalis L. has became scarce in central and south Europe. The reasons for A. vernalis threatened are manyfold. The low seeds germination rate is significant. According to our investigation the main cause of fruit destruction is fungal infection. From the surface of the fruits, collected in Deliblato Sands, the following micromycetes has been isolated and determinated: Fusarium solani (Mart) Sacc., Fusarium sporotrichioides Sherb., Alternaria sp. and Drechslera sp. Histologycal analysis showed the presence of conidiomata and conidia Phoma sp. in the seeds.
AB  - Gorocvet je zeljasta biljka sušnih predela. U centralnoj i južnoj Evropi, kao tipično stepska biljka, ima ograničeno rasprostranjenje, dok je u istočnoj Evropi brojnost populacija u opadanju. U centralnoj i južnoj Evropi ova biljka postaje sve ugroženija zbog preterane eksploatacije od strane čoveka, u medicinske svrhe, kao i zbog narušavanja njenog staništa. A. vernalis je kao ugrožena biljka uključena u crvene knjige. Jedan od razloga ugroženosti A. vernalis je niska sposobnost klijavosti semena. Prema našim istraživanjima glavni uzrok destrukcije plodova je infekcija gljivama. Sa površine plodova A. vernalis, sakupljenih u Deliblatskoj peščari, izolovane su i determinisane sledeće mikromicete: Fusarium solani, Fusarium sporotrichioides, Alternaria sp., Drechslera sp. Histološki preseci inficiranih plodova pokazuju značajne promene: perikarp i semena su razoreni micelijom i plodonosnim telima gljive iz roda Phoma, prema mišljenju mikoloških eksperata (Kew Garden, Engleska) koji su potvrdili identifikaciju. Ovi rezultati donose nova pitanja, ideje i rešenja o konceptu ugroženosti biljnih vrsta.

AB  - The antifungal activity of Aniba rosaeodora, Laurus nobilis, Sassafras albidum and Cinnamomum zeylanicum essential oils were investigated against 17 micromycetes. Among the tested fungal species were food poisoning, spoilage fungi, plant and animal pathogens. In order to determine fungistatic and fungicidal concentrations (MIC and MFC) macrodilution and microdilution tests were used. Linalool was the main component in the essential oil of A. rosaeodora, while 1.8-cineole was dominant in L. nobilis. In sassafras essential oil safrole was the major component and in the oil of C. zeylanicum the main component was trans-cinnamaldehyde. AB  - Commercial sample of essential oil of M. officinalis was analysed using GC and GC/MS. The main compounds identified in the essential oil were citronellal (29.3%), limonen (8.84%), geranial (8.70%), a-terpineole (8.02%), a-terpinyl acetate (7.14%) and neral (5.68%). As test organisms the following micromycetes Penicillium ochrochloron, Aspergillus niger Trichoderma viride and bacteria Bacillus subtilis, Staphylococcus epidermidis, Escherichia coli and Micrococcus luteus were used. Microdilution method on microplates 96-system was used for detection of minimum inhibition concentration (MIC), minimum bactericidal (MBC) and fungicidal concentration (MFC). Essential oil did not show antibacterial effect, but the oil of M. officinalis exhibited significant antifungal activity against tested micromycetes.
AB  - Analiziran je hemijski sastav komercijalnog uzorka etarskog ulja matičnjaka (Melissa officnalis L) i njegova antibakterijska i antifungalna aktivnost. GC i GC-MS analizom konstatovane su najzastupljenije komponente: citronelal (29.3%), limonen (8.84%), geranial (8.70%), a-terpineol (8.02%), a-terpinil acetat (7.14%) i nerol (5.68%). Za ispitivanje biološke aktivnosti korišćeni su test organizmi, mikromicete: Aspergillus niger, Penicillium ochrochloron Trichoderma viride i bakterijski sojevi: Bacillus subtilis, Escherichia coli, Micrococcus luteus i Staphylococcus epidermidis. AB  - In this article we investigated antibacterial and antifungal activities of limonene against 7 bacterial and 19 fungal species. Antibacterial activity and Candida albicans were investigated by on TLC plates, while antifungal activity was tested by micro- and macrodilution tests. Limonene showed antimicrobial activities against all microorganisms tested. The best results were obtained against Micrococcus luteus, Streptococcus epidermidis, S. aureus and Salmonella typhimurium, in concentration of 5 ml. Limonene exhibited antifungal activity in concentration of 8.0 - 13.0 ml/ml in macrodilution and 6.0-10.0 ml/ml in microdilution method.
AB  - U ovom radu je ispitivano antibakterijsko i antifungalno dejstvo limonena na 7 bakterijskih i 19 gljivičnih vrsta. Antibakterijska aktivnost i Candida albicans su testirani bioautografskim testom na TLC pločama, dok je antifungalna mikro- i makrodilucionom metodom. Limonen je pokazao antimikrobnu aktivnost na sve testirane organizme. Najbolje dejstvo je imao na Micrococcus luteus, Streptococcus epidermidis, S. aureus i Salmonella typhimurium, u koncentraciji od 5 ml. Za utvrđivanje sastava etarskog ulja kleke korišćeni su GC i GC/MS. Najzastupljenije komponente u ovom ulju su a-pinen (40.33%), b-mircen (13.72%), limonen (5.61%) i sabinen (5.38%). Test organizmi korišćeni u eksperimentu su bile mikromicete: Penicillium ochrochloron, Aspergillus niger, Trichoderma viride i bakterijski sojevi Bacillus subtilis, Staphylococcus epidermidis, Escherichia coli i Micrococcus luteus.
