AB  - Within the past decade, our research group placed great effort in
exploration of the impact of treated/untreated wastewaters on aquatic
ecosystems as less than 13 % of collected municipal wastewaters are
processed before their release to receiving waters in Serbia. Having in mind
that more than 90% of the rivers within the territory of Serbia belong to the
Danube River Basin, our major focus is naturally on the large transnational
waters such as the Danube and Sava rivers. Depended on the concept of the
study and hydro-morphological characteristics of the sites, various
approaches have been employed for in situ assessment of eco/geno-toxicity
which use different organisms in passive and active biomonitoring. Aquatic
organisms of different trophic levels have diverse life strategies, metabolism
pathways, and consequently, they have a different response to pollutant
pressure. In eco/geno-toxicological studies we have successfully employed
several species of freshwater mussels (Unio tumidus, Unio pictorum and
Sinanodonta woodiana) as well as few economically important fish species
(common carp (Cyprinus carpio), freshwater bream (Abramis brama), bleak
(Alburnus alburnus, chub (Squalius cephalus)).Comet assay is our primary
choice for the assessment of DNA damage considering its sensitivity and
cost effectiveness. Selection of certain types of tissues for the assessment
(such as blood or haemolymph), which do not require sacrificing of the
animals or additional manipulation in preparation of cell suspension coupled
with mini-gel format of the slides enables high-throughput screening of the
genotoxic potential in situ. Additionally, cryopreservation of the blood
samples directly onsite expands the research area as it overcomes issues
related to safe transportation of the samples to the laboratory. Data
obtained so far indicated that the genotoxic response in the studied
indicator organisms significantly differ which is understandable considering
the difference in uptake, accumulation and physiological responses.

AB  - More frequent application of biological parameters beside the conventional chemical parameters in national
monitoring for marine ecosystem assessment is supported by UNEP/MAP. Accordingly, to evaluate the
possible effects of pollution by determination of trace element content and genotoxicity assessment in
marine organisms within the Boka Kotorska Bay, Montenegro (southern Adriatic), we carried a field study
which included transplantation of the seabream (Sparus aurata) from aquaculture farm (Orahovac) to more
impacted sites (Dobrota and port of Tivat), situated in vicinity of the main ports. On the sampling site
Orahovac, the group of seabream specimens were placed in closed fish trap on 5 m depth (O1), another fish
trap was placed in Dobrota, while the third fish trap with seabreams was placed at the site port of Tivat.
The additional group was sampled from standard aquaculture cage on sampling site (O2) to reduce possible
bias in genotoxicity data due to confined space within the fish trap. After two weeks of exposure, blood
from fish heart was taken, afterwards muscle tissues were prepared for trace element determination.
Genotoxicity was measured by DNA damage induction based on comet (single cell gel electrophoresis)
assay parameter – Tail intensity (TI%), while trace element (As, Cd, Pb, Hg, Cr, Cu, Fe, Mn, Ni, Zn and
V) content was determined by ICP. Mean values of TI% obtained from seabream specimens at the sampling
site Orahovac were: 4.26 ± 0.17 – O1 and 2.56 ± 0.11 – O2, while TI% values for transplanted specimens
on the sites Dobrota and port of Tivat were 6.38 ± 0.17 and 11.06 ± 0.23, respectively. Statistically
significant differences (p<0,05) of TI% were observed between all specimens groups. The group of
seabream from port of Tivat showed the highest TI% (higher level of DNA damage), most probably caused
by marine pollution. Obtained trace element concentrations in seabream tissues were significantly lower in
comparison to values recommended by FAO, EC and national legislatives of Croatia, Spain and Turkey.
Since transplanted fish showed a significant response in a relatively short exposure period, our results
support the introduction of seabream caging as monitoring tool to reveal marine pollution by multiple
biomarker approach. Considering the significant share of aquacultured seabream in human consumption,
higher availability for sampling and convenience in application for molecular biomarker analyses in the
same samples, such an integrated monitoring approach would contribute to more credible data.

AB  - This study was conducted on the Danube River locality Višnjica, exposed to the discharge of the largest wastewater collector in Serbia's capital, Belgrade. Concentrations of metals and metalloids (Al, Fe, Cr, As, Sr, Mn, Cd, Zn, Mo, Cu, Li, Ni, B, Co, Pb, and Ba) and histopathological alterations were investigated in different tissues of common bream during one representative month of each season in 2014. This is the first study in which these two biomarkers were assessed parallelly in common bream. The highest concentrations of examined elements were noticed in gills and the lowest in muscle. Statistically significant differences in element concentrations between different seasons were noticed only in gills for Al, Cu, and Fe. Concentrations of As and Pb in fish muscle were below the maximum acceptable concentrations (MAC). The histopathological index (HI) of the liver showed higher values in comparison to the HI of the gills. Histopathological index of the gills had a significantly higher score in November in comparison to August. The liver HI had the highest score in April, and the lowest in August, while the total histopathological index had the highest score in November, and the lowest in August, both without significant differences between the months. This study endorses gills and liver as reliable organs for studying accumulation and histopathology as biomarkers of environmental changes. A faster reaction of the gills was confirmed since seasonal variations of both biomarkers were observed in this organ. Common bream proved as a good indicator of the state of organisms in polluted environments.

AB  - Water quality in the Danube River Basin is largely influenced by the inputs of pollutants - particularly
excessive nutrients, organic material, and hazardous substances, which affects the whole ecosystem
at various levels. The major goal of this study was to evaluate the level of genotoxic potential at the
site at Danube River, Novi Banovci, which is under significant anthropogenic pressure of untreated
municipal and industrial waste waters.
DNA damage in situ was assessed in 5 specimens of white bream (Blicca bjoerkna) erythrocytes, liver
and gills cells with comet assay and in erythrocytes with micronucleus test. Additionally, genotoxicity
of native water samples collected from the site and upstream and downstream of the site was
evaluated by the comet assay on HepG2 cells. Microscopic images of comets were scored using Comet
IV Computer Software (Perceptive Instruments, UK) and tail intensity was chosen as parameters to
assess the DNA damage.
Comet assay on white bream showed that gills were the most affected tissue, while liver showed the
smallest values. Micronuclei frequency was low in general, with mean of 1 micronuclei in 5000 scored
erythrocytes, with no significant correlation to the values of comet assay in erythrocytes. Comet assay
on HepG2 cells showed significant difference between values of negative control samples and samples
of water from effluent site and downstream of wastewater discharge. There was no significant
difference between negative control and upstream samples of water.
Our results showed that great impact of municipal and industrial wastewaters on the river ecosystem.
The application of in vitro and in situ tests gives a better insight into genotoxic effects. More
importantly, because Serbia still doesn’t have waste water treatme nt plants, this kind of research
points out the imperative for implementation of this facilities.

AB  - Due to the increasing industrialization, metals are discharged into all spheres of the environment, in particular, in river courses, which leads to the need for constant monitoring. Metals do not degrade into harmless end products; they are very persistent and have high potential for accumulation in biota. Metals in the fish body are accumulated in different amounts in the tissue specific matter. In relation to the biotic conditions and fish metabolism, the load of food, or the stage of the reproductive cycle, the seasonal variation of metal levels can be expected. Because of that, the objective of our present study was to analyze 15 metals and metalloids in liver, gills, muscle, and gonads of European chub (Squalius cephalus) throughout the 4 seasons, autumn, winter, spring, and summer. The specimens were collected from two rivers, Pestan and Beljanica at the Kolubara basin, and their concentrations were determined with inductively coupled plasma optical emission spectrometry (ICP-OES). Specimens from both rivers have shown similarities in metal accumulation like the highest accumulation of majority of elements in gills, lowest accumulation of majority of elements in muscle (except for Hg), and higher accumulation of some elements in summer (Cu, Fe, Zn). In addition, Cu and Fe showed affinity for liver, while Ba, Cr, Sr, and Zn were specific for gills. Also, Al, B, Fe, Ni, and Pb did not show significant differences in concentrations among different seasons in all investigated tissues.

AB  - The life‐history traits and population dynamics of the black bullhead Ameiurus melas (Rafinesque) were studied monthly from August 2009 to December 2012 in Lake Sava in Belgrade (Serbia). This period included a mass mortality event in May 2011 and subsequent population recovery during 2012. Repeated systematic mass removals of both juveniles and adults (using fyke nets) will be required if future fishery management measures aimed at limiting the population size are to be effective.

AB  - The Velika Morava River is the greatest national Serbian river and the significant tributary of the Danube River. The major problems in the Velika Morava River Basin (VMRB) represent untreated industrial and municipal wastewaters. In this study, the level of genotoxic potential at the sites along the VMRB was evaluated by parallel in vitro and in situ approach. Within in vitro testing, genotoxicity of native water samples collected from the sites in VMRB was evaluated by SOS/umuC test on Salmonella typhimurium TA1535/pSK1002 and by the comet assay on HepG2 cells. DNA damage in situ was assessed in bleak (Alburnus alburnus) erythrocytes by the comet (alkaline and Fpg-modified comet) and micronucleus assays. Additionally, the concentration of heavy metals in fish tissue was measured and this data, compiled with the data of the physico-chemical parameters measured in water, was used as a measure of the pollution pressure at the sites. Results showed that applied in vitro tests with native water samples are less sensitive in comparison with in situ tests and should be taken with precaution when making predictions on the status of the ecosystem. Within applied battery of in situ assays differential sensitivity of assays was observed where alkaline comet assay showed the highest potential in differentiation of the sites based on genotoxic potential. Integrated biomarker response showed that usage of the battery of bioassays provides better insight in a genotoxic effects in animals, and consequently, that the holistic approach is more suitable for this type of study.

AB  - The major objective of this study was to investigate the impact of wastewaters on the level of genotoxicity and the possible reflection of genotoxicity on the genetic variability in chub (Squalius cephalus) collected from the sites under different pollution pressures along the Sava River. To emphasize the impact of wastewaters, this study was carried out in a season with low water discharge. The level of genotoxic potential at the sites was studied by alkaline and Fpg-modified comet assay in fish blood, while RAPD technique was employed on the fish muscle tissue. The variations in the genotoxic potential were observed along the Sava River based on the level of DNA damage in blood of the specimens collected at the selected sites. Clustering of the impacted sites was also observed based on RAPD fingerprints. Comparison of the data obtained in RAPD and the data from the comet assay indicated the possible reflection of genotoxicity on the genetic variability in chub collected from the sites with different pollution pressure. This is a preliminary study which reports the differentiation of the population of chub along the Sava River based on RAPD analysis.

AB  - Sponges in the large rivers within the Danube River Basin (DRB) have not been adequately studied. Hence, the aim of this work was to undertake an investigation on the distribution of sponge species in the Danube and Sava rivers. Out of 88 localities covered by the study, sponges were found at 25 sites only (46 samples in total). By using morphological (light and scanning electron microscopy) and genetic (28S rDNA sequencing) analysis, four species were determined: Ephydatia fluviatilis (Linnaeus, 1759), Spongilla lacustris (Linnaeus, 1759), Eunapius fragilis (Leidy, 1851), Trochospongilla horrida Weltner, 1893. In the Danube, the predominant species was found to be E. fluviatilis making approximately 80% of collected samples, while in the Sava River S. lacustris dominated, representing 46% of the river sponges. Our work represents one of the few studies on freshwater sponges within the DRB from long stretches of the large lowland rivers (more than 2500 km of the Danube River and about 900 km of the Sava River). Moreover, molecular analysis for the identification of freshwater sponges was applied on the material collected from a wide area, thus contributing to the systematic studies on the distribution and abundance of the European freshwater invertebrate fauna in general.

Analysis and forecast of Pontic shad (Alosa immaculata) catch in the Danube River

AB  - The relationship between the Lower Danube River level and Romanian annual catches of Pontic shad (Alosa immaculata, Bennett 1835) were analyzed. For analysis of long term data on the Danube River water level and Pontic shad catch, combinations of different methods were applied using statistical programs, SPSS 13.0 and MATLAB 6. Periodograms, containing cyclic patterns, were obtained using Fourier analysis. Significant oscillations were determined with Fisher-Whittle’s tests and residuals were calculated after subtracting these significant oscillations from the original signals. Autoregressive moving average (ARMA) models of residuals were finally applied. Results indicated that river water levels, and especially those in May, greatly explained the fluctuations of Pontic shad catch. Annual landings varied greatly and appeared to be cyclic. Varying river flow was considered to be one of the most important factors that cause fluctuations in the size of populations. Forecast indicates gradual increase of the catch in the next decade, followed by a decrease in other decades. Estimated as a vulnerable species of fish by the IUCN, development of the forecasting model of the future catch oscillations could be very helpful to regulate fishing efforts towards the sustainable use of stocks and species conservation.

AB  - Introduction. The contamination of water bodies by sewage or manure is generally determined by fecal indicator bacteria. Specific differentiation between sources of contamination is of particular importance, since the health risk to humans is usually considered higher from human, than from animal fecal contamination. Quantitative PCR (qPCR)-based assays for analysis of human- or animal-associated genetic Bacteroidetes fecal markers have gained increased popularity in the field of microbial source tracking (MST).
Aim. The aim of the present study was to assess the level and source of microbial pollution in the Sava River Basin (SRB).
Materials and methods. The sampling at the SRB was performed in September 2015, at 15 locations. Additional samples were collected from 4 wastewater outlets. Quantification of total coliforms, E. coli and enterococci was performed with Colilert Quanti-Tray 2000 and MPN approach. The human-associated BacHum and HF183II, the ruminant-associated BacR and the pig-associated Pig2Bac fecal markers were enumerated by quantitative PCR (qPCR).
Results. According to the concentrations of fecal indicator bacteria, the river samples were scattered within the classes I and II (slight to moderate pollution). According to all monitored indicators, samples collected at 3 wastewater outlets were excessively contaminated. The results of MST revealed the presence of human-associated fecal markers BacHum and HF183II at 13 locations. The ruminant-associated BacR and the pig-associated Pig-2-Bac markers were not detected. High correlation was observed between the standard fecal indicators and human associated fecal markers.
Conclusions. The results reveal human origin of fecal pollution in the SRB and indicates the urgent need for effective wastewater treatment plants in water management.

AB  - Introduction. The SOS/umuC assay is used for assessment of water genotoxicity. Salmonella typhimurium TA1535/pSK1002 is used in this test, but the results can be extrapolated on higher eukaryotic organisms with the introduction of enzymatic S9 fraction in experimental procedure. The test is standardised for the determination of the genotoxic potential of water and wastewater (ISO/DIS 13829, 2000). Aim. Sensitivity of the assay was challenged by using parallel in situ and in vitro approach in evaluation of the genotoxic potential in the basins of significant tributaries of the Danube River: the Sava River and the Velika Morava River.
Materials and methods. Within in vitro testing, native water samples were analysed by SOS/umuC test. DNA damage in situ was assessed in bleak (Alburnus alburnus) erythrocytes by the comet and micronucleus assays. The concentration of heavy metals in fish tissue and the data of the physico-chemical parameters measured in water were used as a measure of the pollution pressure at the sites.
Results. Results showed that applied in vitro tests with native water samples are less sensitive in comparison with in situ tests. None of 20 investigated samples showed genotoxic potential in SOS/umuC assay while in situ analyses indicated variation of genotoxic potential among the investigated sites.
Conclusions. The results of our study point towards low sensitivity of the SOS/umuC test when processing un-concentrated (native) water samples; the results of in vitro tests should be taken with precaution when making predictions on the status of the ecosystem.

AB  - Biomarkers attract increasing attention in environmental studies, as a tool for detection of exposure and effects of pollution, from both natural and anthropogenic sources. This study aims to assess the impact of multiple stressors during distinctive seasons, covering also extreme hydrological events (extensive flooding in the mid May 2014), on different levels of biological organization in the liver and gills of three closely related freshwater breams. Our previous study on DNA damage in blood cells of these specimens showed increased DNA damage in June 2014, one month after the flooding event. As a continuation of that research, the present study was conducted. As a biomarker of exposure DNA damage was measured by applying the alkaline comet assay, while histopathological alterations were monitored as a biomarker of effect. Additionally, concentrations of metals and metalloids in gills, liver and muscle were assessed. Sampling of fish tissues was performed in 2014, during winter (January and February), spring (March and early June) and summer (late June, July and August). Significant seasonal difference in DNA damage was observed for both tissues. During spring and summer the level of DNA damage in gills was significantly higher when compared to the liver. Histopathological analyses showed higher frequency of alterations in gills during spring, and in liver during summer, but without a significant seasonal difference. Gills had the highest concentration of metals and metalloids during the spring and summer, and liver during winter. Muscle was the least affected tissue during all three seasons. This study highlighted the importance of the multiple biomarker approach and the use of different fish tissues in assessment of surface water pollution.

AB  - The typology and quantity of the marine litter was investigated in the shallow coastal area of southeast Adriatic Sea (Montenegro). Surveys were performed during the period 2012-2014 and 47 sites were analysed during underwater visual surveys by SCUBA diving. Litter items were classified in seven categories (plastic, metal, rubber, paper, clothing, glass and others) according to the nature of the material. Plastic litter was dominant (54%) followed by metal items (23%), and the density of marine litter was from 0 to 16.6/1000m 2 with an average of 2.49/1000m2. As expected, a greater abundance of marine litter was found in the Boka Kotorska Bay and close to urbanised areas.

AB  - Freshwater sponges, compared to their marine counterparts have not been studied extensively in the past. The
standard approach in their identification is microscopy analysis of the sponge skeletal structures, combined with
the, ever more popular, genetic analysis. The aim of this paper is to briefly describe the use of light microscopy
and scanning electron microscopy (SEM), two methods applied in sponge ultrastructure analysis, as well as to
present preliminary results on identification of samples from the Danube River. Based on the reviewed material,
the species Spongilla lacustris (Linnaeus 1759) and Ephydatia fluviatilis (Linnaeus 1759) were identified.
This paper describes sample preparation and highlights some of the skeletal characteristics crucial for the
identification of these two freshwater sponge species, widespread in Europe, Asia and North America.

(Unio pictorum/Unio tumidus) and Sinanodonta woodiana. The comet assay was used for the assessment of DNA damage. The research was performed on 34 out of 68 sites analysed within the Joint Danube Survey 3 - the world's biggest river research expedition of its kind in 2013. During research, 2285 river kilometres were covered with an average distance of 68km between the sites. The complex data set on concentrations of various substances present in water, suspended particulate matter and sediment on investigated sites gave the opportunity to identify the groups of xenobiotics which mostly affect the studied biomarker - DNA damage. The highest levels of DNA damage were recorded in the section VI (Panonnian Plain), which is under the impact of untreated wastewater discharges. Both positive and negative influences of the large tributaries on the level of genotoxicity in the Danube River were evident. Significant correlation in response was detected between the studied species of freshwater mussels. The level of DNA damage in mussels correlated with concentrations of compounds from the group of hazardous priority substances (polycyclic aromatic hydrocarbons), persistent organic pollutants (dioxins) and emerging pollutants (Oxazepam, Chloridazon-desphenyl).

AB  - Extreme hydrological events, such as water scarcity and flooding, can modify the effect of other stressors present in aquatic environment, which could result in the significant changes in the ecosystem functioning. Presence and interaction of various stressors (genotoxic pollutants) in the environment can influence the integrity of DNA molecules in aquatic organisms which can be negatively reflected on the individual, population and community levels. Therefore, in this study we have investigated the impact of flooding, in terms of genotoxicity, on organisms belonging to different trophic levels. The study was carried out on the site situated in the lower stretch of the Sava River which faced devastating effects of severe flooding in May 2014. The flooding occurred during our field experiment and this event provided a unique opportunity to assess its influence to the environment. The in situ effects of this specific situation were monitored by measuring physical, chemical and microbiological parameters of water, and by comparing the level of DNA damage in coelomocytes and haemocytes of freshwater worms Branchiura sowerbyi, haemocytes of freshwater mussels Unio tumidus and blood cells of freshwater fish Abramis bjoerkna/Abramis sapa, by means of the comet assay. Our study indicated that the flooding had a significant impact on water quality by decreasing the amount and discharge rate of urban wastewaters but simultaneously introducing contaminants from the nearby fly ash disposal field into river by runoff, which had diverse effects on the level of DNA damage in the studied organisms. This indicates that the assessment of genotoxic pollution in situ is strongly affected by the choice of the bioindicator organism.

AB  - Coupled measurements of nitrate (NO3-), nitrogen (N), and oxygen (O) isotopic composition (δ15NNO3 and δ18ONO3) were used to investigate the sources and processes of N cycling, while the microbial source tracking (MST) method was used to identify microbiological pollution in the surface water of the Sava River Basin (SRB) in autumn in 2014 and 2015 during high and low water discharge. Atmospheric nitrate deposition or nitrate-containing fertilizers were found not to be significant sources of riverine nitrate in the SRB. The ranges of isotope values suggest that NO3- in the SRB derives from soil nitrification, sewage, and/or manure, which were further supported by MST analysis. Microbiological indicators show the existence of hotspots of fecal pollution in the SRB, which are human associated. Long-term observations indicate persistent fecal contamination at selected locations caused by continuous discharge of untreated wastewaters into the SRB.

In total, 12 sites were chosen in compliance with the goals of GLOBAQUA project dealing with the effects of multiple stressors on biodiversity and functioning of aquatic ecosystems. The genotoxic potential was assessed using a complex battery of bioassays performed in prokaryotes and aquatic eukaryotes (freshwater fish). Battery comprised evaluation of mutagenicity by SOS/umuC test in Salmonella typhimuriumTA1535/ pSK1002. The level of DNA damage as a biomarker of exposure (comet assay) and biomarker of effect (micronucleus assay) and the level of oxidative stress as well (Fpg-modified comet assay) was studied in blood cells of bleak and spirlin (Alburnus alburnus/ Alburnoides bipunctatus respectively). Result indicated differential sensitivity of applied bioassays in detection of genotoxic pressure. The standard and Fpg-modified comet assay showed higher potential in differentiation of the sites based on genotoxic potential in comparison with micronucleus assay and SOS/umuC test. Our data represent snapshot of the current status of the river which indicates the presence of genotoxic potential along the river which can be traced to the deterioration of quality of the Sava River by communal and industrialwastewaters. The major highlight of the study is that we have provided complex set of data obtained from a single source (homogeneity of analyses for all samples).

To give an insight into the nature of the genotoxic activity we employed the battery of prokaryotic and eukaryotic assays. Mutagenicity in water samples was evaluated by SOS/umuC test in Salmonella typhimurium TA1535/pSK1002. The level of DNA damage as a biomarker of exposure (comet assay) and biomarker of effect (micronucleus assay) and the level of oxidative stress as well (Fpg - modified comet assay) were studied in blood cells of Salmo cenerinus Nardo, 1847 and Salmo marmoratus Cuvier, 1829. Within the applied bioassays, comet assay showed the highest potential for discriminating the sampling sites which are under lesser extent of pressure (sampling sites 1-Barnes at Bresimo and 4-Noce downstream S. Giustina) from the sites under high pressure (sampling sites 5-Noce at Mezzolombardo and 6/7-Adige upstream and downstream municipality of Trento). Significant correlation between the standard and Fpg - modified comet assay indicated that oxidative stress could be a major contributor to observed DNA damage in collected specimens.

AB  - Element concentrations in selected fish species from different trophic levels were analyzed. The following fish species were analyzed: common nase (Chondrostoma nasus), roach (Rutilus rutilus), freshwater bream (Abramis brama), barbel (Barbus barbus), Prussian carp (Carassius gibelio), chub (Squalius cephalus), European perch (Perca fluviatilis), wels catfish (Silurus glanis) and northern pike (Esox lucius). Fish were collected from the Meduvršje Reservoir (West Morava River Basin, western Serbia) during 2012, and samples of liver, muscle and gills were analyzed for As, B, Ba, Cd, Co, Cr, Cu, Fe, Hg, Li, Mn, Mo, Ni, Pb, Sr, and Zn using inductively coupled plasma optical emission spectrometry (ICP-OES). The liver and gills had the highest measured element concentrations, with Cu, Zn, Fe and Sr being the most prominent. The bioaccumulation of metals was species-specific, with the accumulation exhibiting the following trends: Prussian carp > northern pike > freshwater bream > European perch > chub > common nase > barbel > roach > wels catfish; Li > Pb > Z n > Cu > Fe > Ba > Mn > Sr > Cr > As > Mo > Ni > B, in all examined species; for Co, Cd and Hg, the bioaccumulation factor (BAF) was 0. Results of this study point to the tissue-specific differences in element concentrations, and to distinct differences between fish species regarding the accumulation patterns: common nase, with the highest accumulation observed in the liver, and Prussian carp, with the highest accumulation observed in the gills.

AB  - Genotoxicity monitoring of the lower stretch of the Sava River was performed by the combined approach of in situ assessment of genotoxicity and active biomonitoring of two species of mussels from the Unionidae family, Unio pictorum and Unio tumidus. Genotoxic response was studied using comet assay on hemocytes. For active biomonitoring, the mussels were acclimated to controlled laboratory conditions for 10 days and then exposed at two sites in the Sava River in the area of the city of Belgrade. Hemolymph of exposed specimens of each species was taken after 7, 14, and 30 days of exposure. For in situ assessment, the mussels were collected from five sites in the lower flow of the Sava River. The mussels were sampled immediately after the acclimation served as controls in both types of monitoring procedures. The results of our studies indicated the presence of genotoxic pollution at all studied sites at the Sava River. The level of DNA damage varied at different sites depending on the source and level of pollution. The response to genotoxic pollution was evident at the site in the urban area of Belgrade city, as well as at the sites far from the large urban settlements, suggesting that the lower flow of the Sava River is under pollution pressure.

AB  - Data on the microbiological quality of the freshwater systems under the anthropogenic influence, such as the Sava River, are of the major importance for the water resource management. Furthermore, analyses of the microbial quality of fish meat provide information of the fish as a valuable food resource from the investigated river basin. The health status of the fish, including dynamics of infection and biodiversity of endoparasites, is important bioindicator of changes in the ecosystem structure and function. For the ecosystem-based approach to the Sava River management, investigations of microbiological quality of the Sava River water and the meat of the European chub as the bioindicator organism, as well as dynamics of infection/biodiversity of intestinal parasites Acanthocephala, were performed. The survey comprised the data collected in periods 2005, 2006 and 2012. Microbiological investigation of water was performed in 2006 and 2012, while microbiological analyses of fish meat and ichthyo-parasitological investigation took place during 2005–2006. A high number of heterotrophic bacteria were recorded during 2006 survey, confirmed by the distinctly higher values of the three faecal indicators (total coliform, E. coli and enterococci), and indicated poor water quality downstream of the cities Zagreb and Velika Gorica, as a result of the municipal sewage outlets. The results from 2012 survey indicated the existence of moderate to critical faecal and organic pollution in all samples. Accumulation of the bacteria in the European chub meat was mainly uniform along the watercourse within standards and limitations for the human consumption. Sampling sites downstream cities of Zagreb and Velika Gorica were characterised with the lower prevalence and abundance of two common species of the chub intestinal acanthocephalan parasites, Pomphorhynchus laevis and Acanthocephalus anguillae. Poor microbiological quality of the water and lower distribution of chub intestinal parasites were related to the anthropogenic influence, downstream of the urban areas.

   short-term (96 h) tests to 5-FU (0.004, 0.04, 0.4, 4 and 40 mu M), ET
   (0.004. 0.04, 0.4 and 4 mu M) and CdCl2 (0.004, 0.04, 0.4, 4 and 40 mu
   M). Mortality of worms was observed only for CdCl2 (4 and 40 mu M). Cell
   viability lower than 70 \% was detected for 5-FU (0.4, 4 and 40 mu M),
   ET (4 mu M) and CdCl2 (0.4 and 4 mu M). All tested substances induced
   significant increase of DNA damage except 0.004 mu M of ET. L.
   udekemianus being sensitive to all tested substances indicates that it
   can be used in ecogenotoxicology studies. Concern should be raised to
   cytostatics, especially to 5-FU, since concentration of 0.004 mu M
   induced DNA damage is similar to ones detected in wastewaters.
