AB  - BACKGROUND
According to World Health Organization, 70-95% of the population chooses folk medicine as their primary
approach for health maintenance. In addition to their healing properties and application in folk medicine,
Lamiaceae plants are often valued in cookery as spices and food preservatives.
OBJECTIVES
Hence, the aim of this study was to examine the antibiofilm activity of 12 ethanolic extracts of Lamiaceae species
on biofilm formation and the degradation of existing biofilm of Pseudomonas aeruginosa PAO1 (ATCC 15692).
METHODS
The crystal violet staining method was used to evaluate the newly formed bacterial biofilms. Four concentrations
(double dilutions) of plant extracts (starting conc. 2500 μg/mL), solvent control (ethanol, starting conc.15%),
and positive control (streptomycin, starting conc. 12.5 μg/mL) were tested.082
RESULTS
All ethanol extracts showed antibiofilm activity. However, the strongest activity was observed for Hyssopus
officinalis, Melissa officinalis, Mentha piperita, and Ocimum basilicum, where the viability of bacteria in the
biofilm after treatment was about 60% mainly at each tested concentration. Teucrium chamaedrys extract
exhibited the strongest activity by degrading about 60% of biofilm (about 40% viability) at all concentrations
except at the lowest tested one. Promising results were also observed for M. piperita (at each of the tested
concentrations) and O. basilicum (at the highest tested concentration), where the viability of bacteria in the
biofilm after treatment was reduced by about 40%. The remaining extracts showed a slightly lower effect on
the degradation of the previously formed biofilm of P. aeruginosa PAO1.

AB  - Within the past decade, our research group placed great effort in
exploration of the impact of treated/untreated wastewaters on aquatic
ecosystems as less than 13 % of collected municipal wastewaters are
processed before their release to receiving waters in Serbia. Having in mind
that more than 90% of the rivers within the territory of Serbia belong to the
Danube River Basin, our major focus is naturally on the large transnational
waters such as the Danube and Sava rivers. Depended on the concept of the
study and hydro-morphological characteristics of the sites, various
approaches have been employed for in situ assessment of eco/geno-toxicity
which use different organisms in passive and active biomonitoring. Aquatic
organisms of different trophic levels have diverse life strategies, metabolism
pathways, and consequently, they have a different response to pollutant
pressure. In eco/geno-toxicological studies we have successfully employed
several species of freshwater mussels (Unio tumidus, Unio pictorum and
Sinanodonta woodiana) as well as few economically important fish species
(common carp (Cyprinus carpio), freshwater bream (Abramis brama), bleak
(Alburnus alburnus, chub (Squalius cephalus)).Comet assay is our primary
choice for the assessment of DNA damage considering its sensitivity and
cost effectiveness. Selection of certain types of tissues for the assessment
(such as blood or haemolymph), which do not require sacrificing of the
animals or additional manipulation in preparation of cell suspension coupled
with mini-gel format of the slides enables high-throughput screening of the
genotoxic potential in situ. Additionally, cryopreservation of the blood
samples directly onsite expands the research area as it overcomes issues
related to safe transportation of the samples to the laboratory. Data
obtained so far indicated that the genotoxic response in the studied
indicator organisms significantly differ which is understandable considering
the difference in uptake, accumulation and physiological responses.

AB  - More frequent application of biological parameters beside the conventional chemical parameters in national
monitoring for marine ecosystem assessment is supported by UNEP/MAP. Accordingly, to evaluate the
possible effects of pollution by determination of trace element content and genotoxicity assessment in
marine organisms within the Boka Kotorska Bay, Montenegro (southern Adriatic), we carried a field study
which included transplantation of the seabream (Sparus aurata) from aquaculture farm (Orahovac) to more
impacted sites (Dobrota and port of Tivat), situated in vicinity of the main ports. On the sampling site
Orahovac, the group of seabream specimens were placed in closed fish trap on 5 m depth (O1), another fish
trap was placed in Dobrota, while the third fish trap with seabreams was placed at the site port of Tivat.
The additional group was sampled from standard aquaculture cage on sampling site (O2) to reduce possible
bias in genotoxicity data due to confined space within the fish trap. After two weeks of exposure, blood
from fish heart was taken, afterwards muscle tissues were prepared for trace element determination.
Genotoxicity was measured by DNA damage induction based on comet (single cell gel electrophoresis)
assay parameter – Tail intensity (TI%), while trace element (As, Cd, Pb, Hg, Cr, Cu, Fe, Mn, Ni, Zn and
V) content was determined by ICP. Mean values of TI% obtained from seabream specimens at the sampling
site Orahovac were: 4.26 ± 0.17 – O1 and 2.56 ± 0.11 – O2, while TI% values for transplanted specimens
on the sites Dobrota and port of Tivat were 6.38 ± 0.17 and 11.06 ± 0.23, respectively. Statistically
significant differences (p<0,05) of TI% were observed between all specimens groups. The group of
seabream from port of Tivat showed the highest TI% (higher level of DNA damage), most probably caused
by marine pollution. Obtained trace element concentrations in seabream tissues were significantly lower in
comparison to values recommended by FAO, EC and national legislatives of Croatia, Spain and Turkey.
Since transplanted fish showed a significant response in a relatively short exposure period, our results
support the introduction of seabream caging as monitoring tool to reveal marine pollution by multiple
biomarker approach. Considering the significant share of aquacultured seabream in human consumption,
higher availability for sampling and convenience in application for molecular biomarker analyses in the
same samples, such an integrated monitoring approach would contribute to more credible data.

AB  - This study was conducted on the Danube River locality Višnjica, exposed to the discharge of the largest wastewater collector in Serbia's capital, Belgrade. Concentrations of metals and metalloids (Al, Fe, Cr, As, Sr, Mn, Cd, Zn, Mo, Cu, Li, Ni, B, Co, Pb, and Ba) and histopathological alterations were investigated in different tissues of common bream during one representative month of each season in 2014. This is the first study in which these two biomarkers were assessed parallelly in common bream. The highest concentrations of examined elements were noticed in gills and the lowest in muscle. Statistically significant differences in element concentrations between different seasons were noticed only in gills for Al, Cu, and Fe. Concentrations of As and Pb in fish muscle were below the maximum acceptable concentrations (MAC). The histopathological index (HI) of the liver showed higher values in comparison to the HI of the gills. Histopathological index of the gills had a significantly higher score in November in comparison to August. The liver HI had the highest score in April, and the lowest in August, while the total histopathological index had the highest score in November, and the lowest in August, both without significant differences between the months. This study endorses gills and liver as reliable organs for studying accumulation and histopathology as biomarkers of environmental changes. A faster reaction of the gills was confirmed since seasonal variations of both biomarkers were observed in this organ. Common bream proved as a good indicator of the state of organisms in polluted environments.

abstract = "This study was conducted on the Danube River locality Višnjica, exposed to the discharge of the largest wastewater collector in Serbia's capital, Belgrade. Concentrations of metals and metalloids (Al, Fe, Cr, As, Sr, Mn, Cd, Zn, Mo, Cu, Li, Ni, B, Co, Pb, and Ba) and histopathological alterations were investigated in different tissues of common bream during one representative month of each season in 2014. This is the first study in which these two biomarkers were assessed parallelly in common bream. The highest concentrations of examined elements were noticed in gills and the lowest in muscle. Statistically significant differences in element concentrations between different seasons were noticed only in gills for Al, Cu, and Fe. Concentrations of As and Pb in fish muscle were below the maximum acceptable concentrations (MAC). The histopathological index (HI) of the liver showed higher values in comparison to the HI of the gills. Histopathological index of the gills had a significantly higher score in November in comparison to August. The liver HI had the highest score in April, and the lowest in August, while the total histopathological index had the highest score in November, and the lowest in August, both without significant differences between the months. This study endorses gills and liver as reliable organs for studying accumulation and histopathology as biomarkers of environmental changes. A faster reaction of the gills was confirmed since seasonal variations of both biomarkers were observed in this organ. Common bream proved as a good indicator of the state of organisms in polluted environments.",

AB  - Salvia officinalis (sage) and Salvia rosmarinus (previously Rosmarinus officinalis, rosemary) are commonly used medicinal plants from the Lamiaceae family, with strong antioxidant abilities. This study was carried out in several stages to comprehensively and gradually explore the antioxidant and antitumor mechanisms of their ethanolic extracts using different experimental approaches. Their in vitro antioxidant activity was assessed using ABTS assay. Plasmid pUC19 E. coli XL1-Blue was used to evaluate their genoprotective activity, while SOS/umuC assay on Salmonella typhimurium TA1535/pSK1002 and Comet assay on human lung fibroblast cell line (MRC-5) were employed to examine their antigenotoxic potential. Furthermore, their antitumor mechanisms were analyzed on the colorectal cancer cell line (HCT-116) by testing their impact on the cells’ viability (MTT assay), as well as the production of reactive oxygen species (ROS) (NBT test) and nitric oxide (NO) (Griess assay). The results showed that sage extract had slightly higher ABTS-scavenging and antigenotoxic activities in the bacterial model when compared to the rosemary extract, however, opposite results were obtained for the acellular model. Moreover, both extracts protected the MRC-5 cells from the induced genomic damages. As for tumor cells, sage extract reduced their viability by approximately 10 %. Additionally, both extracts significantly reduced the ROS production, while also increasing the NO production, especially the rosemary extract. These results delivered novel information about the nature of antioxidant effects of sage and rosemary ethanolic extracts. They provided substantial protection of the acellular, bacterial and healthy human cellular models by employing radical-scavenging and antigenotoxic mechanisms. At the same time, both extracts exhibited antitumor activity towards colorectal cancer cells in vitro that could be mediated by inducing an increase in NO production. With that being said, sage and rosemary ethanolic extracts represent potent genoprotective, antioxidant and antitumor agents. AB  - Water quality in the Danube River Basin is largely influenced by the inputs of pollutants - particularly
excessive nutrients, organic material, and hazardous substances, which affects the whole ecosystem
at various levels. The major goal of this study was to evaluate the level of genotoxic potential at the
site at Danube River, Novi Banovci, which is under significant anthropogenic pressure of untreated
municipal and industrial waste waters.
DNA damage in situ was assessed in 5 specimens of white bream (Blicca bjoerkna) erythrocytes, liver
and gills cells with comet assay and in erythrocytes with micronucleus test. Additionally, genotoxicity
of native water samples collected from the site and upstream and downstream of the site was
evaluated by the comet assay on HepG2 cells. Microscopic images of comets were scored using Comet
IV Computer Software (Perceptive Instruments, UK) and tail intensity was chosen as parameters to
assess the DNA damage.
Comet assay on white bream showed that gills were the most affected tissue, while liver showed the
smallest values. Micronuclei frequency was low in general, with mean of 1 micronuclei in 5000 scored
erythrocytes, with no significant correlation to the values of comet assay in erythrocytes. Comet assay
on HepG2 cells showed significant difference between values of negative control samples and samples
of water from effluent site and downstream of wastewater discharge. There was no significant
difference between negative control and upstream samples of water.
Our results showed that great impact of municipal and industrial wastewaters on the river ecosystem.
The application of in vitro and in situ tests gives a better insight into genotoxic effects. More
importantly, because Serbia still doesn’t have waste water treatme nt plants, this kind of research
points out the imperative for implementation of this facilities.

AB  - The main objective of this research was to evaluate the impact of methanolic, ethanolic and aqueous extracts of Origanum majorana L., Origanum vulgare L., Teucrium chamaedrys L., Teucrium montanum L., Thymus serpyllum L. and Thymus vulgaris L. (Lamiaceae) on the effects of free radicals using different model systems. The extracts were characterized on the basis of the contents of total phenolics, phenolic acids, flavonoids and flavonols, and also using high-performance liquid chromatography with diode-array detection. Antioxidant activity in vitro was assessed using DPPH assay. The genoprotective properties were tested using plasmid relaxation assay on pUC19 E. coli XL1-Blue, while SOS/umuC assay on Salmonella typhimurium TA1535/pSK1002 and Comet assay on human lung fibroblasts were used to assess the antigenotoxicity of the extracts. Ethanolic extracts had the most phenolics (up to 236.20 mg GAE/g at 0.5 mg/mL), flavonoids (up to 42.47 mg QE/g at 0.5 mg/mL) and flavonols (up to 16.56 mg QE/g at 0.5 mg/mL), and they exhibited the highest DPPH activity (up to 92.16% at 0.25 mg/mL). Interestingly enough, aqueous extracts provided the best protection of plasmid DNA (the lowest IC50 value was 0.17 mg/mL). Methanolic extracts, on the other hand, most efficiently protected the prokaryotic DNA, while all the extracts had a significant impact against genomic damages inflicted on human fibroblasts. O. vulgare extracts are considered to be the most promising in preserving the overall DNA integrity against oxidative genomic damages. Moreover, HPLC-DAD analysis highlighted rosmarinic acid as the most abundant in the investigated samples (551.45 mg/mL in total in all the extracts), followed by luteolin-7-O-glucoside (150.19 mg/mL in total), while their presence correlates with most of the displayed activities. The novelty of this study is reflected in the application of a prokaryotic model for testing the antigenotoxic effects of Lamiaceae species, as no previous reports have yet been published on the genoprotective potential of these species.

Chemical characterization of Lamiaceae plant extracts – the quest for novel immunomodulators

AB  - The physiological imbalance between the production and accumulation of reactive oxygen species (ROS), known as oxidative stress, is responsible for developing and progression of a plethora of diseases – cancer, among others. Since plants are recognized as potential antioxidant agents, the aim of this study was to chemically characterize ethanolic extracts of 18 species from Lamiaceae family using liquid chromatography-mass spectrometry (LC-MS), followed by testing the antioxidant activity using ABTS assay. Biocompatibility of the investigated extracts was assessed by MTT assay on normal human lung fibroblasts (MRC-5). Finally, antiproliferative activity was evaluated using MTT assay on human colorectal cancer (HCT-116) cell line, while the effects of extracts on superoxide anion radical (O2•-) production were tested using NBT assay. Melissa officinalis, Mentha piperita, Origanum majorana, O. vulgare, and Thymus vulgaris showed the highest antioxidant activity against ABTS radicals. LC-MS analysis indicated that these extracts mostly contain caffeic, protocatechuic and p-coumaric acids. The extracts were biocompatible with MRC-5 cell line. Interestingly, Lavandula angustifolia and Ocimum basilicum extracts, which were less active against ABTS radicals, showed a significant antiproliferative effect against HCT-116. Moreover, L. angustifolia and T. vulgaris extracts significantly stimulated the production of O2•- in HCT-116, indicating potential antitumor activity. The extracts that expressed antiproliferative effect and stimulated ROS production contain higher amounts of rosmarinic, caffeic and chlorogenic acids, naringin, luteolin, apigenin and cirsimaritin, indicating that some of them, or more probably their synergistic effects, are responsible for the extracts’ ability to moderate the production of ROS in HCT-116 cells.
