AB  - Mushrooms are valued worldwide for their nutritional, organoleptic and chemical properties. The aim of this study was to determine the chemical composition (free sugars, organic acids, fatty acids, tocopherols and phenolic compounds) and bioactivity of three wild mushrooms (Lactarius piperatus, Lactarius quietus and Lactarius vellereus) from Serbia. Chemical analysis was performed with HPLC-RI and UFLC-PDA (for hydrophilic compounds) and with GC-FID and HPLC-FP (for lipophilic compounds). The analysis of phenolic compounds was performed by UFLC-DAD. Biological activities were evaluated using three different assays (microdilution, TBARS and SRB assays). The results showed that the fruiting bodies were rich in mannitol and trehalose. The main organic acids were oxalic acid and citric acid. As for lipophilic components, stearic, oleic and linoleic acids and β-tocopherol dominated in all the species studied. In addition, the methanolic and ethanolic extracts obtained showed antibacterial, antibiofilm and antioxidant properties. As for cytotoxicity, the extracts were not toxic or only moderately toxic toward different tumor cell lines. According to the results, the selected Serbian mushrooms are a rich source of bioactive compounds, and due to their good biological potential, they can be further exploited as functional ingredients beneficial to human health (antimicrobial agents, antioxidants).

AB  - In recent years, the interest in products of natural origin has boosted the exploitation and use of plants as food and sources of bioactive compounds, especially wild plants widely used in different cultures for several purposes. Commelina erecta is a wild edible plant (WEP) traditionally used as food and medicine, about which few studies exist. Thus, this study aimed at enhancing the knowledge about its nutritional, chemical and bioactive profile, considering different plant parts and development stages, in order to increase its inclusion in the diet of South American communities. The nutritional profile was found to be similar to other WEP frequently consumed in Brazil. Thirteen phenolic compounds (HPLC-DAD-ESI/MS) were tentatively identified, with apigenin, luteolin and quercetin derivatives being the most abundant. Fructose and oxalic acid were the major sugar and organic acid, respectively, in the aerial parts of C. erecta, and four isoforms of tocopherols were also identified. Regarding the plant's antioxidant activity, the EC50 values varied between 18.4 and 1060 µg/mL in the inhibition of lipid peroxidation assay (TBARS) and between 53 and 115 µg/mL in the oxidative haemolysis inhibition (OxHLIA) assay. The hydroethanolic extract obtained from stems at the flowering stage also presented anti-inflammatory activity. In general, all the extracts evidenced promising antimicrobial activity. Altogether, these results reinforce the traditional use of this plant species as food and medicine to support the diet of needier populations and also promote food sovereignty and sustainability.

AB  - Herein, we report the experimental evaluation of the antimicrobial activity of seventeen new (Z)-methyl 3-(4-oxo-2-thioxothiazolidin-5-ylidene)methyl)-1H-indole-2-carboxylate derivatives. All tested compounds exhibited antibacterial activity against eight Gram-positive and Gram-negative bacteria. Their activity exceeded those of ampicillin as well as streptomycin by 10-50 fold. The most sensitive bacterium was En. Cloacae, while E. coli was the most resistant one, followed by M. flavus. The most active compound appeared to be compound 8 with MIC at 0.004-0.03 mg/mL and MBC at 0.008-0.06 mg/mL. The antifungal activity of tested compounds was good to excellent with MIC in the range of 0.004-0.06 mg/mL, with compound 15 being the most potent. T. viride was the most sensitive fungal, while A. fumigatus was the most resistant one. Docking studies revealed that the inhibition of E. coli MurB is probably responsible for their antibacterial activity, while 14a-lanosterol demethylase of CYP51Ca is involved in the mechanism of antifungal activity. Furthermore, drug-likeness and ADMET profile prediction were performed. Finally, the cytotoxicity studies were performed for the most active compounds using MTT assay against normal MRC5 cells.

AB  - The purpose of this study was to evaluate the surface texture and biofilm adhe sion of veneered or CAD/CAM milled zirconia (partially stabilized with yttrium) after professional
oral hygiene procedures. The samples (4 × 4 mm, thickness 2 mm; n = 72) were separated from
zirconia blanks (3Y-TZP-LA). One group was veenered with ceramics, and the other group of samples
was CAD/CAM milled. Each group had two subgroups: polished and glazed. The samples were
subjected to simulated strokes of professional brushing using abrasive paste and ultrasonic scaling.
The parameters of surface micromorphology and receptivity to biofilm were calculated before and
after simulating the given methods of the professional maintenance of oral hygiene. Scanning electron
microscopy (SEM) was used to evaluate zirconia surface properties. Microbial (bacterial/fungal)
species (Staphylococcus aureus, Streptococcus sanguinis and Candida albicans) were used and cultured
on respective sterilized zirconia surfaces. Colony-forming unit (CFU) counts were used to quantify
the amount of biofilm formation on zirconia samples surfaces. Results: The SEM analysis showed
the greatest change in surface microtopography after the use of ultrasonic scaling on glazed zirconia
samples. Less formation of colonies on the surfaces of CAD/CAM milled zirconia restorations was
observed. Conclusion: Routine methods of oral hygiene professional maintenance can damage the
surfaces of glazed zirconia restorations.

Natural extracts against agricultural pathogens: A case study of Celtis australis L.

AB  - Plant extracts and other plant products have been used as an alternative to synthetic fungicides or an additional way to reduce their use. The choice of plant extracts and their application depends on their functional characteristics, availability, cost-effectiveness and their impact on phytopathogens, and also on the environment. Therefore, the present study aims to assess the potential of Celtis australis methanolic extracts as source of compounds with antifungal activity. Methanolic extracts prepared from leaves and unripe mesocarps of C. australis collected from different localities of Montenegro (Podgorica-PG, Donja Gorica-DG, and Bar-BR), were evaluated for their phenolic compounds composition as well as antifungal and cytotoxic properties. Obtained results revealed that extracts contain various bioactive constituents including phenolic acids, flavonoids and their derivatives. The predominant phenolic acid was ferulic acid, identified in leaves samples from DG (187.97 mg/100g dw), while isoorientin was the most abundant phenolic compound found in all examined samples. Regarding antifungal potential of the tested samples, all but one (prepared from mesocarp BR) possessed higher activity, than Previcur, commercial systemic fungicide intended to control seedling. In vitro studies on HaCaT cell line showed that the extracts had no toxic effect towards the tested cell line. These results lead to the conclusion that methanolic extracts of C. australis can become an alternative to the use of synthetic fungicides in agriculture. Those extracts represent natural biodegradable fungicides and enable a more efficient control of pathogenic fungi.

Water Stress Alleviation Effects of Biostimulants on Greenhouse-Grown Tomato Fruit

AB  - The aim of the present study was to evaluate the effects of three biostimulant products (Nomoren (N), Twin Antistress (TW), x-Stress (XS) and control treatment (C: no biostimulants added)) on the nutritional value, chemical composition and bioactive properties of greenhouse tomato fruit grown under full (W+: 100% of field capacity) and deficit irrigation (W–: 70% of field capacity) conditions. Fat content was the highest for the fully irrigated plants that received no biostimulants (CW+), while proteins and carbohydrates and energetic value were the highest in the XSW+ treatment. The content of the main detected sugars (fructose, glucose and trehalose) varied depending on the irrigation and biostimulant treatment. The highest amounts of individual and total organic acids and tocopherols were recorded in fully irrigated plants treated with Twin Antistress (TW), whereas the lowest overall values were observed under deficit irrigation for plants that received the XS treatment. The most abundant fatty acids were palmitic (27.5–36.0%) and linoleic acid (27.4–35.4%), followed by oleic (9.2–21.2%), linolenic (5.4–13.1%) and stearic acid (5.3–6.8%). Moreover, the highest values of β-carotene and lycopene were recorded for the CW- and NW+ treatments, respectively. The TWW+ showed the highest antioxidant activity for both assays tested (TBARS and OxHLIA). Most of the tested extracts showed lower antibacterial activity against the tested bacteria compared to the positive controls. On the other hand, CW+, XSW+ and XSW- treatments showed higher antifungal activity (MIC values) than positive controls. In conclusion, each biostimulant product had a different effect on the determined characteristics depending on the level of irrigation. Therefore, more research is needed to better identify the mechanisms of action and the physiological processes, after which the tested biostimulants may be used to standardize the application of such products in tomato cultivation.

Supercritical fluid extraction of Chicory reveals its antimicrobial, antibiofilm and wound healing potentials

AB  - Chicory (Cichorium intybus L.) is an important industrial crop cultivated mainly to extract the
dietary fiber inulin. This study aimed to investigate antimicrobial, antibiofilm and wound healing
potentials of the extract obtained by supercritical fluid extraction (SFE). Antibacterial activity
of extract ranged from 0.25 to 4.00 mg/mL regarding minimal inhibitory concentra-tions (MICs),
while minimal bactericidal concentrations (MBCs) were noted within the range of 0.5 to 8.00 mg/
mL. The extract has shown promising antifungal potential with MICs rang-ing 0.125- 0.75 mg/
mL and MFCs 0.25-1.0 mg/mL against Candida species. Incubation with the extract has reduced
the ability of both C. albicans and P. aeruginosa to attach to the sur-face and begin the process
of biofilm formation. Concentration equal to previously deter-mined MIC has almost completely
(>90% inhibition) reduced the biofilm biomass formed by P. aeruginosa. Even twice as lower concentrations
(0.5 MIC) has limited the biofilm forming ability and induce more than 80% inhibition
in P. aeruginosa as bacterial biofilm model or-ganism. Impact on fungal biofilm was less profound
and MIC concentration was able to re-duce the biofilm formation for less than 60%. HaCaT (human
immortalized keratinocytes cell line) cells were treated with IC20 (24 h) concentration of chicory
SFE extract. It is estimated that SFE extract has slight effect on wound closure in keratinocyte
monolayer, when com-pared to untreated control. Since, Chicory SFE fraction possessed excellent
antibacterial po-tential against E. coli and K. pneumoniae and antifungal activity against Candida
species these results are considered very important in the future inventions

AB  - Emerging antimicrobial resistance and side effects associated with antibiotics overuse are driving the need for the novel antimicrobial therapeutics with the natural compounds considered as an appealing alternative. Rosmarinic acid is abundantly present in medicinal plants. This study aimed to elucidate its possible role as an inhibitor of planktonic and biofilm microorganism growth, along with its antifungal mechanisms. In this study, rosmarinic acid showed promising anticandidal (MIC 0.1–0.2 mg/mL) and antibacterial (MIC 0.002->0.8 mg/mL) activity. To some extent it was able to prevent cell attachment and eradicate established biofilms. Rosmarinic acid antifungal mechanisms included reduction of mitochondrial activity, alteration in membrane integrity, and slight inhibition of protease production but not binding to ergosterol. Its antibiofilm activity was moderately linked to the reduction in exopolysaccharide production. Considering its wide antimicrobial and antibiofilm spectrum rosmarinic acid could be further explored as an antimicrobial agent along with range of medicinal plants with rosmarinic acid as the dominant compound. Nevertheless, rosmarinic acid could serve as the basis for the development of antimicrobials employing novel mechanisms of activity.

Evaluation of antimicrobial properties of chicory extracts

AB  - Cichorium intybus L. (Asteraceae), is an important medicinal plant, rich in sesquiterpene
lactones (STLs), inulin, biologically relevant elements, and vitamins which exert potent
pro-health effects on the human organism. The aim of this study was to screen two
extracts of chicory containing STLs, the optimized extract (Ci_SFEopt) and the
supercritical fluid extract (Ci_SFEpur), for their antimicrobial activities against a range
of pathogenic microorganisms. Both extracts showed antibacterial activity, with
Ci_SFEpur being more potent than Ci_SFEopt. The most sensitive species was
Pseudomonas aeruginosa (minimal inhibitory concentration, MIC 0.06 mg/mL).
Ci_SFEpur and Ci_SFEopt extracts demonstrated good antifungal activity against
Candida species (MIC 0.03-1.00 mg/mL). Thin layer chromatography (TLC)
bioautography was used to screen the extracts and provide basic information for the
isolation of bioactive compounds in the future. Ci_SFEopt was active against all tested
microorganisms (Rf values were 0.18-0.88). The Ci_SFEpur extract efficiently inhibited
(Rf 0.76) Staphylococcus aureus, P. aeruginosa and Candida krusei. The identification
of powerful antimicrobial activity of Ci_SFEpur against P. aeruginosa and Candida
species is an important step towards the development of novel antimicrobial therapeutics
and efficient utilization of this widely distributed plant.

abstract = "Cichorium intybus L. (Asteraceae), is an important medicinal plant, rich in sesquiterpene
lactones (STLs), inulin, biologically relevant elements, and vitamins which exert potent
pro-health effects on the human organism. The aim of this study was to screen two
extracts of chicory containing STLs, the optimized extract (Ci_SFEopt) and the
supercritical fluid extract (Ci_SFEpur), for their antimicrobial activities against a range
of pathogenic microorganisms. Both extracts showed antibacterial activity, with
Ci_SFEpur being more potent than Ci_SFEopt. The most sensitive species was
Pseudomonas aeruginosa (minimal inhibitory concentration, MIC 0.06 mg/mL).
Ci_SFEpur and Ci_SFEopt extracts demonstrated good antifungal activity against
Candida species (MIC 0.03-1.00 mg/mL). Thin layer chromatography (TLC)
bioautography was used to screen the extracts and provide basic information for the
isolation of bioactive compounds in the future. Ci_SFEopt was active against all tested
microorganisms (Rf values were 0.18-0.88). The Ci_SFEpur extract efficiently inhibited
(Rf 0.76) Staphylococcus aureus, P. aeruginosa and Candida krusei. The identification
of powerful antimicrobial activity of Ci_SFEpur against P. aeruginosa and Candida
species is an important step towards the development of novel antimicrobial therapeutics
and efficient utilization of this widely distributed plant.",

AB  - Veliki broj makrogljiva predstavlja nedovoljno iskorišćen izvor jedinjenja koja mogu imati potencijalnu primenu u medicini, farmaciji i različitim granama privrede. Pored toga što imaju povoljan odnos makronutrijenata, važan su izvor visoko cenjenih polisaharida, lektina, terpenoida, peptida, itd. Ova jedinjenja su odgovorna za mnogobrojne biološke aktivnosti uključujući antineurodegenerativnu, imunomodulatornu, antimikrobnu, antihiperglikemijsku aktivnost. Danas su u upotrebi različiti dijetetski proizvodi na bazi makrogljiva koji se koriste za prevenciju i ublažavanje simptoma različitih oboljenja. Iako ovi proizvodi ne mogu u potpunosti zameniti komercijalne lekove, u kombinaciji sa njihovom primenom mogu da dovedu do poboljšanja opšteg stanja. Zbog navedenog, autohtone samonikle, ali i komercijalno gajene vrste makrogljiva predstavljaju važan i još uvek nedovoljno istražen izvor bioloških aktivnih supstanci. Štaviše, one imaju ogroman potencijal kao profilaktički agensi kod bolesti čiji je osnovni uzrok povezan sa oksidativnim stresom, bakterijskim i gljivičnim infekcijama kao i pojavom/razvojem različitih tipova tumora. Imajući u vidu da su hranljiva i lekovita svojstva jestivih i lekovitih gljiva hvaljena kako u etnomedicini tako i u naučnim/kliničkim istraživanjima, ova svojstva će biti predstavljena kako bi se procenili novi trendovi i moguća primena jestivih i lekovitih gljiva sa teritorije Srbije u medicini, farmaceutskoj i prehrambenoj industriji.

AB  - Mushrooms are highly appreciated for their nutritional value and therapeutic effects. They have found wide application in pharmacy, medicine and as nutraceuticals, mostly due to the synergistic activity of their bioactive compounds. This study aimed to investigate the chemical composition and antioxidant, antibacterial, antibiofilm, and cytotoxic potential of L. piperatus and L. vellereus wild growing mushrooms from Serbia. Mannitol and trehalose were the most abundant free sugars and malic and citric acid were the major organic acids detected. The four tocopherol isoforms were found in L. piperatus, and the β-tocopherol was the only tocopherol isoform found in the L. vellereus fruiting body. Regarding phenolic compounds, cinnamic and protocatechinic acids were identified in the prepared methanolic and ethanolic extracts, which displayed antioxidant activity through the inhibition of thiobarbituric acid reactive substances (TBARS) formation (IC50 0.30-1.39 mg/mL). Antibacterial activity of Lactarius ethanolic and methanolic extracts was investigated against bacteria isolated from tonsils from the patient with chronic tonsillitis. All tested extracts demonstrated promising results regarding antibacterial and antibiofim activity. In both extracts, the most promising activity was observed for Streptococcus spp. (0.40–6.25 mg/mL as the minimal inhibitory concentration). Ethanolic extracts of tested mushrooms showed a stronger potential to inhibit the formation of Staphylococcus aureus biofilm then methanolic. L. vellereus ethanolic extract showed highest inhibition activity (86%) against S. aureus biofilm formation. Citotoxic potential of L. piperatus and L. vellereus extracts were examined against four tumor cell lines (HeLa, MCF-7, HepG2, NCI-H460) and porcine liver primary culture - PLP2. The most promising cytotoxic effect was caused by the L. vellereus ethanolic extract on cervical carcinoma cell line (HeLa). However, extensive screening of biological activities is needed in future studies that will allow the exploitation of these mushrooms at an industrial level, taking advantage of its benefits for human health.

AB  - Chronic tonsillitis (CT) is a global health issue which can impair patient’s quality of life and has an important socioeconomic impact due to the nonrational use of antibiotics, increased antimicrobial resistance and frequent need for surgical treatment. In order to isolate and identify the causing agents of CT, a total of 79 postoperative palatine and adenoid tissue samples were obtained from the ENT Clinic, KBC Zvezdara, Belgrade, Serbia. Culture identification was performed by MALDI-TOF MS and the Staphylococcus aureus isolates were tested for biofilm forming capability and antibiotic susceptibility. Additionally, a histological examination of palatine and adenoid tissue was performed in order to detect the presence of CT-causing bacteria. The slight majority of participants were females with median age of 28 years for adult patients (group I) and 6 years for children (group II). Analysis of the incidence of bacteria isolated from tissue samples in both groups showed the highest prevalence of S. aureus, Streptococcus oralis and Streptococcus parasanquinis. In addition to interfollicular hyperplasia, colonies of species S. aureus were detected in histological material. The presence of biofilm might be the reason for the recurrence of infection. Therefore, searching for a new treatment of CT is of great importance.

AB  - The chemical composition and biological activity of essential oils (EOs) from leaves of Origanum minutiflorum O. Schwarz & P.H.Davis, and seeds of Coriandrum sativum L. were investigated. Chemical analyses of EOs were performed and the major components were carvacrol (81.5%) and linalool (69.6%), respectively. The antimicrobial activity was assessed against several bacteria originating from the tonsillar tissue. Activities of EOs against Staphylococcus aureus biofilm were investigated, as well as the effect of the mixture of these EOs and antibiotics against the pathogen. The antioxidant activity of both EOs was determined by TBARS assay, and examined wild oregano EO showed better activity. Also, cytotoxicity of EOs was evaluated in vitro and both EOs showed potential to inhibit further proliferation of tumor cells. This study reported for the first time the effects of EOs on chronic tonsillitis causative pathogens, supporting their role as a natural bioactive therapeutic agent with possible antimicrobial applications.

AB  - Herein, we report computational and experimental evaluations of the antimicrobial activity of twenty one 2,3-diaryl-thiazolidin-4-ones. All synthesized compounds exhibited an antibacterial activity against six Gram-positive and Gram-negative bacteria to different extents. Thus, the MIC was in the range of 0.008–0.24 mg/mL, while the MBC was 0.0016–0.48 mg/mL. The most sensitive bacterium was S. Typhimurium, whereas S. aureus was the most resistant. The best antibacterial activity was observed for compound 5 (MIC at 0.008–0.06 mg/mL). The three most active compounds 5, 8, and 15, as well as compound 6, which were evaluated against three resistant strains, MRSA, P. aeruginosa, and E. coli, were more potent against all bacterial strains used than ampicillin. The antifungal activity of some compounds exceeded or were equipotent with those of the reference antifungal agents bifonazole and ketoconazole. The best activity was expressed by compound 5. All compounds exhibited moderate to good drug-likeness scores ranging from −0.39 to 0.39. The docking studies indicated a probable involvement of E. coli Mur B inhibition in the antibacterial action, while CYP51 inhibition is likely responsible for the antifungal activity of the tested compounds. Finally, the assessment of cellular cytotoxicity of the compounds in normal human MRC-5 cells revealed that the compounds were not toxic. View Full-Text

AB  - If chronic tonsillitis (CT) is treated surgically, the long-term risks of infections and respiratory diseases are
higher, making of high importance to find an alternative for tonsillectomy. In order to isolate and identify
the causing agents of CT, a total of 79 postoperative palatine and adenoid tissue samples were obtained
from the ENT Clinic, KBC Zvezdara. A total of 82 bacterial species were isolated and identified by MALDI-TOF.
They belong to the genera Streptococcus, Staphylococcus, Micrococcus, Rothia, Enterobacter and Stenotrophomonas.
Analysis of the incidence of bacteria isolated from tissue samples showed the highest prevalence of Streptococcus
oralis (27/33%), followed by S. aureus (17/21%) and Streptococcus parasanquinis (16/20%). Histological analysis
of tissue samples was conducted in order to detect the presence of CT-causing bacteria. In addition to interfollicular
hyperplasia, colonies of species S. aureus and Stenotrophomonas maltophilia were detected in histological material.
The presented results confirm that the colonizers of tonsils tissues can be both G (+) and G (-) bacteria. The
presence of S. aureus in the tonsillar tissue even after the antibiotic therapy may be related to its ability to
form biofilm. Our study demonstrated that among 17 S. aureus isolates, 13 displays more pronounced biofilm
forming ability than the reference strain S. aureus ATCC 11632, and only 4 isolates display lower biofilm forming ability.
The presence of biofilm might be the reason for the recurrence of infection. Therefore, a searching for new
antibiofilm therapeutics in CT therapy is of great importance.

AB  - Hesperetin is the aglycone of citrus flavonoid hesperidin. Due to the limited information regarding hesperetin antimicrobial potential and emerging need for novel antimicrobials, we have studied its antimicrobial activity (microdilution assay), antibiofilm activity with different assays in two models (mono- and polymicrobial biofilm), and toxicity (MTT and brine shrimp lethality assays). Hesperetin inhibited growth of all Candida isolates (minimal inhibitory concentration, MIC, 0.165 mg/mL), while it’s inhibitory potential towards Staphylococcus aureus was lower (MIC 4 mg/mL). Hesperetin (0.165 mg/mL) reduced ability of Candida to form biofilms and moderately reduced exopolysaccharide levels in biofilm matrix. Effect on the eradication of 24 h old C. albicans biofilms was promising at 1.320 mg/mL. Inhibition of staphylococcal biofilm formation required higher concentrations of hesperetin (<50% inhibition with MIC 4 mg/mL). Establishment of polymicrobial C. albicans-S. aureus biofilm was significantly inhibited with the lowest examined hesperetin concentration (1 mg/mL) in crystal violet and CFU assays. Hesperetin toxicity was examined towards MRC-5 fibroblasts (IC50 0.340 mg/mL) and in brine shrimp lethality assay (LC50 > 1 mg/mL). Hesperetin is efficient in combating growth and biofilm formation of Candida species. However, its antibacterial application should be further examined due to the cytotoxic effects provoked in the antibacterial concentrations

Rosmarinic acid–Modes of antimicrobial and antibiofilm activities of common plant polyphenol

AB  - Emerging antimicrobial resistance and side effects associated with antibiotics overuse are driving the need for the novel antimicrobial therapeutics with the natural compounds considered as an appealing alternative. Rosmarinic acid is abundantly present in medicinal plants. This study aimed to elucidate its possible role as an inhibitor of planktonic and biofilm microorganism growth, along with its antifungal mechanisms. In this study, rosmarinic acid showed promising anticandidal (MIC 0.1–0.2 mg/mL) and antibacterial (MIC 0.002->0.8 mg/mL) activity. To some extent it was able to prevent cell attachment and eradicate established biofilms. Rosmarinic acid antifungal mechanisms included reduction of mitochondrial activity, alteration in membrane integrity, and slight inhibition of protease production but not binding to ergosterol. Its antibiofilm activity was moderately linked to the reduction in exopolysaccharide production. Considering its wide antimicrobial and antibiofilm spectrum rosmarinic acid could be further explored as an antimicrobial agent along with range of medicinal plants with rosmarinic acid as the dominant compound. Nevertheless, rosmarinic acid could serve as the basis for the development of antimicrobials employing novel mechanisms of activity.

abstract = "Emerging antimicrobial resistance and side effects associated with antibiotics overuse are driving the need for the novel antimicrobial therapeutics with the natural compounds considered as an appealing alternative. Rosmarinic acid is abundantly present in medicinal plants. This study aimed to elucidate its possible role as an inhibitor of planktonic and biofilm microorganism growth, along with its antifungal mechanisms. In this study, rosmarinic acid showed promising anticandidal (MIC 0.1–0.2 mg/mL) and antibacterial (MIC 0.002->0.8 mg/mL) activity. To some extent it was able to prevent cell attachment and eradicate established biofilms. Rosmarinic acid antifungal mechanisms included reduction of mitochondrial activity, alteration in membrane integrity, and slight inhibition of protease production but not binding to ergosterol. Its antibiofilm activity was moderately linked to the reduction in exopolysaccharide production. Considering its wide antimicrobial and antibiofilm spectrum rosmarinic acid could be further explored as an antimicrobial agent along with range of medicinal plants with rosmarinic acid as the dominant compound. Nevertheless, rosmarinic acid could serve as the basis for the development of antimicrobials employing novel mechanisms of activity.",

Immature blades (B3) had a higher content of phenolic compounds (178 mg/g extract) and a greater ability to inhibit the formation of thiobarbituric acid reactive substances (IC50 of 1.61 µg/mL). Samples at more advanced growth stages revealed a greater capacity to inhibit oxidative hemolysis (B8, IC50 of 25 and 47.4 µg/mL for Δt of 60 and 120 min, respectively) and higher cytotoxic (B8-B13, GI50 between 7.1 and 17 µg/mL), anti-inflammatory (B13, IC50 of 10 µg/mL), and antibacterial activities. In turn, the antifungal activity varied depending on the tested fungi. All these results suggest that maturity influences the phenolic composition and bioactive properties of cardoon blades, which reveal great potential for the development of bioactive ingredients for food and pharmaceutical applications, among others.

AB  - Cytinus hypocistis (L.) L. is a comestible holoparasite with great potential for cosmeceutical application. Although its high tannin content has been associated with its bioactive and inhibitory enzyme properties, this is the first report establishing a relationship between parasite and host (Halimium lasianthum (Lam.) Greuter) phenolic profile and bioactive properties. Thus, five extracts (aerial and root extracts of non-parasited and parasited H. lasianthum and C. hypocistis) were evaluated. The tentative identification of both species comprises 39 phenolic compounds. Hydrolysable tannins and flavonoids were the main identified groups in C. hypocistis and H. lasianthum extracts, respectively. Regarding bioactivities, C. hypocistis exhibited excellent antioxidant results both in Oxidative Haemolysis (OxHLIA) and inhibition of Thiobarbituric Acid Reactive Substances Formation (TBARS). The tested extracts presented antimicrobial inhibition, anti-inflammatory activity, and effective cytotoxicity against tumour cells. C. hypocistis exhibited the lowest cytotoxicity on a non-tumour cell line. Principal Component Analysis (PCA) was a suitable approach to analyse differences among samples, explaining up to 67% of data variability and suggesting no similarities between parasite and host phenolic composition and bioactivities. Therefore, this comparative study emphasises the significance of both species as a source of biologically active compounds.

Nutritional properties, chemical composition and biological properties of Craterellus cornucopioides

AB  - Craterellus cornucopioides (L.) Pers. (Cantharellaceae), or horn of plenty, is an edible fungi. It is also known as the black chanterelle, black trumpet, crna truba (Serbian), trompette de la mort (French), trombetta dei morti (Italian) or trumpet of the dead, djondjon (Haitian). C. cornucopioides growing wild in Serbia was chemically characterized in nutritional value and individual compounds (sugars, fatty acids, tocopherols, organic acids and phenolic compounds). Antioxidant (scavenging activity, reducing power and inhibition of lipid peroxidation), antibacterial and antifungal activities of its methanolic extract were evaluated under in vitro conditions. C. cornucopioides was showed to be rich in carbohydrates (79.32 g/100g dw), followed by proteins (8.36 g/100g dw), ash (9.43 g/100g dw) and fat (2.90 g/100g dw). The most abundant sugar compound was identified as mannitol (14.16 g/100g dw); four tocopherol isoforms (α-, β-,γ- and δ-tocopherols) were detected. Unsaturated fatty acids predominated over saturated fatty acids, with oleic and linoleic acids as the most dominant ones. Five organic acids were quantified: oxalic, citric, quinic, and fumaric and malic acids. The methanolic extract of the mushroom showed in vitro antioxidant and antimicrobial activities. Methanolic extract successively inhibited growth of pathogenic bacteria.

Given that modern phytopreparation intended for use on human skin requires, above all, knowledge of its chemical composition, the goal of this study was to make a screening of the composition of aqueous and methanolic extracts of the petals of P. tenuifolia L. and to examine them for various skin-beneficial properties. The extracts were prepared by maceration, ultrasound-assisted, and microwave-assisted extraction procedures. The chemical profiling was conducted by the use of UHPLC-LTQ-OrbiTrap MS and UHPLC/MS, and spectrophotometric methods for the determination of total polyphenol and total flavonoid contents. The biological activities entailed antioxidant ABTS, DPPH, CUPRAC (Cupric Ion Reducing Antioxidant Capacity), and FRAP (Ferric Reducing Antioxidant Power) assays, antimicrobial (antibacterial and antifungal) and antibiofilm activities, cytotoxicity, wound healing potential, as well as the adhesion and invasion of Staphylococcus lugdunensis. The results showed that the petals are rich in phenolic acids and flavonoids, which are commonly associated with numerous biological activities. The aqueous extracts were more efficient in the majority of the bioactivity assays then the methanolic ones, whereas the optimal extraction method varied between the assays. This study is the first step towards the safe use of the aqueous extracts of P. tenuifolia petals for therapeutic skin treatments.

AB  - BACKGROUND Infectious diseases represent a significant global strain on public health security and impact on socio-economic stability all over the world. The increasing resistance to the current antimicrobial treatment has resulted in the crucial need for the discovery and development of novel entities for the infectious treatment with different modes of action that could target both sensitive and resistant strains. METHODS Compounds were synthesized using the classical organic chemistry methods. Prediction of biological activity spectra was carried out using PASS and PASS-based web applications. Pharmacophore modeling in LigandScout software was used for quantitative modeling of the antibacterial activity. Antimicrobial activity was evaluated using the microdilution method. AutoDock 4.2® software was used to elucidate probable bacterial and fungal molecular targets of the studied compounds. RESULTS All compounds exhibited better antibacterial potency than ampicillin against all bacteria tested. Three compounds were tested against resistant strains MRSA, P. aeruginosa and E. coli and were found to be more potent than MRSA than reference drugs. All compounds demonstrated a higher degree of antifungal activity than the reference drugs bifonazole (6-17-fold) and ketoconazole (13-52-fold). Three of the most active compounds could be considered for further development of the new, more potent antimicrobial agents. CONCLUSION Compounds 5b (Z)-3-(3-hydroxyphenyl)-5-((1-methyl-1H-indol-3-yl)methylene)-2-thioxothiazolidin-4-one and 5g (Z)-3-[5-(1H-Indol-3-ylmethylene)-4-oxo-2-thioxo-thiazolidin-3-yl]-benzoic acid as well as 5h (Z)-3-(5-((5-methoxy-1H-indol-3-yl)methylene)-4-oxo-2-thioxothiazolidin-3-yl)benzoic acid can be considered as lead compounds for further development of more potent and safe antibacterial and antifungal agents.

Individual stereoisomers of verbenol and verbenone express bioactive features

AB  - Naturally occurring terpene core compounds have been used extensively in both pharmaceutical and cosmetic industry. However, since chirality of these compounds has profound influence on the level of their bioactivity, the aim of the present study was to assess broad spectrum activities of four possible stereoisomers of monoterpene alcohol verbenol and both enantiomers of monoterpene ketone verbenone. Antimicrobial activity was evaluated against different bacterial and fungal strains of clinical relevance. Tested compounds exhibited antibacterial activity within MICs range of 0.6–2.5 mg mL−1, antifungal activity within MICs range of 0.08–0.6 mg mL−1, and anticandidal activity with MICs 0.3–1.2 mg mL−1. While there was no difference in antibacterial activity between the enantiomers of verbenone, this was not the case with tested verbenol enantiomers. Furthermore, the compounds expressed no cytotoxicity to human keratinocyte cell line HaCaT at the tested concentrations. Interestingly, compounds contributed to the downregulation of interleukin IL-6 levels in HaCaT cells exposed to bacterial agent Staphylococcus lugdunensis. According to our knowledge, this study is the first that comparatively describes biological activities of stereoisomers of verbenol and enantiomers of verbenone. Current findings highlighted the importance of stereochemical purity of monoterpenoids in relation to their unique biological features.

Selected terpenoids as obstructers of Candida virulence

AB  - Terpenoids are the most complex class of natural products with over 80000 identified representatives found in plants, microorganisms, insects, and marine invertebrates. Their anticandidal potential, especially impact on fungal virulence is, however, not fully explored yet. We aimed to investigate anticandidal, antibiofilm, and antihyphal properties of selected terpenoids (borneol, thymol, geraniol, and carvacrol). The strongest antifungal potential towards Candida albicans strains could be noticed for borneol (lowest minimal inhibitory concentration, MIC, 0.06 mg/mL) followed by thymol (lowest MIC 0.12 mg/mL). On the other hand, MIC of geraniol was up to 8 mg/mL. Non-albicans Candida strains were most susceptible to thymol (MIC 0.12 mg/mL towards C. krusei H1/16) and borneol (MIC 0.12 mg/mL towards C. parapsilosis ATCC 22019). When the terpenoids were applied in their previously determined minimal inhibitory concentrations the strongest reducing impact on C. albicans ATCC 10231 biofilm forming ability was observed for carvacrol (60% inhibition), followed by geraniol (56% inhibition). Lower concentration of terpenoids (0.5 MIC) led to similar impairment of biofilm formation – the strongest impact was noticed for geraniol (54% inhibition), followed by carvacrol (47% inhibition) as determined by crystal violet assay. MTT antibiofilm assay highlighted great potential of carvacrol and geraniol to reduce viability of Candida cells in biofilms. Inhibition of hyphal formation could be noticed only upon application of carvacrol (14.29% of hyphal cells compared to the untreated control with 50.67% hyphae) suggesting wide antivirulence potential exhibited by this molecule. Other compounds explored in this study have displayed high MIC values (geraniol), low antibiofilm potential (thymol, borneol) or lack of antihyphal properties (all except carvacrol). Potential of carvacrol to reduce fungal virulence should be further studied on molecular level and in in vivo assays.
