AB  - In traditional medicine, the Mediterranean plant Vitex after the last injection. Pituitary glands were excised, agnus-castus L., Familia Verbenacee (Vac) has been weighed and processed for the stereological analysis. used since ancient times to alleviate the symptoms of ACTH-, TSH- and PRL-producing cells were identified with aging. We have already shown that the essential oil of polyclonal rabbit antisera (ACTH, TSHß, PRL respectively).

AB  - Andropause, the culminating phase of male ageing, is characterized by deregulation of the hypothalamic-pituitarygonadal axis and low circulating free testosterone. The aim of this study was to investigate the immunohistomorphometric characteristics of the pituitary gonadotropic i.e. follicle-stimulating hormone (FSH) and the luteinizing hormone (LH) producing cells after testosterone application in a rat model of the andropause. Middle-aged Wistar rats were divided into orchidectomized (ORX; n=8) and testosterone treated orchidectomized (ORX+T; n=8) groups. Testosterone propionate (5 mg/kg b.m. /day) was administered for three weeks, while the ORX group received the vehicle alone. Immunohistochemically stained FSH and LH cells underwent morphometric and optical density-related analysis, while circulating concentrations of the sex steroids were measured by immunoassays. Serum concentrations of testosterone and estradiol were significantly (p<0.05) increased by 24 and 2.7 fold respectively, compared to the ORX group. The volume of FSH and LH cells was significantly (p<0.05) decreased by 51.3% and 56.6% respectively, in comparison with ORX rats. Relative volume density of FSH and LH cells was also significantly (p<0.05) decreased by 54.0% and 72.8% respectively, compared to the ORX group. Results related to the optical density of gonadotropic cells (reflecting their hormonal content) were in line with the morphometric findings i.e. this parameter of FSH and LH cells was significantly (p<0.05) decreased by 25.7% and 16.2% respectively, in comparison with ORX rats. Conclusion: In conclusion, applied testosterone increased the serum concentrations of sex steroids, as well as it decreased morphometric parameters and optical density of gonadotropic cells in ORX rats.

Genistein Increase Thyroid Hormone and 7 alpha-hydroxycholesterol concentrations in the liver of middle-aged male rats

AB  - Obesity, a major public health problem, is associated with hypercholesterolemia and other metabolic disorders, which are potent risk factors for athero-thrombotic disease, a leading cause of mortality worldwide. Advances in both dietary and pharmacological interventions contribute significantly to prevention and treatment of modifiable risk factors. Consumption of purified soybean isoflavones was reported to reduce plasma and liver cholesterol levels. The precise mechanism has not been established, but it has been suggested that these effects occur through an increase in bile acid excretion. The initial and rate limiting step in the classical pathway of hepatic synthesis of bile acids from cholesterol is the enzymatic addition of a 7α hydroxyl group by cholesterol 7α-hydroxylase forming 7α-hydroxycholesterol. Thyroid hormones have been shown to up-regulate expression of this enzyme. We previously reported that subcutaneously administered genistein impaired thyroid functioning in middle-aged rats, but at the same time increased expression of T3-activated genes and increased deiodinase type 1 enzyme activity in the liver. In this study we aimed to further determine effects of genistein on concentrations of thyroid hormones, total cholesterol and 7α-Hydroxycholesterol in the liver. Thirteen-month-old male Wistar rats were injected subcutaneously with 35 mg/kg of genistein, while controls received vehicle alone daily during 4 weeks. For determination of iodothyronines liquid chromatography-mass spectrometry was applied, while total cholesterol and 7α-hydroxycholesterol were determined by gas chromatography/mass spectrometry. Our study results clearly demonstrate that genistein increased (P<0.05) concentration of T4 and T3 in the liver of middle-aged male rats. In line with this, concentration of 7α-hydroxycholesterol also increased (P<0.05), but the total cholesterol levels remained unchanged. In conclusion, genistein increased hepatic availability of thyroid hormones, in accordance with detected increase in 7α-hydroxycholesterol level in the liver. The obtained data brought new light on the mechanisms involved in biological processes in the liver induced by genistein.

Influence of Increased Incubation Temperature on Breast Muscle Development of Broilers After Hatching

AB  - One of the goals ofrnodern poultry production is improving the quality and quantity of
broiler meat. Recent research is directed to find the relation between incubation factors and
muscle development, which can affect on meat characteristics. The aim of this study was
to examine the influence of increased incubation temperature on development of breast
muscles of broilers in days after hatching. Eight hundred broiler eggs (Ross 308) were
divided in four equal groups (200 per each group) and placed in four incubators. Group
A represents control group with incubation temperature 378°C. In groups B, C, D during
the 16‘“, 17‘“ and 18th day of incubation temperature was increased to 38.5°C, 395°C and
41°C reSpectively, for 3 hours each day. On the 7‘1‘, as well as on the 14‘“ day after hatching,
10 chickens from each group were sacrificed. Samples of breast muscle (M pectoralis
superficialis) were taken for histological examination of diameter of muscle cells, volume
density of connective tissue in the muscle and nucleo-cytoplasmic ratio of muscle cells.
Measurements showed that diameter of breast muscle cells on 7L11 day after hatching in
treated groups was decreased in average by 6.36pm compared to control group, while on
 14m day after hatching in group in which incubation temperature was increased to 41°C,
it was decreased in average by 5.84pm compared to other groups. In all groups, on 14‘h
day after hatching, diameter of breast muscle cells was increased in average by 6.71pm,
while nucleo—cytoplasmic ratio of breast muscle cells was decreased in average by 0.035,
compared to 7”h day after hatching. Lower diameter of breast muscle cells in treated groups
on 7‘“ day after hatching can be explained by higher proliferation of myoblasts due to
applied treatments. During the proliferation, growth of cells is reduced, which is reflected
on lower diameter of muscle cells on 7"1 day after hatching. It can be assumed that increased
temperature during incubation affect on increased growth of muscle cells in postnatal
period of development, which is in agreement with approximately equal diameters on 14*
day after hatching between control group and groups in which incubation temperature was
increased to 385°C and 395°C. It seems that increased temperature on 41"C had negative
effect on growth and development of muscle cells, which results in decreased diameter of
breast muscle cells on 14‘h day afier hatching in group which incubation temperature was
increased to 41°C.

AB  - Diosgenin is bioactive, steroidal saponin from Dioscorea viliosa (Wild Yam) and appears to be novel promising substance for the treatment of menopausal symptoms. In addition, diosgenin may manipulate the endogenous steroid hormone levels and may affect the numerous oxidative stress pathways/molecules. Since decline in the steroid hormone levels as well as increase in oxidative stress in the ovaries play an integral part of the menopause, this study aimed to investigate the effects of diosgenin on production of estradiol and progesterone, as well as to investigated the effects of diosgenin on the expression of genes encoding the antioxidant enzymes (Cu, Zn-superoxide dismutase (CuZnSOD), Mn-superoxide dismutase (MnSOD), glutathione peroxidase (GPX), and catalase (CAD) in ovary of acyclic-middle aged female rats. Middle-aged (l3-months-old) acyclic female Wistar rats in constant diestrus stage, which represent tractable model to human menopause, were divided into control and diosgenin treated groups.Diosgenin (100 mgfl<g b.w.lday) was orally applied for 4 weeks, while C group received the vehicle alone. In our experimental model, diosgenin administration to acyclic middle-aged rats significantly increased estradiol and progesterone serum concentrations when compared to the corresponding control. Gene expression analysis revealed that diosgenin significantly down-regulate all examined enzymes involved in antioxidative defense. The down-regulation were especially pronounced in CuZnSOD and CAT, indicating that the diosgenin may decrease the pool of reactive oxygen species in the ovary directly due and may act as free radical scavengers. Therefore, our results indicate beneficial potential of diosgenin in the middle-aged rat ovary.

AB  - Aging is a complex process characterized by a progressive decline in cellular
function, alterations of the endocrine system and the increased incidence of
thyroid diseases. Citrus flavanones naringenin (NAR) and hesperetin (HES) may
contribute to maintenance of health at old age however they may also affect
thyroid hormone economy. The aim of this study was to test weather NAR or HES
administration alter thyroid functioning. NAR or HES were administrated orally
(15 mg/kg) to male 24-month-old Wistar rats during 4 weeks. Control groups
received vehicle, sunflower oil. Immunohistochemical staining of thyroglobulin
(Tg), thyroxine (T4) bound to Tg (T4-Tg) and sodium iodide sysmporter (NIS)
were performed. Immunopositivity of Tg or T4-Tg in thyroid follicles were
measured as intensity of optical density (OD) or relative intensity of fluorescence
(RIF). Thyroid stimulating hormone (TSH) and total T4 concentrations were
measured in serum. After citrus flavanones treatments, thyroids showed Tg and
T4-Tg immunopositivity mainly in the colloid of the follicles. The obtained
results showed increased (P ! 0.05) intensity of OD for Tg and T4-Tg RIF after
both flavanones treatments, while NIS expression did not change. Serum TSH
increased (P ! 0.05) only after NAR, although T4 remained unchanged after both
treatments. In conclusion, citrus flavanones induced stronger expression of
thyroid-specific proteins in gland of old-aged males. However, only NAR
elevated serum TSH, being more potent than HES. This alteration is in line with
preserved capacity of the thyroid to compensate the mild interference of
flavanones treatments with thyroid hormone production in old-aged males.

AB  - Calcitonin (CT) is a hypocalcemic hormone produced by thyroid C-cells that acts as an bone antiresorptive agent. The other endocrine cell population in thyroid, called follicular cells produce thyroid hormones (TH) which also affect bone turnover. In this study, we evaluated the effects of salmon CT administration on structure and function of both CT and TH producing thyroid cells as well as trabecular bone microarchitecture in orchidectomized (Orx) middle-aged rats. Fifteen-month-old male Wistar rats were either Orx or sham-operated (SO). One group of Orx animals were injected subcutaneously with synthetic salmon CT (Orx + CT; 100 IU/kg b.w.) every second day for 6 weeks. The rats from SO and second Orx group received the same volume of vehicle alone by the same schedule. The peroxidase–antiperoxidase method was applied for localization of CT in C-cells. CT-immunopositive thyroid C-cells, thyroid follicular epithelium, interstitium and colloid were evaluated morphometrically. An ImageJ public domain image processing program was used to measure bone histomorphometric parameters of the proximal tibial specimens. Blood serum samples were analyzed for CT, osteocalcin (OC) and thyroxine (T4), and urine samples fot calcium (Ca2+) concentration. We found a significant decrease in the Vc and Vv of thyroid C-cells after CT treatment compared to both SO and Orx. The Vv of the colloid was higher, while VV of the follicular epithelium was lower after CT treatment compared to Orx. Analysis of trabecular microarchitecture showed that salmon CT administration significantly increases of cancellous bone area (B.Ar), trabecular thickness (Tb.Th), and trabecular number (Tb.N) whereas trabecular separation (Tb.Sp) was significantly decreased. CT treatment markedly elevated serum CT, but serum OC, T4 and urinary Ca2+ concentrations were lower than in the Orx group. These findings indicate that administration of salmon CT inhibited calcitonin-producing thyroid C-cells and changes the structure of the thyroid gland indicating hypoactivity.

AB  - Objective: Estrogen replacement therapy remains current as a therapeutic approach to treat menopausal symptoms and may significantly affect hormone-producing cells in the female pituitaries. The aim of this study was to examine the histological parameters of pituitary mammotrophs and prolactin secretion after chronic estradiol treatment in ovariectomized adult female rats, reflecting premature menopause. Materials and Methods: In this experimental study, adult female Wistar rats were divided into non-ovariectomized (C), ovariectomized (OVX) and estradiol-treated ovariectomized (OVX+E) groups. Estradiol dipropionate [0.625 mg/kg body mass per day] was administered for four weeks, while the C and OVX groups received vehicle alone. Mammotrophs were identified by the peroxidase-antiperoxidase (PAP) immunohistochemical procedure, while prolactin concentrations were measured by the non-isotopic two-step assay (Delfia) method. Comparison of the differences between groups was performed using one-way analysis of variance (ANOVA) and Tukay (honest significant difference) HSD test. Results: Ovariectomy caused significant (P < 0.05) decreases in mammotroph optical density (OD), volume density (V V ) and number per mm2 by 29, 27 and 34%, respectively, in comparison with the C females. In the OVX+E group, significant (P < 0.05) increases in OD, cell volume, V V and number of mammotrophs per mm2 by 181, 15%, 5.8-fold and 5.2-fold, respectively, were observed when compared to OVX animals. The serum prolactin concentration in OVX females was significantly (P < 0.05) decreased by 14% in comparison to the C group, while in OVX+E females, prolactin levels were significantly (P < 0.05) increased by 53% compared to the OVX controls. Conclusion: Estradiol supplementation in ovariectomized females is followed by stimulatory histological and secretory changes of the mammotrophs. These results could serve as indicators of possible prolactinome development upon estradiol application in premature menopausal subjects.

AB  - Elevated blood level of vitamin D is associated with multiple health benefits. However, vitamin D deficiency is common among the elderly population. Despite multiple cellular targets in both adjacent thyroid C cells and chief parathyroid cells, little is known about how vitamin D affects thyroid follicles and their function in thyroid hormone production. In this study we administered 50mg of vitamin D3 (that corresponds to 2000IU/daily), during three weeks, to orchidectomized middle-aged male rats, which we used as a model of andropause with osteoporosis. After animals’ decapitation, thyroid tissues were histologically analyzed and serum concentrations of total T4 and TSH were determined by the corresponding rat ELISAs. In comparison with the controls, which received the same amount of the sterile olive oil, thyroids of Vit.D3-treated rats were characterized by interstitial C cell hyperplasia, while the follicular tissue remained preserved.

AB  - Introduction: Genistein is widely used as an alternative therapeutic for various pathological conditions in ageing male and exerts an inhibitory effect on the steroidogenic enzyme families, thus affecting hormonal homeostasis. The aim of this study was to examine the effects of genistein on histological features of the adrenal cortex zona reticularis and the following dehydroepiandrosterone (DHEA) secretion in orchidectomized adult male rats, the most accessible model reflecting andropause.
Material and Methods: The experimental adult Wistar male rats were divided into three groups (n=7): sham-operated (SO), orchidectomized (Orx) and genistein-treated orchidectomized (Orx+G) groups. Genistein (30 mg/kg b.m./day) was administered subcutaneously for three weeks, while the control groups received the vehicle alone. All animal were sacrificed by decapitation 24h after the last injection. The adrenal cortex was analyzed histologically, while serum concentration of DHEA was determined by immunoassay.
Results: Histological analysis revealed that there were no significant changes in adrenal cortex zona reticularis after orchidectomy and orchidectomy followed with genistein treatment. Circulating DHEA concentration was significantly (p<0.005) increased by 97.2% in Orx animals compared to the SO group. In Orx+G group serum DHEA was significantly (p<0.005) decreased by 41.5% in comparison with Orx rats. Serum concentration of DHEA upon genistein application to orchidectomized animals was insignificantly (p>0.05) changed by 15.3% when compared to the SO group.
Conclusion: These data, observed in orchidectomized adult rats, indicate that genistein has an inhibitory effect on the adrenal cortex zona reticularis secretory function.

AB  - Morphological characteristics of skeletal muscles of slow- and medium-growing strains of chickens are very important for meat quality and comparison with fast-growing strains. The aim of this paper was to evaluate morphological parameters of breast and thigh muscles of slow- and mediumgrowing strains in a free-range system. The slow-growing strains used in the experiment were autochthonous breeds Sombor crested and Banat naked neck, and the medium-growing strain was Red-bro. The tissue samples were taken from the thigh muscle and muscles of the breast of 10 chickens of each breed. Samples were stained with hematoxylin - eosin and enzyme succinate - dehydrogenase (SDH). The following morphological parameters were observed: diameter of muscle cells, nucleo-cytoplasmic ratio of muscle cells, volume density of connective tissue within the muscle and the presence of red, white and intermediate muscle cell types. Between strains, the type of muscle or genotype didn’t have significant effects on diameters of muscle cells and nucleo-cytoplasmic ratio in the muscle cells. Results indicated that genotype had significant effect on volume density of the connective tissue in breast muscles. Red muscle cells were, in all strains, significantly more represented in m. biceps femoris than m. pectoralis superficialis. Genotype had significant effect on ratio between connective tissue and muscle cells and no significant effects on other morphological parameters. [Projekat Ministarstva nauke Republike Srbije, br. AB  - Estradiol and human chorionic gonadotropin (hCG) are very important in controlling the secretory activity of hormone producing cells in the female rat pituitary glands. The aim of the present study was to examine the morphometric parameters of immunohistochemically la be led ACTH cells in juvenile (16th day) female rat pituitaries after treatment with five doses of estradiol dipropionate (EDP) and two doses of hCG during the neonatal period of life. The controls were treated on the same schedule with an equivalent volume of vehicle. All animals were sacrificed 24 h after the last treatment. ACTH-producing cells were studied using the peroxidase-antiperoxidase immunohistochemical procedure. The absolute and relative pituitary weights were increased (p<0.05) only in the EDP treated group by 120.0% and by 121.1% respectively, in comparison with the controls. In this group, the volume of ACTH cells, volume of their nuclei and volume density were significantly decreased (p<0.05) by 6.4%, 33.3% and 46.2% respectively, compared to the corresponding controls. After treatment with hCG, there were no significant (p>0.05) changes neither in the volume of ACTH cells nor in the volume of their nuclei, in comparison with the controls. On the basis of the results obtained in our study, it can be concluded that EDP, injected into female rats during the neonatal period of life, has an inhibitory effect on the immunohistomorphometric parameters of ACTH cells, but such an effect is not clearly expressed after treatment with hCG.
AB  - Estradiol i humani horionski gonadotropin (hCG) veoma su važni u kontroli sekretorne aktivnosti ćelija hipofize koje proizvode hormone. Cilj ove studije je bio ispitati morfometrijske parametre imunohistohemijski obeleženih ACTH ćelija hipofize ženki pacova, u juvenilnom periodu (16. dan), nakon tretmana sa pet doza estradiol-dipropionata (EDP) ili dve doze humanog horionskog gonadotropina (hCG). Kontrole su bile tretirane na istovetan način sa odgovarajućom količinom rastvarača. Sve životinje su bile žrtvovane 24 h nakon poslednjeg tretmana. ACTH ćelije su bile imunohistohemijski obeležene metodom peroksidaza-antiperoksidaza (PAP). Dobijeni rezultati ukazuju na značajno povećanje (p<0,05) apsolutne i relativne mase hipofize za 120% i 121,1% kod ženki tretiranih sa EDP u odnosu na kontrolu. U ovoj grupi ženki pacova volumen ACTH ćelija i njihovih jedara, kao i njihova volumenska gustina su bili značajno smanjeni (p<0,05) za 6,4%, 33,3% i 46,2% u poređenju sa kontrolom. Nakon tretmana hCG-om, volumen ACTH ćelija, kao i volumen njihovih jedara, nisu bili značajno (p>0,05) promenjeni u odnosu na kontrolu. Na osnovu dobijenih rezultata možemo zaključiti da EDP, injeciran ženkama pacova tokom neonatalnog perioda života, ima inhibitorni efekat na imuno histomorfometrijske parametre ACTH će lija, što nije tako jasno izraženo nakon tretmana hCG-om.
