AB  - The effects of chronic treatments with either SRIH-14 or octreotide on the adrenal medulla of male Wistar rats were examined. Adult males received subcutaneous (s.c.) injections of 20 μg/100 g body weight of either SRIH-14 or octreotide twice a day for 28 consecutive days. The absolute weights and the absolute volumes of the adrenal glands significantly (p<0.05) decreased after either treatment. The adrenal medulla was analyzed by histological and stereological methods using newCAST. Compared to the control, the relative volumes of the vascular tissues significantly (p < 0.05) decreased - by 40% and 25% in the SRIH-14- and octreotide-treated groups, respectively. In the SRIH-14- and octreotide-treated groups the relative volumes of chromaffin and interstitial tissue increased by 6% and 5% (p < 0.05), respectively. These findings show that both SRIH-14 and octreotide affect the morphological characteristics of the adrenal zona medullaris in a similar manner.

AB  - Glucocorticoids and thyroid hormones control many aspects of fetal development. AB  - Glukokortikoidi i tiroidni hormoni kontrolišu brojne aspekte razvoja fetusa. Koristeći imunocitohemijui stereološke metode ispitivali smo efekte primene deksametazona (Dks) tokom gestacije na TSH ćelije hipofize fetusa pacova starih 19 dana. Doze od 1.0, 0.5 i 0.5 mg Dks/kg tm/dan primenjivane su gravidnim ženkama tri uzastopna dana, počev od 16. dana gestacije. Tretman Dks kod gravidnih ženki uzrokuje značajno smanjenje broja TSH ćelija fetusa po jedinici površine i njihove volumenske gustine u poređenju sa odgovarajućim kontrolama. Naši rezultati pokazuju da maternalna primena Dks inhibira multiplikaciju TSH ćelija kod fetusa starih 19 dana.

AB  - Neonatal female rats (3-7 days old) were injected subcutaneously twice daily with 20 μg of SRIH-14/100g b.w. for five consecutive days. The animals were sacrificed in the peripubertal (38 days old) or adult (80 days old) periods of life. Control rats were treated in the same way with identical volumes of saline. SRIH-14 led to significant reduction (p<0.05) of the absolute and relative volumes of the ZG in peripubertal animals, by 11 and 22%, respectively in comparison with the controls. The total number and volume of ZG cells and their nuclei were also significantly decreased (p<0.05) in peripubertal SRIH-14-treated rats as compared to the controls. There were no significant (p>0.05) changes in the morphometric parameters of adult adrenal cortex after SRIH-14 treatment compared with control females. It can be concluded that subcutaneously applied SRIH-14 in the neonatal period inhibits growth of adrenal ZG cells in female rats only up to the peripubertal stage, while the adrenal gland cortex recovers before mature adulthood is reached.
AB  - Ženke pacova od 3-7. dana starosti (neonatalni period) su subkutano tretirane dva puta dnevno sa SRIH-14 u dozi od 20 μg na 100 g telesne mase tokom pet uzastopnih dana. Životinje su bile žrtvovane u peripubertalnom (38. dana) ili odraslom (80. dana) periodu života. Kontrolne grupe su na identičan način tretirane fiziološkim rastvorom. U poređenju sa kontrolnim vrednostima tretman sa SRIH-14 je značajno smanjio (p<0.05) apsolutni (-11%) i relativni (-22%) volumenzone glomeruloze (ZG) u peripubertalnih životinja. Ukupan broj i zapremina ćelija ZG i njihovih jedara takođe su značajno smanjeni (p<0.05) u pacova tretiranih sa SRIH-14 u peripubertalnom periodu života u poređenju sa kontrolnim ženkama. U kori nadbubrežne žlezde odraslih ženki pacova nisu zapažene (p>0.05) promene morfometrijskih parametrima posle SRIH-14 tretmana u poređenju sa odgovarajućom kontrolom. Na osnovu dobijenih rezultata možemo zaključiti da je subkutano tretiranje ženki pacova u neonatalnom periodu sa SRIH-14 inhibiralo rast zone glomeruloze u peripubertalnih pacova.

AB  - The effects of multiple somatostatin (SRIH-14) administration on the pituitary–ovarian axis were examined in peripubertal rats. Female Wistar rats received subcutaneously, two daily doses of 20 μg SRIH-14 per 100 g body weight (b.w.) for five consecutive days (from the 33rd to the 37th day of life). Follicle-stimulating (FSH), luteinizing (LH) and somatotropic (GH) cells were examined by the peroxidase–anti-peroxidase immunocytochemical method. Changes in cell volumes, volume densities and number per unit area (mm2) of FSH-, LH- and GH-immunoreactive cells were evaluated by stereology and morphometry. Serum FSH and LH levels were determined by RIA. Ovaries were analyzed by simple point counting of follicles. The volumes and volume densities of FSH-, LH- and GH-immunoreactive cells were significantly decreased while their numbers per mm2 remained unchanged. SRIH-14 induced a significant decrease in serum FSH and LH levels. In the ovary, SRIH-14 induced an increase in the number of primordial follicles, followed by a reduction in the number of small healthy growing follicles and absence of preovulatory follicles. The number of atretic follicles was unchanged. We concluded that treatment with SRIH-14 during the peripubertal period markedly inhibited pituitary FSH, LH and GH cells. In the ovary, SRIH-14 acted by inhibiting folliculogenesis without affecting atretic processes.

AB  - The effects of estradiol dipropionate (EDP) or calcium glucoheptonate (Ca) on the morphology and stereology of the PRL cells in 14-month-oldWistar female rats were studied. The animals were treated daily with EDP in the dose of 0.625 mg/kg b.w. or calcium glucoheptonate (Ca; 11.4 mg/kg b.w.) for two weeks. The controls were injected with vehicle alone by the same schedule. Mammotrophs (PRL cells) were immunocytochemically localized by the PAP method. Serum concentration of PRL was significantly increased (p<0.05) by 17% after estradiol treatment, but in Ca-treated females this parameter was insignificantly (p>0.05) changed by 2% compared to controls. Based on these results, it can be concluded that EDP expresses a strong stimulatory effect on the morphology and function of pituitary PRL cells.

Aciklične ženke su svakodnevno tokom dve nedelje dobijale 0,625 mg/kg tm EDP-a ili 11,4 mg/kg tm Ca. PRL ćelije su imunocitohemijski obeležavane PAP metodom. AB  - This study was to evaluate the effect of androgen deficiency on thyroid immunoreactive C-cells and bone structure and function in a male orchidectomized middle-aged rat model. Fifteen-month-old male Wistar rats were divided into orchidectomized (Orx) and the sham-operated control (Sham) group. In the Orx group significant decreases (P < 0.05) were found in the volume of C cells (by 14%), their relative volume density (by 13%) and serum calcitonin concentration (by 54%) compared to the controls. Analyses of trabecular microarchitecture of the proximal tibia metaphysis showed that Orx induced marked decreases of cancellous bone area, trabecular thickness and trabecular number (by 52, 20 and 19% respectively; P < 0.05), whereas trabecular separation was increased by 27% (P < 0.05). In Orx rats, serum osteocalcin concentration was increased by 119% (P < 0.05), while serum calcium and phosphorus were 6 and 14% (P < 0.05) lower, respectively, compared to the levels in the Sham. In addition, urine calcium content was considerably higher (by 129%; P < 0.05) in Orx animals. These findings indicate that the androgen deficiency caused by Orx in middle-aged rats modulated the structure of C cells and diminished secretion of calcitonin. Histomorphometrical and biochemical analyses demonstrated a decrease of cancellous bone mass and increased bone turnover.

AB  - Nutritional supplements containing soybean phytoestrogens, the isoflavones genistein (G) and daidzein (D), are increasingly used as alternative therapy for osteoporosis, cancer, and cardiovascular and other diseases with a frequency that increases with advancing age. In this study we examined the effects of subcutaneous administration of either G or D on serum lipid levels in orchidectomized (Orx) and intact (IA) middle-aged male rats, which are experimental models of andropause. Sixteen-month-old Wistar rats were treated with 10 mg/kg and 30 mg/kg of either G or D. The control groups received testosterone, estradiol, or vehicle for 3 weeks, after which the total serum cholesterol (TC), low-density lipoprotein (LDL-C), high-density lipoprotein (HDL-C), and total triglycerides (TT) were measured. Compared with the matching vehicle-treated controls, the higher doses of G and D and testosterone treatment significantly (P < 0.05) lowered the TC and lipoprotein cholesterol levels. The greatest effect was observed regarding LDL-C in both Orx and IA males after G and D treatments, in which LDL-C decreased by more than 30%. The lower isoflavone doses induced a significant cholesterol-lowering effect (P < 0.05) only in the Orx group. Like the estradiol treatment, the higher doses of G and D increased the TT levels in both rat models by more than 50% (P < 0.05). The lower doses of isoflavones increased TT only in the Orx group. In male middle-aged rats, injections of higher doses of G and D decreased the serum cholesterol levels, as did testosterone injection, and brought about an increase in serum triglycerides similar to that observed after estradiol treatment.

AB  - This study describes the development of ACTH and GH cells in 19- and 21-day-old rat fetuses using immunohistochemistry and morphometric measurements. Between days 19 and 21 of pregnancy, the total volume of fetal ACTH cells was unchanged, while their volume density and number per unit of area decreased significantly. ACTH-like immunopositivity in the pars intermedia increased during the examined period. The cell volume, volume density and number of GH cells per unit of area all markedly increased in parallel with fetal development, i.e., from gestational days 19 to 21. GH-like immunopositivity is demonstrated in the pars intermedia of 21-day-old fetuses for the first time.
AB  - Prezentovano istraživanje opisuje razvoj ACTH i GH ćelija hipofize fetusa pacova, neposredno pred rođenje korišćenjem imunohistohemije i morfometrijskih merenja. Od 19. do 21. dana gestacije volumen ACTH ćelija fetusa bio je nepromenjen, dok su volumenska gustina i broj ćelija po jedinici površine značajno smanjeni. Intenzitet ACTH imunopozitivnosti u pars intermedia povećavn je tokom ispitivanog perioda. Volumen GH ćelija, volumenska gustina i brojnost po jedinici površine značajno su povećani tokom završnog perioda fetalnog razvoja, tj. od 19. do 21. dana gestacije. GH imunopozitivnost prvi put je demonstrirana u ćelijama pars intermedia kod fetusa starih 21 dan.

AB  - This study examined the effects of intracerebroventricularly (i.c.v.) administered somatostatin (SRIH-14 orSRIH-28) on growth and function of pituitary somatotropes (GH cells). Female rats were subjected to intracerebroventricular administration of three 1g doses of SRIH-14 or SRIH-28 every second day. Blood samples were collected for hormone analyses and pituitaries were removed for histological and morphometric evaluation, five days after the last injection. Compared to control animals, SRIH treatment decreased (p<0.05) all morphometric measurements obtained for GH cells. The concentration of serum growth hormone (GH) in SRIH-14- or SRIH-28-treated groups was lower (p<0.05) by 69% and by 61% respectively, in comparison to control rats. These findings suggest that centrally administered somatostatin is specifically involved in the control of growth and secretory activity of GH cells. Thus, pharmacological manipulation of SRIH receptors reached from cerebrospinal fluid may alter the systemic effects of GH.
AB  - Ispitivani su efekti intracerebroventrikularno (i.c.v.) ubrizganih somatostat-ina (SRIH-14 ili SRIH-28) na rast i funkciju somatotropnih ćelija hipofize ženki pacova. Sve eksperimentalne životinje su primale po 1μg SRIH-14 ili SRIH-28 rastvorenog u 5μL fiziološkog rastvora, svaki drugi dan. Kontrole su na isti način injecirane fiziološkim rastvorom. Životinje su žrtvovane petog dana po poslednjoj primljenoj dozi. Krv je sakupljana za određivanje koncentracije hormona rasta (GH) u serumu, a hipofize su pripremane za histološku i morfometrijsku analizu. Dobijeni rezultati pokazuju da SRIH tretirane životinje pokazuju značajno smanjenje (p<0.05) svih ispitivanih morfometrijskih parametara (volumen ćelija i jedara, volumenska gustina) u odnosu na kontrolu. Koncentracija GH u serumu SRIH-14 i SRIF-28 tretiranih ženki takodje je značajno smanjena(p<0.05)za69%odnosno 61% u poređenju sa kontrolnim ženkama. Na osnovu ovako dobijenih rezultata može se zaključiti da oba aplikovana somatostatina inhibiraju rast i sekretornu aktivnost somatotropnih ćelija hipofize.

The controls received an equivalent volume of saline in the same manner according to the same schedule. The pituitary glands were excised and used for immunohistochemical and morphometric evaluation. The results obtained demonstrated a somatostatin-related decrease in relative pituitary weight. The morphometric analyzes showed decreased volumetric density of TSH-immunoreactive cells in SRIH-treated groups, but this difference was more expressed in SRIH-14- treated animals than in those receiving SRIH-28. On the basis of these results it can be concluded that centrally administered somatostatins inhibit TSH cells.
AB  - Ispitivani su efekti intracerebroventrikularno (i.c.v.) injiciranih sintetičkih somatostatina (SRIH-14 i SRIH-28) na hipofizne TSH ćelije adultnih ženki pacova. Životinje su i.c.v. injicirane satri doze od po 1 μg SRIH-14 ili -28 rastvorenog u 5 pi fiziološkog rastvora svaki drugi dan, a žrtvovane 5 dana posle poslednje injekcije. Kontrole su tretirane odgovarajućom dozom fiziološkog rastvora. Hipofize su izolovane i pripremljene za imunocitohemijska i morfometrijska ispitivanja. Oba somatostatina izazivaju smanjenje relativne težine hipofize. Morfometrijska analiza pokazuje da je u obe tretirane grupe životinja volumenska gustina TSH-imunoreaktivnih ćelija smanjena te razlike su značajnije izražene u grupi životinja tretiranih sa SRIH-14 u odnosu na one tretirane sa SRIH-28.
