AB  - Background: A rapid increase in the turnover of malignant cells modulates the enzymes level in circulation and may be a prognostic sign of disease progression. We evaluated the activity of serum lactate dehydrogenase and superoxide dismutase in erythrocytes of breast cancer patients immediately after surgery and before adjuvant therapy. Methods: Fourteen newly operated breast cancer patients were included in the study. Lactate dehydrogenase and superoxide dismutase activity were estimated using standard tests for evaluation of enzyme activity. Results: Activity of lactate dehydrogenase was in normal range in all but one sample obtained from patient at clinical status IIIB. Superoxide dismutase activity was elevated in 11 out of 14 patients, and was 2 to 8 fold higher compare to control values. The highest activity of superoxide dismutase was found in samples of two patients at clinical status I. Conclusion: Determination of lactate dehydrogenase and superoxide dismutase activity might be useful in clinical follow up of breast cancer patients.

AB  - Cell culture K562 samples were treated with fullerenol (C6o(OH)24) at a concentration of 10 nmol/mL and thereafter irradiated with X-rays (24Gy). The activity of gamma-glutamyltransfrease (γ-GT), total superoxide-dismutase (SOD) and glutathion-peroxidase (GSH-Px) was determined 1, 24 and 48 hours after irradiation. Irradiation induces an increase in the activity of all the investigated enzymes. Fullerenol in the applied dose decreased the γ-GT activity 24 and 48 h after irradiation. The total SOD activity is increased in both pretreated groups except in the irradiated group at the 48th hour. Treatment with fullerenol before irradiation increased GSH-Px activity in irradiated groups and decreased it in the control groups.
AB  - U uzorcima ćelijske kulture humane eritroleukemije (K562) ozračenim X-zracima doze 24 Gy i predtretiranim fulerenolom (C60(OH)24) koncentracije 10 nmol/mL određivana je aktivnost gama-glutamiltransferaze (γ-GT), ukupne superoksid-dismutaze (SOD) i glutation-peroksidaze (GSH-Px) 1, 24 i 48 sati nakon zračenja. Iradijacija povećava aktivnost sva tri ispitivana enzima. U 24-om i 48-om satu nakon zračenja zapaža se sniženje aktivnosti γ-GT u grupama predtretiranim fulerenolom. Fulerenol utiče na povišenje aktivnosti SOD u obe tretirane grupe, osim u grupi ozračenih uzoraka u 48-om satu gde je došlo do sniženja aktivnosti. Predtretman fulerenolom u zračenim grupama povećava, a u neozračenim eksperimentalnim grupama snižava aktivnost GSH-Px.

AB  - Postoji veli ki broj podataka o pokusajima uspostavljanja pouzdanog animalnog modela atcroskleroze, ali jos n ije poznat  dovoljno  dobar  analog  humanoj  aterosk lerozi. Kao prilog ovim is trazivanjima, pokusali smo da proce­ nimo doprinos  reaktivnih  vrsta  kiseonika  (ROS)  u  ranoj ate rogenezi i  prevcntivnu  ulogu  cndogenog  antioksida ­ cio nog zastitnog sis tema (AOS). Odrasli muzjaci Wistar albin o pacova, te les ne masc  oko  400  g,  korisceni  su  za sp rovodenje specijalne ate rogenc dijctc  koja se sas toja la od  smcse  masti  ( buter,  holesterol)  i  drugih   sup stanc i ( ho ln a kise li na, holi n hlorid, tiourac i l ). Kontrolnc i eks­ per imenta lne zivo tin je su bi le srnestcne u plasticne  ka­ veze  i  imale  s u  dostu pne  peleti ranu   hranu  i  vodu po volji. Eksperimentalne zivotinje su dobijale svakoga dana u toku 8 nedeija po 5 g aterogene smese, sondom, intragastricno. Na  kraju  dijete  sve zivotinje  su zrtvovane i sakupljena je krv za odredivanje (1) lipidnog statusa: koncentracija triglicerida (TG), ukupnog holesterola (CH). LDL holesterola i HDL holesterola u plazmi, od­ nosa  koncentracija  LDL i HDL  holesterola  (LDL/HDL) i (2) antioksidacionog statusa u krvi: aktivnost kata­ laze u eritrocitima (Er CAT), aktivnost glutation-S-tran­ sferaze (GST) u  plazmi,  koncentracija  glutationa  (GSH) u plazmi i koncentracija vitamina E (Vit. E) u plazmi. Uzimani su isecci abdominalne aorte za histolosku veri­ fikaciju ateroskle roze. Razlike izmedu vrednosti kon­ trolnih i eksperimentalnih zivotinja testirane su  stati­ sticki.
Analiza hitoloskih praparata aorte pokazala je normalnu gradu arterijskog zida u kontrolnih zivotinja , dok su u eksperimentalnih zivotinja nadene inicijalne promene u intimi (zadebljanje i  celijska akumulacija). Vrednosti svih pracenih varijabli lipidnog i  antioksidacionog  sta­ tusa u kontrolnih zivotinja bile su normalne. U eksperi­ inentalnih zivotinja postojalo je statisticki znacajno po­ vecanje koncentracije TG  (p < 0,05),  koncentracije  CH (p < 0,01), koncentracije LDL (p < 0,01) i odnosa LDL/HDL (p < 0,01) u poredenju sa kontrolnim zivoti­ njama . Koncentracija HDL bila je nepromenjena (p > 0,05). Aktivnost Er CAT i koncentracija  GSH  u plazmi bile su povecane  u  eksperimentalnih  zivotinja  u odnosu na kontrolne,  ali  ne znacajno (p > 0,05). Aktivnost  GST  u plazmi nije bila promenjena (p > 0,05). Samo kon­ centracija Vit. E u  plazmi eksperimentalnih  zivotinja bila je statisticki znacajno veca  u poredenju  sa  kontrol­ nim zivotinjama (p < 0,01). Mozemo zakljuciti da anti­ oksidacioni status u krvi nije bio u korelaciji sa lipidnim statusom  u toku rane aterogeneze  u odraslih  pacova.

Efekti deprenila, rezerpina i njihove kombinacije na aktivnost antioksidativnih enzima u mozgu pacova

AB  - Antioxidant enzyme activities: superoxide dismutases (CuZn SOD and Mn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) and glutathione reductase (GR) were determined in different brain regions: striatum, hippocampus, hypothalamus with thalamus, as well as in the rest of the brain of rats treated subchronically with L-deprenyl, after a single dose of reserpine and in animals subchronically treated with L-deprenyl and then with reserpine. Our results show that reserpine (p < 0.02), as well as, the combination of deprenyl + reserpine (p<0.01) induced a significant decrease in CAT activity in the rest of the brain and a significant decrease in GSH-Px activity in the hypothalamus with thalamus (p<0.05). Deprenyl expressed no significant effect on antioxidant enzyme activities in the examined brain regions. In experimental group treated with deprenyl and then with reserpine a significant decrease of CuZn SOD activity (p<0.05) in the hippocampus, Mn SOD activity (p<0.05) in the striatum and an increase of CnZn SOD activity (p<0.01) in the rest of the brain were observed. Our results support the opinion that depletion of neuronal catecholamine pool (due to the treatment with reserpine) has a direct influence on antioxidant enzyme activities, while deprenyl probably exerts its effects through interaction with some trophic factors.
AB  - Aktivnost antioksidativnih enzima: superoksid dismutaze (CuZn SOD i Mn SOD), katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-S-trans-feraze (GST) i glutation reduktaze (GR) u strijatumu, hipokampusu, hipotalamusu sa talamusom, kao i ostatku mozga, praćena je u eksperimentima subhroničnog tretiranja pacova L-deprenilom, akutnog rezerpinom i u kombinaciji oba. Naši rezultati pokazuju da rezerpin (r<0.02), kao i kombinacija deprenila i rezeršša (r<0.01) dovode do smanjenja aktivnosti CAT u ostatku mozga i aktivnosti GSH-Px (r< 0.05) u hipotalamusu sa talamusom. Deprenil ne izaziva značajne promene enzimske aktivnosti u ispitivanim regionima mozga. U eksperimentalnoj grupi tretiranoj deprenilom, pa rezerpinom dolazi do značajnog smanjenja aktivnosti CuZn SOD (p< 0.005) hipokampusa, aktivnosti Mn SOD strijatuma (p<0i)5) i povećnja aktivnosti CuZn SOD (r<0.01) ostatka mozga. Ovi rezultati ukazuju da iscriljenje pula kateholamina (usled tretmana rezerpinom) ima direktan uticaj na aktivnost antioksidativnih enzima, dok su za efekte deprenila značajne interakcije sa nekim trofičkim faktorima.

abstract = "Antioxidant enzyme activities: superoxide dismutases (CuZn SOD and Mn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) and glutathione reductase (GR) were determined in different brain regions: striatum, hippocampus, hypothalamus with thalamus, as well as in the rest of the brain of rats treated subchronically with L-deprenyl, after a single dose of reserpine and in animals subchronically treated with L-deprenyl and then with reserpine. Our results show that reserpine (p < 0.02), as well as, the combination of deprenyl + reserpine (p<0.01) induced a significant decrease in CAT activity in the rest of the brain and a significant decrease in GSH-Px activity in the hypothalamus with thalamus (p<0.05). Deprenyl expressed no significant effect on antioxidant enzyme activities in the examined brain regions. In experimental group treated with deprenyl and then with reserpine a significant decrease of CuZn SOD activity (p<0.05) in the hippocampus, Mn SOD activity (p<0.05) in the striatum and an increase of CnZn SOD activity (p<0.01) in the rest of the brain were observed. Our results support the opinion that depletion of neuronal catecholamine pool (due to the treatment with reserpine) has a direct influence on antioxidant enzyme activities, while deprenyl probably exerts its effects through interaction with some trophic factors., Aktivnost antioksidativnih enzima: superoksid dismutaze (CuZn SOD i Mn SOD), katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-S-trans-feraze (GST) i glutation reduktaze (GR) u strijatumu, hipokampusu, hipotalamusu sa talamusom, kao i ostatku mozga, praćena je u eksperimentima subhroničnog tretiranja pacova L-deprenilom, akutnog rezerpinom i u kombinaciji oba. Naši rezultati pokazuju da rezerpin (r<0.02), kao i kombinacija deprenila i rezeršša (r<0.01) dovode do smanjenja aktivnosti CAT u ostatku mozga i aktivnosti GSH-Px (r< 0.05) u hipotalamusu sa talamusom. Deprenil ne izaziva značajne promene enzimske aktivnosti u ispitivanim regionima mozga.
