Assessment of genoprotective effects of Gentiana lutea extracts prepared from plants grown in field and in vitro

AB  - Medicinal plant Gentiana lutea is considered as an official drug for gastrointestinal disorders in many pharmacopoeias. Its uncontrolled overexploitation led to the protection regime and encouraged development of in vitro culture. The aim of this work was to chemically characterize root and leaf methanol extracts of plant grown in plantation and in vitro conditions, and to determine their antigenotoxicity against heterocyclic aromatic amines IQ and PhIP, the food-borne mutagens that are formed during high temperature cooking of protein-reach foods. High yields and growth ratios were determined for in vitro plants. Higher content of polyphenols and flavonoids were determined in aerial parts, while UPLC-MS/MS analysis pointed at the richness of in vitro grown shoot extract (GLvS) in active constituents, and high amounts of gentiopicroside in all tested extracts. Notable amounts of sweroside were detected in plantation root extract (GLR) and in GLvS, and of loganic acid in plantation leaf extract (GLL) and GLvS. Two experimental models for genotoxicity/antigenotoxicity study, i.e. SOS/umuC test with metabolic activation (addition of rat liver S9 fraction) and alkaline comet assay on hepatocellur carcinoma HepG2 cells indicated dual effects: genotoxic of high and antigenotoxic of lower non-genotoxic doses. While in vitro extracts were genotoxic in both models, plantation extracts were active only in SOS/umuC with S9. In contrast, inhibition of genotoxicity was observed in both models for all extracts (up to 83 %). Results of DPPH assay indicated that antioxidativity could be responsible for antigenotoxic properties. In conclusion, genoprotective effect encourages further investigation of antigenotoxicity, while high growth ratio and richness of GLvS in active compounds justify the use of in vitro cultivation method.

Bioactivity of Juniperus communis essential oil and post-distillation waste: Assessment of selective toxicity against food contaminants

AB  - Previously chemically characterized Juniperus communis essential oil (EO) and post-distillation waste (PDW) were tested for cytotoxicity and antimicrobial activity against food contaminants. Microdilution assay showed that PDW induced moderate antifungal (minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) values, ranging between 0.118-0.900 mg mL-1), and an antibacterial effect against Listeria monocytogenes (MIC and minimum bactericidal concentration (MBC) were 0.39 and 0.74 mg mL-1, respectively). Combinations of EO/PDW with selected antibiotics induced synergistic antilisterial activity in the checkerboard assay. The MTT assay determined that cytotoxicity against colon cancer cells was high for the EO but negligible for PDW (IC50 values were 0.087-0.106 and 1.450-6.840 mg mL-1, respectively). The selectivity indices indicated high selectivity of PDW against tested fungi and L. monocytogenes. In the adhesion-inhibition assay, PDW reduced in vitro adhesion of L. monocytogenes to colon cells (29-62% of inhibition). In conclusion, PDW exhibited an antimicrobial effect against important food spoilage and poisoning fungi and L. monocytogenes, and also reduced in vitro adhesion of L. monocytogenes to colon cells. The results indicate that J. communis PDW could be considered as natural preservative against food spoilage and poisonous fungi, and as an adjuvant to conventional therapy of listeriosis

AB  - The success of antioxidant therapy in hyperthyroidism implies that disease is mediated by oxidative stress, which is known as one of the causing agents of ageing, degenerative diseases, and cancer. The main objective of our study was to determine possible protective effects of methanolic extract of N. rtanjensis in triiodothyronine (T3)-induced DNA breaks of human lymphocytes under in vitro conditions, based upon plant antioxidant capacity related to its phytochemical profile, mainly its polyphenolic complex. The total phenolic and flavonoid content and the antioxidant activity using in vitro 1,1-dyphenyl-2- picrylhydrazyl reagent (DPPH) was determined in methanolic extracts of plant leaves and flowers. The phenolic compound content of 62.73±1.80mg of GaA/g, exhibited solid antioxidant activity (IC50= 112.59±0.95μg/ml). The antigenotoxic activity of 0.2, 0.5 and 1.0mg/ml N. rtanjensis methanol extracts mixture with 100µM of T3 was studied in human lymphocytes in vitro using the Comet assay. It is supposed that the antigenotoxicity of N. rtanjensis methanol extracts was caused by high presence of chlorogenic acid, rosmarinic acid and rutin, all known as efficient antioxidant bioactive compounds, which were determined by ultrahigh-pressure liquid chromatograph with MS/MS Mass Spectroscopy (UHPLC/-HESI-MS / MS).

AB  - In this study we assessed the level of genotoxic pollution along the Danube River by measuring the level of DNA damage in the haemocytes of freshwater mussels of Unio sp. (Unio pictorum/Unio tumidus) and Sinanodonta woodiana. The comet assay was used for the assessment of DNA damage. The research was performed on 34 out of 68 sites analysed within the Joint Danube Survey 3 - the world's biggest river research expedition of its kind in 2013. During research, 2285 river kilometres were covered with an average distance of 68km between the sites. The complex data set on concentrations of various substances present in water, suspended particulate matter and sediment on investigated sites gave the opportunity to identify the groups of xenobiotics which mostly affect the studied biomarker - DNA damage. The highest levels of DNA damage were recorded in the section VI (Panonnian Plain), which is under the impact of untreated wastewater discharges. Both positive and negative influences of the large tributaries on the level of genotoxicity in the Danube River were evident. Significant correlation in response was detected between the studied species of freshwater mussels. The level of DNA damage in mussels correlated with concentrations of compounds from the group of hazardous priority substances (polycyclic aromatic hydrocarbons), persistent organic pollutants (dioxins) and emerging pollutants (Oxazepam, Chloridazon-desphenyl).

Presence and interaction of various stressors (genotoxic pollutants) in the environment can influence the integrity of DNA molecules in aquatic organisms which can be negatively reflected on the individual, population and community levels. Therefore, in this study we have investigated the impact of flooding, in terms of genotoxicity, on organisms belonging to different trophic levels. The study was carried out on the site situated in the lower stretch of the Sava River which faced devastating effects of severe flooding in May 2014. The flooding occurred during our field experiment and this event provided a unique opportunity to assess its influence to the environment. The in situ effects of this specific situation were monitored by measuring physical, chemical and microbiological parameters of water, and by comparing the level of DNA damage in coelomocytes and haemocytes of freshwater worms Branchiura sowerbyi, haemocytes of freshwater mussels Unio tumidus and blood cells of freshwater fish Abramis bjoerkna/Abramis sapa, by means of the comet assay. Our study indicated that the flooding had a significant impact on water quality by decreasing the amount and discharge rate of urban wastewaters but simultaneously introducing contaminants from the nearby fly ash disposal field into river by runoff, which had diverse effects on the level of DNA damage in the studied organisms. This indicates that the assessment of genotoxic pollution in situ is strongly affected by the choice of the bioindicator organism.

AB  - AimsIn the present work, the Libyan wild-growing Thymus capitatus
   essential oil (EO) was evaluated for its biological properties.
   Methods and ResultsCarvacrol (6819\%) and thymol (1229\%) were found to
   be the main compounds of the oil. Antioxidant properties, determined by
   2,2-diphenylpicrylhydrazyl (DPPH) assay, revealed that IC50 values were
   119, 403 and 105gml(-1) for oil, thymol and carvacrol respectively.
   Microdilution method showed strong antibacterial and especially
   antifungal potential. Tetrazolium (MTT) colorimetric assay indicated
   moderate cytotoxicity towards human cell lines MRC-5, HCT 116 and HT-29
   (IC50=30-150gml(-1)). In adhesion-inhibition assay oil and main
   compounds reduced adhesion of Escherichia coli and Listeria
   monocytogenes on colon cells HT-29 (51 and 39\% of inhibition against
   L.monocytogenes and E.coli respectively).
   ConclusionsEssential oil of Th.capitatus showed moderate cytotoxic
   activity, together with excellent antimicrobial effect, in particular
   against fungi, and significant potential to reduce pathogen colonization
   in colon.
   Significance and Impact of the StudyThis is the first report that EO of
   Th.capitatus could protect against colonization of pathogens to colon
   epithelium. Thymus capitatus from Libya should be recognized as possible
   new source of natural antioxidants, antimicrobials as well as possible
   source of new chemotherapeutics.

AB  - Genotoxicity monitoring of the lower stretch of the Sava River was performed by the combined approach of in situ assessment of genotoxicity and active biomonitoring of two species of mussels from the Unionidae family, Unio pictorum and Unio tumidus. Genotoxic response was studied using comet assay on hemocytes. For active biomonitoring, the mussels were acclimated to controlled laboratory conditions for 10 days and then exposed at two sites in the Sava River in the area of the city of Belgrade. Hemolymph of exposed specimens of each species was taken after 7, 14, and 30 days of exposure. For in situ assessment, the mussels were collected from five sites in the lower flow of the Sava River. The mussels were sampled immediately after the acclimation served as controls in both types of monitoring procedures. The results of our studies indicated the presence of genotoxic pollution at all studied sites at the Sava River. The level of DNA damage varied at different sites depending on the source and level of pollution. The response to genotoxic pollution was evident at the site in the urban area of Belgrade city, as well as at the sites far from the large urban settlements, suggesting that the lower flow of the Sava River is under pollution pressure.

AB  - Data on the microbiological quality of the freshwater systems under the anthropogenic influence, such as the Sava River, are of the major importance for the water resource management. Furthermore, analyses of the microbial quality of fish meat provide information of the fish as a valuable food resource from the investigated river basin. The health status of the fish, including dynamics of infection and biodiversity of endoparasites, is important bioindicator of changes in the ecosystem structure and function. For the ecosystem-based approach to the Sava River management, investigations of microbiological quality of the Sava River water and the meat of the European chub as the bioindicator organism, as well as dynamics of infection/biodiversity of intestinal parasites Acanthocephala, were performed. The survey comprised the data collected in periods 2005, 2006 and 2012. Microbiological investigation of water was performed in 2006 and 2012, while microbiological analyses of fish meat and ichthyo-parasitological investigation took place during 2005–2006. A high number of heterotrophic bacteria were recorded during 2006 survey, confirmed by the distinctly higher values of the three faecal indicators (total coliform, E. coli and enterococci), and indicated poor water quality downstream of the cities Zagreb and Velika Gorica, as a result of the municipal sewage outlets. The results from 2012 survey indicated the existence of moderate to critical faecal and organic pollution in all samples. Accumulation of the bacteria in the European chub meat was mainly uniform along the watercourse within standards and limitations for the human consumption. Sampling sites downstream cities of Zagreb and Velika Gorica were characterised with the lower prevalence and abundance of two common species of the chub intestinal acanthocephalan parasites, Pomphorhynchus laevis and Acanthocephalus anguillae. Poor microbiological quality of the water and lower distribution of chub intestinal parasites were related to the anthropogenic influence, downstream of the urban areas.

AB  - Genotoxic response of freshwater mussels U. tumidus and U. pictorum to
   environmental stress was studied using comet assay on hemocytes and gill
   cells. The mussels were acclimated to controlled laboratory conditions
   for 10 days, and then exposed at 4 sites in the Sava and Danube rivers
   in the area of the city of Belgrade. Samples of each species were taken
   after 7, 14, and 30 days of exposure. The mussels sampled immediately
   after acclimation served as controls. Genotoxic response in both species
   was induced earlier at sites receiving untreated wastewaters from the
   city's main collectors (7 days), than at the site receiving only
   domestic wastewaters from small settlements located upstream from the
   city (30 days). There was a correlation between the comet tail intensity
   values in tissues of exposed mussels and the concentrations of zinc,
   copper, iron, and arsenic at the exposure sites. The genotoxic responses
   in both tissues of U. pictorum and in hemocytes of U. tumidus were
   similar, while U. tumidus gill cells failed to exhibit significant
   genotoxic response at two sites. These findings, together with higher
   mortality of U. tumidus at the most polluted sites, promote U. pictorum
   as a model for genotoxicity monitoring in freshwater environments.

AB  - This study was undertaken to investigate the potential of the freshwater mussel Sinanodonta woodiana for detection of genotoxic pollution of the environment. Study was performed at two sites in the Velika Morava River, from May 2010 to February 2011. The alkaline comet assay on haemocytes was used, and the olive tail moment (OTM) was chosen as a measure of DNA damage. The specimens held on acclimation under controlled laboratory conditions for 10 d were used as a control. Chemical analysis revealed the presence of phosphates and increased concentrations of zinc, copper and nickel at both sites during the entire sampling period. The values of OTM in mussels collected from the environment, significantly correlated with the concentration of zinc (r = 0.6248), temperature (r = 0.7006) and dissolved oxygen (r = 0.7738). Seasonal variations in genotoxic response were observed, with the highest OTM values obtained during summer months. Preliminary results of the in vitro study indicated the effect of water temperature on genotoxic response to zinc and cadmium in S. woodiana suggesting that the presence of genotoxic pollutants during months with lower temperature could be under-estimated. Obtained results indicate that S. woodiana could be a valuable tool for active biomonitoring of aquatic environments and emphasizes the importance of seasonal genotoxic monitoring with this organism. AB  - In this study, we investigated the level of sanitary pollution and organic contamination of the Velika Morava River, the largest river in central Serbia. Samples of water for microbiological analysis were collected at 5 sites, monthly, from April 2010 to February 2011. Sanitary analysis, i.e. enumeration of total and fecal coliforms and intestinal enterococci, indicated moderate to critical fecal contamination, while organic load assessment (oligotroph to heterotroph ratio, index of phosphatase activity) revealed the category of moderately polluted water. We also investigated seasonal variations of these groups of bacteria and the factors that could contribute to these differences. Our results showed that the micro- biological quality of the water in the Velika Morava River during different seasons is affected by numerous factors such as unequal loading of wastewaters, solar irradiation, and relations of flow/dilution and rainfall/runoff.

AB  - The aim of this work was to examine the impact of urban wastewaters on the water quality of the Danube River in Serbia. Samples of water and sediments for microbiological analysis and genotoxicity monitoring were collected from 6 sites during spring and/or autumn 2010. Sanitary analysis, i.e. enumeration of total and fecal coliforms and intestinal enterococci, indicated moderate to critical fecal contamination, while organic load assessment (oligotroph to heterotroph ratio, index of phosphatase activity) revealed the category of moderately polluted water. Mercury-resistant bacteria were detected in all water samples, with high numbers at locations positioned downstream of Belgrade. There was no correlation of the microbiological parameters of the sediment and water samples. Genotoxicity monitoring, performed by the comet assay on hemocytes of mussels Sinanodonta woodiana, indicated a significant increase of DNA damage in mussels collected from the studied sites compared with the control group.

Antimutagenic activity of essential oil and crude extract of Phlomis fruticosa

AB  - The present study was carried out to investigate the antimutagenic activity of essential oil and crude extract of Phlomis fruticosa. This species belongs to the family Lamiaceae which includes a number of species with medical and pharmaceutical properties. The Escherichia coli K12 reversion assay for identifying antimutagens was used. The number of spontaneous and UV-induced Arg(+) revertants, as well as cell survival was monitored in the presence of non-toxic concentrations of the essential oil. The number of UV-induced revertants was slightly increased in the presence of essential oil. The ethanol extract demonstrated a significant effect on viable cells. Study on pharmacological and phytochemical activities as well as antimutagenetic potential against some chemical mutagens of the essential oils, total extract and their different fractions and constituents of Ph. fruticosa is in progress.

AB  - The chemical composition of essential oils from three Micromeria species: M. dalmatica Benth., M. albanica (Griceb. ex K. Mal) Silic and M. thymifolia (Scop.) Fritsch were investigated by GC and GC-MS and their antibacterial and antifungal activities against seven fungal and six bacterial species were evaluated. Biological assays showed strong fungitoxicity of oils from all three Micromeria spp., particularly M. albanica, against all fungi tested. Essential oils of these species also exerted antibacterial effect against Streptococcus aureus, Escherichia coli, Bacillus subtilis and Micrococcus luteus at low concentrations. Higher concentrations of essential oil of M. albanica and M. dalmatica were active against Gram-negative Pseudomonas aeruginosa, which could be due to the high content of piperitenone oxide. Biological assays showed strong fungitoxicity of oils from all three Micromeria spp., particularly M. albanica, against all fungi tested. Essential oils of these species also exerted antibacterial effect against Streptococcus aureus, Escherichia coli, Bacillus subtilis and Micrococcus luteus at low concentrations. Higher concentrations of essential oil of M. albanica and M. dalmatica were active against Gram-negative Pseudomonas aeruginosa, which could be due to the high content of piperitenone oxide.
