AB  - The use of anticoagulant rodenticides (ARs) is one of the most commonly employed management methods for
pest rodents. ARs compete with vitamin K (VK) required for the synthesis of blood clotting factors in the liver,
resulting in inhibition of blood coagulation and often animal death due to hemorrhage. Besides rodents (target
species), ARs may affect non-target animal species and humans. Out of hemostasis disturbance, the effects of ARs
may be related to the inhibition of proteins that require VK for their synthesis but are not involved in the
coagulation process, to their direct cytotoxicity, and their pro-oxidant/proinflammatory activity. A survey of the
cellular and molecular mechanisms of these sublethal/asymptomatic AR effects is given in this review. Data from
field, clinical, and experimental studies are presented. Knowledge of these mechanisms might improve hazard
characterization and identification of potential ecotoxicological risks associated with ARs, contributing to a safer
use of these chemicals.

AB  - While the effect of gut microbiota and/or inflammation on a distant body site, including the lungs (gut–lung axis), has been well characterized, data about the influence of lung microbiota and lung inflammation on gut homeostasis (lung–gut axis) are scarce. Using a well-characterized model of pulmonary infection with the fungus Aspergillus fumigatus, we investigated alterations in the lung and gut microbiota by next-generation sequencing of the V3–V4 regions of total bacterial DNA. Pulmonary inflammation due to the fungus A. fumigatus caused bacterial dysbiosis in both lungs and gut, but with different characteristics. While increased alpha diversity and unchanged bacterial composition were noted in the lungs, dysbiosis in the gut was characterized by decreased alpha diversity indices and modified bacterial composition. The altered homeostasis in the lungs allows the immigration of new bacterial species of which 41.8% were found in the feces, indicating that some degree of bacterial migration from the gut to the lungs occurs. On the contrary, the dysbiosis occurring in the gut during pulmonary infection was a consequence of the local activity of the immune system. In addition, the alteration of gut microbiota in response to pulmonary infection depends on the bacterial composition before infection, as no changes in gut bacterial microbiota were detected in a rat strain with diverse gut bacteria. The data presented support the existence of the lung–gut axis and provide additional insight into this mechanism.

Impact of gut microbiota on immune reactions relevant to lung pathologies

AB  - Bacterial microbiota of the gastrointestinal tract is known to prevent the invasion of
pathogenic microorganisms and regulate intestinal permeability, digestion, metabolism, and
immune response. It affects function, homeostasis, and disease outcomes in the gastrointestinal
tract and extra-intestinal sites such as the lungs. This review summarizes the currently available
knowledge regarding the gut-lung axis. The association of bacterial composition and/or dysbiosis
in the gut with asthma, chronic obstructive lung disease, cystic fibrosis, recurrent respiratory tract
infections, and lung cancer in humans is highlighted, as well as data obtained from animal models
of pulmonary inflammation, which indicated that modulation of immune system activity lies at
the base of this interaction. Additionally, the potential use of prebiotics, probiotics, and postbiotics
in the treatment of lung inflammation is presented.
AB  - Poznato je da bakterije prisutne u gastrointestinalnom traktu imaju ulogu u sprečavanju
invazije patogenih mikroorganizama, regulaciji propustljivosti creva, varenju hrane, metabolizmu
i imunskom odgovoru. Ove bakterije utiču na funkciju, održavanje homeostaze i ishod bolesti
kako u gastrointestinalnom traktu, tako i u udaljenim organima kao što su pluća. Ovaj pregledni
rad sumira trenutno dostupna znanja o osi creva-pluća. Prikazana je veza između bakterijskog
sastava i/ili disbioze u crevima sa različitim bolestima kod ljudi kao što su astma, hronična
opstruktivna bolest pluća, cistična fibroza, rekurentne infekcije respiratornog trakta i karcinom
pluća, kao i podaci dobijeni u životinjskim modelima inflamacije pluća koji su pokazali da
modulacija aktivnosti imunskog sistema leži u osnovi ove interakcije. Potencijalna upotreba
prebiotika, probiotika i postbiotika u terapiji inflamacije u plućima je takođe prikazana.

AB  - Since the realization that the lungs are not sterile but are normally inhabited by various bacterial species, studies have been conducted to define healthy lung microbiota and to investigate whether it changes during lung diseases, infections, and inflammation. Using next-generation sequencing, we investigatedbacterial microbiota from whole lungs in two rat strains (previously shown to differ in gut microbiotacomposition) in a healthy state and during pulmonary infection caused by the opportunistic fungusAspergillus fumigatus. No differences in alpha diversity indices and microbial composition between DAand AO rats before infection were noted. Fungal infection caused dysbiosis in both rat strains, characterizedby increased alpha diversity indices and unchanged beta diversity. The relative abundance ofgenera and species was increased in DA but decreased in AO rats during infection. Changes in lungmicrobiota coincided with inflammation (in both rat strains) and oxidative stress (in DA rats). Disparateresponse of lung microbiota in DA and AO rats to pulmonary fungal infection might render these two ratstrains differentially susceptible to a subsequent inflammatory insult.

AB  - Кадмијум је глобално присутан токсични метал који изазива бројне штетне ефекте у организму и заузима седмо место на листи приоритетних супстанци од којих је неопходно пронаћи оптималне начине заштите. Наше пређашње студије су показале да егзополисахарид изолован из соја Lactiplantibacillus plantarum BGAN8 (EPS-AN8) показује висок афинитет за везивање јона кадмијума у воденом раствору и остварује значајан ниво in vitro заштите Caco-2 ћелија од његових токсичних ефеката. Имајући у виду да је за општу популацију најзаступљенији унос кадмијума исхраном, у овој студији је праћен паралелни ефекат уноса кадмијума (кроз воду) и EPS-AN8 (кроз храну) у DA пацовима. Након 30 дана третмана, утврђено је да је унос EPS-AN8 допринео сниженом нивоу депонованог метала у бубрезима, јетри и крви, а повећаном садржају у фекалном материјалу. Такође, микрографије дуоденума, бубрега и јетре су указале на ниже степене кадмијумом индукованих оштећења. Праћењем активности ензима који учествују у антиоксидативној заштити (CAT, GST), терминалних производа липидне пероксидације (MDA) и продукције проинфламаторних цитокина (IL-1β, TNFα, IFN-γ) у хомогенатима дуоденума, утврђен је нижи ниво оксидативног стреса и инфламације, што представља додатни показатељ заштите коју остварује EPS-AN8. С обзиром да орални унос кадмијума доводи до значајних промена у цревној микробиоти, изоловали смо тоталну DNK из дуоденума и секвенцирали 16S rDNK ампликон на Illumina NovaSeq платформи. Примећено је да унос EPS-AN8 ублажава појаву промена у релативној заступљености бактеријских родова и врста карактерисичних за излагање кадмијуму, попут пораста броја опортунистичких патогених бактерија и смањења бројности лактобацила. Остварени заштитни ефекти указују на снажан потенцијал примене EPS-AN8 у условима високе контаминације кадмијумом.

AB  - The importance of issues associated with urban/commensal rats and mice (property damage, management costs,
and health risks) press upon research on these animals. While the demography of commensal rodents is mostly
studied, the need for understanding factors influencing their natural morbidity/mortality is also stressed. In this
respect, more attention is expected to be paid to immunity, the physiological mechanism of defense against host
survival threats (pathogens, parasites, diseases). Commensal rats and mice carry numerous pathogens that evoke
diverse immune responses. The state of immunity in commensal house mice is studied in great detail, owing to
the use of laboratory strains in biomedical research. Because commensal rats are, compared to mice, carriers
of more zoonotic agents, rats’ immunity is studied mainly in that context. Some of these zoonotic agents cause
chronic, asymptomatic infections, which justified studies of immunological mechanisms of pathogen tolerance
versus clearance regulation in rats. Occurrence of some infections in specific tissues/organs pressed upon analysis
of local/regional immune responses and/or immunopathology. A survey of immunological activity/responses in
commensal rats is given in this review, with mention of existing data in commensal mice. It should throw some
light on the factors relevant to their morbidity and lifespan, supplementing the knowledge of commensal rodent
ecology.

The little data of the effect of pulmonary fungal infection on lung and
gut microbiota exist, thus aim of our study was to examine the effect of
Aspergillus fumigatus infection on bacterial homestasis in these organs in
Dark Agouti (DA) and Albino Oxford (AO) rats known to develop different
immune responce to the fungus.
Methods:
16S rRNA amplicon sequencing of the V3-V4 hypervariable region.
Enzyme-linked immunosorbent assay for cytokine content (IL-10, IL-6, TNF,
IL-1beta, IL-17 i IFN-gamma) in lung and gut homogenats, and plasma (IL-
6, IL-17, TNF). Myeloperoxidase, catalase, glutathione S-transferase and
content of reduced glutathione in lung homogenates.
Results:
No initial differences in lung bacterial microbiota between healthy DA
and AO rats exists, but A. fumigatus infection resulted in increase in
alpha diversity (without effect on bacterial composition) in both strains.
Changes in relative abundance at genus and species level were expressed
differently (increased in DA, decreased in AO). Dysbiosis in lung coincided
with increase of IFN-gamma, IL-17 and MPO in both strains, and increase
oxidative stress solely in DA. Gut inflammation was noted in both strains,
while bacterial dysbiosis only in DA.
Conclusions:
Pulmonary fungal infection resulted in bacterial dysbiosis in lung (in both
rat strains), and gut (solely in DA). Noted lung and gut inflammation in
these two rat strains, coexisted with different effects on bacterial
homeostasis, which could be due to different genetic background or
bacterial composition prior infection.

AB  - Cadmium (Cd) is a highly toxic metal that is distributed worldwide. Exposure to it is
correlated with a vast number of diseases and organism malfunctions. Exopolysaccharides (EPS)
derived from Lactiplantibacillus plantarum BGAN8, EPS-AN8, previously showed great potential for
the in vitro protection of intestinal cells from this metal. Here, we investigated the potential of food
supplemented with EPS-AN8 to protect rats from the hazardous effects of Cd exposure. After thirty
days of exposure to lower (5 ppm) and higher (50 ppm)-Cd doses, the administration of EPS-AN8 led
to decreased Cd content in the kidneys, liver, and blood compared to only Cd-treated groups, whereas
the fecal Cd content was strongly enriched. In addition, EPS-AN8 reversed Cd-provoked effects on
the most significant parameters of oxidative stress (MDA, CAT, GST, and GSH) and inflammation
(IL-1 , TNF- , and IFN-
) in the duodenum. Moreover, micrographs of the duodenum were in line
with these findings. As the gut microbiota has an important role in maintaining homeostasis, we used
16S rRNA amplicon sequencing and investigated the effects of Cd and EPS-AN8 on one part of the
microbiota presented in the duodenum. Although Cd decreased the growth of lactobacilli and mostly
favored the blooming of opportunistic pathogen bacteria, parallel intake of EPS-AN8 reversed those
changes. Therefore, our results imply that EPS-AN8 might be extremely noteworthy in combatting
this toxic environmental pollutant.

AB  - Objective The effect of oral cadmium (Cd) intake to influence contact skin allergies was examined,
since it is known that Cd is a heavy metal that affects many tissues, including the skin, in which it
disturbs homeostasis, thus resulting in inflammation and injury.
Methods Male rats were evoked with experimental contact hypersensitivity reaction (CHS) to hapten
dinitrochlorobenzene (DNCB), after prolonged (30 day) oral exposure to an environmentally relevant Cd
dose (5 ppm). The ear cell population was analyzed with flow cytometry. Cytokine production by ear
skin cells and the activity of skin-draining lymph node (DLN) cells were measured using enzyme-linked
immunosorbent assay (ELISA).
Results Orally acquired Cd (5 ppm) increased CHS intensity only in Dark Agouti (DA) rats by affecting
inflammatory responses in both the sensitization (an increase of IFN-γ and IL-17 cytokine production)
and challenge (an increase of CD8+ and CD4+ cell number and TNF, IFN-γ and IL-17 cytokine production)
phases. An increased CHS reaction was seen in Albino Oxford (AO) rats only at a high Cd dose (50 ppm),
during the challenge phase (an increase of CD8+ and CD4+ cell number and TNF, IFN-γ and IL-17 cytokine
production).
Conclusion These novel data indicate that oral Cd intensifies the skin response to sensitizing chemicals
such as DNCB.

Интеракција загађивача са ћелијама имунског система може довести до поремећаја имунолошки-посредоване ткивне хомеостазе са штетним здравственим ефектима. Тешки метали спадају у најопасније загађиваче међу којима је кадмијум (Cd) један од најтоксичнијих, с обзиром на податке који показују да је токсичан при ниским концентрацијама и на студије које га повезују са имунолошки-посредованим поремећајима. Кадмијум најчешће доспева у организам оралним путем, уношењем контаминиране хране и воде, након чега се путем крви дистрибуира до свих органа где се депонује. На пацовском моделу продужене оралне примене Cd (дозе релевантне за срединску изложеност) показали смо да Cd остварује имуномодулаторне ефекте локално (у цревном тракту) али и у удаљеним ткивима плућа и коже. Оштећење ткива и инфламација (повећан оксидативни стрес и продукција проинфламаторних цитокина) запажени су у сва три органа. Поремећај имунске хомеостазе у цревима огледа се и у стимулацији ефекторског имунског одговора (пролиферација, IFN-γ, IL-17) у чворовима који дренирају ову регију. Инфламација у кожи и повећана оксидативна и проинфламаторна активност епидермних ћелија коинцидирају са повећаном реактивношћу овог ткива на контактни алерген. Утичући на активност имунског система орално примењен Cd може да мења његову функцију и последично осетљивост на развој болести, што указује да Cd може представљати озбиљну претњу по здравље људи.

AB  - In addition to physical and chemical insults originating from nature (heavy metals, radiation, electromagnetic field), human beings are exposed to various agents as a result of progress in industry and technology (noise, pesticides, pharmaceuticals, personal care products, food additives, etc.) whose number continuously increases. Some of these agents may have adverse effects on the environment and human health and therefore every new product must be examined for its toxic potential and health risk. The immune system is an important homeostatic system that is included in response to various pathogens, but in other processes as well including wound healing and elimination of damaged cells and tumors. Due to its involvement in various physiological processes, alternation in immune system activity, as a consequence of exposure to xenobiotics, may cause an increase in the incidence of infections, tumors, allergies, and autoimmune diseases. Various tests that exist for the evaluation of immunotoxicity are grouped in two separate sets, tier one (experiments directed to measure the effect of xenobiotics on general immune parameters i.e. basic morphological and functional tests) and tier two(experiments aimed to more specifically define the cellular and biochemical mechanisms of toxicity). Evaluation of potential adverse effects of xenobiotics on the immune system is now incorporated into standard hazard identification and has improved human health risk assessment.

AB  - Cadmium (Cd) in food and drinking water presents a health risk to the general population. We have shown previously that orally‐acquired Cd affects basal immune homeostasis in the skin. In this study, we examined the effect of 30‐days oral exposure of inflammatory disease‐prone Dark Agouti (DA) rats to two environmentally relevant Cd doses (5 and 50 ppm) on contact hypersensitivity reaction (CHS) induced by topical 0,4% dinitrochlorobenzene (DNCB). Both Cd doses increased proinflammatory epidermal cell response (IL‐1, TNF and IL‐6 production) to DNCB sensitization, as well as epidermal cells’ potential to stimulate naïve lymphocytes ex vivo (increased IFN‐γ and IL‐17 production in co‐cultures). The proinflammatory milieu of epidermal cells induced by sensitization was accompanied by increased hapten‐specific production of IFN‐γ (at a lower Cd dose) and IL‐17 (at both Cd doses) by draining lymph node (DLN) cells, compared to Cd non‐treated animals. During the challenge phase of CHS, oral Cd increased ear swelling response and skin inflammation (edema, mononuclear and neutrophil cell infiltration) at both Cd doses, what correlated with increased innate (TNF) and hapten‐specific effector (IFN‐γ, IL‐17) cytokine response by ear cells. Even in Albino Oxford (AO) rats generally less prone to inflammation, oral Cd increased the proinflammatory response of epidermal cells following sensitization, however, DLN cell responses were absent. Ear swelling response to hapten challenge was observed in AO individuals which acquired a higher Cd dose. Presented data imply the potential of food‐ and water‐borne Cd to be risk factors for skin disease development and/or its exacerbation.

Owing to its immunomodulatory potential, orally acquired Cd affects antimicrobial immune defense in several organs, including the lungs. While there are data concerning Cd and viral and bacterial pulmonary infections, effects on fungal infections are not studied yet. In the present study, the effect of the Cd (5 mg/L for 30 days, in drinking water, the average daily Cd intake 0.641 ± 0.089 mg/kg) on the immune response of rats to pulmonary A. fumigatus infection was examined. Data obtained showed that orally acquired cadmium does not affect the elimination of the fungus in immunocompetent rats owing to the preservation of some aspects of innate immune responses (lung leukocyte infiltration and NBT reduction) and an increase in other (increased numbers of mucus-producing goblet cells, MPO release). Cd does not affect an IFN-γ response in lung leukocytes during the infection (despite suppression of cytokine production in cells of lung-draining lymph nodes), while it stimulates IL-17 and suppresses IL-10 response to the fungus. As a result, the elimination of the fungus occurs in a milieu with the prevailing proinflammatory response in Cd-exposed animals that preserved fungal elimination from the lungs, though with more intense injury to the lung tissue. Therefore, the proinflammatory microenvironment in the lungs created by Cd that sustains inflammatory/immune response to the fungus to which humans are exposed for a lifetime, raises a concern of orally acquired Cd as a risk factor for the development of chronic low-grade pulmonary inflammation.

The involvement of AhR signaling in the effects of Cd on the interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF) lung leukocyte response was investigated in vitro using the receptor antagonist CH-223191. Results Cd increased CYP1B1 ( in vivo and in vitro) and CYP1A1 ( in vitro) mRNA, indicating AhR involvement in the action of Cd. In response to Cd, lung leukocytes increased IL-6 and decreased TNF at the gene expression and protein levels, but decreased IL-1β production due to reduced NLRP3. The AhR antagonist CH-223191 abrogated the observed effects of Cd on the cytokine response. The absence of AhR reactivity and cytokine response to Cd of leukocytes from the lungs of a rat strain that is less sensitive to Cd toxicity coincided with a high AhR repressor mRNA level. Conclusion AhR signaling is involved in the lung leukocyte proinflammatory cytokine response to Cd. The relevance of the AhR to the cytokine response to Cd provides new insight into the mechanisms of Cd immunotoxicity.

AB  - Cadmium (Cd) represents a unique hazard because of the long biological half-life in humans (20-30 years). This metal accumulates in organs causing a continuum of responses, with organ disease/failure as extreme outcome. Some of the cellular and molecular alterations in target tissues can be related to immune-modulating potential of Cd. This metal may cause adverse responses in which components of the immune system function as both mediators and effectors of Cd tissue toxicity, which, in combination with Cd-induced alterations in homeostatic reparative activities may contribute to tissue dysfunction. In this work, current knowledge concerning inflammatory/autoimmune disease manifestations found to be related with cadmium exposure are summarized. Along with epidemiological evidence, animal and in vitro data are presented, with focus on cellular and molecular immune mechanisms potentially relevant for the disease susceptibility, disease promotion, or facilitating development of pre-existing pathologies.

However, it is clear that Cd can affect cells of the immune system and can modulate some immune responses. Due to the complex nature of the immune system and its activities which are determined by multiple interactions, the underlying mechanisms involved in the immunotoxicity of this metal are still vague. Here, the current knowledge regarding the interaction of Cd with cells of the immune system, which may affect immune responses as well as potential mechanisms of consequent biological effects of such activities, is reviewed. Tissue injury caused by Cd-induced effects on innate cell activities depicts components of the immune system as mediators/effectors of Cd tissue toxicity. Cd-induced immune alterations, which may compromise host defense against pathogenic microorganisms and homeostatic reparative activities, stress this metal as an important health hazard.

Since our previous study has showed that orally acquired Cd affects skin, the contribution of genetic background to dermatotoxicity of oral cadmium was examined in two rat strains, Albino Oxford (AO) and Dark Agouti (DA), which differed in response to chemicals. While similar accumulation of Cd in the skin of both strains was noted, the skin response to the metal differed. DA rat individuals mounted antioxidant enzyme defense in the skin already at lower Cd dose, in contrast to AO rats which reacted to higher metal dose solely (and less pronounced), implying higher susceptibility of DA strain to Cd dermatotoxicity. Epidermal cells from both strains developed stress response, but higher intensity of antioxidant response in AO rats implied this strain`s better ability to defend against Cd insult. Cd induced epidermal cells' proinflammatory cytokine response only in DA rats. Increased IL-10 seems responsible for the lack of response in AO rats. Differences in the pattern of skin/epidermal cell responsiveness to cadmium give a new insight into repercussion of genetic variability to dermatotoxicity of orally acquired cadmium, bearing relevance for variations in the link between dietary cadmium and inflammation-based skin pathologies.

AB  - In last two decades, there has been a strong trend in the application of lactic acid bacteria as adjunctive cultures to control growth of spoilage and pathogenic bacteria in food. One of the most important properties that contribute to the application of these bacteria is the production of antimicrobial molecules. Lactococcus lactis subsp. lactis BGBU1-4, isolated from traditional brined cheese, produces thermostable bacteriocin named lactolisterin BU, with broad spectrum of activity against spoilage bacteria and foodborne pathogens. In this study, effect of strain BGBU1-4, as adjunct culture, on the numbers of Listeria monocytogenes ATCC19111 and Staphylococcus aureus LMM322 in artificially contaminated Quark-type, soft acid coagulated cheese, was examined. In addition, we analyzed influence of BGBU1-4 on chemical and sensory properties of the cheese, as well as immunological response of Albino oxford rats fed with Quark-type of cheese made using BGBU1-4 as adjunct culture. Results of this study revealed antibacterial potential of strain BGBU1-4 against L. monocytogenes ATCC19111 and S. aureus LMM322 in Quark-type cheese during 21 days of storage at 4 °C. Also, it was noticed the ability of BGBU1-4 to control the spontaneously grown yeasts and molds. Chemical composition and pH values of cheese containing BGBU1-4 were unchanged in comparison to control. The sensory quality scores showed that there was difference between cheese with and without adjunct culture in terms of flavor and oral texture, while for the odor and appearance no differences between two cheese variants were scored. Results of the immunological response of Albino rats fed with Quark-type cheese containing BGBU1-4 indicate absence of systematic inflammation. However, increased pro-inflammatory cytokines content (IL-1β, IL-6, IL-17) in intestine of rats fed with cheese containing BGBU1-4, concomitantly with unchanged anti-inflammatory cytokines suggests disruption of gut homeostasis and inflammation in this tissue. The changes caused by BGBU1-4 are reversible, system returns into homeostasis seven days after cessation of feeding with cheese containing BGBU1-4.

However, increased pro-inflammatory cytokines content (IL-1β, IL-6, IL-17) in intestine of rats fed with cheese containing BGBU1-4, concomitantly with unchanged anti-inflammatory cytokines suggests disruption of gut homeostasis and inflammation in this tissue. The changes caused by BGBU1-4 are reversible, system returns into homeostasis seven days after cessation of feeding with cheese containing BGBU1-4.",

Iako je poznato da topikalna primena Cd može uticati na vijabilnost ćelija kože, gotovo da nema podataka o efektu Cd na homeostatske procese ovog tkiva, uključujući efekte na imunski sistem kože, naročito nakon oralnog unosa. Efekti produženog izlaganja (30 dana) sredinski bitnim dozama Cd (5 i 50 ppm), ispitani su kod Dark Agouti (DA) i Albino Oxford (AO) pacova, koji se razlikuju u imunskom odgovoru na različite stimuluse. Pored efekta Cd na oksidativnu i imunsku/inflamatornu aktivnost kože, ispitana je reaktivnost kože na dodatni stimulus, dinitrohlorobenzen (DNCB) u reakciji kontaktne preosetljivosti. Uprkos sličnom deponovanju Cd u koži/ćelijama kože, kod DA soja već na 5ppm Cd zapaženo je oštećenje tkiva i veći intenzitet/različit obrazac oksidativnog i imunskog odgovora, ukazujući na veću osetljivost ovog soja na dermatotoksične efekte Cd. Iako su EĆ oba soja razvile stres odgovor na prisustvo Cd, intezivniji antioksidativni odgovor (povećanje GSH, viša ekspresija gena za MT-1 i -2, proteina za Nrf2, kao i ekspresije gena AHR puta) uz prisustvo apoptoze samo kod AO soja, ukazuje na bolju sposobnost zaštite ovog soja od štetnog efekta Cd. Proinflamatorni odgovor zapažen je samo kod EĆ DA soja (povećanje IL-1β, TNF i IL-6), a povećanje IL-10 kao potencijalnog mehanizma zaštite kod AO soja. Ipak, nakon topikalne primene DNCB razvija se inflamatorni odgovor kod oba soja, sugerišući da se kod AO pacova probijaju zaštitni mehanizmi koji su se održavali nakon primene samog Cd (makar na 50 ppm Cd). Ova studija može biti korisna prilikom ispitivanja veze oralnog unosa Cd i različite osetljivosti kože na razvoj zapaljenskih reakcija i patoloških stanja.
AB  - Harmful effects of environmental Cd exposure, which affect different organs, are being increasingly recognized. Although it is known that topical Cd administration may affect skin cell viability, there is a paucity of data concerning Cd effect on the homeostatic processes of this tissue, including effects on the skin's immune system, especially after oral intake. The effects of prolonged exposure (30 days) to the two environmentally relevant Cd doses (5 and 50 ppm) were examined in Dark Agouti (DA) and Albino Oxford (AO) rats, which differ in the immune responses to different stimuli. In addition to the Cd effects on the oxidative and immune/inflammatory skin activity, the reactivity of skin after additional dinitrochlorobenzene (DNCB) stimulus during contact hypersensitivity reaction (CHS), was examined. Despite similar Cd deposition in skin/skin cells, tissue damage and a higher intensity/different pattern of oxidative and immune responses were observed in DA strain already at 5 ppm Cd, indicating a higher sensitivity of this strain to the dermatotoxic Cd effects. Although epidermal cells (ECs) of both strains developed stress response to the Cd presence, a more intensive antioxidative response (an increase of GSH, a higher level of gene expression for MT-1 and MT-2, proteins for Nrf2, as well as the genes of AHR pathway) with the presence of apoptosis only in AO strain, indicates a better protection capability of this strain from deleterious Cd effect. The proinflammatory response was observed only in the ECs of DA strain (an increase in IL-1β, TNF and IL-6), as well as IL-10 increase in AO strain as a potential protection mechanism. Nevertheless, after topical administration of DNCB, an inflammatory response develops in both strains, indicating the breakthrough of protective mechanisms that persisted in AO rats after oral Cd intake (at least at 50 ppm Cd). This study may be useful for examining the relationship between oral Cd intake and different skin sensitivity to the development of inflammatory responses and pathological conditions.

AB  - The gut microbiome is very important for hosts’ proper essential functions, as microbiota influence both near and distant organs and disruption at any level of this complex relation can be underlying cause for many diseases. Both intestinal bacterial population and their metabolic products are involved in immune system development, activity and maintenance of immune homeostasis. Previous findings revealed differences in immune reactivity in Albino Oxford (AO) and Dark Agouti (DA) rat strains in various diseases models (such as experimental autoimmune encephalomyelitis, rheumatoid arthritis, contact hypersensitivity reaction, pulmonary aspergillosis etc.), and in response to xenobiotics such as cadmium, warfarin, etc.
The aim of present study was to investigate microbial composition of gut (duodenum, jejunum, coecum and colon) and lungs of AO and DA rats using DGGE method. Although similar number of bacterial species were noted in both tissue and lumen of duodenum, bacterial composition differ between these two strains (solely 76.2% and 44.4% species were common in both strains tissue and lumen, respectively), while greater variability was noted in DA rats. Similar results were noted in jejunum. In contrast to duodenum and jejunum, higher number of bacterial species were detected in coecum (content) and colon (tissue and content) of AO rats. Around 50% of detected bacteria were present in both gut segments of both strains. Analysis of DGGE bands obtained from lung tissue revealed similar number of bacteria in both strains, but solely 54.5% were common. Further investigations will be directed to identification of bacterial species and try to connect observed differences in microbial composition with different immune reactivity in AO and DA rats.

AB  - Cadmium (Cd) is a heavy metal widely spread in the environment and significant water and food contaminant. This metal exerts toxic effects in various tissues thus representing great threat to human health, and our previous study showed that oral consumption of Cd (in water for 30 days) increased the metal deposition and exerted immunomodulatory effects in lung leukocytes. Although the most studied mechanisms of Cd toxicity include oxidative stress and inflammation, recent studies have indicated that this metal can activate aryl hydrocarbon receptor (AHR) and exert effects on AHR-regulated genes (i.e. CYPs). AHR represents a link between environmental toxicants and immune response as high receptor expression is noted in immune cells and barrier tissues, thus the aim of presented study was to investigate if activation of AhR by Cd is associated with metals’ immunomodulatory effects. Treatment of lung leukocytes with Cd in vitro (non-toxic doses) caused an increase in mRNA levels for AHR, CYP1B1 and CYP1A1, but co-treatment with metal and AHR antagonist CH223191 indicated that higher Cd doses (5 and 10 μM) can activate CYPs directly while a lower dose (1 μM) exerted effects on CYPs expression through activation of AHR. Low Cd dose induced increased production of IL-6 and decreased TNF and IL-1β by lung leukocytes, compared to controls. Gene expression data revealed unchanged mRNA for IL-6, decreased TNF, but increased IL-1β. Lower IL-1β protein level despite increased mRNA, was a consequence of decreased mRNA for NLRP3, a component of inflammasome that is involved in processing of pro-IL-1β in IL-1β. All noted effects were abolished in the presence of CH223191. Data obtained indicate that immunomodulatory effects of low Cd dose are mediated through AHR activation.

AB  - Adverse effects of non-occupational exposure to cadmium (Cd) are increasingly
acknowledged. Using a rat model of oral Cd exposure in drinking water we have shown
that skin is a target for this metal. Due to contribution of individual variability to the
intensity of cadmium toxicity, dermatotoxicity of two environmentally relevant Cd
doses (5 and 50 ppm) was examined in individuals of two rat strains, Albino Oxford
(AO) and Dark Agouti (DA), which differ in response to chemicals. A dose-dependent
accumulation of Cd in the skin/epidermal cells was noted in both strains, and although
there were no strain differences in the Cd accumulation, the degree of skin response to
the metal differed. Signs of skin damage were evident in both strains, but response to
injury was more pronounced in DA. Individuals of DA rats responded by an increase
in the levels of antioxidant defense enzymes in the skin already at lower dose, in
contrast to AO (which reacted to higher dose solely), implying higher sensitivity of DA
strain to Cd-induced toxicity. Epidermal cells from both strains developed stress
response, however increased GSH, and higher metallothionein/MT-1 and MT-2
mRNA, Nrf2 protein, apoptosis, Ahr and Cyp genes in AO, depicting this strain`s
ability to better defend against Cd insult. Epidermal cells` IL-1β, TNF and IL-6
response was induced by Cd in DA, while pro-inflammatory cytokine production was
unchanged in AO (though increased following stimulation with S. epidermidis), with
increased IL-10 as a possible underlying mechanism. T cells from non-exposed rats
produce more IFN-γ and IL-17 in co-culture with epidermal cell from Cd–exposed DA
rats what strengthens the view that this strain is more prone to metal’s dermatotoxicity.
These data give a new insight into repercussion of genetic variability to toxicity of
cadmium acquired by the skin via gut, bearing relevance for variations in the link
between dietary cadmium and inflammation-based skin pathologies.

AB  - Cadmium (Cd) is one of the most cytotoxic agents and environmental contaminants, which can cause serial health problems in various organs such as liver, kidney, testis, bone, nervous tissue and immune system. Furthermore, Cd is classified as a human and animal carcinogen agent. Despite the fact that Cd accumulates in the skin, there is a lack of evidence about its involvement in skin cancer biology. Skin cancer is one of the most common cancers worldwide, with melanoma as the most lethal type. Based on that, we set up the study to examine the effect of cadmium on the B16 melanoma cell line, measuring viability by MTT and crystal violet (CV) tests, proliferation rate (CFSE), ROS production (DHR), migration (scratch test) and adhesion on matrigel. The results showed that non-cytotoxic doses of cadmium (ranging from 0,3-2,5µM) increased viability, proliferation, and migration of B16 cells, while at the same time slightly promoted ROS production. Otherwise, pronounced oxidative stress induced by higher doses (ranging from 5-20µM) led to cell death. Summary, these results showed that low doses of cadmium could upregulate B16 melanoma cell line growth and migration probably through moderate ROS/RNS generation and thus their ability to modulate relevant signaling pathways involved in cancer progression. Considering that, underlying mechanisms of Cd impact on melanoma cell lines proliferation, migration and invasiveness needs to be clarified in vitro, as well as its relevance for in vivo system.
