AB  - The genus Nepeta, belonging to the Lamiaceae family, encompasses a diverse group of plants with significant
biological activities attributed mainly to their iridoid compounds. This review provides a comprehensive analysis
of recent research on iridoid biosynthesis, regulation, and evolutionary aspects within the Nepeta genus. The biological
activities of Nepeta species, including repellent, phytotoxic, antimicrobial, and cytotoxic effects, have been extensively
investigated, highlighting the potential applications of iridoids. Over the past decade, significant progress has been
made in elucidating the molecular basis of iridoid biosynthesis and regulation, thanks to advancements in transcriptomics,
genomics, and metabolomics. The presence of distinct chemotype groups within Nepeta has been revealed,
characterized by their ability to produce both iridoid aglycones (nepetalactones) and glycosylated iridoids (IAs and
IGs), exclusively produce IGs, or lack iridoids. The identification of key enzymes involved in iridoid biosynthesis, such
as geraniol synthase (GES) and iridoid synthase (ISY), has played a crucial role in understanding the pathway. Furthermore,
the evolutionary aspects of the iridoid biosynthesis loss in some of the Nepeta taxa, and the association of iridoid
presence and content with the expression levels of specific genes, have been investigated. However, several areas remain
to be explored, including the final steps of iridoid aglycones biosynthetic branch, the production of iridoid glucosides,
the role of transcription factors in fine-tuning of iridoid biosynthesis, and the intricate interplay between biosynthetic
enzymes. Continued research in these areas will deepen our understanding of iridoid metabolism in Nepeta and unlock
their full potential in various fields, including pharmaceuticals, agriculture, and natural product-based industries.

AB  - Nepeta L. is the largest genus of the Lamiaceae family. The species of this genus are famous
for their biopesticidal potential and medicinal properties. Within the genus, we found
considerable variability in the qualitative and quantitative content of iridoid and phenolic
compounds. Using state-of-the-art metabolomics techniques (UHPLC/LTQ-Orbitrap MS,
GC/MS, UHPLC/DAD/(+/-)HESI-MS2), we comprehensively assessed the overall diversity of iridoids and phenolics in the selected chemodiverse Nepeta taxa at the intra-species level.
To reconstruct phylogenetic relations among the selected chemodiverse taxa from across the
Nepeta genus, genomic ITS molecular marker and two maternally inherited plastid DNA
regions (trnL-trnF and rbcL) were analyzed. A consensus phylogenetic tree was constructed
to interpret the distribution of the most abundant iridoid and phenolic compounds. It was
not possible to uncover clear regularities between phylogenetic relationship and chemotype
affiliation.

AB  - Introduction: Recent intensive research on the genus Nepeta has resulted in increasingly accumulated
information regarding the biosynthetic pathways and chemical evolution of iridoids; however, several in-
triguing aspects remain yet to be resolved. Our objective was to deeply investigate the molecular back-
ground of the diversity of iridoid compounds within the genus Nepeta, as well as regulative mechanisms
of their biosynthesis.
Methods: Leaves of greenhouse-grown plants were analysed for the composition of iridoid aglycones
(IAs) and iridoid glucosides (IGs) adopting non-targeted (UHPLC/LTQ Orbitrap MS n
, GC/MS) and targeted
(UHPLC/DAD/(±)HESI-MS 2 ) metabolomics approaches. Following RNA-Seq, transcriptomes of phylodi-
verse Nepeta taxa were searched for the presence/absence of iridoid-related biosynthetic genes. Co-ex-
pression patterns (qPCR) of biosynthetic genes and transcription factors were determined following
plants’ exposure to various environmental stresses.
Results: The comparison of metabolite composition among phylodiverse Nepeta taxa provided a sys-
tematic understanding of qualitative and quantitative composition of iridoids in leaves of the selected
Nepeta taxa. Mining of RNA-Seq data in search for iridoid biosynthesis-related genes pointed to signifi-
cant differences between taxa producing both IAs and IGs and those producing only IGs. Comparative
metabolomics and gene co-expression analysis provided new information about mechanisms of regu-
lation of iridoid biosynthesis.
Conclusion: Integration of data from several techniques analysed by different methods resulted in iden-
tifying key genes involved in the regulation of metabolic flux through the iridoid biosynthetic pathway
and offered an explanation why some Nepeta taxa produce only IGs, while others produce both IAs and
IGs. The obtained results pointed to new gene targets for improving iridoid production through biotech-
nology-based approaches.

AB  - Data on the distribution and diversity of iridoid glycosides (IGs) and iridoid aglycones (IAs) across the genus Nepeta (fam. Lamiaceae) are fragmentary; only rarely these two subgroups of iridoids are simultaneously analysed. Our ambition was to fulfil these knowledge gaps by adopting state-of-the-art metabolomics techniques (untargeted LC-HRMS and GC/MS) to comprehensively acquire, analyse and interpret the overall diversity of iridoids in selected phyllo-diverse Nepeta taxa, and thus facilitate further reconstruction of the molecular background of this diversity. Analyzed taxa were robustly grouped into three major chemotypes: (A) producers of both IGs and IAs; (B) taxa exclusively producing IGs; and (C) iridoid-lacking taxa. Furthermore, Nepeta species belonging to the chemotype A displayed significant heterogeneity in their IAs composition, and were roughly classified into several sub-chemotypes based on the stereochemistry of the major nepetalactone. No clear regularities between chemotype affiliation and phylogenetic relatedness, reconstructed adopting DNA barcoding tools, was revealed. Transcriptomes of leaves of distinguished Nepeta chemotypes were mined for the transcripts of iridoid-related biosynthetic genes, to facilitate the assessment of the factors responsible for the presence/absence of iridoids, or specific subgroups of iridoids, in these plants. Integration of metabolomics and transcriptomics data pointed to several key genes in the biosynthetic pathway as significant determinants of the iridoid diversity within the genus: geraniol synthase (GES), iridoid synthase (ISY), NAD-dependent nepetalactol-related short-chain-dehydrogenases/reductases (NEPSs), and latex protein-like genes (MLPLs). This study provided a framework to revisit some of the iridoid-metabolism-related ecological and chemical evolution theories, which is the course of our further work.

AB  - Numerous species belonging to the genus Nepeta are appreciated producers of various
phenolic and iridoid compounds that have tremendously vast applicative value in
pharmacology and agriculture but are also highly esteemed as chemotaxonomical markers.
Nepeta nuda is a widespread Eurasian species, found in forest edges and clearings. We have
collected leaf samples from 12 populations across the central Balkan Peninsula and
investigated their genetic and phytochemical variations. We used 3 genomic and 7 transcriptome-derived microsatellite markers and revealed relatively low variation within the
populations. Both qualitative metabolite profiling of methanol extracts (Q-TOFF LC/MS) and
essential oils (GC/MS) analysis pointed out to high inter- and intra-population variations,
especially regarding the content of iridoids. A UHPLC-QQQ-MS/MS approach targeted towards
quantification of the most abundant iridoids and phenolics was highly efficient in the
diversification of N. nuda populations.

AB  - Nepeta L. holds a prominent position as the largest genus in the Lamiaceae family and serves as the exclusive representative of the iridoid-lacking Nepetoidae subfamily which produces iridoids. The genus showcases a rich chemodiversity, encompassing taxa both with and without iridoids. Our study investigates the genetic basis of iridoid diversity using omics-guided and functional genomics approaches. We identified functional iridoid synthases (ISY) in iridoid-producing N. rtanjensis (NrIS2) and N. sibirica L. (NsIS), as well as in iridoid-lacking N. nervosa L. (NnIS). Remarkably, N. nervosa carries a dormant iridoid biosynthetic platform, suggesting a loss of iridoid production during its evolutionary history. Moreover, we explore regulatory mechanisms through comparative iridoid profiling and co-expression analysis of biosynthetic genes and transcription factors (TFs) under various stress conditions (e.g., dehydration, UV − B radiation, pathogens) or elicitors (MeJA). These mechanisms influence plant productivity and the presence/absence of iridoids or their specific groups (iridoid aglycones and iridoid glycosides). Our research offers valuable insights into the molecular mechanisms driving iridoid biosynthesis and the chemical evolution of iridoids within Nepeta. This study enhances our understanding of the intricate relationship between genetics and the environment, providing comprehensive insights into iridoid metabolism, chemical evolution, and ecology.

AB  - The genus Nepeta is characterized by a fascinating diversity of iridoid compounds. Some species within the genus are characterized by the presence of both iridoid glycosides (IGs) and iridoid aglycones (IAs), while others produce exclusively IGs, or completely lack iridoids. The absence of certain iridoids does not necessarily imply the absence of gene transcripts dedicated to the particular steps of their biosynthetic pathway. Studying the biosynthetic pathways of iridoids in chemodiverse Nepeta species, while taking into account tissue-specific profiles of metabolites and relevant genes expression, may give us further insight into the mechanisms of regulation of the biosynthesis of these compounds. In this work we performed a comparative transcriptomic analysis of glandular trichomes and abraded leaves of Nepeta rtanjensis, an IAs- and IGs –rich species, and of IGs-producing N. grandiflora. The results of this analysis provided us with very informative data that can be used for parallel study of iridoid-related biosynthetic genes (BGs) and transcription factors (TFs) of the two chemodiverse taxa. On the other hand, differentially expressed genes in the investigated tissues, enabled us to generate a list of candidate genes for yet unelucidated steps of the iridoid metabolic pathway. These results combined with state-of-the-art metabolomics data bring us closer to understanding the molecular background of iridoid diversity within the genus Nepeta, and point to the factors involved in the regulation of the two distinct branches of the iridoid biosynthetic pathway, leading to either IGs or IAs.

AB  - Within a large iridoids-lacking clade of Nepetoidae (Lamiaceae), the genus Nepeta stands out as an exclusive producer of these atypical monoterpenes with a unique stereochemistry. The genus Nepeta exhibits a high diversity of iridoids, with nepetalactones (NLs) predominating; however, a few Nepeta species lack these compounds. Comparative metabolomics, transcriptomics, gene co-expression, and phylogenetic analysis were conducted on the trans,cis-NL-rich N. rtanjensis Diklić & Milojević, the cis,trans-NL-dominant N. sibirica L., and the iridoid-lacking N. nervosa Royle & Bentham, revealing significant structural and functional differences in their iridoid synthases (ISY). N. rtanjensis was found to have two orthologues of ISY (NrISY1 and NrISY2), while N. sibirica and N. nervosa each had only one ISY (NsISY and NnISY, respectively). Although the phylogenetic analysis grouped NrISY1, NsISY, and NnISY with non-active Family 1 isoforms, organ-specific expression highlighted NrISY2, NsISY, and NnISY as promising ISY candidates. In vitro assays confirmed their function and NrISY2 was the most efficient in converting 8-oxogeranial to cis,trans-nepetalactol. However, 3D modeling and molecular docking analyses suggested that NnISY may have a unique cyclase activity. Likely, under in vivo conditions, the NnISY lacks substrates to catalyze, as a result of the loss of upstream geraniol synthase (GES) activity, which results in the loss of iridoids in N. nervosa. Presumably, some of the downstream pathway genes (NEPSs and MLPLs), essential for the NL biosynthesis, also underwent structural and activity changes in this species. Our ongoing work is conducted towards testing these robust hypotheses and re-establishing iridoid biosynthesis in N. nervosa.

AB  - The genus Nepeta, one of the largest genera of the Lamiaceae family (subfamily Nepetoidae, tribe Nepeteae) comprises around 250 herbaceous, perennial, rarely annual species, native to temperate Europe, Asia, North Africa, and the mountainous region of tropical Africa. Phytochemical investigations have revealed monoterpenes (iridoids), diterpenes, triterpenes, as well as phenolics, including flavonoids, to be the major specialized metabolites in these remarkable plants. The chemical richness of terpenoids and phenolics in catmints plays functional roles in nature, facilitating complex interactions with insects, phytopathogens, and other co-occurring plants. On the other hand, Nepeta species are used widely by humans as medicines, flavor additives, fragrances, and toys for cats, and their extensively studied bioactivities are mainly assigned to iridoid monoterpenoids nepetalactones. These compounds are proven repellents against insects (e.g. mosquitos, cockroaches, ticks, flies, mites, termites), and promising bioherbicides against a vast array of weeds (e.g. amaranth, ragweed, goosefoot, dandelion).
In spite of the plenitude of fundamental research that has documented the vast bioactive potential of Nepeta species, their commercial exploitation is restrained. There are several important factors that make the commercialization of Nepeta sp. most challenging, including limited bioresources, limited availability of bioactive compounds in purified forms, challenges to optimize the cultivation and standardize the quality of plant material, low number of studies focusing on the effects on non-target organisms and on the mutual relationships of individual compounds, instability and relatively low retainability of volatile nepetalactones, and others.
The challenge ahead is to provide sustainable sources of bioactive compounds of Nepeta sp. and thus establish a background for their commercialization. One of the solutions might be the creation of alternative sources of these biomolecules through synthetic biology approaches, thereby developing microbial or plant systems producing bioactive compounds of interest. Essential prerequisites to achieve this is the comprehensive and simultaneous acquiring, analyzing and interpreting the overall diversity of specialized metabolites within the genus Nepeta, reconstruction of the molecular background of the chemical diversity and elucidation of the biosynthetic pathways, as well as adopting the biotechnology tools to scale-up the production of the compounds of interest.

AB  - Iridoids, a class of atypical monoterpenes, exhibit exceptional diversity within the Nepeta genus (subfam. Nepetoidae, fam. Lamiaceae).The majority of these plants produce iridoids of the unique stereochemistry, with nepetalactones (NLs) predominating; however, a few Nepeta species lack these compounds. By comparatively analyzing metabolomics, transcriptomics, gene co-expression, and phylogenetic data of the iridoid-producing N. rtanjensis Diklić & Milojević and iridoid-lacking N. nervosa Royle & Bentham, we presumed that one of the factors responsible for the absence of these compounds in N. nervosa is iridoid synthase (ISY). Two orthologues of ISY were mined from leaves transcriptome of N. rtanjensis (NrPRISE1 and NrPRISE2), while in N. nervosa only one (NnPRISE) was identified, and it was phylogenetically closer to the representatives of the Family 1 isoforms, designated as P5βRs. Organ-specific and MeJA-elicited profiling of iridoid content and co-expression analysis of IBG candidates, highlighted NrPRISE2 and NnPRISE as promising candidates for ISY orthologues, and their function was confirmed using in vitro assays with recombinant proteins, after heterologous expression of recombinant proteins in E. coli and their His-tag affinity purification. NrPRISE2 demonstrated ISY activity both in vitro and likely in planta, which was supported by the 3D modeling and molecular docking analysis, thus reclassification of NrPRISE2 to NrISY is accordingly recommended. NnPRISE also displays in vitro ISY-like activity, while its role under in vivo conditions was not here unambiguously confirmed. Most probably under in vivo conditions the NnPRISE lacks substrates to act upon, as a result of the loss of function of some of the upstream enzymes of the iridoid pathway. Our ongoing work is conducted towards re-establishing the biosynthesis of iridoids in N. nervosa.

AB  - Nepeta nuda L. (familija Lamiaceae) je, pored N. rtanjensis i N. cataria, jedna od tri vrste roda Nepeta koje rastu u Srbiji, naseljavajući prvenstveno sušna staništa na obodima šuma planinskih masiva. Poput većine predstavnika ovog roda, vrstu N. nuda karakteriše prisustvo različitih specijalizovanih metabolita, prvenstveno iridoida i fenolnih jedinjenja. Listovi jedinki N. nuda, prikupljeni su na različitim lokalitetima u istočnoj Srbiji: Izvor (Paraćin), Rtanj, Ozren i Barta Berilovac (Stara planina). Kvalitativni i kvantitativni hemijski sastav prikupljenog biljnog materijala analiziran je UHPLC/DAD/(±)HESI-MS2 metodom, usmerenom na dominantna bioaktivna jedinjenja iz grupa iridoida i polifenola. Rezultati metaboličkog profilisanja iridoida pokazali su da jedinke iz analiziranih populacija dominantno produkuju iridoidni glikozid - epideoksiloganinsku kiselinu, dok aglikoni iridoida nisu detektovani. Predstavnici svih ispitivanih populacija od fenolnih jedinjenja dominantno sintetišu ruzmarinsku kiselinu, dok su u tragovima detektovana jedinjenja kao što su kafeinska kiselina, izokvercitrin i astragalin. Sadržaj analiziranih jedinjenja varira između analiziranih populacija N. nuda. Populacija sa lokaliteta Balta Berilovac izdvaja se po visokom sadržaju epideoksiloganinske kiseline, što je čini interesantnom za dalja ispitivanja bioloških aktivnosti.
AB  - Nepeta nuda L. (фамилија Lamiaceae) је, поред N. rtanjensis и N. cataria, једна од три врсте рода Nepeta које расту у Србији, насељавајући првенствено сушна станишта на ободима шума планинских масива. Попут већине представника овог рода, врсту N. nuda карактерише присуство различитих специјализованих метаболита, првенствено иридоида и фенолних једињења. Листови јединки N. nuda, прикупљени су на различитим локалитетима у источној Србији: Извор (Параћин), Ртањ, Озрен и Барта Бериловац (Стара планина). Квалитативни и квантитативни хемијски састав прикупљеног биљног материјала анализиран је UHPLC/DAD/(±)HESI-MS2 методом, усмереном на доминантна биоактивна једињења из група иридоида и полифенола. Резултати метаболичког профилисања иридоида показали су да јединке из анализираних популација доминантно продукују иридоидни гликозид - епидеоксилоганинску киселину, док агликони иридоида нису детектовани. Представници свих испитиваних популација од фенолних једињења доминантно синтетишу рузмаринску киселину, док су у траговима детектована једињења као што су кафеинска киселина, изокверцитрин и астрагалин. Садржај анализираних једињења варира између анализираних популација N. nuda. Популација са локалитета Балта Бериловац издваја се по високом садржају епидеоксилоганинске киселине, што је чини интересантном за даља испитивања биолошких активности.

Sadržaj analiziranih jedinjenja varira između analiziranih populacija N. nuda. AB  - The aim of this study was to determine qualitative composition of methanol-soluble metabolites in leaves
from in vitro-grown plants of phylogenetically distinct Nepeta species producing iridoids (N. sibirica L., N.
spicata Benth., N. argolica Bory & Chaub., and N. rtanjensis Diklić & Milojević) and iridoid-lacking N. nervosa
Royle ex Benth., adopting UHPLC-LTQ OrbiTrap MS technique. Totally, 52 compounds were identified
based on their monoisotopic masses, MSn fragmentation, and previously reported MS data and they were
clustered into four major groups: phenolic acid derivatives (23 compounds), flavonoids (10 compounds),
iridoids (13 compounds), and 6 compounds belonging to other classes. Both phenolic and iridoid profiles
of the tested species were found to be very diverse regarding both qualitative content of compounds and
their relative representation in extracts. Rosmarinic, 5-O-cafeoylquinic, and caffeic acids were found to be
the dominant phenolics in all analyzed species, especially in N. argolica, N. rtanjensis, and N. nervosa.
Iridoids were present in the form of glucosides and the most abundant compounds from this group were
epideoxyloganic acid, geniposide, and aucubin. Although iridoids were not detected in N. nervosa extracts,
this species was revealed as a rich source of phenolic acids and flavonoids. Conversely, N. spicata was found
to produce iridoids but negligible amounts of phenolic compounds. Extracts of two closely related endemic
species originating from the Balkan Peninsula, N. argolica and N. rtanjensis, were very similar in both
phenolic and iridoid profiles. The UHPLC-LTQ OrbiTrap MS method, in combination with chemometrics
data processing, was proved to be simple, rapid, and accurate strategy for unambiguous taxonomic
determination and chemical differentiation of Nepeta species.

AB  - Род Nepeta (Lamiaceae) обухвата око 300 биљних врста које продукују различите специјализоване метаболите, при чему представља скоро једини род подфамилије Nepetoidae који производи иридоиде. Биљке рода Nepeta продукују иридоидне гликозиде и једноставне иридоиде – агликоне. Ова једињења се сматрају значајним за различите биолошке активности врста рода Nepeta, укључујући антимикробно, антитуморско, алелопатско и репелентно дејство. Извршена је детаљна фитохемијска анализа у циљу идентификације и квантификације иридоида одабраних представника различитих врста рода Nepeta: N. rtanjensis, N. laevigata, N. grandiflora, N. nervosa, N. parnassica, N. ernesti-mayeri, N. nuda, N. sibirica, N. stewartiana, N. nervosa. На основу добијених резултата извршено је раздвајање представника овог рода у неколико карактеристичних хемотипова, а на основу присуства иридоидних гликозида и/или иридоидних агликона. Спроведена су детаљна истраживања биосинтетског пута у оквиру рода Nepeta, са посебним акцентом на откривању ензима одговорних за реакције модификације иридоидног скелета (нпр. оксидације, редукције, гликозилације). Поред тога, описани су неки од механизама регулације биосинтезе иридоида током одговора биљака на абиотичке и биотичке факторе. Добијени резултати уз примену различитих биотехнолошких метода омогућиће таргетиране измене метаболизма у циљу повећане продукције иридоида од значаја.

Biljke roda Nepeta produkuju iridoidne glikozide i jednostavne iridoide – aglikone. Ova jedinjenja se smatraju značajnim za različite biološke aktivnosti vrsta roda Nepeta, uključujući antimikrobno, antitumorsko, alelopatsko i repelentno dejstvo. Izvršena je detaljna fitohemijska analiza u cilju identifikacije i kvantifikacije iridoida odabranih predstavnika različitih vrsta roda Nepeta: N. rtanjensis, N. laevigata, N. grandiflora, N. nervosa, N. parnassica, N. ernesti-mayeri, N. nuda, N. sibirica, N. stewartiana, N. nervosa. AB  - Many Nepeta species (fam. Lamiace, subfam. Nepetoideae, tribe Nepetae) are well known for their
pharmacological properties and are widely used in folk medicine as diuretic, diaphoretic, anti-tussive, antispasmodic, anti-asthmatic, febrifuge and sedative agents, against gastrointestinal and respiratory
hyperactive disorders (Formisano et al., 2011). They also show biopesticide potential and display
behavioural effect on cats. Various biological activities of Nepeta species are closely related to their main
secondary metabolites, among which terpenes and phenolics predominate. Two herbaceous perennial
Nepeta species investigated within the present study are widely distributed in EuroAsia. N. cataria L. is
spreaded from Central Europe to the Iranian plateaus and in Central Asia, and N. nuda L. grows throughout
Europe and European part of Russia, the Crimea, in the Dnieper area and on Carpathians in Ukraine, as well
as in Siberia (Boikova and Grishkina, 2019).
Untargeted metabolomics approach adopting UHPLC/LTQ Orbitrap−MSn revealed iridoids and phenolic
acids as the major groups of compounds in methanol extracts of both N. cataria and N. nuda.
UHPLC/(±)HESI−MS2 profiling of major compounds from these groups of metabolites revealed that
rosmarinic acid was the major phenolic acid in both species, while from the group of iridoids, 1,5,9-
epideoxyloganic acid predominated. Methanol extract of N. cataria was rich in iridoid aglycone cis,transnepetalactone. Based on the bioautography assays, methanol extracts of N. cataria and N. nuda show
moderate α-amylase activity, and display significant antioxidative activity, which is primarily ascribed to
high content of rosmarinic acid.

AB  - Nepeta nuda L. (family Lamiaceae) is a widespread species characterized by the prevalence of bioactive specialized metabolites from the groups of iridoids and phenolic. Analyzed plants were grown in vitro and in greenhouse from seeds collected from eastern Serbia (“Debeli Lug”), obtained from other research groups (“Rhodopes”, Bulgaria), or commercially purchased (“Jellito”, Germany). The chemical composition of N. nuda leaves was analyzed using the UHPLC/DAD/(±)HESI±МS2 method, targeted towards dominant iridoid compounds. Both in vitro and greenhouse grown plants are rich in iridoid glycoside 1,5,9-epideoxyloganic acid (1,5,9– eDLA), and among analysed accessons N. nuda “Debeli Lug” was pointed out as the most productive one. When it comes to iridoid aglycones, it is noticeable that N. nuda “Jelitto” biosynthetizes cis, trans- isomer of nepetalactone (NL) while N. nuda “Rhodopes” and N. nuda “Debeli Lug” possess cis,cis-NL. Relative expression of early biosynthetic genes- EBGs (NnGPPS, NnGES, NnG80, Nn8HGO), in plants grown in vitro, is much higher in N. nuda “Debeli Lug“ accession, compared to N. nuda “Rhodopes” and N.nuda “Jellito”. In greenhouse grown plants, there is no particular regularity when it comes to the expression of EBGs in three N. nuda accessions. NEPS enzymes seem to be one of the key factors determining the metabolic flux and productivity of N. nuda. While plants grown in vitro and in greenhouse both express NnNEPS2, only plants grown in vitro express NnNEPS1.2. It is also noticeable that N. nuda “Debeli Lug“ accession displays the highest NnNEPS2 expression, and the lowest NnNEPS1.2 expression.

AB  - Drough-stress conditions lead to excessive production of reactive oxygen species (ROS) and
oxidative damage in plants. Plants display various defense mechanisms for survival, including the
activation of antioxidant enzymes and the production of low molecular weight non-enzymatic
antioxidants like phenolics. In this work, roots of four weeks old Nepeta nervosa plants were exposed to PEG solutions with osmotic pressure of −3 MPa for one, three and six days. Fresh weight
(FW) and relative water content (RWC) of N. nervosa plants were significantly reduced compared
to the corresponding control - up to 68% (FW) and up to 35% (RWC). In leaves, the content of rosmarinic and chlorogenic acids, dominant phenolic compounds, as well as of less abundant ferulic
and caffeic acids, was dependent on the duration of PEG-induced drought stress. The content of
rosmarinic acid in control plants achieved an average value of 18.27 µg/mg FW, while at PEG-treatments it increased by 355% (3 days of PEG-treatment). Among all examined antioxidative enzymes
in leaves of PEG treated N. nervosa plants, superoxide dismutases (SOD) were the most active (an
increase of 853% on day 6). PEG treatment also caused an increase in peroxidase (POX) activity in
leaves of N. nervosa plants (108% increase on day 3). Since catalase (CAT) activity was significantly decreased on PEG treatment, except on day 1, it could be presumed that H2O2 elimination is
performed mostly by POX. This work is a useful contribution to the development of a strategy for
overcoming oxidative stress in Nepeta species, especially in arid and semiarid regions.

AB  - Nepeta L. is the largest genus of the Lamiaceae family, and the exclusive representative of the
subfamily Nepetoidae which produces iridoids. However, the genus Nepeta comprises both taxa
producing iridoids (iridoid aglycones-IAs and/or iridoid glucosides-IGs), and those lacking iridoids.
Combining omics-guided approach and functional genomics in elucidating the genetic background
of the iridoid diversity within the genus Nepeta, we provide novel insights into molecular
mechanisms driving the iridoid biosynthesis, and thus into chemical evolution of iridoids within
the genus Nepeta. Multidisciplinary experimental approach is here explained on the example
of iridoid synthases (IS), functionally characterized by in vitro enzymatic assays using recombinant
proteins heterologously expressed in Escherichia coli. Functional ISs are expressed in both
iridoid-producing N. rtanjensis (NrIS2) and N. sibirica L. (NsIS), as well as in iridoid-lacking N. nervosa
L. (NnIS). N. nervosa possesses an iridoid biosynthetic platform, but inactive one, most likely due
to the “switching off” or “silencing” of some gene(s) upstream in the pathway, which suggests that
this species has lost the ability to produce iridoids during the evolution. Furthermore, comparative
iridoid profiling and co-expression analysis of biosynthetic genes and transcription factors (TFs)
during the exposure of chemodiverse Nepeta taxa to environmental stresses (e.g. dehydration,
UV−B radiation, pathogens) or elicitors (MeJA), facilitated the explanation of regulatory mechanisms
determining the productivity of plants, as well as of factors responsible for the presence/

AB  - Trichoderma spp. have been extensively explored and used in agriculture due to their well-known biological control mechanisms. Fungi of the genus Trichoderma produce secondary metabolites which affect plant metabolism by stimulating the production of defense-related compounds and increasing the antioxidant capacity through enhanced polyphenol content and elevated activity of antioxidant enzymes, a result of the excess production of reactive oxygen species. In this work, Nepeta sibirica plants were grown in vitro on solid medium treated with two Trichoderma strains (T. viride and T. harzianum). After three and six days of treatment, leaves were metabolically profiled for major phenolic acids (chlorogenic, caffeic, and rosmarinic acid), and iridoids content (cis,trans-nepetalactone and 1,5,9-epideoxyloganic acid), in parallel with the expression analysis of nepetalactone biosynthetic genes and regulatory genetic elements-transcription factors (TFs). Both fungal strains induced changes in phenolic acids production, while only T. harzianum induced elevated levels of iridoids. Biosynthetic genes GPPS, and IS, as well as TF MYC2, were the only genes with expression levels not affected by the treatments with micro fungi. The response of antioxidant enzymes in N. sibirica leaves was also studied. The most active antioxidant enzyme following the infection with T. viride and T. harzianum was peroxidase (POX). Catalase (CAT) and superoxide dismutase (SOD) activity were also affected by the treatments. Results indicate a possibility of using Trichoderma infection in N. sibirica to elicit the production of biologically active defense compounds.

AB  - Nepeta species (fam. Lamiace, subfam. Nepetoideae, tribe Nepetae) are well known for their
pharmacological properties, biopesticide potential and behavioral effects on cats. Different biological activities of Nepeta species are closely related to their secondary metabolites, among which
iridoids predominate. Research objective of this study was to determine profiles of methanol-soluble iridoids in leaves of greenhouse-grown plants of 10 phylodiverse Nepeta species: N. grandiflora
M. Bieb., N. laevigata (D. Don) Hand.-Mazz., N. nervosa Royle ex Benth., N. stewartiana Diels, N. nuda
L., N. parnassica Heldr. & Sart., N. rtanjensis Diklić & Milojević, N. ernesti-mayeri Diklić & V. Nikolić, N.
cataria L., and N. sibirica L. Untargeted metabolomics approach adopting UHPLC/LTQ Orbitrap MSn
analysis in a negative ionization mode revealed the presence of totally 15 iridoids in samples, of
which 13 were glucosides and two were aglycons of iridoids. Iridoid compounds were identified
based on their monoisotopic masses, MSn
 fragmentation, and previously reported MS data. The
results indicated that the iridoid profiles of the investigated species are very diverse both in terms
of qualitative content of the compounds and their relative abundance in extracts. Among analysed Nepeta species, Balkan Peninsula endemics (N. rtanjensis, N. parnassica, and N. ernesti-mayeri)
and N. cataria, are especially rich in isomers of 1,5,9-epideoxyloganic acid. 5-Deoxylamiol, nepetariaside, nepetanudoside B, and ajugol were detected in majority of analyzed species. The UHPLC-LTQ OrbiTrap MS analysis, in combination with chemometric data processing, has proven to
be an efficient tool for chemical differentiation between Nepeta taxa, and could aid the efforts to
unravel the chemical evolution within the genus.

AB  - The genus Nepeta L. comprises medicinal plant species that synthetize terpenes and phenolics of therapeutic value. Considerable variability in qualitative and quantitative content of iridoid
monoterpenoids has been detected within the genus Nepeta, which comprises iridoid produc-ing
and non-producing species. Nepeta rtanjensis Diklić & Milojević is an endemic plant of the Balkan Peninsula, which produces both iridoid aglycones (IAs) and iridoid glucosides (IGs). N. nervosa
Royle ex Benth., endemic to the Himalayas of Kashmir, India, and Paki-stan, is iridoid-lacking species. This study aimed to comparatively analyze the effect of me-thyl jasmonate (MeJA), an elicitor of terpene production in many plant species, on iridoid bio-synthesis at the molecular level.
Iridoid content in MeJA-treated N. rtanjensis and N. nervosa plants under in vitro conditions was
analysed in parallel with the co-expression patterns of iridoid related biosynthetic genes (GPPS,
GES, G80, 8HGO, 7DLGT, IS, and NEPS), major latex protein-like gene (MLPL), and genes coding for
different transcription factors (MYC2, YABBY5, COI1, and JAZ3). In general, the exogenous application of 250 µM MeJA for 2 h, 24 h and 72 h resulted in an increased accumulation of targeted IAs
(trans, cis-nepetalactone and 5,9-dehydronepetalactone) and IGs (1,5,9-epideoxyloganic acid) in
leaves of N. rtanjensis, as a result of elevated expression of the key biosynthetic genes. Interestingly,
although the majority of iridoid-related biosynthetic genes and TFs were identified in transcriptome of N. nervosa leaves, and the expression of some of them (NEPS and JAZ3) is up-regulated
follow-ing MeJA treatment, no iridoids were accumulated in leaves of N. nervosa.
