AB  - Pollution of surface waters still represents one of the major environmental challenges in the Republic of Serbia, where wastewaters are discharged directly into the recipients without proper treatment. Within the last decade, we have assessed the microbiological water quality of more than 100 sites situated at different water bodies in Serbia. Quality was determined using faecal indicator bacteria, while microbial source tracking was employed to assess the source of pollution. Almost 50 % of the investigated sites were characterized by critical or even higher level of faecal contamination indicating that untreated wastewaters indeed represent significant pollution pressure on surface waters. Human-associated markers were prevalent in samples from the majority of contaminated sites but the source of pollution was not exclusively human-associated. 
The impact on the water quality of the Danube River was demonstrated in our previous research conducted at the whole river level within the Joint Danube Surveys. In the river stretch from Novi Sad to its confluence with the Velika Morava River, all the midstream samples were critically polluted. In this section, the highest level of pollution was recorded downstream of Belgrade. As ultimate recipients of wastewaters, Danube and its largest tributary Sava currently represent the only solution for disposing of wastewaters originating from the Serbian capital‘s 1,700,000 inhabitants. Such kind of disposal rises additional issues such as antimicrobial resistance and presence of infectious agents in water. For instance during COVID-19 pandemic, we have demonstrated that SARS-CoV-2 RNA can be detected even in surface waters of the Danube River at the sites receiving high wastewater loads from Belgrade which was the unique case for the Basin. 
Despites its extreme importance in this case, wastewater-based epidemiology is neglected in our country and hereby we would like to emphasize the need for implementation of program of such kind in Serbia.

– akumulacije) na teritoriji Specijalnog rezervata prirode „Uvac“ obuhvatila je određivanje
koncentracije ukupnih koliforma i Escherichia coli pomoću Colilert-18 sistema. U slučaju
reka uzimani su površinski uzorci vode (dubina 0,2 m), dok su u slučaju akumulacija uzeti
uzorci sa više dubina. Najveći pritisci fekalnog zagađenja zabeleženi su na lokalitetima
Uvac-Čedovo i Vapa-Čedovo (umeren ekološki status – III klasa). Na rekama Tip 6
zabeležena je I ili II klasa zavisno od datuma uzorkovanja.

reserve „Uvac“ included determination of total coliforms and Escherichia coli
concentration using the Colilert-18 system. Surface water samples (depth 0.2 m) were taken
for rivers, while for reservoirs samples were taken from several depths. The highest
pressures of faecal pollution were recorded at sites Uvac-Čedovo and Vapa-Čedovo
(moderate ecological status, III class). In the case of the remaining analysed sites, I or II
class was recorded depending on the sampling date.

AB  - Assessment of impact of pollution in the environmental studies requires multi-endpoints approach to properly
link cause and effects of pollution with focus on chemical pollution. In this sense systematic weight-of-evidence
approach (WoE) with multiple lines of evidence (LoEs) is preferable to apply. The WoE approach highlights the
importance to identify strengths and weaknesses of used LoEs. Therefore, in this study we have tested efficacy
of genotoxicological endpoints as one of the LoEs in the in situ assessment of pollution effects in the freshwater
ecosystems using Alburnus alburnus (bleak) as a bioindicator species. Additional LoEs that were used in the
study are: component-based methods for the assessment of SumTU in water based on monitoring data of the
Serbian Environmental Protection Agency (SEPA), effect based methods employing in vitro genotoxicological
analyses of Joint Danube Survey 4 (JDS4) water extracts and field derived species inventories for the
assessment and indication of ecological status/potential based on SEPA and JDS4 data. The study was
conducted within the JDS4 campaign at nine sampling sites at the Tisa, Sava, Velika Morava and Danube rivers
in the Republic of Serbia. In the case of three sampling sites, the results were uniform, meaning that all four
LoEs pointed to pollution pressure. The differences in the LoEs outcomes for other sites indicated the
importance of multiple LoEs approach for proper identification of ecological impact. In the case of current
study we have identified comet and micronucleus assay to be appropriate for the genotoxicological assessment
in the in situ studies due to high sensitivity in discrimination of sites in relation to pollution intensity, while
RAPD analysis to be more suitable for controlled ex situ investigations.

AB  - Pollution of natural water bodies is one of the biggest challenges in developing countries, such as
Serbia, where wastewater is discharged directly into the recipients without proper treatment and
poses serious threats to surface water quality, general safety and environmental health. Numerous
pathogenic microorganisms, bacteria, viruses, protozoa and helminthes may reach surface waters
and eventually contaminate groundwater. Despites its extreme importance in this case,
implementation of wastewater-based epidemiology is a challenge for developing countries, as the
majority of households are not connected to sewerage systems. Within our research, we have placed
major focus on recipients of wastewaters as an alternative of targeting raw wastewater. During the
fourth COVID-19 wave in Serbia that started in late February 2021 multiple water samples were
collected at 12 sites at Sava and Danube Rivers in the Belgrade city area which are under the impact
of untreated wastewaters.
SARS-CoV-2 RNA was quantified using RT-qPCR with primer sets targeting nucleocapsid (N1 and N2)
and envelope (E) protein genes. Microbiological (standard fecal indicator bacteria and microbial
faecal source tracking markers), epidemiological, physico-chemical and hydro-morphological
parameters were analysed in parallel. Out of 44 samples analyzed, 31 were positive for at least one
of the target regions of SARS-CoV-2. The results indicated that surveillance of SARS-CoV-2 RNA in
surface waters in context with the large amount of epidemiological and environmental metadata can
be used as epidemiological early-warning tool in countries with poor wastewater treatment.

AB  - Lake Ludaš is a protected nature reserve in Serbia that has been the focus of scientific interest, due to
perpetual water quality deterioration that has led to consistent cyanobacterial blooms for almost five decades. To describe the possible effects of cyanotoxins on fish (Carrasius gibelio) inhabiting Lake Ludaš, several examinations were performed in November of 2018, including: assessment of water quality (physical and chemical parameters, as well as water microbiology), and assessment of several tissues of C. gibelio (micronucleus test, comet assay, and histopathological analyses). The water quality of Lake Ludaš has been categorized as class III, indicating moderate pollution. Physical and chemical parameters from November of 2018 seemed to be within acceptable levels. Examined fish tissues showed signs of DNA damage (highest in the blood, followed by the liver and gills), and histological alterations (in the liver
and gills). Although other xenobiotics can induce similar changes, cyanotoxins could be the most likely cause of the observed alterations in C. gibelio tissues. The present study demonstrates the need to monitor this important wetland of international significance, since the living organisms in Lake Ludaš are showing signs of environmental stress suggesting that the preservation and natural balance of this ecosystem could be in danger.

AB  - Environmental studies which aim to assess the ecological impact of chemical and other types of pollution should employ a complex weight-of-evidence approach with multiple lines of evidence (LoEs). This study focused on in situ genotoxicological methods such as the comet and micronucleus assays and randomly amplified polymorphic DNA analysis as one of the multiple LoEs (LoE3) on the fish species Alburnus alburnus (bleak) as a bioindicator. The study was carried out within the Joint Danube Survey 4 (JDS4) at nine sites in the Danube River Basin in the Republic of Serbia. Out of nine sampling sites, two were situated at the Tisa, Sava, and Velika Morava rivers, and three sites were at the Danube River. The three additionally employed LoEs were: SumTUwater calculated based on the monitoring data in the database of the Serbian Environmental Protection Agency (SEPA) (LoE1); in vitro analyses of JDS4 water extracts employing genotoxicological methods (LoE2); assessment of the ecological status/potential by SEPA and indication of the ecological status for the sites performed within the JDS4 (LoE4). The analyzed biomarker responses in the bleak were integrated into the unique integrated biomarker response index which was used to rank the sites. The highest pollution pressure was recorded at JDS4 39 and JDS4 36, while the lowest was at JDS4 35. The impact of pollution was confirmed at three sites, JDS4 33, 40, and 41, by all four LoEs. At other sampling sites, a difference was observed regarding the pollution depending on the employed LoEs. This indicates the importance of implementing a comprehensive weight-of-evidence approach to ensure the impact of pollution is not overlooked when using only one LoE as is often the case in environmental studies.

AB  - In this study, the possible 'vector effect' within the exposure of Mediterranean mussels (Mytilus galloprovincialis) to polystyrene microplastics with adsorbed fluoranthene was investigated by applying the multibiomarker approach. The major focus was placed on genotoxicological endpoints as to our knowledge there are no literature data on the genotoxicity of polystyrene microparticles alone or with adsorbed fluoranthene in the selected experimental organisms. DNA damage was assessed in haemocytes by comet assay and micronucleus test. For the assessment of neurotoxicity, acetylcholinesterase activity was measured in gills. Glutathione S-transferase was assessed in gills and hepatopancreas since these enzymes are induced for biotransformation and excretion of lipophilic compounds such as hydrocarbons. Finally, differences in physiological response within the exposure to polystyrene particles, fluoranthene, or particles with adsorbed fluoranthene were assessed by the variation of heart rate patterns studied by the noninvasive laser fibre-optic method. The uniform response of individual biomarkers within the exposure groups was not recorded. There was no clear pattern in variation of acetylcholinesterase or glutathione S-transferase activity which could be attributed to the treatment. Exposure to polystyrene increased DNA damage which was detected by the comet assay but was not confirmed by micronucleus formation. Data of genotoxicity assays indicated differential responses among the groups exposed to fluoranthene alone and fluoranthene adsorbed to polystyrene. Change in the heart rate patterns within the studied groups supports the concept of the Trojan horse effect within the exposure to polystyrene particles with adsorbed fluoranthene.

DNA damage was assessed in haemocytes by comet assay and micronucleus test. For the assessment of neurotoxicity, acetylcholinesterase activity was measured in gills. Glutathione S-transferase was assessed in gills and hepatopancreas since these enzymes are induced for biotransformation and excretion of lipophilic compounds such as hydrocarbons. Finally, differences in physiological response within the exposure to polystyrene particles, fluoranthene, or particles with adsorbed fluoranthene were assessed by the variation of heart rate patterns studied by the noninvasive laser fibre-optic method. The uniform response of individual biomarkers within the exposure groups was not recorded. There was no clear pattern in variation of acetylcholinesterase or glutathione S-transferase activity which could be attributed to the treatment. Exposure to polystyrene increased DNA damage which was detected by the comet assay but was not confirmed by micronucleus formation. Data of genotoxicity assays indicated differential responses among the groups exposed to fluoranthene alone and fluoranthene adsorbed to polystyrene. Change in the heart rate patterns within the studied groups supports the concept of the Trojan horse effect within the exposure to polystyrene particles with adsorbed fluoranthene.",

According to World Health Organization, 70-95% of the population chooses folk medicine as their primary
approach for health maintenance. In addition to their healing properties and application in folk medicine,
Lamiaceae plants are often valued in cookery as spices and food preservatives.
OBJECTIVES
Hence, the aim of this study was to examine the antibiofilm activity of 12 ethanolic extracts of Lamiaceae species
on biofilm formation and the degradation of existing biofilm of Pseudomonas aeruginosa PAO1 (ATCC 15692).
METHODS
The crystal violet staining method was used to evaluate the newly formed bacterial biofilms. Four concentrations

AB  - Within the Joint Danube Survey 4 (JDS4) (2019) surface water samples
were collected at 24 localities along the Danube River and were extracted by
horizon large volume solid-phase extraction (LVSPE) technique. LVSPE
technique provided 25,000x relative enrichment factor (REF) of the river
samples. The bioactivity of the LVSPE samples was investigated employing
prokaryotic SOS/umuC assay (S9+ and S9-) and battery of bioassays
analyzing different endpoints – cytotoxicity (MTS assay), genotoxicity
(comet assay), cell cycle analyses of ZFL cells and embryo toxicity (zFET) in
eukaryotic models (zebrafish liver cell line – ZFL and Danio rerio embryos –
zebrafish). In the case of the SOS/umuC assay none of the tested samples
(REF100) has induced genotoxic effect. The cytotoxic effect was detected for
11 samples at REF100 (MTS assay). The comet assay results indicated
genotoxic potential for nine samples – one in the Upper Danube, six in the
Middle Danube and two in the Lower Danube. The most potent were
JDS4-37 and JDS4-41 samples collected at two sites in the Serbian part of
the Danube River. These samples were analyzed at REF13. The analysis of
the cell cycle was performed in the case of eight samples for which were
previously recorded genotoxic effect. For one sample (JDS4-41) was found
the G1 phase cell cycle arrest. The zFET assay was performed for all the
samples at REF100 and there was no embryo toxicity or teratogenicity
recorded. The results obtained within this study indicate a higher sensitivity
of eukaryotic models compared to prokaryotic SOS/umuC assay. Likewise, it
was found that most of the genotoxic samples originate from the middle
part of the Danube.

The increasing bacterial resistance to antibiotics poses a serious global threat. For that reason, the research into plant-derived antibacterial agents has become progressively more interesting, particularly in regard to their capacity to prevent the delay or inability of wound closure and healing.
OBJECTIVES
The aim of this study was to evaluate i) the antibacterial potential of ethanolic extracts (70% ethanol) of 18
Lamiaceae species against four Gram-negative and three Gram-positive bacterial strains; ii) the effect of these extracts on Pseudomonas aeruginosa PAO1 infection of human lung fibroblasts (MRC-5), and iii) the effect on MRC-5 cell migration.
METHODS
The antibacterial activity was tested by minimal inhibitory concentration (MIC) assay. The effect on bacterial infection of MRC-5 cells was determined using the invasion assay, while the cell migration was assessed with Scratch assay.
RESULTS
The tested Gram-positive bacteria, especially Bacillus subtilis, are more sensitive to the effects of ethanolic extracts than the Gram-negative bacteria. Moreover, the tested extracts significantly inhibited the invasion of Pseudomonas aeruginosa PAO1 during MRC-5 infection, and most of them also displayed promising stimulating potential on MRC-5 migration in a disrupted cell monolayer. Finally, the results of IBR (integrated biomarker response) analysis highlighted Salvia officinalis ethanolic extract as the most active one, which is in accordance with the existing data suggesting that this plant represents an effective antibacterial and wound-healing agent commonly used in traditional medicine. Altogether, these results are expected to foster further studies that will focus on combating bacterial resistance to antibiotics using natural products.

AB  - The middle section of the Danube River basin in the previous three Joint
Danube Surveys (JDS) was found to be under intensive pollution, mainly
affected by wastewaters. As a consequence of the lack of wastewater
treatment plants, complex xenobiotics mixtures may get into surface water
and decrease water quality and organisms’ health. Some xenobiotics may
induce different types of DNA damage. Hence, DNA damage could be a
useful biomarker in the detection of “early warning signals” of organisms’
exposure to genotoxic compounds, while the usage of bioassays batteries
can provide better insight into a genotoxic potential. This study was
conducted in 2019, during the JDS4 expedition, to assess the genotoxic
potential of nine sites on the section of the Danube River which goes
through Serbia and its major tributaries: the Sava, Tisza and Velika Morava.
Blood and muscle of Alburnus alburnus (bleak) were used for genotoxicity
assessment by comet, micronucleus and RAPD assays. In comet assay, the
highest level of DNA damage was recorded at the Velika Morava mouth (JDS
39) and on the Danube River, downstream Radujevac (JDS 41). The highest
frequency of micronucleus was recorded at the Sava mouth (JDS 36) and on
the Danube River, downstream Pančevo (JDS 37). The lowest level of DNA
damage in both tests was recorded on the Sava, site Jamena (JDS 35). The
RAPD analysis distinct three major groups: Tisza mouth (JDS 33) and sites
on the Sava (JDS 35, JDS 36), then two sites on the Danube River (JDS 37,
JDS 41) and one site on the Danube River, Ram (JDS 40) with the Velika
Morava mouth (JDS 39). Analyzed bioassays showed different sensitivity
where the comet assay had the highest potential in discrimination of sites
based on genotoxicity. Bleak was proved to be a reliable bioindicator in eco/

Danube Surveys (JDS) was found to be under intensive pollution, mainly
affected by wastewaters. As a consequence of the lack of wastewater
treatment plants, complex xenobiotics mixtures may get into surface water

AB  - Within the past decade, our research group placed great effort in
exploration of the impact of treated/untreated wastewaters on aquatic
ecosystems as less than 13 % of collected municipal wastewaters are
processed before their release to receiving waters in Serbia. Having in mind
that more than 90% of the rivers within the territory of Serbia belong to the
Danube River Basin, our major focus is naturally on the large transnational
waters such as the Danube and Sava rivers. Depended on the concept of the
study and hydro-morphological characteristics of the sites, various
approaches have been employed for in situ assessment of eco/geno-toxicity
which use different organisms in passive and active biomonitoring. Aquatic
organisms of different trophic levels have diverse life strategies, metabolism
pathways, and consequently, they have a different response to pollutant
pressure. In eco/geno-toxicological studies we have successfully employed
several species of freshwater mussels (Unio tumidus, Unio pictorum and
Sinanodonta woodiana) as well as few economically important fish species
(common carp (Cyprinus carpio), freshwater bream (Abramis brama), bleak
(Alburnus alburnus, chub (Squalius cephalus)).Comet assay is our primary
choice for the assessment of DNA damage considering its sensitivity and
cost effectiveness. Selection of certain types of tissues for the assessment
(such as blood or haemolymph), which do not require sacrificing of the
animals or additional manipulation in preparation of cell suspension coupled
with mini-gel format of the slides enables high-throughput screening of the
genotoxic potential in situ. Additionally, cryopreservation of the blood
samples directly onsite expands the research area as it overcomes issues
related to safe transportation of the samples to the laboratory. Data
obtained so far indicated that the genotoxic response in the studied
indicator organisms significantly differ which is understandable considering
the difference in uptake, accumulation and physiological responses.

Prokaryotic assays are of great interest in eco/geno-toxicology as they are short-term, simple and cost effective. Application of strains of Salmonella typhimurium with modifications such as increased cell wall permeability and lack of excision repair system is common for the detection of genotoxicity. Various eukaryotic models have been developed lately, such as fish derived cell lines, which can mimic the response of the aquatic organisms. Considering different level of cell organization among these models, difference in sensitivity is expected, yet only few studies address this topic.
OBJECTIVES
The major objective was to investigate genotoxic potential of extracts of surface water of the Danube
and to compare the results obtained in applied prokaryotic and eukaryotic models.
METHODS
Surface water samples from 24 sites along the Danube River were extracted by LVSPE Horizon sampler providing relative enrichment factor (REF) of the extracts of 25,000x. For representative prokaryotic model the SOS/umuC assay with S. typhimurium TA1535/pSK1002 was used. Eukaryotic model comprised assessment of cytotoxicity by MTS test and assessment of genotoxicity by comet assay in ZFL (ZebraFish Liver) cells.
RESULTS
While none of the tested samples has induced SOS response, the result of MTS assay indicated that 11 of 24 samples were cytotoxic. In comet assay, genotoxic potential was found in 9 of 24 samples. Results indicated higher sensitivity of used eukaryotic model in comparison with prokaryotic. Applied prokaryotic assay is often suggested to be used as prescreening tool in diagnostics of genotoxic potential which by our opinion could be misleading.

AB  - More frequent application of biological parameters beside the conventional chemical parameters in national
monitoring for marine ecosystem assessment is supported by UNEP/MAP. Accordingly, to evaluate the
possible effects of pollution by determination of trace element content and genotoxicity assessment in
marine organisms within the Boka Kotorska Bay, Montenegro (southern Adriatic), we carried a field study
which included transplantation of the seabream (Sparus aurata) from aquaculture farm (Orahovac) to more
impacted sites (Dobrota and port of Tivat), situated in vicinity of the main ports. On the sampling site
Orahovac, the group of seabream specimens were placed in closed fish trap on 5 m depth (O1), another fish
trap was placed in Dobrota, while the third fish trap with seabreams was placed at the site port of Tivat.
The additional group was sampled from standard aquaculture cage on sampling site (O2) to reduce possible
bias in genotoxicity data due to confined space within the fish trap. After two weeks of exposure, blood
from fish heart was taken, afterwards muscle tissues were prepared for trace element determination.
Genotoxicity was measured by DNA damage induction based on comet (single cell gel electrophoresis)
assay parameter – Tail intensity (TI%), while trace element (As, Cd, Pb, Hg, Cr, Cu, Fe, Mn, Ni, Zn and
V) content was determined by ICP. Mean values of TI% obtained from seabream specimens at the sampling
site Orahovac were: 4.26 ± 0.17 – O1 and 2.56 ± 0.11 – O2, while TI% values for transplanted specimens
on the sites Dobrota and port of Tivat were 6.38 ± 0.17 and 11.06 ± 0.23, respectively. Statistically
significant differences (p<0,05) of TI% were observed between all specimens groups. The group of
seabream from port of Tivat showed the highest TI% (higher level of DNA damage), most probably caused
by marine pollution. Obtained trace element concentrations in seabream tissues were significantly lower in
comparison to values recommended by FAO, EC and national legislatives of Croatia, Spain and Turkey.
Since transplanted fish showed a significant response in a relatively short exposure period, our results
support the introduction of seabream caging as monitoring tool to reveal marine pollution by multiple
biomarker approach. Considering the significant share of aquacultured seabream in human consumption,
higher availability for sampling and convenience in application for molecular biomarker analyses in the
same samples, such an integrated monitoring approach would contribute to more credible data.

Genotoxicity was measured by DNA damage induction based on comet (single cell gel electrophoresis)
assay parameter – Tail intensity (TI%), while trace element (As, Cd, Pb, Hg, Cr, Cu, Fe, Mn, Ni, Zn and
V) content was determined by ICP. Mean values of TI% obtained from seabream specimens at the sampling
site Orahovac were: 4.26 ± 0.17 – O1 and 2.56 ± 0.11 – O2, while TI% values for transplanted specimens
on the sites Dobrota and port of Tivat were 6.38 ± 0.17 and 11.06 ± 0.23, respectively. Statistically
significant differences (p<0,05) of TI% were observed between all specimens groups. The group of
seabream from port of Tivat showed the highest TI% (higher level of DNA damage), most probably caused
by marine pollution. Obtained trace element concentrations in seabream tissues were significantly lower in
comparison to values recommended by FAO, EC and national legislatives of Croatia, Spain and Turkey.
Since transplanted fish showed a significant response in a relatively short exposure period, our results
support the introduction of seabream caging as monitoring tool to reveal marine pollution by multiple
biomarker approach. Considering the significant share of aquacultured seabream in human consumption,
higher availability for sampling and convenience in application for molecular biomarker analyses in the
same samples, such an integrated monitoring approach would contribute to more credible data.",

AB  - U ovom radu ispitivan je stepen kontaminacije vode i plavne zone reke Save na primeru ispusta otpadnih voda kod Beogradskog sajma korišćenjem mikrobioloških indikatora fekalnog zagađenja. Analiza broja spora klostridija rađena je membranskom filtracijom u skladu sa standardom ISO 14189:2013. Rezultati mikrobioloških analiza ukazuju na visok stepen kontaminacije vode i plavnog područja na ispitivanom lokalitetu, kao i na potencijalni rizik koje zabeleženo fekalno zagađenje može predstavljati po zdravlje osoba koje ispitivano područje koriste za rekreaciju.
AB  - In this paper, the level of contamination of the Sava River water and floodplain was asssessed using microbiological indicators of fecal pollution. Wastewater outlet near the Belgrade Fair was chosen as a case study. Quantification of total coliforms, faecal coliforms and fecal enterococci was performed using the Colilert-18 and Enterolert-E systems. The analysis of the number of clostridian spores was carried out by membrane filtration in accordance with the ISO 14189 standard. The results of microbiological analyzes indicate a high degree of contamination of the water and floodplain area at the investigated location, as well as the potential risk that detected fecal pollution may represent for the health of persons who use the investigated area for recreation.

AB  - Rak je vodeći uzrok smrti ljudi širom sveta sa 10 miliona umrlih u 2020. godini
zbog čega je jedan od imperativa naučne zajednice pronalazak efikasnijih
hemioterapeutika. Više od polovine lekova prirodnog je porekla, a budući da
okeani čine oko 70% Zemljine površine, marinski ekosistem je odlična osnova za
dobijanje novih lekova. Hinoni avarol i avaron su redoks par izolovan iz
mediteranskog sunđera Disidea avara i pokazuju raznovrsnu biološku aktivnost
poput antimikrobne i antitumorske. Кao i kod većine marinskih organizama,
najveće probleme za njihovu primenu predstavljaju slab prinos i cena
eksploatacije. Rešenje problema može biti sinteza jedinjenja sličnih po
hemijskoj strukturi i delovanju avarolu/avaronu poput terc-butilhinona (TBQ),
a sintezom alkiltio i ariltio derivata dodatno povećanje njegove aktivnosti.
Uporedno je testiran antibakterijski, toksični, citotoksični, genotoksični,
antioksidativni i antineurodegenerativni potencijal TBQ i njegovih derivata.
Generalno, derivati TBQ pokazuju jaču biološku aktivnost. Najjača
antibakterijska aktivnost uočena je na S. aureus i B. subtilis. Modifikacije
povećavaju citotoksičnost i genotoksičnost na humanim ćelijskim linijama dok
derivat 2-terc-butil-5,6-(etileneditio)-1,4-benzohinon pokazuje najjaču
aktivnost i na osnovu γH2AX testa dovodi do dvolančanih prekida DNК
molekula. Sva ispitivana jedinjenja snažno zaustavljaju ćelijski ciklus u G0/G1
fazi. Dodatno, jedinjenja pokazuju dobar antioksidativni i anttineurodegenerativni potencijal.

због чега је један од императива научне заједнице проналазак ефикаснијих
хемиотерапеутика. Више од половине лекова природног је порекла, а будући да

Synthesis and biological activity of alkylthio and arylthio derivatives of tert-butylquinone

AB  - Biological activity of 2-tert-butyl-1,4-benzoquinone (TBQ) and its derivatives, 2-tert-butyl-5-(2-propylthio)-1,4-benzoquinone, 2-tert-butyl-5- -(propylthio)-1,4-benzoquinone, 2-tert-butyl-5,6-(ethylenedithio)-1,4-benzoquinone, 2-tert-butyl-5-(phenylthio)-1,4-benzoquinone and 2-tert-butyl-6-(phenylthio)- 1,4-benzoquinone, were tested for their antioxidant, antibacterial, toxic, cytotoxic and genotoxic potential. Using the DPPH test, all derivatives showed good antioxidant activity, better than ascorbic acid, and the 2-tert- -butyl-5-(propylthio)-1,4-benzoquinone derivative showed the strongest effect. Better antibacterial potential was observed against Gram-positive bacteria in the broth microdilution method in which the 2-tert-butyl-5-(phenylthio)-1,4- -benzoquinone derivative showed the strongest activity (MIC = 15.6 μM). The results of toxicity tests, using the Brine shrimp test, indicated that the derivatives lose their toxic potential compared to TBQ, except for 2-tert-butyl-6- -(phenylthio)-1,4-benzoquinone, which showed a 3 times stronger effect. Cytotoxicity was assessed by the MTT assay in 24 and 72 h treatments in MRC-5, HS 294T and A549 cell lines in threefold decreasing gradient (11, 33 and 100 μM). Modifications potentiate the cytotoxic effect, and the strongest effect was observed with the 2-tert-butyl-5,6-(ethylendithio)-1,4-benzoquinone derivative. In addition, the genotoxic potential was examined in the MRC-5 cell line using the comet assay. All tested derivatives of TBQ showed a genotoxic effect at all applied subtoxic concentrations. In general, the chemical modifications of TBQ enhanced its biological activity.

abstract = "Biological activity of 2-tert-butyl-1,4-benzoquinone (TBQ) and its derivatives, 2-tert-butyl-5-(2-propylthio)-1,4-benzoquinone, 2-tert-butyl-5- -(propylthio)-1,4-benzoquinone, 2-tert-butyl-5,6-(ethylenedithio)-1,4-benzoquinone, 2-tert-butyl-5-(phenylthio)-1,4-benzoquinone and 2-tert-butyl-6-(phenylthio)- 1,4-benzoquinone, were tested for their antioxidant, antibacterial, toxic, cytotoxic and genotoxic potential. Using the DPPH test, all derivatives showed good antioxidant activity, better than ascorbic acid, and the 2-tert- -butyl-5-(propylthio)-1,4-benzoquinone derivative showed the strongest effect. Better antibacterial potential was observed against Gram-positive bacteria in the broth microdilution method in which the 2-tert-butyl-5-(phenylthio)-1,4- -benzoquinone derivative showed the strongest activity (MIC = 15.6 μM). The results of toxicity tests, using the Brine shrimp test, indicated that the derivatives lose their toxic potential compared to TBQ, except for 2-tert-butyl-6- -(phenylthio)-1,4-benzoquinone, which showed a 3 times stronger effect. Cytotoxicity was assessed by the MTT assay in 24 and 72 h treatments in MRC-5, HS 294T and A549 cell lines in threefold decreasing gradient (11, 33 and 100 μM). Modifications potentiate the cytotoxic effect, and the strongest effect was observed with the 2-tert-butyl-5,6-(ethylendithio)-1,4-benzoquinone derivative. In addition, the genotoxic potential was examined in the MRC-5 cell line using the comet assay. All tested derivatives of TBQ showed a genotoxic effect at all applied subtoxic concentrations. In general, the chemical modifications of TBQ enhanced its biological activity.",

AB  - Wastewater-based epidemiology (WBE) surveillance of COVID-19 and other future outbreaks is a challenge for developing countries as most households are not connected to a sewerage system. In December 2019, SARS-CoV-2 RNA was detected in the Danube River at a site severely affected by wastewaters from Belgrade. Rivers are much more complex systems than wastewater systems, and efforts are needed to address all the factors influencing the adoption of WBE as an alternative to targeting raw wastewater. Our objective was to provide a more detailed insight into the potential of SARS-CoV-2 surveillance in Serbian surface waters for epidemiological purposes. Water samples were collected at 12 sites along the Sava and Danube rivers in Belgrade during the fourth COVID-19 wave in Serbia that started in late February 2021. RNA was concentrated using Amicon Ultra-15 centrifugal filters and quantified using RT-qPCR with primer sets targeting nucleocapsid (N1 and N2) and envelope (E) protein genes. Microbiological (faecal indicator bacteria and human and animal genetic faecal source tracking markers), epidemiological, physicochemical and hydromorphological parameters were analysed in parallel. From 44 samples, SARS-CoV-2 RNA was detected in 31, but only at 4 concentrations above the level of quantification (ranging from 8.47 × 103 to 2.07 × 104 gc/L). The results indicated that surveillance of SARS-CoV-2 RNA in surface waters as ultimate recipients could be used as an epidemiological early-warning tool in countries lacking wastewater treatment and proper sewerage infrastructure. The performance of the applied approach, including advanced sampling site characterization to trace and identify sites with significant raw sewage influence from human populations, could be further improved by adaptation of the methodology for processing higher volumes of samples and enrichment factors, which should provide the quantitative instead of qualitative data needed for WBE.

AB  - U Srbiji se prerađuje manje od 13% otpadnih voda,1 što za posledicu ima
ispuštanje velikih količina neprečišćenih voda u vodotokove. Ravničarske reke
su pod značajnim pritiscima ovih izvora zagađenja, s obzirom da delom ili
čitavim tokom protiču kroz naseljene oblasti, kao i oblasti u kojima se nalaze
industrijska postrojenja. U analizi uticaja zagađenja na akvatične organizme,
oštećenje DNК molekula predstavlja značajan biomarker ranog upozorenja jer
može ukazati na uticaj ksenobiotika na biološke sisteme pre nego što se odrazi
na čitav organizam i/ili populaciju. U skladu sa tim, cilj ovog istraživanja je
procena osetljivosti široko rasprostranjene vrste Alburnus alburnus (uklija)
prilikom detekcije genotoksičnog efekta, kako na prostorno bliskim
lokalitetima na Savi, tako na i prostorno udaljenim na Dunavu, Tisi, Savi i
Velikoj Moravi. Za poređenje lokaliteta korišćen je integrisani odgovor
biomarkera (IBR) koji je objedinio pojedinačne biomarkere: vijabilnost ćelija
krvi, nivo DNК oštećenja ćelija krvi detektovanog u alkalnom komet i
mikronukleus testu i indeks zagađenja metalima/prioritetnim supstancama.
Integrisanjem pojedinačnih odgovora biomarkera u jedinstvenu IBR vrednost
omogućeno je jednostavnije poređenje lokaliteta. Dobijeni rezultati su ukazali da
su odgovori biomarkera uklija specifični za lokalitet. U skladu sa rezultatima,
utvrđeno je da uklija može biti pouzdan bioindikator u diskriminaciji i
udaljenih i prostorno bliskih lokaliteta koji su pod različitim uticajem
zagađenja.

AB  - Velika, Zapadna, Južna Morava, kao i Sava, Kolubara i Barička reka su pod značajnim uticajem komunalnih i industrijskih otpadnih voda. Ciljevi studije su procena genotoksičnog potencijala analiziranih lokaliteta i ispitivanje osetljivosti primenjenih biotestova i pristupa istraživanja. Studija slučaja Velika Morava obuhvatila je veliko ispitivano područje na osam lokaliteta na Velikoj, Zapadnoj i Južnoj Moravi, a u studiji Sava ispitivano je tri prostorno bliska lokaliteta koji su pod različitim pritiscima zagađenja. U in silico pristupu na osnovu literaturnih podataka analiziran je toksični/genotoksični potencijal jedinjenja detektovanih u vodi. Ex situ/in vitro pristup obuhvatio je ispitivanje genotoksičnog potencijala pomoću SOS/umuC testa, kao i primenom alkalnog komet testa na HepG2 ćelijskoj liniji. U in situ/in vivo istraživanju analizirano je DNK oštećenje ćelija krvi vrste Alburnus alburnus (uklija) pomoću alkalnog, Fpg-modifikovanog komet testa i mikronukleus testa. Rezultati su pokazali različitu osetljivost zavisno od upotrebljenih testova i pristupa, ali i različit odgovor kod uklija u zavisnosti od tipa zagađenja na lokalitetima. In silico i ex situ/in vitro pristupi su pokazali niži stepen osetljivosti u odnosu na in situ/in vivo testove, a alkalni komet test je pokazao najveći potencijal u diskriminaciji lokaliteta. Utvrđeno je da uklija može biti pouzdan bioindikator u ekogenotoksikološkim istraživanjima velikih ravničarskih reka.

abstract = "Velika, Zapadna, Južna Morava, kao i Sava, Kolubara i Barička reka su pod značajnim uticajem komunalnih i industrijskih otpadnih voda. Ciljevi studije su procena genotoksičnog potencijala analiziranih lokaliteta i ispitivanje osetljivosti primenjenih biotestova i pristupa istraživanja. Studija slučaja Velika Morava obuhvatila je veliko ispitivano područje na osam lokaliteta na Velikoj, Zapadnoj i Južnoj Moravi, a u studiji Sava ispitivano je tri prostorno bliska lokaliteta koji su pod različitim pritiscima zagađenja. U in silico pristupu na osnovu literaturnih podataka analiziran je toksični/genotoksični potencijal jedinjenja detektovanih u vodi. Ex situ/in vitro pristup obuhvatio je ispitivanje genotoksičnog potencijala pomoću SOS/umuC testa, kao i primenom alkalnog komet testa na HepG2 ćelijskoj liniji. U in situ/in vivo istraživanju analizirano je DNK oštećenje ćelija krvi vrste Alburnus alburnus (uklija) pomoću alkalnog, Fpg-modifikovanog komet testa i mikronukleus testa. Rezultati su pokazali različitu osetljivost zavisno od upotrebljenih testova i pristupa, ali i različit odgovor kod uklija u zavisnosti od tipa zagađenja na lokalitetima. In silico i ex situ/in vitro pristupi su pokazali niži stepen osetljivosti u odnosu na in situ/in vivo testove, a alkalni komet test je pokazao najveći potencijal u diskriminaciji lokaliteta. Utvrđeno je da uklija može biti pouzdan bioindikator u ekogenotoksikološkim istraživanjima velikih ravničarskih reka.",

AB  - Metoda nasumično umnožene polimorfne DNK (RAPD) je poslednjih godina našla primenu u ekogenotoksikologiji. U okviru dve studije slučaja na Savi i Dunavu, za RAPD analizu korišćeno je mišićno tkivo jedinki Alburnus alburnus (uklija) uzorkovanih sa ukupno devet lokaliteta. RAPD rezultati istraživanja na Savi ukazuju na grupisanje jedinki sa zagađenih lokaliteta (UB i UK), u odnosu na jedinke sa manje zagađenog lokaliteta, SZ. RAPD rezultati studije slučaja na Dunavu ne pokazuju jasno grupisanje uklija u odnosu na jačinu pritisaka što ukazuje na neophodnost dodatnih analiza.
AB  - In recent years the randomly amplified polymorphic DNA method (RAPD) is frequently used in ecogenotoxicology. Within two case studies on the Sava River and the Danube River, muscle tissue of Alburnus alburnus (bleak) specimens sampled from nine sites was used for RAPD analysis. The RAPD results on the Sava River study indicate the grouping of specimens from polluted sites (UB and UK) as opposed to specimens from the less polluted site, SZ. The RAPD results of the Danube River case study do not show a clear bleak grouping in relation to pollution pressures, indicating that additional analyses are needed.

AB  - This research was aimed to assess the potential of Glechoma hederacea, Hyssopus officinalis, Lavandula angustifolia, Leonurus cardiaca, Marrubium vulgare and Sideritis scardica (Lamiaceae) methanolic, ethanolic and aqueous extracts against the damaging effects of oxidative stress using different experimental models. The chemical characterization was done spectrophotometrically by quantifying total phenolics, phenolic acids, flavonoids and flavonols in the extracts, as well as by employing HPLC-DAD technique. Moreover, DPPH assay was used to assess the extracts' radical scavenging potential. Genoprotective properties of the extracts were evaluated using plasmid pUC19 Escherichia coli XL1-Blue, whereas their antigenotoxic potential was determined using Salmonella typhimurium TA1535/pSK1002 and normal human lung fibroblasts. All of the extracts showed antioxidant activity in DPPH assay. Furthermore, the results have shown that aqueous extracts provided the best protection for plasmid DNA, while alcoholic extracts most effectively contributed to the preservation of prokaryotic DNA. Additionally, each of the tested samples significantly protected the eukaryotic cells against genomic damages. Finally, despite not showing exceptional results in DPPH assay, S. scardica extracts are regarded as the most favorable in maintaining the integrity of DNA, which might be due to high quantities of phenolics such as quercetin (up to 17.95 mg g-1), naringin (up to 5.07 mg g-1) and luteolin-7-O-glucoside (up to 3.54 mg g-1). Overall, this comprehensive concept highlights the ability of these Lamiaceae species to safeguard the DNA from reactive oxygen species, to curtail the inflicted damage and also improve the efficiency of the DNA repair mechanisms, while emphasizing the importance of polyphenols as their active principles.

AB  - This study was designed to evaluate the genoprotective, antigenotoxic, as well as antitumor potential of methanolic, ethanolic, and aqueous extracts of Melissa officinalis, Mentha × piperita, Ocimum basilicum, Rosmarinus officinalis, Salvia officinalis, and Satureja montana (Lamiaceae), in different model systems. The polyphenols in these extracts were quantified both spectrophotometrically and using HPLC-DAD technique, while DPPH assay was used to assess the antioxidant activity. The genoprotective potential was tested on pUC19 Escherichia coli XL1-blue, and the antigenotoxicity on Salmonella typhimurium TA1535/pSK1002 and human lung fibroblasts, while the antitumor activity was assessed on colorectal cancer cells. Rosmarinic acid, quercetin, rutin, and luteolin-7-O-glucoside were among the identified compounds. Methanolic extracts had the best DPPH-scavenging and SOS-inducing activities, while ethanolic extracts exhibited the highest antigenotoxicity. Additionally, all extracts exhibited genoprotective potential on plasmid DNA. The antitumor effect was mediated by modulation of reactive oxygen species (ROS), nitric oxide (NO) production, and exhibition of genotoxic effects on tumor cells, especially with O. basilicum ethanolic extract. Generally, the investigated extracts were able to provide antioxidant protection for the acellular, prokaryotic, and normal human DNA, while also modulating the production of ROS and NO in tumor cells, leading to genotoxicity toward these cells and their decrease in proliferation.

Mapping of the microbiological water quality of surface waters in Serbia overlooked by the National monitoring programme

AB  - Surface waters in Serbia are under high anthropogenic pressure. One of the major problems is untreated municipal and industrial wastewaters. Unfortunately, Serbia processes only 5 % of wastewaters before discharging. As a consequence, pollutants such as metals and metalloids from industrial wastewaters, pharmaceuticals, compounds from personal care products, etc. directly endure into surface water. Microbial faecal pollution, as an indicator of presence of human or/and animal pathogens, due to health hazard limits water usage for drinking, recreation, irrigation, etc. In regulations, faecal coliforms, with Escherichia coli as dominant representative, are widely used as faecal indicator bacteria. The aim of this study was to investigate the microbiological al water quality in the Republic of Serbia at the sites which are not routinely investigated within the national monitoring program. In this purpose 78 sites situated on canals, mountain springs, as well as lowland rivers were selected. Defined Substrate Technology was used for determination Most Probable Number (MPN) of E. coli using Colilert-18 System. The water classification system, developed for the Danube River (Kavka et al., 2006) was used. 
The results indicated that more than 47 % of selected sites are under critical (21.79 %) or strong (25.64 %) faecal pollution. On the other hand, about 32 % (32.05 %) of sites are little polluted and 20.51 % of sites showed moderate pollution. The increasing levels of faecal pollution detected on sites situated downstream of settlements indicated discharge of untreated municipal wastewaters directly into surface water. In further research, the focus will be placed on the determination of the origin of pollution by the employment of microbial source tracking technique. Obtained data will be used for modelling and predicting the effect of detected contamination on the water quality of the major water bodies in the Republic of Serbia

AB  - Uvođenje metoda uslovljenim efektima (eng. effect based methods), uključujući i genotoksikološke metode, u programe monitoringa i naučna istraživanja na velikim ravničarskim rekama, omogućilo bi sveobuhvatniju procenu stanja ekosistema i odnosa između samog hemijskog i ekološkog statusa. Osnovni ciljevi disertacije su ispitivanje osetljivosti in vitro i in vivo testova u ekogenotoksikologiji i formulisanje smernica za njihovu primenu u istraživanjima velikih ravničarskih reka. Studija je obuhvatila ukupno 17 lokaliteta u slivovima Velike Morave, Save i Dunava. Za procenu genotoksičnog efekta nativnih i koncentrisanih uzoraka rečne vode, in vitro pristup obuhvatio je SOS/umuC test, alkalni komet test na HepG2 i ZFL ćelijama, kao i analizu ćelijskog ciklusa na ZFL ćelijama. In vivo pristup je za procenu genotoksičnosti obuhvatio alkalni, Fpg modifikovani komet test, kao i mikronukleus test na ćelijama krvi Alburnus alburnus (uklija). Dodatno, ispitivana je osetljivost RAPD analize u detekciji genotoksičnog efekta. U in silico pristupu na osnovu literature analiziran je toksični/genotoksični potencijal hemijskih parametara koji su detektovani u vodi i u uklijama, a integrativnim metodama vršena je kvantifikacija pritiska zagađenja. Biotestovi su pokazali različitu osetljivost u detekciji genotoksičnog efekta. U in vitro istraživanju koncentrisani uzorci rečne vode su se pokazali pogodnim za testiranje toksičnosti/genotoksičnosti, a ZFL ćelije kao dobar model sistem. U in vivo istraživanju A. alburnus se pokazao kao pouzdan bioindikator. Najveća osetljivost u detekciji genotoksičnog efekta u ćelijama krvi dobijena je u alkalnom komet testu. In silico analizom ne može se izvršiti potpuna predikcija genotoksičnog efekta ksenobiotika na organizme.
AB  - The implementation of effect based methods, including genotoxicological methods, in monitoring and research programs on large lowland rivers, could provide comprehensive assessment of ecosystem quality and relationship between chemical and ecological status itself. The main objectives of the dissertation are examination the sensitivity of in vitro and in vivo tests in ecogenotoxicology and guidelines formulation for their application in studies of large lowland rivers. The study included a total of 17 sites in the Velika Morava, Sava and Danube River basins. To assess the genotoxic effect of native and concentrated river water samples, the in vitro approach included the SOS/umuC test, alkaline comet test on HepG2 and ZFL cells, and cell cycle analysis on ZFL cells. The in vivo approach for genotoxicity assessment included the alkaline, Fpg modified comet and micronucleus test on Alburnus alburnus (bleak) blood cells. Also, the sensitivity of RAPD analysis in the detection of genotoxic effects was examined. Within in silico approach, based on the literature, the toxic/genotoxic potential of chemical parameters detected in water and bleaks was analyzed while the quantification of pollution pressure was performed by integrative methods. Bioassays showed different sensitivity in detecting the genotoxic effect. Within in vitro study, concentrated river water samples proved to be a suitable for toxicity/genotoxicity testing, and ZFL cells a good model system. Within in vivo research, bleak proved to be a reliable bioindicator. The highest sensitivity in the detection of genotoxic effect in blood cells was obtained in the alkaline comet test. A complete prediction of the genotoxic effect of xenobiotics on organisms cannot be performed by in silico analysis.
