Hematoprotective effects and antioxidant properties of β-glucan and vitamin C against acetaminophen-induced toxicity: an experimental study in rats.

AB  - Acetaminophen is widely used as an over-the-counter analgesic and antipyretic drug. The aim of the present study was to investigate the pro-oxidative effects of acetaminophen (300 mg/kg/day i.p.) and antioxidative effects of β-glucan (4 mg/kg/day i.p.) and/or vitamin C (100 mg/kg/day i.p.) on the blood parameters of treated rats. After 3 days of treatment, hematological and parameters of redox status were measured. Exposure of rats to acetaminophen caused significant changes in some hematological parameters and the glutathione redox cycle, leading to an increased concentration of oxidative stress parameters and the formation of lipid peroxidation, while the activities of antioxidant enzymes were decreased. Administration of β-glucan and/or vitamin C reduced lipid peroxidation and restored the levels of examined hematological and oxidative stress parameters and improved the activities of antioxidant enzymes. Obtained results demonstrated that acetaminophen has significant pro-oxidative effects and may disrupt redox balance in blood of rats, while the combination of β-glucan and/or vitamin C amplified the antioxidant defense potential and exhibited a strong hematoprotective activity against acetaminophen-induced toxicity. Therefore, β-glucan and vitamin C co-treatment may be a promising therapeutic option for the treatment of acute acetaminophen hematotoxicity.

AB  - Excessive use of organophosphate insecticides, including fenitrothion (FNT) can cause detrimental consequences in non-target organisms. Selenium (Se) and vitamin C (Vit C) possess protective abilities against various toxic compounds due to their antioxidative properties. Accordingly, the aim of the present study was to examine the possible ameliorative effects of Se and Vit C in hepatotoxicity induced by FNT. For the purpose of this study, male Wistar albino rats were divided into control and groups treated with Se (0.5 mg/kg b.w, as Na2SeO3) and Vit C (100 mg/kg b.w), FNT (20 mg/kg b.w) and FNT in cotreatment with Se and Vit C for 30 days. The current data showed a reduction in absolute and relative liver weight after FNT administration. Increased activities of liver enzymes (AST, ALT, ALP, LDH and GGT) indicated liver damage. FNT alone caused significant alterations in biochemical parameters (glucose and total bilirubin). Elevation in LPO level along with decreased activities of antioxidant enzymes (SOD, CAT, GSH-Px) and GSH content reflected the presence of oxidative stress. Coadministration of FNT with Se and Vit C exhibited hepatoprotective role confirmed by reduction of oxidative stress levels and restoration in the values of examined parameters. Because of their beneficial effects, Se and Vit C may be used in reducing injuries caused by pesticides.

AB  - Fenitrothion is widely used organophosphate pesticide in agriculture and health programs, but besides, it causes several toxic effects. The present study was designed to evaluate the possible protective effects of selenium (0.5 mg/kg b.w.) and vitamin C (100 mg/kg b.w) on altered haematological, biochemical and oxidative stress parameters in the blood of rats orally treated with fenitrothion (20 mg/kg b.w) for 30 days. Fenitrothion caused changes in body weight, food and water intake, and some haematological and biochemical parameters. Fenitrothion altered the glutathione redox status (GSH and GSSG) and decreased activity of antioxidant enzymes (GSH-Px, GST, SOD and CAT), leading to a lipid peroxidation. Selenium and vitamin C, by improving the activity of antioxidants, reduced oxidative stress and a lipid peroxidation, maintaining the values of examined parameters to optimal levels. Therefore, selenium and vitamin C could be useful in providing protection of exposed non-target organisms including people from fenitrothion.

Paracetamol-induced changes of haemato-biochemical and oxidative stress parameters in rat blood: protective role of vitamin C and betaglucan

Exposure of rats to paracetamol caused changes of some haematological parameters (R BCs count, Hb concentration, Ht value and WBCs count), suggesting that the paraceta mol induced haematotoxicity. Paracetamol reduced serum total protein (TP), album in and globulin, while increased alanine aminotransferase (ALT), aspartate aminotran sferase (AST) and lactate dehydrogenase (LDH) activities compared to the cont rol. The results indicate that paracetamol is led to significant decrease in the c oncentration of Na + and K + and increase of Ca 2+ in the serum compared to the control. Coadministra tion of vitamin C and β -glucan with paracetamol reversed these changes of haematol ogical and biochemical parameters and diminished the toxic effects of paracetamol. Th e obtained results indicated that the concentration of LPO in erythrocytes significantly increased in, while the concentration of GSH significantly decreased in the group treated with paracetamol compared to control group. Coadministration of vitamin C and β -glucan with paracetamol reversed paracetamol-induced alterations in these oxidative stress parameters. This study suggests that paracetamol has significant prooxidative effec ts and may disrupt oxidant/antioxidant balance in erythrocytes. AB  - We investigated the biochemical and ultrastructural changes in the liver of the freshwater fish, European perch (Perca fluviatilis), in response to Aphanizomenon flos-aquae bloom in the Gruža Reservoir, Serbia. The activities of total manganese- and copper zinc-containing superoxide dismutase (Tot SOD, Mn-SOD, Cu/Zn-SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and biotransformation phase II enzyme glutathione-S-transferase (GST), as well as concentrations of total glutathione (GSH) and sulfhydryl (-SH) groups were examined before and during the bloom period. Mn-SOD activity was significantly higher, while the activities of Cu/Zn-SOD, CAT and GSH-Px and the concentration of the -SH groups were significantly lower during the bloom. The ultrastructure of the liver revealed necrotic and apoptotic damage to the hepatocytes during the bloom period. Our work represents the first study to report the influences of an Aphanizomenon flos-aquae bloom in the Gruža Reservoir on antioxidant biomarkers and on histopathological alterations in the liver of the freshwater fish European perch (Perca fluviatilis).

AB  - The role of oxidative stress in cisplatin (CP) toxicity and its prevention by pretreatment with selenium (Se) was investigated. Male Wistar albino rats were injected with a single dose of cisplatin (7.5 mg CP/kg b.m., i.p.) and selenium (6 mg Se/kg b.m, as Na2SeO3, i.p.) alone or in combination. The results suggest that CP intoxication induces oxidative stress and alters the glutathione redox status: reduced glutathione (GSH), oxidized glutathione (GSSG) and the GSH/GSSG ratio (GSH RI), resulting in increased lipid peroxidation (LPO) in rat liver. The pretreatment with selenium prior to CP treatment showed a protective effect against the toxic influence of CP on peroxidation of the membrane lipids and an altering of the glutathione redox status in the liver of rats. From our results we conclude that selenium functions as a potent antioxidant and suggest that it can control CP-induced hepatotoxicity in rats.
AB  - U našoj studiji ispitivana je uloga oksidacionog stresa u toksičnosti cisplatina (SR) i njegova prevencija pretretmanom selenom (Se). Mužjaci Wistar albino pacova su inicirani jednom dozom cisplatina (7.5 mg CP/kg t.m., i.p.) i selena (6 mg Se/kg t.m, kao Na2SeO3, i.p.) pojedinačno ili u kombinaciji. Rezultati pokazuju da intoksikacija SR uzrokuje oksidacioni stres i promenu glutation redoks statusa: redukovanog (GSH), oksidovanog (GSSG) i GSH/GSSG indeksa (GSH RI), kao i povećanje lipidne peroksidacije (LPO) u jetri pacova. Tretman Se koji je prethodio tretmanu SR pokazao je zaštitne efekte protiv toksičnog delovanja SR na peroksidaciju lipida membrane i promenu glutation redoks statusa u jetri pacova. Na osnovu naših rezultata zaključujemo da Se, deluje kao snažan antioksidans i da može imati ulogu u kontroli SR indukovane hepatotoksičnosti kod pacova.

AB  - Ascorbic acid (Vit C) is a reducing agent that efficiently quenches potentially damaging free radicals. The aim of this study was to investigate effects of high acute dose of Vit C on redox and antioxidative status in blood of rats. Our study included two groups of Wistar albino rats: I - untreated group (control); II - group treated with Vit C (500 mg/kg bw, i.p.). In all cases animals were sacrificed after three days of treatment and blood samples were taken at the day of sacrifice. Concentration of Heinz bodies (HB) was determined in whole blood, while the concentration of reactive oxygen and nitrogen species (ROS and RNS), as well as, parameters of antioxidative defense system were followed in plasma and red blood cells (RBC). Our results showed that Vit C treatment induced higher levels of peroxynitrites and lipid peroxidation in plasma. In RBC, Vit C induced higher level of HB, GSSG/2 GSH ratio and glutathione peroxidase activity, while decreased glutathione reductase activity. In conclusion, acute high doses of Vit C act as prooxidant and induced oxidative stress that resulted in lipid peroxidation in plasma and hemoglobin oxidation in RBC. .

AB  - In this experiment, we compared the in vivo effects of phenylhydrazine (PHZ) and bleeding treatment on the redox status and glutathione antioxidative mechanism parameters in the plasma and red blood cells (RBC) of rats. Results showed a lower level of reactive oxygen species (ROS), a higher level of lipid peroxidation and the effective antioxidative role of the glutathione system in the blood of bleeding rats. PHZ-treatment induced higher concentrations of ROS and an accumulation of oxidized glutathione in the plasma, while the glutathione system showed a satisfactory antioxidative capacity in the RBC of rats. When comparing the two anemic groups, the PHZ-treated rats showed marked oxidative stress in the plasma.

Rezultati pokazuju niži nivo reaktivnih vrsta kiseonika (ROS), viši nivo lipidne peroksidacije i efikasnu antioksidacionu ulogu glutationskog sistema u krvi iskvavljenih pacova. Tretman PHZom prouzrokovao je više koncentracije ROSa i akumulaciju oksidovanog glutationa u plazmi, dok je glutationski sistem pokazao efikasan antioksidativni kapacitet u RBCu pacova. AB  - In order to investigate daily changes of hematological parameters in bleeding-induced anemia, we treated Wistar albino male rats by daily bleeding (1.5-2 mL of blood from the tail vein for eight days). Blood samples were taken before (on day zero) and on the first to eighth days of bleeding. The values of hematocrit, hemoglobin, and erythrocyte count decreased significantly after the second, sixth, and second days of bleeding, respectively. The number of leuko­cytes and platelets, as well as Heinz body levels, increased significantly after the third and second days of treatment. The percentage of reticulocytes increased significantly from the second day and attained the maximum level (32.55 ± 0.96%) on the eighth day.
AB  - U cilju ispitivanja dnevnih promena hematoloških parametara u toku anemije indukovane dnevnim krvarenjem, tretirali smo Wistar albino pacove dnevnim krvarenjem (1.5-2 ml krvi iz repne vene u toku 8 dana). Uzorci krvi uzimani su pre (0 dan) i 1-8. dana krvarenja. Vrednosti za hematokrit, hemoglobin i broj eritrocita značajno opadaju nakon drugog, šestog i drugog dana krvarenja. Broj leukocita i trombocita, kao i nivo formiranja Heinz-ovih telašaca raste značajno nakon trećeg i drugog dana tretmana. AB  - Specimens of a marine freshwater fish - hake (Merluccius merluccius L.) - were collected at the localities of Platamuni and Valdanos (Montenegro, South Adriatic) in the spring of 2003. Our results showed higher concentrations of LPO, Vit C, and Vit E in the liver in comparison with white muscle. The concentration of LPOin both tissues was higher, while that of Vit E was lower at Valdanos compared to Platamuni. These differences in parameters of oxidative stress are partly due to differences in temperature and the concentrations of nitrites, nitrates, and detergents in the waters of Valdanos compared to Platamuni.
AB  - Tokom 2003. godine uzorkovane su jedinke morske ribe - oslića (Merluccius merluccius L.) iz voda sa lokaliteta Platamuni i Valdanos (Crna Gora, južni Jadran) u prolećnom periodu. Dobijeni rezultati su pokazali veće koncentracije LP, Vit Ci Vit E u jetri oslića u odnosu na belo mišićno tkivo. Koncentracija LP je bila veća, a Vit E manja u oba ispitivana tkiva oslića u vodama lokaliteta Valdanos u poređenju sa Platamuni. Ovakve promene parametara oksidacionog stresa posledica su promena temperature, kao i povećanih koncentracija nitrita, nitrata i deterdženata u vodama lokaliteta Valdanos u odnosu na Platamune.

AB  - Fish are the most important organisms in the biomonitoring of aquatic ecosystems. A number of pollutants may cause the oxidative stress in the organism of fish. The monitoring of parameters of oxidative stress in tissues of fish is a good biomarker in the assessment of the condition of environment. Catch of commercially important species of fish, hake (Merluccius merluccius L.) was carried out in front of sea-port Bar (South Adriatic) at the end of May 2005. The parameters of oxidative stress: concentrations of lipid peroxides (LP) and reduced glutathione (GSH) were determined in liver and white muscle. Physical-chemical parameters (salinity, concentration of dissolved oxygen and temperature) as well as the concentrations of nitrites, nitrates and detergents in the water of investigated locality were determined. The obtained results showed the tissue specificity that was the consequence of different metabolic and antioxidative activities. In the liver of hake, the increased concentration of LP was found in comparison to the white muscle, whereas in white muscle the increased concentration of GSH was recorded in comparison to the liver. These results suggest that the intensity of oxidative stress was higher in the liver in comparison to the white muscle, which is the consequence of increased presence of pollutants in water of the investigated locality. .

AB  - Cadmium (Cd) has been recognized as being among the most toxic environmental and industrial pollutants, one that is pres­ ent in soils, sediments, air, and water (S to e pp 1 er, 1991) . After penetrating into the organism, mostly through the respir­ atory and gastrointestinal tracts, Cd accumulates in the liver and kidneys, as well as in other tissues and organs, causing many metabolic, histological, and pathological changes, such as neph­ rotoxicity, cardiotoxicity, increased lipid peroxidation and hem­ orrhagic lesions of seminal tubules. In tissues, Cd also binds to proteins of low molecular mass, producing metallothioneins by the induction of metallothionein mRNA synthesis (G e o r g e et al., 1996). Cadmium depletes glutathione and protein-bound sulfhydryl groups, resulting in enhanced production of reactive oxygen species (ROS). These ROS result in increased lipid per­ oxidation, enhanced excretion of urinary lipid metabolites, modulation of intracellular oxidized states, DNA damage, membrane damage, altered gene expression, and apoptosis (Kim et al., 2003).

Parameters of oxidative stress in fish tissues represent significant biomarkers in the assessment of the status of environment. Specimens of hake (Merluccius merluccius L.) and sea bream (Pagellus erythrinus L.) were collected at the end of May 2005 from the locality in front of sea-port Bar (South Adriatic Sea). Parameters of oxidative stress (concentration of lipid peroxidase - LP and reduced glutathione - GSH) were determined in the liver and white muscle of hake and sea bream. Physical-chemical parameters (salinity, concentration of dissolved oxygen and temperature) were determined in water of the investigated locality. Obtained results showed that the concentration of LP was higher in liver of hake and sea bream in comparison to white muscle. However, significantly higher concentration of GSH was found in white muscle of hake and sea bream in comparison to liver. These changes of oxidative stress parameters point to tissue specificity, which is the consequence of different metabolic and antioxidative activity. Inter-specific differences were also established. Higher concentrations of LP and GSH in liver of hake and in white muscle of sea bream were recorded, due to the changes of physical-chemical parameters of environment. The intensity of oxidative stress in liver of hake and sea bream was higher in comparison to muscle.
AB  - Ribe predstavljaju najznačajnije organizme koji se koriste u biomonitoringu vodenih ekosistema. Parametri oksidacionog stresa u tkivima riba predstavljaju značajne biomarkere u proceni stanja životne sredine. Krajem maja 2005. godine uzorkovane su jedinke oslića (Merluccius merluccius L.) i arbuna (Pagellus erythrinus L.) sa lokaliteta ispred luke Bar (Južni Jadran). Parametri oksidacionog stresa (koncentracija lipidnih peroksida - LP i redukovani glutationa - GSH) su određivani u tkivima jetre i belog mišića oslića i arbuna. U vodi ispitivanog lokaliteta određivani su i fizičko-hemijski parametri (salinitet, koncentracija rastvorenog kiseonika i temperatura). Dobijeni rezultati su pokazali da je koncentracija LP veća u jetri oslića i arbuna u odnosu na belo mišićno tkivo. Međutim, zabeležena je znatno veća koncentracija GSH u belom mišiću i oslića i arbuna u odnosu na jetru. Ovakve promene parametara oksidacionog stresa ukazuju na tkivnu specifičnost, kao posledica različite metaboličke i antioksidacione aktivnosti. Ustanovljene su i interspecijske razlike, zabeležena je veća koncentracija LP i GSH u jetri oslića, kao i u belom mišiću arbuna, kao posledica promena fizičko-hemijskih parametara sredine i prisustva polutanata. Intenzitet oksidacionog stresa u jetri oslića i arbuna je bio viši u odnosu na mišiće.

Parameters of oxidative stress in fish tissues represent significant biomarkers in the assessment of the status of environment. Specimens of hake (Merluccius merluccius L.) and sea bream (Pagellus erythrinus L.) were collected at the end of May 2005 from the locality in front of sea-port Bar (South Adriatic Sea). Parameters of oxidative stress (concentration of lipid peroxidase - LP and reduced glutathione - GSH) were determined in the liver and white muscle of hake and sea bream. Physical-chemical parameters (salinity, concentration of dissolved oxygen and temperature) were determined in water of the investigated locality. Obtained results showed that the concentration of LP was higher in liver of hake and sea bream in comparison to white muscle. However, significantly higher concentration of GSH was found in white muscle of hake and sea bream in comparison to liver. These changes of oxidative stress parameters point to tissue specificity, which is the consequence of different metabolic and antioxidative activity. Inter-specific differences were also established. Higher concentrations of LP and GSH in liver of hake and in white muscle of sea bream were recorded, due to the changes of physical-chemical parameters of environment. The intensity of oxidative stress in liver of hake and sea bream was higher in comparison to muscle., Ribe predstavljaju najznačajnije organizme koji se koriste u biomonitoringu vodenih ekosistema. Parametri oksidacionog stresa u tkivima riba predstavljaju značajne biomarkere u proceni stanja životne sredine. Krajem maja 2005. godine uzorkovane su jedinke oslića (Merluccius merluccius L.) i arbuna (Pagellus erythrinus L.) sa lokaliteta ispred luke Bar (Južni Jadran). AB  - The effects of nitroglycerine (NTG) are mediated by liberated nitric oxide (NO) after NTG enzymatic biotransformation in cells. The aim of this study was to evaluate some products of NTG bio-transformation and their consequences on the redox status of rat erythrocytes and reticulocytes, considering the absence and presence of functional mitochondria in these cells, respectively. Rat erythrocyte and reticulocyte-rich red blood cell (RBC) suspensions were aerobically incubated (2h, 37 degrees C) without (control) or in the presence of different concentrations of NTG (0.1, 0.25, 0.5, 1.0 and 1.5 mM). In rat erythrocytes, NTG did not elevate the concentrations of any reactive nitrogen species (RNS). However, NTG robustly increased concentration of methemoglobin (MetHb), suggesting that NTG bio-transformation was primarily connected with hemoglobin (Hb). NTG-induced MetHb formation was followed by the induction of lipid peroxidation. In rat reticulocytes, NTG caused an increase in the levels of nitrite, peroxinitrite, hydrogen peroxide, MetHb and lipid peroxide levels, but it decreased the level of the superoxide anion radical. Millimolar concentrations of NTG caused oxidative damage of both erythrocytes and reticulocytes. These data indicate that two pathways of NTG bio-transformation exist in reticulocytes: one generating RNS and the other connected with Hb (as in erythrocytes). In conclusion, NTG bio-transformation is different in erythrocytes and reticulocytes due to the presence of mitochondria in the latter.

AB  - The effects of cadmium (Cd, 15 mg/kg/day) on the blood antioxidant defense system and the possible protective effects of selenium (Se, 7 μg/kg/day) were studied. Chronic Cd administration resulted in increased activities of copper, zinc superoxide dismutase (CuZn SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) in red blood cells (RBC), as well as glutathione S-transferase (GST) activity and vitamin E (Vit E) concentration in the plasma. Se induced a significant increase in GSH-Px and GST activities, as well as concentrations of vitamin C and E. Thus, the mechanism by which Se decreases Cd toxicity in rats, seems to rely on the protection of the antioxidant defense system in blood, possibly by forming non-toxic cadmium selenide. The accumulation of Cd in RBC also decreased significantly upon co-exposure to Se. The obtained results show, that Cd exhibit toxic effects on the antioxidant defense system in the blood of rats and that Se demonstrates the protective effect against Cd toxicity.
AB  - U ovom radu ispitivan je efekat kadmijuma (Cd, 15 mg/kg/danu) na antioksidacioni zaštitni sistem i moguća zaštitna uloga selena (Se, 7 μg/kg/danu). Posle hroničnog tretmana Cd značajno se povećavaju aktivnosti bakar, cink superoksid dismutaze (CuZn SOD), katalaze (CAT) i glutation peroksidaze (GSH-Px) u crvenim krvnim ćelijama (RBC), kao i aktivnost glutation S-transferaze (GST) i koncentracije vitamina E (Vit E) u plazmi. Selen značajno povećava aktivnosti enzima GSH-Px i GST, kao i koncentracije vitamina C i E. Mehanizam kojim selen smanjuje toksičnost Cd kod pacova odnosi se na zaštitu antioksidacionog sistema u krvi, posebno formiranje ne-toksičnih kadmium-selenita. Akumulacija Cd u RBC takođe je značajno smanjena posle ko-tretmana selenom. AB  - The aim of our study was to investigate the copper and zinc containing superoxide dismutase (CuZn SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cells (RBCs), as well as phase II biotransformation enzyme glutathione-S-transferase (GST) activity in the plasma of male, two months old Wistar albino rats. The rats were divided into two experimental groups: control animals and animals treated with olive oil (100 μL of virgin olive oil, i.m., every fifth day during 30 days). The obtained results show that olive oil induced a significant decrease of CuZn SOD, CAT and GSH-Px activities in RBCs (p<0.05) and significant increase of GST activity in the plasma (p<0.05) in relation to the control animals. The results show that olive oil influences the activity of investigated enzymes in the blood of rats and in that way improves the overall antioxidative defense.
AB  - Cilj našeg rada bio je da se odrede aktivnosti bakar cink sadržavajuće superoksid-dismutaze (CuZn SOD), katalaze (CAT) i glutation-peroksidaze (GSH-Px) u eritrocitima, kao i enzima faze II biotransformacije glutation-S-transferaze (GST) u plazmi mužjaka dva meseca starih Wistar albino pacova tretiranih maslinovim uljem. Pacovi su podeljeni u dve grupe i to kontrolna i grupa tretirana maslinovim uljem (100 μL of maslinovog ulja, i.m., svaki peti dan tokom 30 dana). Rezultati pokazuju da maslinovo ulje izaziva značajno smanjenje aktivnosti CuZn SOD, CAT i GSH-Px (p<0.05) u eritrocitima, kao i povećanje aktivnosti GST u plazmi pacova (p<0.05) u odnosu na kontrolu. Dobijeni rezultati pokazuju da maslinovo ulje utiče na aktivnost ispitivanih enzima u smislu poboljšanja antioksidacione zaštite.

AB  - Fish are the most important organisms used in biomonitoring of aquatic ecosystems. Parameters of oxidative stress in fish tissues represent significant biomarkers in the assessment of the status of environment. Specimens of hake (Merluccius merluccius L) and red mullet (Mullus barbatus L) were collected from the locality in front of sea-port Bar (South Adriatic Sea) at the end of May 2005. Activity of the enzymes of the antioxidative system (superoxide dismutase - SOD and catalase - CAT) and parameters of oxidative stress (lipid peroxidation - LP and reduced glutathione - GSH content) were determined in the liver of hake and red mullet. Obtained results showed that the activities of SOD and CAT were significantly lower in the liver of hake than in red mullet. Higher concentrations of LP and GSH in liver of hake were recorded, due to the changes of physical-chemical parameters of environment. The intensity of oxidative stress in liver of hake was higher in comparison to red mullet.
AB  - Ribe predstavljaju najznačajnije organizme koji se koriste u biomonitoringu vodenih ekosistema. Parametri oksidacionog stresa u tkivima riba predstavljaju značajne biomarkere u proceni stanja životne sredine. Krajem maja 2005. godine uzorkovane su jedinke oslića (Merluccius merluccius L) i trlje (Mullus barbatus L) sa lokaliteta ispred luke Bar (Južni Jadran). Aktivnost enzima antioksidacionog sistema (superoksid dismutaze - SOD i katalaze - CAT) i parametri oksidacionog stresa (koncentracija lipidnih peroksida - LP i redukovanog glutationa - GSH) su određivani u jetri oslića i trlje. Dobijeni rezultati su pokazali da su aktivnosti SOD i CAT značajno manje u jetri oslića nego kod trlje. Zabeležena je veća koncentracija LP i GSH u jetri oslića, kao posledica promena fizičko-hemijskih parametara sredine i prisustva polutanata. Intenzitet oksidacionog stresa u jetri oslića je bio viši u odnosu na trlju.

AB  - Preeclampsia represents a pregnancy specific syndrome characterized by elevated arterial blood pressure and proteinuria. The aim of this study was to evaluate whole blood and erythrocytes energy status in healthy and preeclamptic women. Our study involved 18 healthy (control) and 19 preeclamptic women. Diagnosis of preeclampsia was done according to the recommendation of International Society for the Study of Hypertension in Pregnancy. After blood collection and erythrocytes extraction, the next parameters were followed: concentrations of lactate and adenine nucleotides (ATP, ADP and AMP). Results showed increased concentrations of lactate and ATP in whole blood of preeclamptic, vs. control pregnant women. All parameters of energy status were also elevated in erythrocites of preeclamptic women (elevation of ADP level was significant). This study suggests that preeclamsia was followed by an elevation of all parameters of energy status in whole blood and in erythrocytes, as a result of increased rate of glycolysis. .
AB  - Preeklampsija predstavlja za trudnoću specifičan sindrom koji se karakteriše povišenim krvnim pritiskom i proteinurijom. Cilj istraživanja je određivanje energetskog statusa pune krvi i eritrocita zdravih i trudnica sa preeklampsijom. Ispitivanjem je bilo obuhvaćeno 18 zdravih i 19 trudnica sa preeklampsijom. Dijagnoza preeklampsije je postavljena prema preporukama International Society for the Study of Hypertension in Pregnancy. Nakon uzimanja krvi i ekstrakcije eritrocita praćeni su sledeći parametri: koncentracije laktata i adenin nukleotida (ATP, ADP i AMP). Rezultati su pokazali povećanje koncentracija laktata i ATP u punoj krvi trudnica sa preeklampsijom u odnosu na kontrolne trudnice. U eritrocitima, svi parametri energetskog statusa su bili povećani u preeklampsiji (povećanje ADP nivoa je bilo značajno). Ova studija pokazuje da je preeklampsija praćena povećanjem svih parametara energetskog statusa u punoj krvi i eritrocitima, kao posledica stimulisanog intenziteta procesa glikolize. .

AB  - Preeclampsia represents a pregnancy specific syndrome with high rate of both maternal and neonatal morbidity and mortality. The maternal syndrome is characterized by elevated arterial blood pressure and proteinuria. Several recent studies have reported that there is a disbalance of oxidant/antioxidant system in women with preeclampsia, and this factor might contribute to the pathogenesis of the disease. Our study involved 22 healthy (control) and 20 preeclamptic women. Diagnosis of preeclampsia was done according to the recommendation of International Society for the Study of Hypertension in Pregnancy. After blood collection, the next parameters were followed in erythrocytes: concentration of lipid peroxides (TBARS) non-enzymatic components (reduced glutathione - GSH, oxidized glutathione GSSG) and activities of enzymatic components of AOS (glutathione peroxidase GSH-Px, glutathione reductase - GR and glutathione-S-transferase - GST). Results of this study showed that concentration of TBARS and activity of GST were significantly higher, while activity of GR was significantly lower in erythrocytes of preeclamptic, vs. healthy women. Concentrations of GSH, GSSG and activity GSH-Px did not change significantly. Increased concentration of TBARS and activity of GST, as well as disorder in GSH and GSH-dependent enzymes status were indicators of oxidative stress in erythrocytes of preeclamptic women.
AB  - Preeklampsija je sindrom specifičan za trudnoću sa visokim stepenom morbiditeta i mortaliteta fetusa i majke. Klinički se manifestuje posle 20. nedelje gestacije. Kod majke sindrom se manifestuje povišenim krvnim pritiskom i proteinurijom. Brojne savremene studije pokazuju da je disbalans oksidacionog i antioksidacionog sistema značajan faktor u patogenezi preeklampsije. Istraživanjem je obuhvaćeno 22 zdrave i 20 trudnica sa preeklampsijom. Dijagnoza preeklampsije je postavljena prema preporukama International Society for the Study of Hypertension in Pregnancy. U eritrocitima su praćeni sledeći parametri: koncentracija lipidnih peroksida (TBARS), neenzimskih komponenti (redukovani glutation - GSH, oksidovani glutation - GSSG) i aktivnost enzimskih komponenti AOS (glutation-peroksidaza, GSH-Px, glutation-reduktaza, GR i glutation-S-transferaza, GST). AB  - The effect of cadmium (Cd), coenzyme Q10 (CoQ10) and Cd+CoQ10 on the activities of superoxide dismutases (total SOD), manganese containing superoxide dismutase (Mn SOD) and copper-zinc containing superoxide dismutase (Cu,Zn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-Stransferase (GST), glutathione reductase (GR) and concentrations of ascorbic acid (AsA) and vitamin E (Vit E) in the heart of male Wistar albino rats were studied in comparison to the controls and cadmium treated animals. Cd induces a significant increase of total SOD, Cu, Zn SOD and GSH-Px activities, as well as AsA and Vit E concentrations, but leads to a significant decrease of CAT and GR activities. CoQ10 induces a significant increase of total SOD, Mn SOD, Cu, Zn SOD and GSH-Px activities, as well as AsA and Vit E concentrations. In the same group of animals the activities of CAT, GST and GR were significantly decreased. By concomitant treatment of rats with Cd+CoQ10 the activities of total SOD, Mn SOD and GSH-Px, as well as concentrations of AsA and Vit E were markedly increased. In the same group of animals the activities of Cu, Zn SOD, CAT and GR were significantly decreased. In respect to the Cd treated rats in Cd+CoQ10 partially are reversed changes (Cu,Zn SOD) of antioxidant defense system in the heart.
AB  - Ispitivan je uticaj kadmijuma (Cd), koenzima Q10 (CoQ10) i Cd+ CoQ10 na aktivnost ukupne superoksid dismutaze (Uk SOD), mangan sadržavajuće superoksid dismutaze (Mn SOD), bakar cink sadržavajuće superoksid dismutaze (Cu,Zn SOD), katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-Stransferaze (GST) i glutation reduktaze (GR), kao i na koncentracije askorbinske kiseline (AsA) i vitamina E (Vit E) u srcu mužjaka Wistar albino pacova u poređenju sa kontrolnom grupom i grupom tretiranom sa kadmijumom. Cd u srcu indukuje značajno povećanje aktivnosti Uk SOD, Cu,Zn SOD i GSH-Px, kao i koncentracija AsA Vit E uz značajno smanjenje aktivnosti CAT i GR. CoQ10 izaziva značajno povećanje aktivnosti Uk SOD, Mn SOD, Cu,Zn SOD i GSH-Px, kao i koncentracija AsA i Vit E. U istoj grupi životinja aktivnosti CAT, GST i GR su značajno smanjene. Istovremeni tretman sa Cd+CoQ10 dovodi do značajnog povećanja aktivnosti Uk SOD, Mn SOD i GSH-Px, kao i koncentracija AsA i Vit E. AB  - Cadmium (Cd+2) is an ubiquitous toxic metal that may induce oxidative damage by disturbing the prooxidant-antioxidant balance in the blood. Wistar albino rats (3 months old), were injected with a single dose of CdCh (0.4 mg Cd/kg i.p. and sacrificed after 24h). The hematological parameters: red blood cells count (RBCs), hematocrite (Ht) value and hemoglobin (Hb) concentration were significantly decreased in the blood of Cd treated rats. The activities of enzymes alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in plasma, as well as concentration of blood glucose were significantly increased in animals treated with cadmium in comparison to control values. The treatment with Cd increased lipid peroxidation (LP) and reduced glutathione (GSH) contents in the blood, suggesting that the Cd induced oxidative stress. These results of our study suggested that Cd induced alterations in hemato-biochemical parameters and LP and GSH content.
AB  - Kadmijum (Cd+2) je široko rasprostranjen toksičan metal koji može izazvati oksidaciona oštećenja u krvi remeteći prooksidaciono-antioksidacionu ravnotežu. Wistar albino pacovi (stari 3 meseca), tretirani su pojedinačnim dozama CdCl; (0.4 mg Cd/kg i.p) i žrtvovani posle 24 časa. Hematološki parametri, i to: broj eritrocita (RBCs), hematokritska vredno.st (Ht) i koncentracija hemoglobina (Hb) značajno su smanjeni u krvi tretiranih pacova. Aktivnost enzima alanin aminotransaminaze (ALT) i aspartat aminotransaminaze (AST) u plazmi, kao i koncentracija glukoze u krvi značajno su povećani kod životinja tretiranih kadmijumom u poređenju sa kontrolnim vrednostima. Tretman sa kadmijumom, takođe je doveo do povećanja lipidne peroksidacije (LP) i redukovanog glutationa (GSH). Sve to ukazuje da je kadmijum izazvao oksidacioni stres u krvi pacova. Dobijeni rezultati ukazuju da je Cd indukovao promené u hemato-biohemijskim parametrima, kao i u sadržaju LP i GSH u krvi tretiranih pacova.

AB  - The aim of this study is to establish the effects of cadmium (Cd), coenzyme Q10 (CoQ10) and Cd + CoQ10 on the activities of: Superoxide dismutases (total SOD, manganese containing, Mn SOD, copper zinc containing, CuZn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST), as well as glutathione reductase (GR) in the kidneys of male Wistar albino rats in comparison with control animals. Significantly increased activities of Tot SOD and CuZn SOD in the kidneys of Cd-treated animals were observed. At the same time, the activity of Mn SOD was significantly decreased in the kidneys of animals treated with Cd, as well as with CoQ10. The activity of CAT was significantly lower in rats exposed to CoQ10 and Cd+CoQ10 in respect to the control animals. GSH-Px and GST activities were significantly decreased in all treated groups of animals (Cd, CoQ10 and Cd+CoQ10), while GR activity was not changed. Presented results reveal, that CoQ10 only partially reversed Cd-induced changes of antioxidant defense enzyme activities in kidney by reversing the effect of Cd on Tot SOD and CuZn SOD activities.
AB  - Cilj ovog rada bio je da se objasni efekat kadmijuma (Cd), koenzima Q10 (CoQ10) i Cd + CoQ10 na aktivnost: superoksid dismutaza (ukupne, Uk SOD, mangan sadržavajuće, Mn SOD, bakar cink sadržavajuće, CuZn SOD, katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-S-transferaze (GST) kao i glutation reduktaze (GR) u bubrezima mužjaka Wistar albino pacova u poređenju sa kontrolnom grupom životinja. Dobijeni rezultati pokazuju značajno povećanje aktivnosti Uk SOD i CuZn SOD u bubrezima pacova tretiranih sa Cd. U isto vreme aktivnost Mn SOD je bila značajno smanjena u bubrezima pacova tretiranih sa Cd, kao i tretiranih sa Cd+CoQ10. U poređenju sa kontrolnim životinjama, aktivnost CAT je bila značajno smanjena u bubrezima pacova tretiranih sa CoQ10, kao i tretiranih kombinacijom Cd+CoQ10. Aktivnosti GSH-Px i GST su bile značajno smanjene u bubrezima svih tretiranih grupa životinja (Cd, CoQ10 i Cd+CoQ10), dok se aktivnost GR nije statistički značajno menjala. Iz prikazanih rezultata može se zaključiti, da CoQ10 samo delimično utiče na promené aktivnosti antioksidacionih zaštitnih enzima nastale pod uticajem Cd, kao i da vraća aktivnosti Uk SOD i CuZn SOD na normalne vrednosti.

abstract = "The aim of this study is to establish the effects of cadmium (Cd), coenzyme Q10 (CoQ10) and Cd + CoQ10 on the activities of: Superoxide dismutases (total SOD, manganese containing, Mn SOD, copper zinc containing, CuZn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST), as well as glutathione reductase (GR) in the kidneys of male Wistar albino rats in comparison with control animals. Significantly increased activities of Tot SOD and CuZn SOD in the kidneys of Cd-treated animals were observed. At the same time, the activity of Mn SOD was significantly decreased in the kidneys of animals treated with Cd, as well as with CoQ10. The activity of CAT was significantly lower in rats exposed to CoQ10 and Cd+CoQ10 in respect to the control animals. GSH-Px and GST activities were significantly decreased in all treated groups of animals (Cd, CoQ10 and Cd+CoQ10), while GR activity was not changed. Presented results reveal, that CoQ10 only partially reversed Cd-induced changes of antioxidant defense enzyme activities in kidney by reversing the effect of Cd on Tot SOD and CuZn SOD activities., Cilj ovog rada bio je da se objasni efekat kadmijuma (Cd), koenzima Q10 (CoQ10) i Cd + CoQ10 na aktivnost: superoksid dismutaza (ukupne, Uk SOD, mangan sadržavajuće, Mn SOD, bakar cink sadržavajuće, CuZn SOD, katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-S-transferaze (GST) kao i glutation reduktaze (GR) u bubrezima mužjaka Wistar albino pacova u poređenju sa kontrolnom grupom životinja. Dobijeni rezultati pokazuju značajno povećanje aktivnosti Uk SOD i CuZn SOD u bubrezima pacova tretiranih sa Cd. U isto vreme aktivnost Mn SOD je bila značajno smanjena u bubrezima pacova tretiranih sa Cd, kao i tretiranih sa Cd+CoQ10. U poređenju sa kontrolnim životinjama, aktivnost CAT je bila značajno smanjena u bubrezima pacova tretiranih sa CoQ10, kao i tretiranih kombinacijom Cd+CoQ10. Aktivnosti GSH-Px i GST su bile značajno smanjene u bubrezima svih tretiranih grupa životinja (Cd, CoQ10 i Cd+CoQ10), dok se aktivnost GR nije statistički značajno menjala. Iz prikazanih rezultata može se zaključiti, da CoQ10 samo delimično utiče na promené aktivnosti antioksidacionih zaštitnih enzima nastale pod uticajem Cd, kao i da vraća aktivnosti Uk SOD i CuZn SOD na normalne vrednosti.",
