Antioxidant defense enzyme activities in the liver of red mullet (Mullus barbatus L.) from the Adriatic Sea: The effects of locality and season

AB  - The effects of locality and season on the activity of antioxidant defense enzymes (ADE), namely, total superoxide dismutase (Tot SOD), copper zinc containing superoxide dismutase (CuZn SOD), manganese containing superoxide dismutase (Mn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and biotransformation phase II enzyme glutathione-S-transferase (GST) in the liver of red mullet (Mullus barbatus L.) from the Platamuni and Valdanos localities (Adriatic Sea) in winter and spring season were studied. All enzyme activities were expressed as specific (units/mg proteins) and total (units/g wet tissue) ones. Our results show increased specific and total activities of almost all investigated ADE in winter, with respect to the spring, both in Platamuni and Valdanos. The opposite effect was obtained for the specific and total GSH-Px activity at both localities. The presented results show that the activity of investigated ADE depends mainly on the analyzed season. Also, it was confirmed that biotransformation phase II enzyme GST is the most sensitive indicator of environmental status in the liver of red mullet. The results of Principal Component Analysis (PCA) based on correlations show similar trends in the positive and negative correlations between specific and total ADE in both seasons. At the same time, examination of seasonal patterns of ADE revealed clear differences between the investigated localities in winter and spring for specific and total activities. From the presented results, it can be concluded that seasonal variations of ADE should be incorporated into interpretation of further biomonitoring studies of red mullet.

AB  - Fish are the most important organisms in the biomonitoring of aquatic ecosystems. A number of pollutants may cause the oxidative stress in the organism of fish. The monitoring of parameters of oxidative stress in tissues of fish is a good biomarker in the assessment of the condition of environment. Catch of commercially important species of fish, hake (Merluccius merluccius L.) was carried out in front of sea-port Bar (South Adriatic) at the end of May 2005. The parameters of oxidative stress: concentrations of lipid peroxides (LP) and reduced glutathione (GSH) were determined in liver and white muscle. Physical-chemical parameters (salinity, concentration of dissolved oxygen and temperature) as well as the concentrations of nitrites, nitrates and detergents in the water of investigated locality were determined. The obtained results showed the tissue specificity that was the consequence of different metabolic and antioxidative activities. In the liver of hake, the increased concentration of LP was found in comparison to the white muscle, whereas in white muscle the increased concentration of GSH was recorded in comparison to the liver. These results suggest that the intensity of oxidative stress was higher in the liver in comparison to the white muscle, which is the consequence of increased presence of pollutants in water of the investigated locality. .

AB  - The effects of selenium ( Se) on antioxidant defense system in liver and kidneys of rats with cadmium (Cd)-induced toxicity were examined. Cd exposure (15 mg Cd/kg b.m./day as CdCl2 for 4 weeks) resulted in increased lipid peroxidation ( LP) in both organs (p < 0.005 and p < 0.01). Vitamin C (Vit C) was decreased in the liver ( p < 0.005), whereas vitamin E ( Vit E) was increased in the liver and kidneys ( p < 0.005 and p < 0.05) of Cd-exposed animals. Superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities were decreased in both tissues ( p < 0.05 and p < 0.005), whereas catalase (CAT) activity was decreased only in liver ( p < 0.005). Glutathione S-transferase (GST) increased in both tissues ( p < 0.005 and p<0.01). Treatment with Se (0.5 mg Se/kg b.m./day as Na2SeO3 for 4 weeks) significantly increased liver and kidneys SOD and GSH-Px activities ( p < 0.05 to p < 0.005), as well as CAT and GST activities only in the liver ( p < 0.01). In animals exposed to Se, both the concentrations of Vit C ( p < 0.01) and Vit E (p < 0.005) were increased in both tissues. Co-treatment with Se resulted in reversal of oxidative stress with significant decline in analyzed tissues Cd burden. Our results show that Se may ameliorate Cd-induced oxidative stress by decreasing LP and altering antioxidant defense system in rat liver and kidneys and that Se demonstrates the protective effect from cadmium-induced oxidative damage.

AB  - Recent investigations in biomonitoring include the monitoring of biochemical and physiological parameters to evaluate the condition of environment. Parameters of oxidative stress are significant biomarkers of environmental pollution of the sea. Specimens of marine fish - hake (Merluccius merluccius) was collected from the localities of Platamuni and Valdanos (Sauth Adriatic) in winter   and   spring   during   2003   year.   Concentrations   of antioxidant compounds such as Vitamin E (Vit E) and Vitamin C (Vit C) as well as, lipid peroxidation (LP) were determined in the liver and white muscle of the hake. Physical chemical parameters of water of investigated  localities  were also determined: salinity, temperature, oxygen saturation and concentrations of nitrites, nitrates and detergent in both seasons.
The obtained results showed higher concentrations of LP, Vit E and Vit C in liver of hake in comparison to white muscle. Concentrations of LP and Vit C were increased, while the concentrations of Vit E were decreased in winter period in comparison to spring. Concentrations of LP were increased while the concentrations of Vit E were decreased in both investigated tissues of hake from the waters of locality Valdanos in respect to Platamuni. These changes of parameters of oxidative stress are, in part, the consequence of changes of temperature as well as, concentrations of nitrites, nitrates and detergents in water of investigated localities and in particular season.
In conclusion, the concentrations of parameter of oxidative stress in investigated tissues of hake represent a good marker in the biomonitoring of environmental pollution. The intensity of oxidative stress was increased in the liver of hake in comparison to white muscle, in winter period ill respect to spring and in waters of Valdanos in comparison to Platamuni

AB  - Cadmium (Cd) is  an  ubiquitous,  nonessential  element  that  has  recentl y  raised concerns due to its accumulation in the environment. The toxicity of Cd as an industrial  pollutant  a  food  contaminant and  as  one  of  the  major  components   in  cigarette smoke has been well established. A number of nutrient s have been shown to interact with Cd and alter its cellular effects. Several protective agents, including conzyme Q10 (CoQ10) and vitamin E (Vit E) play an important role in detoxification of  endogenous  and  exogenous  compounds.  Our  study  investigated  the possible protective  effects  of  coenzyme Q10  (CoQ 10)  and  vitamin   E  (Vit  E)  alone  or  in cornbrnation against cadmium (Cd) induced alterations of antioxidant defense system  in the rat heart. Male Wistar rats were injected with a single dose of CdCl2 (0.4 mg Cd/kg  i.p.),  CoQ10 (20  mg CoQ 1 0/kg  i. m.) and  Vit E (20  lU  Vit  E/kg, i. m.), a lone or in combination. Acute  intoxication  of  rats with Cd  were followed  by significantly  increased activity of antioxidant defense enzymes (CuZn SOD, GSH-Px, GST and GR), while the activity of Mn SOD  was decreased in the heart. The treatment with Cd significantly decreased Vit C and Vit E concentrations . Treatment with CoQ10+Vit E reversed  Cd-induced  alterations  in Vit C and  Vit  E concentrations. Our study suggests that the prooxidative effect of Cd is respon si ble for  alterations in prooxidant-ant ioxidant balance in the heart. The obtained results support the assumption   that   CoQ10  and Vit E functions   cooperatively with endogenous antioxidants and diminished some toxic effects of Cd in rat hearts .

AB  - After enzymic biotransformation, molsidomine (MO) acts via the metabolite 3-morpholmosydnonimine (SIN-1) through spontaneous liberation of nitric oxide (NO) and superoxide (O-2(.-)). The aim of this study was to compare the effects of MO and its active metabolite SIN-1 on the redox status of rat erythrocytes and reticulocytes. Rat erythrocyte as well as reticulocyte-rich red blood cell (RBC) suspensions were aerobically incubated (2h, 37 degrees C) without (control) or in the presence of different concentrations of MO or SIN-1. In rat erythrocytes, biotransformation of MO resulted in the production of NO and nitroxyl (NO-). Endogenous superoxide anion (O-2(.-)) participated in peroxynitrite generation. SIN-1 simultaneously liberated NO and O-2(.-), which formed peroxynitrite (at least in part), but the liberated NO predominantly reacted with haemoglobin, forming methaemoglobin in erythrocytes. In reticulocytes, MO and SIN-1 caused an increase in the levels of both nitrite and 3-nitrotyrosine (an indicator of peroxynitrite), whereas they decreased the level of O-2(.-). In reticulocytes, MO was metabolized into SIN-1 which led to the generation of NO, which reacted with O-2(.-) (endogenous or exogenous) forming reactive nitrogen species. In conclusion, there are two metabolic pathways for MO biotransformation: one causing NO and NO- generation predominantly in erythrocytes and the other, via SINA metabolism, in reticulocytes. AB  - Background and Purpose: In order to assess exposure to effects of environmental pollutants on aquatic oiganisms, several different biochemical and physiological parameters were studied. The activity of antioxidant defense enzymes was measured in the Mediterranean Sea shrimp (Parapenaeus longirostris) and compared with the presence of polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) in their sea water environment. Materials and Methods: The activity of the antioxidant defence enzymes superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione reductase (GR), as well as glutathione-S transferase (GST) as a phase II detoxification enzyme, was measured in the abdominal muscle of the shrimp in winter and spring at four different locations in the Montenegrine costal region of the Adriatic Sea. The results were correlated to the presence of PCBs and PAHs in the sea water at the time of sampling. Results and Conclusions: There was no obvious direct correlation between antioxidant defense activity and the presence and concentration of PCBs and PAHs in the sea water: The activities of SOD, GSH-Px and GR showed seasonal changes and therefore seasonality should be incorporated into the interpretation of antioxidant changes in future biomonitoring studies. Generally, SOD and GR activities were lower and GSH-Px was higher in winter when compared to spring. However these effects were not uniform and statistical trends were influenced by local environment. There were no seasonal changes in GST activity, but it's activity varied depending on the location, but without correlation to the presence of PCBs and PAHs.

AB  - Cadmium (Cd) is a commonly occurring environmental contaminant which after being intake into the organism accumulates mostly in the liver and kidneys. Recent studies on mammals have shown that Cd stimulates formation of superoxide anion radicals, hydrogen peroxide and hydroxyl radicals. As a consequence, enhanced lipid peroxidation, DNA damage, altered calcium and sulfhydryl homeostasis, as well as marked disturbances of antioxidant defense system occur.
Coenzyme Q10 (CoQ10) in its reduced form (CoQ10H2) is a powerful antioxidant. As an antioxidant it is exploited either directly upon superoxide or indirectly on lipid radicals. CoQ10 may prevent both the initiation and propagation of lipid peroxidation, protects DNA from oxidation causing by lipid peroxidation and protects organism from oxidative stress induced by various toxic agents. 
	The activities of Total SOD, Mn SOD, CuZn SOD, CAT, GSH-Px, GR and phase II biotransformation enzyme-GST, as well as Vit C and Vit E concentrations were analyzed in the liver, heart and kidneys of male, two months old Wistar albino rats treated with Cd (200 mg CdCl2 x H2O, in drinking water during 30 days), CoQ10 (20 mg/kg CoQ10 dissolved in olive oil, i.m., every fifth day), concomitantly treated with Cd and CoQ10 (200 mg CdCl2 x H2O, in drinking water during 30 days + 20 mg/kg CoQ10 dissolved in olive oil, i.m., every fifth day) and in control animals (C). 
	 This study suggested that Cd influences antioxidative defense enzyme activities in the liver, heart and kidneys of rats in various ways, especially depending on its metabolic activity. CoQ10 only partially (and tissue-specific) reversed Cd-induced changes on these enzymes. At the same time, CoQ10 exerts very strong beneficial effects on Vit C and Vit E by elevating its concentrations and thus induced an increase in antioxidative defense potential in investigated tissues of rats treated with Cd.

AB  - Specimens of the carps (Cyprinus carpio L.) were adapted to the oxygen concentration of 6.5mg/L water (100%) as control (C) group. The first (H1) and second (H2) experimental groupswere exposed to progressive reduction of O2 to 4.2 mg/L (55%) during 115 hours and 2.3 mg/L(30%) during 230 hours, respectively. Acute hypoxia decreased glucose (H1) and pyruvate (H1and H2) concentrations in the blood of carps, while lactate level was significantly increased(H2). Concentration of red blood cells (RBCs) ATP was decreased in both (H1 and H2)groups of fish. Concomitant increase of ADP concentration was observed in (H2) group offish. The decrease of ATP/ADP ratio in (H2) group was also found. In carps of (H1) group,the activities of superoxide dismutase (SOD) and catalase (CAT) in RBCs were increasedwhereas glutathione-S-transferase (GST) activity in the plasma was increased in both groupsof fish. Glutathione peroxidase (GSH-Px) activity and the concentration of reducedglutathione (GSH) were decreased in the same conditions. Increased concentration of lipidperoxides (LP) in the blood of (H1 and H2) groups of animals in relation to the controls wasalso found. In conclusion, acute hypoxia induced a significant alteration of antioxidantenzyme activities and lipid peroxide concentration in the blood of carps (Cyprinus carpio L.)under influence of progressive reduction of oxygen concentration. All specified changes aredependent from duration of hypoxic conditions. The presented results suggest that during the evolution carps developed a well defined metabolic regulation and antioxidant defense systemfor the protection from acute hypoxia.

AB  - Spiny cheek crayfish (Orconectes limosus L.) were found in the Serbian part of the River
Danube (August 2004, at 1112 km of the water course). A description of the biology, origin and distribution of this species, as well as the potential impact to the aquatic ecosystems is presented and discussed. This is the first record of this invasive crayfish in Serbian
part of the River Danube. In addition, our results could indicate that there is potential risk
for the expansion of O. limosus in other regional waters in the future.

AB  - Cadmium (Cd) has been recognized as being among the most toxic environmental and industrial pollutants, one that is pres­ ent in soils, sediments, air, and water (S to e pp 1 er, 1991) . After penetrating into the organism, mostly through the respir­ atory and gastrointestinal tracts, Cd accumulates in the liver and kidneys, as well as in other tissues and organs, causing many metabolic, histological, and pathological changes, such as neph­ rotoxicity, cardiotoxicity, increased lipid peroxidation and hem­ orrhagic lesions of seminal tubules. In tissues, Cd also binds to proteins of low molecular mass, producing metallothioneins by the induction of metallothionein mRNA synthesis (G e o r g e et al., 1996). Cadmium depletes glutathione and protein-bound sulfhydryl groups, resulting in enhanced production of reactive oxygen species (ROS). These ROS result in increased lipid per­ oxidation, enhanced excretion of urinary lipid metabolites, modulation of intracellular oxidized states, DNA damage, membrane damage, altered gene expression, and apoptosis (Kim et al., 2003).

AB  - We investigated the activities of glutathione peroxidase (GSH-Px) glutathione reductase (GR) and phase II biotransformation enzyme glutathione-S-transferase (GST) in the hepatopancreas, the gills and muscle of Noble crayfish (Astacus astacus-AST), Stone crayfish (Austropotamobius torrentium-AUS) and Spynicheek crayfish (Orconectes limosus-ORC) from the South Morava River, Krajkovačka River and Serbian part of the Danube. Specific GSH-Px activity was lower in the hepatopancreas of AUS than in ORC and in the muscle of AST than in AUS and ORC. We obtained differences of total GSH-Px activity in the muscle between AST and AUS and between AST and ORC. Specific and total GR activity was higher in the hepatopancreas of AST than in other species. Specific and total activity of GR in the gills and in the muscle of crayfish species was higher in AUS then in AST and ORC. The specific and total GST activity was lower in the hepatopancreas of AST and ORC than in AUS. Specific and total GST activity in the gills was higher in AUS than in AST. The lowest specific and total GST activity was detected in the muscle of ORC compared to the AST and AUS. Our results show that specific and total enzyme activities of GSH-Px, GR and GST have species and tissue specifics between crayfish species and represent biomarkers of environmental influences on these organisms.

AB  - Fish are the most important organisms used in biomonitoring of aquatic ecosystems. Parameters of oxidative stress in fish tissues represent significant biomarkers in the assessment of the status of environment. Specimens of hake (Merluccius merluccius L.) and sea bream (Pagellus erythrinus L.) were collected at the end of May 2005 from the locality in front of sea-port Bar (South Adriatic Sea). Parameters of oxidative stress (concentration of lipid peroxidase - LP and reduced glutathione - GSH) were determined in the liver and white muscle of hake and sea bream. Physical-chemical parameters (salinity, concentration of dissolved oxygen and temperature) were determined in water of the investigated locality. Obtained results showed that the concentration of LP was higher in liver of hake and sea bream in comparison to white muscle. However, significantly higher concentration of GSH was found in white muscle of hake and sea bream in comparison to liver. These changes of oxidative stress parameters point to tissue specificity, which is the consequence of different metabolic and antioxidative activity. Inter-specific differences were also established. Higher concentrations of LP and GSH in liver of hake and in white muscle of sea bream were recorded, due to the changes of physical-chemical parameters of environment. The intensity of oxidative stress in liver of hake and sea bream was higher in comparison to muscle.
AB  - Ribe predstavljaju najznačajnije organizme koji se koriste u biomonitoringu vodenih ekosistema. Parametri oksidacionog stresa u tkivima riba predstavljaju značajne biomarkere u proceni stanja životne sredine. Krajem maja 2005. godine uzorkovane su jedinke oslića (Merluccius merluccius L.) i arbuna (Pagellus erythrinus L.) sa lokaliteta ispred luke Bar (Južni Jadran). AB  - Background and Purpose: Recent investigations in biomonitoring include the follow-up of biochemical and physiological parameters in order to estimate the condition of the environment. Oxidative stress parameters are significant biomarkers of ecological pollution of the sea. Materials and Methods: Specimens of red mullet (Mullus barbatus) were collected from the seawater at the localities of Platamuni and Valdanos (in the Adriatic Sea) both in winter and spring. The concentrations of lipid peroxides (LP), vitamin E (vit E) and vitamin C (vit C) were determined both in the liver and white muscle. Salinity, temperature, oxygen saturation, concentrations of nitrates and nitrites and detergents were determined in the seawater at the time of specimen collection. Results and Conclusions: The obtained results showed higher concentration of LP, vit E and vit C in the liver of red mullet when compared to white muscle. The concentrations of LP and vit C were higher, whereas the vit E concentration was lower in winter than in spring. In both the liver and white muscle (from Valdanos), higher concentration of LP and lower concentration of vit E were found when compared to those from Platamuni. Such changes in the oxidative stress parameters were in correlation with seasonal changes in temperature and the concentrations of nitrates, nitrites and detergents in the water. The intensity of oxidative stress in liver of red mullet was higher in comparison to muscles. It was also higher in winter compared to spring, and higher in Valdanos than in Platamuni. In conclusion, parameters of oxidative stress in tissues of red mullet are good markers for the biomonitoring of environment conditions.

AB  - Our study investigated the possible protective effects of coenzyme Q(10) (CoQ(10)) and Vitamin E (Vit E) alone or in combination against cadmium (Cd) induced alterations of antioxidant defense system in the rat heart. Male Wistar rats were injected with a single dose of CdCl(2) (0.4 mg Cd/kg BW i.p.), CoQ(10) (20 mg CoQ(10)/kg BW i.m.) and Vit E (20 IU Vit E/kg BW i.m.), alone or in combination. Acute intoxication of rats with Cd were followed by significantly increased activity of antioxidant defense enzymes (CuZn SOD, GSH-Px, GST and GR), while the activity of Mn SOD was decreased in the heart. The treatment with Cd significantly decreased Vit C and Vit E concentrations. Treatment with CoQ10 and Vit E reversed Cd-induced alterations of antioxidant defense system. The obtained results support the assumption that CoQ10 and Vit E functions cooperatively with endogenous antioxidants and diminished toxic effects of Cd in rat heart. (c) 2006 Elsevier B.V. AB  - The effects of cadmium (Cd, 15 mg/kg/day) on the blood antioxidant defense system and the possible protective effects of selenium (Se, 7 μg/kg/day) were studied. Chronic Cd administration resulted in increased activities of copper, zinc superoxide dismutase (CuZn SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) in red blood cells (RBC), as well as glutathione S-transferase (GST) activity and vitamin E (Vit E) concentration in the plasma. Se induced a significant increase in GSH-Px and GST activities, as well as concentrations of vitamin C and E. Thus, the mechanism by which Se decreases Cd toxicity in rats, seems to rely on the protection of the antioxidant defense system in blood, possibly by forming non-toxic cadmium selenide. The accumulation of Cd in RBC also decreased significantly upon co-exposure to Se. The obtained results show, that Cd exhibit toxic effects on the antioxidant defense system in the blood of rats and that Se demonstrates the protective effect against Cd toxicity.
AB  - U ovom radu ispitivan je efekat kadmijuma (Cd, 15 mg/kg/danu) na antioksidacioni zaštitni sistem i moguća zaštitna uloga selena (Se, 7 μg/kg/danu). Posle hroničnog tretmana Cd značajno se povećavaju aktivnosti bakar, cink superoksid dismutaze (CuZn SOD), katalaze (CAT) i glutation peroksidaze (GSH-Px) u crvenim krvnim ćelijama (RBC), kao i aktivnost glutation S-transferaze (GST) i koncentracije vitamina E (Vit E) u plazmi. Selen značajno povećava aktivnosti enzima GSH-Px i GST, kao i koncentracije vitamina C i E. Mehanizam kojim selen smanjuje toksičnost Cd kod pacova odnosi se na zaštitu antioksidacionog sistema u krvi, posebno formiranje ne-toksičnih kadmium-selenita. Akumulacija Cd u RBC takođe je značajno smanjena posle ko-tretmana selenom. Dobijeni rezultati pokazuju da Cd ispoljava toksične efekte na antioksidacioni zaštitni sistem u krvi pacova i da Se pokazuje zaštitni efekat protiv toksičnosti Cd.

Se induced a significant increase in GSH-Px and GST activities, as well as concentrations of vitamin C and E. Thus, the mechanism by which Se decreases Cd toxicity in rats, seems to rely on the protection of the antioxidant defense system in blood, possibly by forming non-toxic cadmium selenide. The accumulation of Cd in RBC also decreased significantly upon co-exposure to Se. The obtained results show, that Cd exhibit toxic effects on the antioxidant defense system in the blood of rats and that Se demonstrates the protective effect against Cd toxicity., U ovom radu ispitivan je efekat kadmijuma (Cd, 15 mg/kg/danu) na antioksidacioni zaštitni sistem i moguća zaštitna uloga selena (Se, 7 μg/kg/danu). Posle hroničnog tretmana Cd značajno se povećavaju aktivnosti bakar, cink superoksid dismutaze (CuZn SOD), katalaze (CAT) i glutation peroksidaze (GSH-Px) u crvenim krvnim ćelijama (RBC), kao i aktivnost glutation S-transferaze (GST) i koncentracije vitamina E (Vit E) u plazmi. Selen značajno povećava aktivnosti enzima GSH-Px i GST, kao i koncentracije vitamina C i E. Mehanizam kojim selen smanjuje toksičnost Cd kod pacova odnosi se na zaštitu antioksidacionog sistema u krvi, posebno formiranje ne-toksičnih kadmium-selenita. Akumulacija Cd u RBC takođe je značajno smanjena posle ko-tretmana selenom. Dobijeni rezultati pokazuju da Cd ispoljava toksične efekte na antioksidacioni zaštitni sistem u krvi pacova i da Se pokazuje zaštitni efekat protiv toksičnosti Cd.",

AB  - The aim of our study was to investigate the copper and zinc containing superoxide dismutase (CuZn SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cells (RBCs), as well as phase II biotransformation enzyme glutathione-S-transferase (GST) activity in the plasma of male, two months old Wistar albino rats. The rats were divided into two experimental groups: control animals and animals treated with olive oil (100 μL of virgin olive oil, i.m., every fifth day during 30 days). The obtained results show that olive oil induced a significant decrease of CuZn SOD, CAT and GSH-Px activities in RBCs (p<0.05) and significant increase of GST activity in the plasma (p<0.05) in relation to the control animals. The results show that olive oil influences the activity of investigated enzymes in the blood of rats and in that way improves the overall antioxidative defense.
AB  - Cilj našeg rada bio je da se odrede aktivnosti bakar cink sadržavajuće superoksid-dismutaze (CuZn SOD), katalaze (CAT) i glutation-peroksidaze (GSH-Px) u eritrocitima, kao i enzima faze II biotransformacije glutation-S-transferaze (GST) u plazmi mužjaka dva meseca starih Wistar albino pacova tretiranih maslinovim uljem. Pacovi su podeljeni u dve grupe i to kontrolna i grupa tretirana maslinovim uljem (100 μL of maslinovog ulja, i.m., svaki peti dan tokom 30 dana). Rezultati pokazuju da maslinovo ulje izaziva značajno smanjenje aktivnosti CuZn SOD, CAT i GSH-Px (p<0.05) u eritrocitima, kao i povećanje aktivnosti GST u plazmi pacova (p<0.05) u odnosu na kontrolu. Dobijeni rezultati pokazuju da maslinovo ulje utiče na aktivnost ispitivanih enzima u smislu poboljšanja antioksidacione zaštite.

AB  - Fish are the most important organisms used in biomonitoring of aquatic ecosystems. Parameters of oxidative stress in fish tissues represent significant biomarkers in the assessment of the status of environment. Specimens of hake (Merluccius merluccius L) and red mullet (Mullus barbatus L) were collected from the locality in front of sea-port Bar (South Adriatic Sea) at the end of May 2005. Activity of the enzymes of the antioxidative system (superoxide dismutase - SOD and catalase - CAT) and parameters of oxidative stress (lipid peroxidation - LP and reduced glutathione - GSH content) were determined in the liver of hake and red mullet. Obtained results showed that the activities of SOD and CAT were significantly lower in the liver of hake than in red mullet. Higher concentrations of LP and GSH in liver of hake were recorded, due to the changes of physical-chemical parameters of environment. The intensity of oxidative stress in liver of hake was higher in comparison to red mullet.
AB  - Ribe predstavljaju najznačajnije organizme koji se koriste u biomonitoringu vodenih ekosistema. Parametri oksidacionog stresa u tkivima riba predstavljaju značajne biomarkere u proceni stanja životne sredine. Krajem maja 2005. godine uzorkovane su jedinke oslića (Merluccius merluccius L) i trlje (Mullus barbatus L) sa lokaliteta ispred luke Bar (Južni Jadran). Aktivnost enzima antioksidacionog sistema (superoksid dismutaze - SOD i katalaze - CAT) i parametri oksidacionog stresa (koncentracija lipidnih peroksida - LP i redukovanog glutationa - GSH) su određivani u jetri oslića i trlje. Dobijeni rezultati su pokazali da su aktivnosti SOD i CAT značajno manje u jetri oslića nego kod trlje. Zabeležena je veća koncentracija LP i GSH u jetri oslića, kao posledica promena fizičko-hemijskih parametara sredine i prisustva polutanata. Intenzitet oksidacionog stresa u jetri oslića je bio viši u odnosu na trlju.

AB  - Preeclampsia represents a pregnancy specific syndrome characterized by elevated arterial blood pressure and proteinuria. The aim of this study was to evaluate whole blood and erythrocytes energy status in healthy and preeclamptic women. Our study involved 18 healthy (control) and 19 preeclamptic women. Diagnosis of preeclampsia was done according to the recommendation of International Society for the Study of Hypertension in Pregnancy. After blood collection and erythrocytes extraction, the next parameters were followed: concentrations of lactate and adenine nucleotides (ATP, ADP and AMP). Results showed increased concentrations of lactate and ATP in whole blood of preeclamptic, vs. control pregnant women. All parameters of energy status were also elevated in erythrocites of preeclamptic women (elevation of ADP level was significant). This study suggests that preeclamsia was followed by an elevation of all parameters of energy status in whole blood and in erythrocytes, as a result of increased rate of glycolysis. .
AB  - Preeklampsija predstavlja za trudnoću specifičan sindrom koji se karakteriše povišenim krvnim pritiskom i proteinurijom. Cilj istraživanja je određivanje energetskog statusa pune krvi i eritrocita zdravih i trudnica sa preeklampsijom. Ispitivanjem je bilo obuhvaćeno 18 zdravih i 19 trudnica sa preeklampsijom. Dijagnoza preeklampsije je postavljena prema preporukama International Society for the Study of Hypertension in Pregnancy. Nakon uzimanja krvi i ekstrakcije eritrocita praćeni su sledeći parametri: koncentracije laktata i adenin nukleotida (ATP, ADP i AMP). Rezultati su pokazali povećanje koncentracija laktata i ATP u punoj krvi trudnica sa preeklampsijom u odnosu na kontrolne trudnice. U eritrocitima, svi parametri energetskog statusa su bili povećani u preeklampsiji (povećanje ADP nivoa je bilo značajno). Ova studija pokazuje da je preeklampsija praćena povećanjem svih parametara energetskog statusa u punoj krvi i eritrocitima, kao posledica stimulisanog intenziteta procesa glikolize. .

AB  - Preeclampsia represents a pregnancy specific syndrome with high rate of both maternal and neonatal morbidity and mortality. The maternal syndrome is characterized by elevated arterial blood pressure and proteinuria. Several recent studies have reported that there is a disbalance of oxidant/antioxidant system in women with preeclampsia, and this factor might contribute to the pathogenesis of the disease. Our study involved 22 healthy (control) and 20 preeclamptic women. Diagnosis of preeclampsia was done according to the recommendation of International Society for the Study of Hypertension in Pregnancy. After blood collection, the next parameters were followed in erythrocytes: concentration of lipid peroxides (TBARS) non-enzymatic components (reduced glutathione - GSH, oxidized glutathione GSSG) and activities of enzymatic components of AOS (glutathione peroxidase GSH-Px, glutathione reductase - GR and glutathione-S-transferase - GST). Results of this study showed that concentration of TBARS and activity of GST were significantly higher, while activity of GR was significantly lower in erythrocytes of preeclamptic, vs. healthy women. Concentrations of GSH, GSSG and activity GSH-Px did not change significantly. Increased concentration of TBARS and activity of GST, as well as disorder in GSH and GSH-dependent enzymes status were indicators of oxidative stress in erythrocytes of preeclamptic women.
AB  - Preeklampsija je sindrom specifičan za trudnoću sa visokim stepenom morbiditeta i mortaliteta fetusa i majke. Klinički se manifestuje posle 20. nedelje gestacije. Kod majke sindrom se manifestuje povišenim krvnim pritiskom i proteinurijom. Brojne savremene studije pokazuju da je disbalans oksidacionog i antioksidacionog sistema značajan faktor u patogenezi preeklampsije. Istraživanjem je obuhvaćeno 22 zdrave i 20 trudnica sa preeklampsijom. Dijagnoza preeklampsije je postavljena prema preporukama International Society for the Study of Hypertension in Pregnancy. U eritrocitima su praćeni sledeći parametri: koncentracija lipidnih peroksida (TBARS), neenzimskih komponenti (redukovani glutation - GSH, oksidovani glutation - GSSG) i aktivnost enzimskih komponenti AOS (glutation-peroksidaza, GSH-Px, glutation-reduktaza, GR i glutation-S-transferaza, GST). AB  - The activity of the antioxidant defence enzymes superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), glutathione peroxidase (GSH-Px, EC 1.11.1.9), glutathione reductase (GR, EC 1.6.4.2) and the phase 11 biotransformation enzyme glutathione-S-transferase (GST, EC 2.5.1.18) in whole mussels (Mytilus galloprovincialis) were studied. The mussels were collected in winter and in spring at two localities in the Adriatic Sea: Bar Port and Tivat Bay. Our results show that the activities of SOD, GSH-Px and GST were seasonally dependent with higher activities in winter. GR activity was also higher in winter, but only in mussels from Bar Port. In mussels from Tivat Bay, GR activity was lower in winter compared to spring. In addition, a decrease in CAT activity in mussels from Bar Port compared to those from Tivat Bay was found. It can be concluded that seasonal variations should be incorporated into interpretation of biomonitoring studies in mussels. (c) 2005 Published by Elsevier Inc.

AB  - The effect of cadmium (Cd), coenzyme Q10 (CoQ10) and Cd+CoQ10 on the activities of superoxide dismutases (total SOD), manganese containing superoxide dismutase (Mn SOD) and copper-zinc containing superoxide dismutase (Cu,Zn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione-Stransferase (GST), glutathione reductase (GR) and concentrations of ascorbic acid (AsA) and vitamin E (Vit E) in the heart of male Wistar albino rats were studied in comparison to the controls and cadmium treated animals. Cd induces a significant increase of total SOD, Cu, Zn SOD and GSH-Px activities, as well as AsA and Vit E concentrations, but leads to a significant decrease of CAT and GR activities. CoQ10 induces a significant increase of total SOD, Mn SOD, Cu, Zn SOD and GSH-Px activities, as well as AsA and Vit E concentrations. In the same group of animals the activities of CAT, GST and GR were significantly decreased. By concomitant treatment of rats with Cd+CoQ10 the activities of total SOD, Mn SOD and GSH-Px, as well as concentrations of AsA and Vit E were markedly increased. In the same group of animals the activities of Cu, Zn SOD, CAT and GR were significantly decreased. In respect to the Cd treated rats in Cd+CoQ10 partially are reversed changes (Cu,Zn SOD) of antioxidant defense system in the heart.
AB  - Ispitivan je uticaj kadmijuma (Cd), koenzima Q10 (CoQ10) i Cd+ CoQ10 na aktivnost ukupne superoksid dismutaze (Uk SOD), mangan sadržavajuće superoksid dismutaze (Mn SOD), bakar cink sadržavajuće superoksid dismutaze (Cu,Zn SOD), katalaze (CAT), glutation peroksidaze (GSH-Px), glutation-Stransferaze (GST) i glutation reduktaze (GR), kao i na koncentracije askorbinske kiseline (AsA) i vitamina E (Vit E) u srcu mužjaka Wistar albino pacova u poređenju sa kontrolnom grupom i grupom tretiranom sa kadmijumom. Cd u srcu indukuje značajno povećanje aktivnosti Uk SOD, Cu,Zn SOD i GSH-Px, kao i koncentracija AsA Vit E uz značajno smanjenje aktivnosti CAT i GR. AB  - In our experiment we studied the antioxidant enzyme activities (superoxide dismutase-SOD, catalase-CAT, glutathione peroxidase-GSH-Px, glutathione reductase-GR, and glutathione-S-transferase-GST} in hepatopancreas, gills and abdominal muscle of spinycheek crayfish (Orconectes limosus) from the Serbian part of Danube River. The speciments were collected with deep nets and tissue samples were prepared for analisys by standard methods. All enzyme activities were  measured spectrophotometrically and expressed as specific (U/mg proteins) and total (U/g net mass). The results show strong tissue specifity of investigated enzymes. Specific and total SOD activity in hepatopancreas was lower than in gills  and muscle. At the same time, total SOD activity in gills was significantly lower in respect to muscle. The specific and total activity of CAT was significantly higher in hepatopancreas than in gills and muscle, as well as in gills in respect to the muscle. Same trend was obtained for specific and total activity of GSH-Px. In gills of spinycheek crayfish·, specific activity of GR was significantly higher than in hepatopancreas and muscle, as well as in gills in respect to the muscle. Total GR activity was also significantly higher in gills comparing to hepatopancreas and muscle. The specific and total activity of GST was significantly increased in hepatopancreas than in gills and muscle. At the same time, the activity of GST was higher in gills than in muscle. The obtained results show a strong tissue specifity of antioxidant defense enzymes in investigated tissues of spinycheek crayfish which is accompanied with its different metabolic activity.

Accumulation of cadmium in some tissues of rats after chronic treatment with CdCl2

AB  - Cadmium is the important environmental pollutant and major routes of Cd intake are oral exposure and inhalation. From the total amount of food Cd intake, only about 6% is absorbed in the gastrointestinal tract of animals. The aim of our experiment was to determine the rate of Cd accumulation in liver, kidneys, skeletal muscle (musculus gastrocnemius) and testes of two months old Wistar albino male rats. Animals were divided into 2 experimental groups consisting 7 animals and treated in 30 days courses. The first group of animals functioned as controls (C) and the second group was treated with 200 mg CdCh x 5H20 in drinking water during 30 days (Cd). The average intake of 17 mg Cd/mg b.m. was calculated from the water consumed. Cadmium concentration was determined by atomic absorption spectrophotometry in the mixture of nitric and perchloric acid at the wavelength of 228.8 nm, slit 0.1 nm and lamp current 4 mA in the mixture of air and acetylene. The obtained results show a significant accumulation of Cd in all examined tissues of rats (p<0.005) in respect to controls. These results are in accordance with the findings of other authors and show that Cd mainly accumulated in the tissues of high metabolic rate, such as liver and kidneys. In these organs Cd accumulates mainly in the hepatocytes and cells of the proximal and distal renal tubules. In the cells Cd mainly deposits in the cytoplasm, nuclei and nucleoli where disturbs the cell metabolism, protein synthesis and sulfhydryl homeostasis.

AB  - The freshwater crayfishes can be useful organisms in the biomonitoring of environmental conditions. In our experiments we studied vitamin E (Vit E) and sulphydryl group (-SH)  concentrations  in  hepatopancreas,  gills  and  abdominal muscle of three crayfish species: Noble crayfish  (Astacus  astacus)  from  South  Morava river, Stone crayfish (Austropotamobius torrentium) from Krajkovacka river  and Spinycheek crayfish (Orconectes limosus) from Danube river. All species were collected in summer season (August). The  speciments  were  collected  by deep  nets and supematants for analysis were prepared according to the standard methods. Vit E concentration was assayed by standard method using bathophenantroline as reagent, while -SH groups concentration was  measured  by  standard  spectrophotometric method using dithionitrobenzoic acid (DTNB). The  obtained  results  show  significantly lower concentration of vit E in hepatopancreas of Orconectes limosus in respect to Astacus astacus (p<0.05). At the same time, significantly decreased concentration of vit E was detected in gills of Orconectes limosus comparing  to  Astacus astacus and Austropotamobius torrentium (p<0.005). Concentration of -SH groups was significantly increased  in  hepatopancreas  of  Noble  crayfish  in comparison to Stone crayfish (p<0.05) and Spinycheek crayfish (p<0.005). In conclusion,  the concentrations of the  investigated  antioxidant  compounds  (vit  E and
-SH groups) in the selected tissues of freshwater crayfishes are good markers for biomonitoring of environmental conditions. Differences in the investigated nonenzymatic oxidative stress parameters in tissues reflect different metabolic and antioxidative activities, which  can  be the consequence  of species  or tissue specifity,  as well as the consequence  of different environmental  conditions.
